UASB reactor was operated for treating wastewater containing high sulfate to assess their performance, competition between sulfate-reducing bacteria(SRB) and methane-producing bacteria(MPB), and the change in the characteristics of microbial granules according to change of hydraulic retention time(HRT) in the reactor. The reactor was fed with a synthetic moderate strength wastes(glucose, 2000 mgCOD/l) containing high sulfate($2400mgSO_4{^{2-}}/l$). The organic loading rate(OLR) ranged from 1.5 to 3.0 gCOD/l.d as HRT maintained 15 to 30 hrs in the stage I. The COD removal efficiency was between 80 to 92%. During this period, methane yield rapidly decreased from 0.3 to 0.1 1 $CH_4$/gCODremoved. While sulfide concentration in the effluent increased from 80 to 200 mgS/l. This indicates that SRB becomes dominant over MPB at a relatively long HRT in the excess sulfate. When OLR of reactor maintained from 5 to 8 gCOD/l.d in the stage II, methane yield increased from 0.1 to 0.17 1 $CH_4$/gCODremoved regardless of decrease of COD removal efficiency. This indicates that SRB is more sensitive to the change of a short HRT than MPB. In the competition between SRB and MPB, about 30% of the removed COD was utilized by SRB at HRT of 30 hrs during the start-up period, while about 73% was used by SRB at HRT of 15hrs at the final step of second experimental stage. Whereas after shock exposure of OLR about 62% was utilized by SRB at HRT of 5hrs. It indicates that SRB is strongly suppressed by the wash-out of significant dispersed SRB since a large electron flow is distributed to the MPB. In addition, the granulation in the presence of high sulfate is unfavoured at a long HRT because of substrate transport limitations into MPB like Methanothrix spp. which is an important factor in the composition of the granules. Accordingly, granule sizes in the UASB reactor decreased with time due to weak network frame of granules by the decreased activity of MPB.

The Inhibiting activity of newly synthesized phenol (E-series) and triazine (T-series) derivatives was evaluated by using thylakoid membranes extracted from cyanobacteria (Anacystis nidulans $R_2$). There were no significant differences between phenol derivatives and dinoseb to the thylakoid membrane extracted from wild type in the Hill reaction. However, a phenol derivative, E-24 which has no -Cl at phenyl ring, did not show any activity. The longer the length of R substituents was in phenol derivatives, the lower inhibiting activity was in the Hill reaction. Triazine derivatives, T-27, T-28, T-40, T-41, T-47 and T-48 were also compared with diuron and atrazine. Among triazine compounds, T-27 and T-28 showed 10 and 30 times activity as high as atrazine to wild type, respectively. Other triazine derivatives, T-40, T-41, T-47 and T-48 showed low inhibiting activity to wild and mutant type. A structural difference of T-27 and T-28 from T-40, T-41, T-47 and T-48 was the presented of -C-NH-. Both T-27 and T-28 were very closely associated with serine, an amino acid located at the 264th position of D1 protein because of the resistant ratio(R/S) to mutant G-264 were higher than that of atrazine.
The effect of Salicylic acid (SA) on the leaf growth, chlorophyll content, photosynthesis, stomatal conductance in Commelina communis L. was investigated. C. communis which was grown in Hoagland solution containing 1 mM SA during 3 weeks resulted in a significant reduction of leaf length, width and area. 1 mM SA reduced about 10% of the leaf area at 1 week and 2 weeks, but inhibited 22% of the leaf area at 3 weeks. SA also showed great effect on the reduction of chlorophyll content. SA reduced 47% and 53% of chlorophyll content each at 2 weeks and 3 weeks when it was compared with the control. Chlorophyll a/b ratio in SA treated sample was increased at 3 weeks representing that SA was sensitive to chlorophyll b. The treatment of SA did not inhibit the activity of PSII+PSI, PSII and PSI. This result indicates that SA did not show direct effect oil the photosynthetic electron transport activity. However, when C. communis was grown in Hoagland solution containing I mM SA fer long period, SA showed clear inhibition of photosyhthesis in intact leaves. The treatment of SA for 3 weeks showed about 23~65% inhibition of photosynthetic activity at various light intensity (100~1000$\mu$mo1 m$^2$S$^1$). Similar effect was found on stomatal conductance. The treatment of SA caused an almost closure of stomata. Similar effects of stomatal conductance at various light intensity indicate a loss of normal control on stomatal mechanism. These results indicate that the effect of SA on photo-synthetic activity was not by SA itself but by indirect metabolic pathway.
