Inhibitory Effects of the Extracts from Eleutherococcus senticosus Maxim. on Histamine-release from Rat's Mast Cell

  • Eleutherococcus senticosus Maxim. has been successfully used as an oriental medicine for various diseases including allergic disorders. Histamine is a major factor on various allergic responses and it is reported that histamine was released from mast cells by sensitization of allergens. In this study, ethanol extracts from E. senticosus Maxim. were prepared and the composition was analyzed by high performance liquid chromatography. The eleutheroside B as a primary effective component of E. senticosus was contained approximately 225 mg/kg in root bark extracts. The extracts were found to significantly inhibit compound 48/80-induced histamine release form mast cells in dose dependent manner. However the extracts had low cytotoxicity on the mast cells with MTT assay. These results showed that E. senticosus Maxim. extracts may be the effective materials on inflammatory disorders.

생물전환을 통한 음나무발효물의 지표성분 설정 및 동시분석법 검증 (Validation of Simultaneous Analysis Method of Standard Compounds in Fermented Kalopanax pictus Nakai by Bioconversion)

  • The aim of this study was to select compounds for the standardization of fermented Kalopanax pictus Nakai (KP-F), to develop the analysis method using HPLC-PDA and to perform method validation. KP-F is a fermented powder developed to improve the original physiological activities and create a new functionality. Eleutheroside E, Acanthoside B, and Syringaresinol were selected as the standard compounds and developed our own method for simultaneous analysis. The analyte was isolated using C18 column with a gradient elution of 0.05 M phosphoric acid in water and methanol as the mobile phase at a flow rate of 1 mL/min and detected at 210 nm. As a result, all standard compounds showed good linearity with an $R^2$ (coefficient of correlation) of 1.000 and for the limit of detection range of $0.710{\sim}0.831{\mu}g/mL$, and the limit of quantification as $2.150{\sim}2.520{\mu}g/mL$. The precision was RSD (%) of less than 4.80%, while the accuracy was 4.70%>RSD (%) for the range 102.44~110.48%. In conclusion, the developed analysis method is suitable for the detection of Eleutheroside E, Acanthoside B, and Syringaresinol in KP-F.

가시오가피 버섯균사체 발효물의 기능성 식품 소재 개발 (Development of Functional Food Materials from Acanthopanax senticosus-Fermented Mushroom Mycelia)

  • 가시오가피(Acanthopanax senticosus)의 잎과 줄기를 천연물배지로 발효미생물인 영지버섯, 노루궁뎅이버섯 및 상황버섯균사체를 각각 최적조건으로 발효하여 3종의 가시오가피-버섯발효물인 가시오가피 영지버섯균사체 발효물, 가시오가피 노루궁뎅이버섯균사체 발효물, 가시오가피 상황버섯균사체 발효물을 얻었으며 이를 건강기능식품 소재로서 이용하고자 하였다. 우선적으로 배양용 시험관과 실험실용 배양병을 사용하여 버섯균사체별 가시오가피의 천연배지로서 최적혼합비율을 검토한 결과 영지버섯균사체, 노루궁뎅이버섯균사체 및 상황버섯균사체의 가시오가피 줄기와 잎의 혼합비율을 각각 80:20, 50:50, 50:50으로 확립하였으며, 이 조건을 가지고 산업용 배양포트(2,000 mL)에서의 최적배양시간을 검토한 결과 영지버섯균사체 14일, 노루궁뎅이버섯균사체 40일, 상황버섯균사체 30일로 확립하였다. 상기와 같이 최적화된 조건으로 배양된 3종의 가시오가피 버섯균사체 발효물들을 물과 70% 에탄올로 추출, 여과, 농축, 동결건조를 수행하여 가시오가피 버섯균사체 발효물 추출분말인 열수추출물(FM-5111, FM-5121, FM-5131)과 70% 에탄올추출물(FM-5112, FM-5122, FM-5132)을 얻었다. 이 가시오가피 버섯균사체 발효물 추출분말의 eleutheroside B와 eleutheroside E를 분석한 결과, eleutheroside B는 FM-5111 $0.050{\pm}0.003mg/g$, FM-5121 $0.067{\pm}0.001mg/g$, FM-5131 $0.026{\pm}0.001mg/g$, FM-5112 $0.038{\pm}0.002mg/g$, FM-5122 $0.053{\pm}0.005mg/g$, FM-5132 $0.025{\pm}0.001mg/g$의 함량을 나타내었고, eleutheroside E는 FM-5111 $0.052{\pm}0.014mg/g$, FM-5121 $0.047{\pm}0.003mg/g$, FM-5131 $0.078{\pm}0.003mg/g$, FM-5112 $0.045{\pm}0.002mg/g$, FM-5122 $0.040{\pm}0.002mg/g$, FM-5132 $0.080{\pm}0.011mg/g$의 함량을 보였으며, ${\beta}$-glucan의 경우 FM-5111 $4.47{\pm}0.14%$, FM-5121 $3.41{\pm}0.15%$, FM-5131 $4.15{\pm}0.06%$, FM-5112 $5.18{\pm}0.36%$, FM-5122 $3.74{\pm}0.40%$, FM-5132 $4.37{\pm}0.32%$의 함량이 측정되었다.

Analysis of Lignans in Acanthopanax sessiliflorus Fruits and Their Fermented Wine by HPLC

  • High performance liquid chromatography (HPLC) was used for the determination of lignans, eleutherosides B and E, in Acanthopanax sessiliflorus fruits and their fermented wine. The lignans were quantified by a reversed-phase system using a gradient of $H_2O$ and acetonitrile as a mobile phase within 20 min. The analysis was successfully carried out within 20 min. The contents of eleutherosides Band E as main active principles of Acanthopanax species were measured in A. sessiliflorus fruits (1.15 and $8.49\;{\mu}g/mg$, respectively), their fermented wine (0.45 and $1.33\;{\mu}g/mg$, respectively) and wine residues (no detection).

