Studies on the Fine Structure of Caeca in Domestic Geese

  • The objective of this study was to investigate the villus distribution in the caeca of domestic geese based on the fine structure. The caeca of White Roman geese, 14-week old, were sampled and specimens were detected under photomicroscope and scanning electron microscope (SEM). The results indicated that the villi existed at the proximal caecum. The morphologies of these villi showed finger-like, peak-like or tongue-like shapes. The heights of the villi decreased far from the proximal caecum. No villi were found in the middle and distal caecum. It was obvious that the proximal segment was the main portion for absorbing food nutrients in the caeca. The caecal content particles were small and possessed a viscid character. The large particles filtered out at the proximal caecum just like a mesh. The surface of the middle caecum exhibited parallel ridges with no villi. There were band plicae circular shapes found in the middle caecum under scanning electron microsopy.

Electron Microscopic Obsenrations on Micropvle after Sperm Penetration in Rainbow Trout, Oncorhynchus mykiss (정자 침입전후 무지개 송어의 난문에 대한 미세구조적 변화)

  • The time-course process by which spermatozoa penetrates through the micropvle apparatus into the egg cytoplasm of rainbow trout, Oncorhvnchus mvkiss, was examined with transmission and scanning electron microscopy. In the unfertilized egg, the ess surface beneath the inner opening of the micropylar canal did not differ distinctly from the rest of the animal pole area. A spermatozoon attached to the micropvle opening 20 seconds after insemination. In the initial stases of penetration, the spermatozoon still within the micropvlar canal attached perpendicularly at its apical tip to the ess surface, then the sperm head was rapidly engulfed by the folded egg surface with its manly microvilli. A large fertilization cone with microvillus-free surface appeared on the esS surface sutra-rounding the penetrating spermatozoon. The head portion of the penetrating spermatozoon was completely wrapped by the ess surface with only the tail portion visible externally 30 seconds after insemination. The fertilization cone displayed the tail portion of the penetrating spermatozoon on the central portion of its surface 60 seconds after insemination. 150 seconds after insemination, breakdown of the cortical granules elevation were initiated at the animal pole, then completed at the vegetable pole area. The spermatozoon disappeared from the outer surface of the ess before the fertilization cone completely retracted 250 seconds after insemination. In result, the block to polvspermv to permit entry of a sin81e sperm is considered to be mechanical by the rnorpholoSical design of the micropvle and fertilization cone.

  • TEM is a powerful tool for semiconductor material analyses in structure or biological sample in micro structure. TEM observation need to make to coincide specimens for special purpose. in this paper, we have experimented for minimum surface damage on bulk wafer and patterned specimen by various conditions such as accelerating energy, depth of ion beam, ion milling types, and etc. in various specimen preparation methods by FIB (Focus Ion Beam). The optimal qualified specimens are contain low mounts of surface damage(about 5 nm) on patterned specimen.

  • This paper presents a design method for optimizing the focused beam characteristics, which are mainly determined by the condenser lenses in a scanning electron microscopy (SEM) design. Sharply reducing the probe diameter of electron beams by focusing the condenser lens (i.e., the rate of condensation) is important because a small probe diameter results in high-performance demagnification. This study explored design parameters that contribute to increasing the SEM resolution efficiently using lens analysis and the ray tracing method. A sensitivity analysis was conducted based on those results to compare the effects of these parameters on beam focusing. The results of this analysis on the design parameters for the beam characteristics can be employed as basic key information for designing a column in SEM.

Characterization of Silver Nanoparticles Synthesized by Using Marine Isolate Streptomyces albidoflavus

  • Silver nanoparticles production by the green chemistry approach was investigated using an isolated marine actinomycetes strain. The isolated strain was identified as Streptomyces albidoflavus based on chemotaxonomic and ribotyping properties. The strain revealed production of silver nanoparticles both extracellular and intracellularly. Surface Plasmon Resonance analysis with the function of time revealed that particle synthesis by this strain is reaction time dependent. The produced particles were spherical shaped and monodispersive in nature and showed a single surface plasmon resonance peak at 410 nm. Size distribution histograms indicated production of 10-40-nm-size nanoparticles with a mean size of 14.5 nm. FT-IR spectra of nanopartilces showed N-H, C-H, and C-N stretching vibrations, denoting the presence of amino acid/peptide compounds on the surface of silver nanoparticles produced by S. albidoflavus. Synthesized nanoparticles revealed a mean negative zeta potential and electrophoretic mobility of -8.5 mV and -0.000066 $cm^2/Vs$, respectively. The nanoparticles produced were proteinaceous compounds as capping agents with -8.5 mV zeta potential and revealed antimicrobial activity against both Gram-negative and -positive bacterial strains. Owing to their small size, these particles have greater impact on industrial application spectra.