The morphological characteristics, carbon and nitrogen concentrations, stable isotope values and photosynthetic rates of Porphyra yezoensis were studied at the main purple lavers production areas on southwestern coast of Korea. The morphological characteristics of leaf length, leaf width and weight of Porphyra blades were between 11.6~16.3 (average 13.8) cm, 4.6~6.3 (average 5.4) cm, $1.1{\sim}2.6(average\;1.86)g\;DW\;m^{-2}$, respectively. Photosynthetic pigment of Chl a concentration of Porphyra blades was between $2.18{\sim}17.77(average\;9.65)mg\;DW\;Chl\;a\;m^{-2}$. Carbon and nitrogen concentrations of Porphyra blades was between $201{\sim}317(average\;240)mg\;DW\;g^{-1}$, $39.8{\sim}50.0(average\;43.5)mg\;DW\;g^{-1}$ and C/N ratio 5.0~6.7 (average 5.5). The range of average ${\delta}^{13}C$ and ${\delta}^{15}N$ values of Porphyra blades was between - 25.6 to - 24.0 (average - 24.7)‰ for ${\delta}^{13}C$, and 1.3 to 4.1 (average 2.1)‰ for ${\delta}^{15}N$. Photosynthetic characteristics of seaweeds measured by pulse amplitude modulation (PAM) fluorometry was used as an indicator of photosynthetic activity. We use Diving-PAM fluorometry to examine photosynthetic rates of the seaweeds Porphyra yezoensis at each station. Maximum quantum yield of Porphyra blades was between 0.46~0.55 (average 0.52), the variance of the effective PS II maximum quantum yield of the station was broadly similar. Maximum relative electron transport rate (rETRmax) of Porphyra blades was between $4.71{\sim}5.84(average\;5.33){\mu}mol\;electrons\;m^{-2}\;s^{-1}$, the changes of maximum relative electron transport rate (rETRmax) of Porphyra yezoensis were similar to those of PS II maximum quantum yield. Photosynthetic efficiency (${\alpha}$) was between 0.027~0.045 (average 0.036). Minimum saturating irradiance ($E_k$) range was $139{\sim}180(average\;156){\mu}mol\;photons\;m^{-2}\;s^{-1}$. Minimum saturating irradiance ($E_k$) made a difference by station within the area on southwestern coast. Carbon and nitrogen concentrations and photosynthetic rates of Porphyra blades production areas on southwestern coast were broadly similar. The photosynthetic characteristics showed low photosynthetic rates because the low maximum quantum yields and low maximum relative electron transport rate.
To investigate the effects of yellow loess on the microbial community after applying into C. polykrikoides as a red tide centrol method during decomposition process, we conducted this study using microcosm experiments, which consisted of sediment collected from Jinhae and Masan bay. The composition, number of bacteria and respiratory electron transport system activity (ETSA) were analyzed. The number of heterotrophic bacteria examined in the samples of both stations reached maximum value within 12 hrs with $10^7$ cells/dry g, independent with the yellow loess applied. In addition, a differenee in the variation of heterotrophic bacterial composition was not observed by adding the yellow loess, and Vibrio spp. always appeared during the culture periods, However, in day 8 culture, the sulfate reducing bacteria was $3.8\times10^7$ cells/dry g in Masan bay and $5.5\times10^6$ cells/dry g in Jinhae bay samples without yellow loess, and these were 120, 350 fold-and 160, 420 fold-increased when yellow loess was added (1 : 1, 1 : 2). The average ETSA was 6.8$\~$7.6 $\mu$g formazan $h^{-1}$ dry $g^{-1}$ independently with yellow loess in aerobic condition for both samples, but activity was decreased by addition of yellow loess in anaerobic. Thus the addition of yellow loess to marine sediment seems to have an effect to inhibit the anaerobic decomposition process and growth of sulfate reducing bacteria which lead to the bad condition of marine environments.