전처리 및 건조방법에 따른 가시오갈피생약재의 품질변화 (Quality Changes in Eleutherococcus senticosus Cortex Processed by Different Pretreatment and Drying Method)

  • Eleuthero(Eleutherococcus senticosus Maxim.) cortex is well known as a herb medicine for tonic. This study was performed to improve the quality of dried E. senticosus cortex. Investigation of quality factor and contents of efficient compounds under different steaming times and drying methods were performed to determine the proper processing and drying conditions of Eleuthero cortex harvested on March in annual stems. The proper steaming time for peeling bark to make high quality Eleuthero cortex took less than 20 mins. Eleutheroside B and E contents among drying methods were significantly different at 5% level DMRT. The $50^{\circ}C$ heat drying was the most advisable condition for drying, when drying and keeping contents of effective compounds.

탐라오갈피의 침출 중 유용성분의 변화 (Changes in Major Constituents by Soaking of Acanthopanax koreanum with Spirit Solution)

  • 제주자생 탐라오갈피(Acanthopanax koreanum Nakai)를 리큐르 소재로 활용하기 위하여 주정농도를 각각 $30{\sim}95%$로 조절한 다음 0.5 cm 이하로 세절한 뿌리 및 줄기를 700 g/10 l의 비율로 첨가하여 70일간 침출시키면서 이화학적 특성 및 유용성분의 경시적인 변화를 검토하였다. 침출액의 pH 변화는 대체로 $5.5{\sim}6.5$ 범위였으며, 색도 b 값은 주정농도가 낮을수록, 침출기간이 길어질수록 높은 값을 나타내었고, a값은 주정농도가 높을수록 - 값이 증가하였으며 주정농도에 의한 영향이 컸다. 가용성고형물의 변화는 침출기간에 따라 점진적으로 증가하여 주정농도 $30{\sim}70%$ 범위에서 줄기는 $0.6{\sim}0.7%(w/v)$, 뿌리는 $1.0{\sim}1.5%(w/v)$이었다. Eleutheroside B와 E는 침출기간 20일까지 급속히 증가하는 경향이었으며, 줄기가 뿌리보다 함량이 많았다. 그리고 주정농도 30%에서 70%로 높아짐에 따라 eleutheroside B와 E 함량은 크게 증가하였다. Acanthoic acid는 주정농도와 이용부위에 따른 함량변화가 뚜렷하게 나타났으며, 침출 후 $5{\sim}10$일에 급속히 용출되었으며, 줄기에서는 $3{\sim}25\;{\mu}g/ml$, 뿌리에서는 $46{\sim}1,700\;{\mu}g/ml$이 침출되었다. 따라서 탐라오갈피를 이용하여 약용주인 리큐르로 상품화하기 위해서는 뿌리 비율을 높게 하여 주정농도 $60{\sim}80%$ 범위에서 $30{\sim}50$일간 침출시킨 다음 13주간 숙성하여 블렌딩하는 것이 효과적이라고 판단되었다.

  • Acanthopanacis Cortex (Acanthopanax sessiliflorum, Araliaceae, KP VIII), an important Korean medicinal herbal drug, has been widely used as tonic, anti-stress and immuno-enhancing drugs. To monitor the contents of active ingredients (acanthoside D[=eleutheroside E], eleutheroside B, isofraxidin, chlorogenic acid, and caffeic acid) in Acanthopanax sp., we developed the HPLC analysis method and validated. The simultaneous determination of five active ingredients was achieved in a C$\sub$18/ column with an acetonitrile water (containing 1 % phosphoric acid) (15 : 85) mobile phase. (omitted)

Phytochemical Constituents from the Stems of Acanthopanax senticosus

  • Six compounds were isolated from the stems of Acanthopanax senticosus (Araliaceae). Their structures were elucidated as iso-fraxidin, H-sesamin, 5-hydroxymethylfurfural, eleutheroside B, eleutheroside E and an unknown compound by spectral analysis. Of them, 5-hydroxymethylfurfural was isolated for the first time from A. senticosus.

Mass Production of Adventitious Roots of Eleutherococcus sessiliflorus through the Bioreactor Culture

  • This paper reported the establishment of mass production system of adventitious roots of Eleutherococcus sessiliflorus through the shake flask and bio-reactor culture. Induction of adventitious roots was started from the explants of germinated somatic embryos on half-strength Murashing and Skoog (MS) solid medium. The frequency of adventitious root formation was better in the explants comprising the basal hypocotyl parts than root explants alone. Among the different auxins tested (NAA, IBA and IAA), frequency of adventitious root induction was highest on medium with 0.5 mg/L NAA, and produced $16.3\pm1.9$ roots per explant. In shake-flask culture, deletion of $NH_4NO_3$ of MS medium was effective for induction of adventitious root compared with both full and half-strength MS media. Fresh weight increase of induced adventitious roots was performed well in medium with 0.5 mg/L IBA. When adventitious roots produced in shake-flask culture were transferred to 10-liter bioreactor, 5.5 times of fresh weight increase was gained after one month of culture. HPLC analysis revealed that the amount of eleutheroside E and E1 was higher in in vitro cultured adventitious roots than the 3 year-old field cultivated root barks of Eleutherococcus sessiliflorus. The content of eltutheroside B was much lower in adventitious roots than that of field cultivated one.
