Fabrication and Characterization of Porous Silicon-based Urea Sensor Syst (다공질 실리콘을 이용한 요소검출용 바이오 센서 제작)

  • 바이오 마이크로 시스템 및 바이오 MEMS 분야, 특히 실리콘을 기질로 하는 바이오 센서 제작에서 반도체 공정 기술은 센서의 대량 생산과 초소형화를 위해서 반드시 필요한 기술이다. 그러나, 감지전극의 마이크로화에 따른 센서의 감도 및 안정성 저하 문제는 해결해야 할 과제이다. 최근, 다공질 실리콘이 갖는 대면적이 실리콘 기질과 생체 고분자 (예: 단백질, 핵산 등) 간의 결합력을 향상시킬 수 있음이 알려지면서, 바이오 센서 분야에서, 새로운 형태의 드랜스듀서 재료로서의 다공질 실리콘에 대한 논의가 활발히 전개되고 있으며 또한, ISFET (Ion-Selective Field-Effect Transistors) 와는 달리 다공질 실리콘 층은 저항이 크기 때문에 센서 제작 과정에서의 부가적인 절연막을 필요로 하지 않는다. 본 연구에서는, 백금을 증착한 다공질 실리콘 표면에 전도성 고분자로서 Polypyrrole (PPy) 필름과 생체 고분자 물질로서 Urease를 각각 전기화학적으로 흡착하였다. 다공질 실리콘 층의 형성을 위해 테플론 소재의 전기화학 전지에 불산 (49%), 에탄올 (95%), $H_2O$ 혼합 용액을 넣고 실리콘 웨이퍼에 일정시간 수 mA의 산화 전류를 흘려주었으며, 약 $200{\AA}$의 티타늄 박막과 $200{\AA}$의 백금 박막을 RF 스퍼터링하여 작업 전극을 제작하였고, 백금 박막 및 Ag를 기화 증착하여 제작한 Ag/AgCl 박막을 각각 상대 전극과 기준전극으로 하였다. 박막 전극의 표면 분석을 위해 SEM (Scanning Electron Microscopy), EDX (Energy Dispersive X-ray spectroscopy) 등을 이용하였다. 제작된 요소 센서로부터 요소 농도 범위 0.01 mmol/L ${\sim}$ 100 mmol/L에서 약 0.2 mA/decade의 감도를 얻었다.

An Aptamer-Based Electrochemical Sensor That Can Distinguish Influenza Virus Subtype H1 from H5

  • The surface protein hemagglutinin (HA) mediates the attachment of influenza virus to host cells containing sialic acid and thus facilitates viral infection. Therefore, HA is considered as a good target for the development of diagnostic tools for influenza virus. Previously, we reported the isolation of single-stranded aptamers that can distinguish influenza subtype H1 from H5. In this study, we describe a method for the selective electrical detection of H1 using the isolated aptamer as a molecular probe. After immobilization of the aptamer on Si wafer, enzyme-linked immunosorbent assay (ELISA) and field emission scanning electron microscopy (FE-SEM) showed that the immobilized aptamer bound specifically to the H1 subtype but not to the H5 subtype. Assessment by cyclic voltammetry (CV) also demonstrated that the immobilized aptamer on the indium thin oxide-coated surface was specifically bound to the H1 subtype only, which was consistent with the ELISA and FE-SEM results. Further measurement of CV using various amounts of H1 subtype provided the detection limit of the immobilized aptamer, which showed that a nanomolar scale of target protein was sufficient to produce the signal. These results indicated that the selected aptamer can be an effective probe for distinguishing the subtypes of influenza viruses by monitoring current changes.

Effects of ${\gamma}-Aminobutyric$ Acid on Intrinsic Cholinergic Action in Exocrine Secretion of Isolated, Perfused Rat Pancreas

  • ${\gamma}$-Aminobutyric acid (GABA) has been reported to enhance exocrine secretion evoked not only by secretagogues but also by intrinsic neuronal excitation in the pancreas. The pancreas contains cholinergic neurons abundantly that exert a stimulatory role in exocrine secretion. This study was undertaken to examine effects of GABA on an action of cholinergic neurons in exocrine secretion of the pancreas. Intrinsic neurons were excited by electrical field stimulation (EFS; 15 V, 2 msec, 8 Hz, 45 min) in the isolated, perfused rat pancreas. Tetrodotoxin or atropine was used to block neuronal or cholinergic action. Acetylcholine was infused to mimic cholinergic excitation. GABA $(30{\mu}M)$ and muscimol $(10{\mu}M)$, given intra-arterially, did not change spontaneous secretion but enhanced cholecystokinin (CCK; 10 pM)-induced secretions of fluid and amylase. GABA (3, 10, $30{\mu}M$) further elevated EFS-evoked secretions of fluid and amylase dose-dependently. GABA (10, 30, $100{\mu}M$) also further increased acetylcholine $(5{\mu}M)$-induced secretions of fluid and amylase in a dose-dependent manner. Bicuculline $(10{\mu}M)$ effectively blocked the enhancing effects of GABA $(30{\mu}M)$ on the pancreatic secretions evoked by either EFS or CCK. Both atropine $(2{\mu}M)$ and tetrodotoxin $(1{\mu}M)$ markedly reduced the GABA $(10{\mu}M)$-enhanced EFS- or CCK-induced pancreatic secretions. The results indicate that GABA enhances intrinsic cholinergic neuronal action on exocrine secretion via the $GABA_A$ receptors in the rat pancreas.