Morphology and Thermal Properties of PPS/ABS Blends (PPS/ABS 블렌드의 형태학적/열적 특성)

  • In this study, the PPS/ABS blend system was investigated in order to collectively identify the relationship among blend morphology, chemical compatibilization and thermal property. ABS resin was chemically modified by the incorporation of maleic anhydride through reactive extrusion for enhanced compatibilization, and PPS, ABS and the modified ABS were blend by a sing twin screw extruder. The effect of chemical modification of ABS on the morphological, mechanical, and thermal properities of the resulting blend was examined. A strong interaction was observed between PPS and MABS by optical microsopy as well as scanning electron microscopy, exhibiting a well-dispersed morphological feature. The PPS/MABS blend showing a single glass transition temperature was observed in dynamic mechanical analysis, demonstrating a pseudo-homogeneous phase morphology induced by chemical compatibilization. PPS/MABS blend also exhibited an enhanced thermal stability and heat distortion temperature compared with modified PPS/ABS blend.

  • Kartagener's sydrome is an autosomaly inherited recessive condition characterized by situs inversus, bronchiectasis, and chronic sinusitis. And recently it was recognized as a subclass of dyskinetic cilia syndrome which caused by a defect in mucociliary transport owing to immotile or dyskinetic beating of cilia. Electron microsopy of cilia from sperm tails, nasal and bronchial epithelium of patients reveals the partial or complete absence of dynein arms. Our four patients were diagnosed as a Kartagener's syndrome by classic triad. We carried out electron microscopy of cilia of the nasal mucosa. And many other tests were done. One patient had squamous cell carcinoma of the lung, and another one patient revealed features of adult respiratory distress syndrome at admission. All patients improved with conservative therapy such as physiotherapy, bronchodilater, antibiotics except one patient who mechanical ventilation was required. A brief review of literature was made.

  • 산업화 이후, 석탄 석유를 중심으로 한 화석연료가 이산화탄소를대량으로 배출하며 지구 온난화를 야기함에 따라, 석유를 대체할 새로운 에너지원에 대한 관심이 높아지고 있다. 많은 대체에너지 가운데, 청정하고 무한 재생 가능한대체에너지를 이야기할 때, 가장 큰 기대를 받고 있는 것은 태양에너지이며, 이에 보조를 맞춰 태양광 발전에 대한 연구개발이 국내외적으로 활발히 진행되고 있는 실정이다. 태양 전지는 빛 에너지를 직접 전기 에너지로 바꿔주는 소자로, 셀의효율을 높이기 위해서는 최대한 많은 빛을 흡수시킬 수 있는 것이 중요하다. 빛의 반사를 줄이는 방법에는 Texturing 과 Antireflecting coating 이있다. Antireflecting coating은 반도체와 공기의 중간 굴절율을 갖는 박막을 증착하여 측면 반사를 감소시킴으로서 빛의 손실을 감소시키는 역활을 한다. 반사 방지막으로 쓰이는 SiNx는 SiOx의 대체 물질로 굴절률이 약 1.5로서 Si에 쉽게 형성시킬 수 있고, texturing된 Si 표면에 적합하며 반사율을 10 %에서 2 %로 줄일 수 있다. 나아가 고성능의 반사방지막은 박막의 균일도확보 및 passivation 공정이 필수적이라 판단된다. 따라서 본 연구에서는 PECVD 방법으로 SiH4와 NH3 gas 의 비율을 변화시켜 증착한 SiNx 박막의 결정학적 특성을 X-ray Diffraction 분석과 TEM (TransmissionElectron Microsopy) 을 통해 관찰하였으며, XPS (X-rayphotoelectron spectroscopy) 를 통해 화학적결합을 확인하였고, 이를 FT-IR (Fourier Transform-Infrared spectroscopy)를 통해 관찰한 결과와 연관시켜분석하였다. 굴절율의 경우 Ellipsometry를 이용하여측정하였으며 위의 측정을 통하여 SiNx박막의 반사 방지막으로써의 가능성을 확인하였다.