Vertical and temporal variations of active and total bacterial abundance were monthly estimated in Lake Soyang from April 1999 to January 2000. The number of total and respiring bacteria was determined directly under microscope by AODC and CTC methods, respectively. The number of total and active bacteria varied from $2.1{\times}10^5 to 3.1{\times}10^6 $,$cells{\cdot}ml^{-1}$ and $1.8{\times}10^4 to 8.0{\times}10^5 $,$cells{\cdot}ml^{-1}$, respectively. The proportions of respiring bacteria to total cell ranged from 3.7 to 44.2% : The proportions was the highest in November 1999 and the lowest in December 2000. The specific activity of${\beta}$-glucosidase divided by total bacteria was$1.6{\times}10^5\;amol{\cdot}cell^{-1}{\cdot}hr^{-1}$in August and$1.4{\times}10^5\;amol{\times}cell^{-1}{\times}hr^{-1}$in September while the specific activity divided by CTC active bacteria was about$3.6{\times}10^5\;amol{\cdot}cell^{-1}{\cdot}hr^{-1},\;24.0{\times}10^5\;amol{cdot}cell^{-1}{cdot}hr^{-1}$. The specific activity of active bacteria in September was 6.7 times higher than that of August. By these data of active bacteria, the new information of aquatic ecosystem was unveiled.
Gene expression profiling is a useful tool for identifying critical genes and pathways in metabolism. The objective of this study was to determine the major differences in the expression of genes associated with metabolism and metabolic regulation in liver and mammary tissues of lactating cows. We used the Michigan State University bovine metabolism (BMET) microarray; previously, we have designed a bovine metabolism-focused microarray containing known genes of metabolic interest using publicly available genomic internet database resources. This is a high-density array of 70mer oligonucleotides representing 2,349 bovine genes. The expression of 922 genes was different at p<0.05, and 398 genes (17%) were differentially expressed by two-fold or more with 222 higher in liver and 176 higher in mammary tissue. Gene ontology categories with a high percentage of genes more highly expressed in liver than mammary tissues included carbohydrate metabolism (glycolysis, glucoenogenesis, propanoate metabolism, butanoate metabolism, electron carrier and donor activity), lipid metabolism (fatty acid oxidation, chylomicron/lipid transport, bile acid metabolism, cholesterol metabolism, steroid metabolism, ketone body formation), and amino acid/nitrogen metabolism (amino acid biosynthetic process, amino acid catabolic process, urea cycle, and glutathione metabolic process). Categories with more genes highly expressed in mammary than liver tissue included amino acid and sugar transporters and MAPK, Wnt, and JAK-STAT signaling pathways. Real-time PCR analysis showed consistent results with those of microarray analysis for all 12 genes tested. In conclusion, microarray analyses clearly identified differential gene expression profiles between hepatic and mammary tissues that are consistent with the differences in metabolism of these two tissues. This study enables understanding of the molecular basis of metabolic adaptation of the liver and mammary gland during lactation in bovine species.
Myoblast fusion depends on mitochondrial integrity and intracellular $Ca^{2+}$ signaling regulated by various ion channels. In this study, we investigated the ionic currents associated with $[Ca^{2+}]_i$ regulation in normal and mitochondrial DNA-depleted(${\rho}0$) L6 myoblasts. The ${\rho}0$ myoblasts showed impaired myotube formation. The inwardly rectifying $K^+$ current ($I_{Kir}$) was largely decreased with reduced expression of KIR2.1, whereas the voltage-operated $Ca^{2+}$ channel and $Ca^{2+}$-activated $K^+$ channel currents were intact. Sustained inhibition of mitochondrial electron transport by antimycin A treatment (24 h) also decreased the $I_{Kir}$. The ${\rho}0$ myoblasts showed depolarized resting membrane potential and higher basal $[Ca^{2+}]_i$. Our results demonstrated the specific downregulation of $I_{Kir}$ by dysfunctional mitochondria. The resultant depolarization and altered $Ca^{2+}$ signaling might be associated with impaired myoblast fusion in ${\rho}0$ myoblasts.
