본 연구에서는 8족 금속 원소인 osmium을 중심금속으로 4가지의 착물을 합성하였다. 합성한 착물은 ${[Os(bpy)}_2{(ap-im)Cl]}^{+/2+}$, ${[Os(dme-bpy)}_2{(ap-im)Cl]}^{+/2+}$, ${[Os(dmo-bpy)}_2{(ap-im)Cl]}^{+/2+}$, ${[Os(dcl-bpy)}_2{(ap-im)Cl]}^{+/2+}$이다. 합성된 착물을 순환전압전류법을 포함한 다양한 전기화학분석방법을 이용하여 전기적 성질을 조사하여 작용기에 따른 전위의 변화를 다음의 전위구간에서 $E_p$:$-0.06\;V{\sim}0.313\;V$ vs. Ag/AgCl 확인하였다. 합성한 화합물을 전기적 흡착방법으로 고정된 금나노입자(gold nano-particles)를 전극 위에 자기조립방식으로 고정화를 시켰다. 당과 당 분해효소(Glucose Oxidase, GOx)에 의한 촉매반응의 전류를 확인하였고, glucose농도에 따른 변화하는 전류의 양도 확인하였다. 마지막으로 고정된 4가지의 osmium complex는 서로 다른 전위로 인하여 촉매전류의 양이 달라지는 것을 알 수 있었고, 이로 인해 redox complex의 전위가 촉매반응에 미치는 영향을 확인 할 수 있었다.
This paper presents the fabrication and characterization of carbon films pyrolyzed with various photoresists for bioMEMS applications. To verify the usefulness of pyrolyzed carbon films as an electrode material in an electrochemical biosensor developed by the authors, interactions between avidin and biotin on the pyrolyzed carbon film were studied via electrochemical impedance spectroscopy based on electrostatic interactions between avidin and negatively-charged ferricyanide. The pyrolyzed carbon films were characterized using a surface profiler, a precision semiconductor parameter analyzer, a nanoindentor, scanning electron microscopy, and atomic force microscopy. Amine conjugated biotin was immobilized on the electrode using EDC/NHS as crosslinkers after $O_2$ plasma treatment to enhance functional groups on the carbon electrode pyrolyzed at $1000^{\circ}C$ with AZ9260. The detection of avidin binding with different concentrations in a range of 0.75 nM to $7.5\;{\mu}M$ to the pyrolyzed carbon electrode modified with biotin was carried out by measuring the electrochemical impedance change. The results show that avidin binds to the biotin on the electrode not by non-specific interaction but by specific interaction, and that EIS successfully detects this binding event. Pyrolyzed carbon films are a promising material for miniaturization, integration, and low-cost fabrication in electrochemical biosensors.
False smut caused by Ustilaginoidea virens is an important rice fungal disease that significantly decreases its production. In the recent past, conventional methods have been developed for its detection that is time-consuming and need high-cost equipments. The research and development in nanotechnology have made it possible to assemble efficient recognition interfaces in biosensors. In this study, we present a simple, sensitive, and selective oxidized graphene-based geno-biosensor for the detection of rice false smut. The biosensor has been developed using a probe DNA as a biological recognition element on paper electrodes, and oxidized graphene to enhance the limit of detection and sensitivity of the sensor. Probe single-stranded DNA (ssDNA) and target ssDNA hybridization on the interface surface has been quantitatively measured with the electrochemical analysis tools namely, cyclic voltammetry, and linear sweep voltammetry. To confirm the selectivity of the device, probe hybridization with non-complementary ssDNA target has been studied. In our study, the developed sensor was able to detect up to 10 fM of target ssDNA. The paper electrodes were employed to produce an effective and cost-effective platform for the immobilization of the DNA and can be extended to design low-cost biosensors for the detection of the other plant pathogens.
Interfacing functional materials with electrical or biological systems is of prime importance in terms of expanding applicative fields and obtaining high performances of devices. Herein, I report the functionalization of graphenes through supramolecular assembly and their electrochemical applications into fuel cells, supercapacitors, and biosensor devices. The solution processable nanohybridization of graphenes by functional materials such as ionic liquids, polyelectrolytes, block copolymers, and biomaterials, described herein would pave the way to obtain high performances of flexible energy and biosensor devices as well as to overcome the existing technology barriers.