  • Chitosan scaffold is widely applied to drug delivery and tissue engineering. We have developed chitosan scaffolds, with various pore size, by differing freezing temperature and duration of ultraviolet (UV) irradiation, for reconstructing skin equivalent. Chitosan scaffold was coated with type I collagen, fibronectin and basic fibroblast growth factor (bFGF) in various combinations and concentrations, to evaluate the effect of extracellular matrix (ECM) and bFGF on cell adhesion, growth and differentiation of dermal fibroblasts. Human dermal fibroblasts, isolated from newborn foreskin and passaged between 3 and 5, were seeded on the top of scaffolds and cultivated for 2 weeks. We examined the morphology and the secretion of ECM of fibroblasts using scanning electron microsopy (SEM) and histochemistry. A stellate morphology of fibroblasts were seen in all groups. The scaffold coated with either type I collagen and bFGF or type I collagen and fibronectin, however, showed the best condtion of dermal fibroblasts, in that the highest cell number and ECM secretion were seen. On the contrary, scaffolds coated with all three factors, type I collagen, bFGF and fibronectin, showed lower number of cells and ECM secretion than scaffolds with two factors. There was a tendency of dose-dependence in all three factors for fibroblast growth and ECM secretion. In conclusion, we may suggest that chitosan scaffold coated with either type I collagen/bFGF or type I collagen/fibronectin could provide more favorable environment for the growth and differentiation of dermal fibroblasts.

Immunohistochemical study of the Cholinergic Nerve Cells in the Medial Septal Nucelus and Diagonal Band of Broca of the Postnatal and Adult Rats (흰쥐 출생후 발생단계에 따른 전뇌 기저부의 내측중격핵과 대각 Broca대에서 콜린성 신경세포에 대한 면역조직화학적 연구)

  • 출생 후 0일, 7일, 14일, 21일 그리고 성체의 흰쥐 전뇌 기저부의 내측중격핵과 수직 및 수평대각 Broca대에서 choline acetyltransferase(ChAT)에 면역반응을 보이는 신경조직과 세포의 분화를 면역조직화학적 및 전자현미경적 방법을 이용하여 조사하였다. 출생 후 초기와 성체에서 신경세포의 세포질과 수상돌기에서 고루 ChAT 면역반응이 확인되었다. 뇌 기저부의 ChAT 면역반응 신경세포들은 발생에 따른 뇌 크기의 증대와 뇌 조직의 분화에 따라 점차 수적 증가를 보였다. 이 ChAT 면역반응 신경세포들은 세포의 모양과 세포제의 장 · 단축의 비에 따라 6가지 형 즉 1) 월형 2) 난형, 3) 세장형, 4) 방추형. 5) 삼각형, 6) 다각형으로 분류되었다 전뇌 기저 핵에서 원형과 난형 신경세포들의 출현율은 출생 후 0일에서 높았으나 성체로 되면서 감소된 반면, 세장형. 방추형. 삼각형 그리고 다각형 신경세포들의 출현율은 출생 후 0일에서는 낮았으나 성체로 되면서 증가하였다. 모든 핵들에서 ChAT 면역반응 신경세포체의 부피는 출생 후 0일에 996-1,252 Um3로 제일 작았으며, 내측중격핵과 수직대각 Broca대 그리고 수평대각 Broca대에서는 출생 후 21일에 각각 5,061, 5.701, 5,820 um3로 최대치를 보였다. 그후 성체로 되면서 모든 핵에서1,897-2,704 roms로 다시 감소하였다. 전자현미경적 관찰에서 출생 후 21일된 흰쥐 수평대각 Broca대에서 ChAT 면역반응은 핵의 핵질 일부와 핵막 그리고 미토콘드리아와 조면소포체에서 관찰되었다. 이 결과들로 미루어 출생 후 초기 발생단계에서 흰쥐 전뇌 기저부의 내측중격핵과 수직 및 수평대각 Broca대에서 ChAT 면역반응 신경세포들은 축삭과 수상돌기의 형성에 따라 세포의 형과 그 출현율 및 세포제의 크기에서 현저한 변화가 이루어지는 것으로 생각된다.