The fluctuations of biochemical and molecular activities III the harmful dinoflagellate, Cochlodinium polykrikoides, depending on water temperatures, were studied. In genomic DNA concentration, a similar value of 0.6 was shown at $12^{\circ}C$ and $15^{\circ}C$, but significantly increasing DNA from $18^{\circ}C$ (p<0.05), with a maximum of 1.8 at $24^{\circ}C$. After$24^{\circ}C$, the DNA significantly decreased to 0.6. Likely, the concentrations of RNA and total protein were at their highest values of 1.7 and 0.07 g $mL^1$ at $24^{\circ}C$, respectively. In contrast to ONA, RNA and total protein began to increase at $15^{\circ}C$. Oxygen availability between lower and higher temperatures was significantly different and increased from $18^{\circ}C$ according to light intensity, regardless of wavelengths (p<0.05). At $24^{\circ}C$, the highest value of the maximum electron transport rate (ETRmax), ranging from 537.9 (Ch 1) to 602.5 mol electrons $g^{-1}$ Ch1 a $s^{-1}$ (Ch 4), was also shown. Nitrate reductase (NR) and ATPase activities were at their highest values of 0.11 mol $NO_2^-g^{-1}$ Ch1 a $h^{-1}$ and 0.78 pmol 100 $mg^{-1}$$at^2$$4^{\circ}C$, respectively. When the cells cultured at $15^{\circ}C$, NR and ATPase activities significantly increased compared to $12^{\circ}C$ (p<0.05). In an analysis of CHN, the concentration of C and N also significantly increased (p<0.05). However, at $27^{\circ}C$, most of the molecular and biochemical movements were much lower, compared to $24^{\circ}C$. These results suggest that C. polykrikoides is very sensitive biochemical and molecular activities depending on water temperatures. Possibly, it is desirable to estimate at $18^{\circ}C$ the initiation of the massive blooming development of C. polykrikoides. In nature, it will be very difficult to maintain the massive blooms after $24^{\circ}C$ because of the possibility of significantly decreasing the molecular movement and activity of C. polykrikoides.

Vegetation canopy plays an important role in $CO_2$/$H_2$O exchange between the biosphere and the atmosphere by controlling leaf stomata. In this study, rice (Oryza sativa L.), a staple crop in Asia was investigated to formulate its single leaf model of photosynthesis and stomatal conductance. Photosynthesis and stomatal conductance were measured with a portable infrared gas analyzer system. Other plant and meteorological variables were also measured. To evaluate empirical constants in this biochemical leaf model, nonlinear least squares technique was used. The maximum catalytic activity of enzyme and the maximum rate of electron transport were $ 100\mu$㏖ $m^{-2}$$s^{-1}$ and $140 \mu$㏖ m$^{-2}$ s$^{-1}$ (@ 35$^{\circ}C$), respectively. The empirical constants, m and b, associated with stomatal conductance model were 9.7 and $0.06 m^{-2}$$s^{-1}$ , respectively. On a leaf scale, agreements between the modeled and the measured values of photosynthesis and stomatal conductance were on average within 20%, and the simulation of diurnal variation was also satisfactory On a canopy scale, the Simple Biosphere model(SiB2) was tested using the derived parameters. The modeled energy fluxes were compared against the micrometeorologically measured fluxes over a rice canopy. Agreements between the modeled and the measured values of net radiation, sensible heat and latent heat fluxes, and $CO_2$ flux (i.e., net canopy photosynthesis) were on average within 25%.