In this paper, we fabricated a biosensor for detecting hydrogen peroxide and investigated the sensing property. We prepared a viologen and hemoglobin modified gold electrode using self-assembly and layer by layer method. The electrochemical property of the viologen derivative was characterized in 0.1 M $NaClO_4$ electrolyte solution by cyclic voltammetry. The modified electrode showed reversible electrochemical properties and high stability. From the results, the viologen can act as a charge transfer mediator for access to the electrode surface. The catalytic characteristics of the designed sensor proved that hemoglobin has been kept in its natural structure and can retain its biological activity. The designed biosensor showed a fast amperometric response, excellent linearity and low detection limit. In addition, it had high sensitivity, good reproducibility and stability.
본 연구에서는 8족 금속 원소인 오스뮴을 중심금속으로 일차아민을 포함하고 있는 피리딘 (pyridine) 화합물을 배위시켜 착화합물을 합성하였다. 합성된 오스뮴 착화합물은 $[Os(dme-bpy)_2(ap-im)Cl]^{+/2+}$을 순환전압전류법을 포함한 다양한 전기화학분석법을 이용하여 전기적인 성질을 조사하였다. 또한 합성된 일차 아민을 갖는 오스뮴 착화합물을 이용하여 당 측정용 바이오센서를 제작하기 위하여 금 나노입자(Cold nano-particles)를 전기적 흡착방법을 이용하여 스크린 인쇄방법으로 만든 탄소반죽 전극 (Screen Printed Carbon Electrodes, SPEs) 위에 고정화를 시켰다. 당과 당 분해효소(Glucose Oxidase, GOx)를 혼합하여 발생하는 산화촉매전류를 확인하였고, 당 농도에 따라 변화하는 산화촉매전류의 양도 확인하였다. 새롭게 만들어진 바이오센서는 1 mM 과 같은 낮은 농도에서 감응할 수 있는 바이오센서에 응용할 수 있음을 확인하였다.
과산화수소를 이용한 펜톤(Fenton)산화법은 수처리 및 토양 복원분야에서 활용되는 친환경 산화방법이다. 이 방법으로 오염물질을 제거할 때, 오염물의 농도에 따라 과산화수소의 농도를 적절하게 조절하는 것이 상당히 중요하다. 이에 본 연구에서는 HRP (horseradish peroxidase) 효소를 이용한 전기화학적 바이오센서를 제조하고 효소의 활성과 과산화수소의 검출 특성에 대한 연구를 수행하였다. SPCE (Screen Printed Carbon Electrode)의 작업 전극 표면에 키토산과 AuNP를 이용하여 HRP를 전착하였다. 이 후, 전위주사법(CV)과 전기화학적 임피던스 분광법(EIS)을 이용하여 효소의 고정화를 확인하였다. 또한 시간대전류법(CA)과 UV 분광법으로부터 HRP 효소의 활성을 확인하였다. 본 연구에서 제조한 바이오센서를 PBS 전해질에 담그고 과산화수소를 적정하여 CA 분석으로부터 전극에서 발생하는 전류를 측정하였다. 발생 전류는 과산화수소의 농도에 대하여 선형적으로 증가하였으며, 전류로부터 과산화수소의 농도를 예측할 수 있는 검정곡선을 도출하였다.
To date, many researchers have developed a variety of biosensors to detect the biomolecular interactions. Recently, electrochemical biosensors have been attracting great interest as one of key technologies in a ubiquitous healthcare (U-healthcare) system since they are highly sensitive and feasible to miniaturize. Here we overview the current electrochemical biosensors based on strip-type, nanowire/nanotube, field effect transistor (FET), and nanogap electrode.
Multi-wall carbon nano-tube(MWCNT)를 이용해 screen printed carbon electrodes(SPCEs)을 제작하여 혈당센서의 선택성과 감도가 증가됨을 확인 할 수 있었다. 효소촉매반응을 위한 탄소전극으로의 전자이동의 매개체로 8족 금속 원소인 오스뮴을 중심금속으로 일차 아민을 포함하는 피리딘(pyridine) 리간드를 배위시켜 $[Os(dme-bpy)_2(4-aPy)Cl]^{+/2+}$를 합성하였다. 합성된 오스뮴 착물은 순환 전압전류법을 포함한 다양한 전기화학분석방법을 이용하여 전기적 성질을 조사하였다. 전기적 흡착방법을 이용하여 일차 아민을 갖는 착화합물을 전극위에 고정화 하였다. 오스뮴이 고정화된 MWCNT-SPCEs는 일반적인 carbon electrode보다 약 100배가량의 오스뮴이 흡착됨을 확인 할 수 있었다. (${\tau}_0=2.0\;{\times}\;10^{-9}\;mole/cm^2$) 마지막으로 당(Glucose)과 당 분해효소(Glucose Oxidase, GOx)에 의한 촉매반응의 전류를 확인하였고, 당 농도에 따라 선형 변화하는 전류의 양도 확인하였다.