  • In this study geostatistical technique using indicator kriging was performed to evaluate the optimal rock mass classification by integrating the various geophysical information such as borehole data and geophysical data. To get the optimal kriging result, it is necessary to devise the suitable technique to integrate the hard (borehole) and soft (geophysical) data effectively. Also, the model parameters of the variogram must be determined as a priori procedure. Iterative non-linear inversion method was implemented to determine the model parameters of theoretical variogram. To verify the algorithm, behaviour of object function and precision of convergence were investigated, revealing that gradient of the range is extremely small. This algorithm for the field data was applied to a mountainous area planned for a large-scale tunneling construction. As for a soft data, resistivity information from AMT survey is incorporated with RMR information from borehole data, a sort of hard data. Finally, RMR profiles were constructed and attempted to be interpreted at the tunnel elevation and the upper 1D level.

Analysis of Middle and High School Technology Teachers' Recognition and Educational Needs about Engineering Education (중·고등학교 기술교사의 공학교육에 대한 인식 및 교육요구도)

  • The purpose of this study is to analyze middle and high school technology teachers' recognition and educational need about engineering education. For this, we surveyed secondary school technology teachers. The result of this study is as follow. First, most technology teachers perceive that engineering contents are lack in technology education, therefore they recognize the needs of reinforcement of engineering contents. Second, most technology teachers perceive that present technology subject does not give positive affect to students for their own career selection on engineering field. Furthermore, they perceive that if it contained the contents of engineering in technology education, students would experience creative design and problem solving process. Third, most technology teachers perceive that they are able to teach engineering in technology education and they need the change of national curriculum and the development of engineering program contents. Fourth, they perceive that the 7~9 grade is the best grade to start to study engineering in elementary and secondary school. Fifth, they perceive that 'design', 'problem solving' and 'creative thinking' are the priority of educational needs of teaching ability and knowledge about engineering. Sixth, they perceive that the aerospace engineering, the electrical engineering, the electronic engineering, the mechanical engineering, the computer engineering and the environmental engineering are the priority of educational needs of teaching knowledge about engineering fields.

The Poly-Si Thin Film Transistor for Large-area TFT-LCD (대면적 TFT-LCD를 위한 다결정 실리콘 박막 트랜지스터)

  • In this paper, the n-channel poly-Si thin-film transistors (poly-Si TFT's) formed by solid phase crystallization (SPC) on glass were investigated by measuring the electrical properties of poly-Si films, such as I-V characteristics, mobility, leakage current, threshold voltage, and subthreshold slope. It is done to decide to be applied on TFT-LCD with large-size and high density. In n-channel poly-Si TFT with 2, 10, 25$\mu\textrm{m}$ of channel length, the field effect mobilities are 111, 126 and 125 $\textrm{cm}^2$/V-s and leakage currents are 0.6, 0.1, and 0.02 pA/$\mu\textrm{m}$, respectively. Low threshold voltage and subthreshold slope, and good ON-OFF ratio are shown, as well. Thus, the poly-Si TFT’s used by SPC are expected to be applied on TFT-LCD with large-size and high density, which can integrate display panel and peripheral circuit on a large glass substrate.

TID and SEL Testing on OP-Amp. of DC/DC Power Converter (DC/DC 컨버터용 OP-Amp.의 TID 및 SEL 실험)

  • DC/DC switching power converters are commonly used to generate a regulated DC output voltage with high efficiency. The advanced DC/DC converter uses a PWM-IC with OP-Amp. (Operational Amplifier) to control a MOSFET (metal-oxide semiconductor field effect transistor), which is a switching component, efficiently. In this paper, it is shown that the electrical characteristics of OP-Amp. are affected by radiations of ${\gamma}$ rays using $^{60}Co$ for TID (Total Ionizing Dose) testing and 5 heavy ions for SEL (Single Event Latch-up) testing. TID testing on OP-Amp. is accomplished up to the total dose of 30 krad, and the cross section($cm^2$) versus LET($MeV/mg/cm^2$) in the OP-Amp. operation is evaluated SEL testing after implementation of the controller board.

Effect of Zeolite Application on Growth and Yield of Chinese Cabbage and Chemical Properties of Soil Under Greenhouse Cultivation

  • Zeolite may help crop growth, yield increase, and salt removal. Field experiment under greenhouse cultivation was conducted to study the effect of zeolite application on growth and yield of Chinese cabbage (Brassica campestris L.) and soil. Soil was classified as Gyuam series (coarse silty, mixed, nonacid, mesic family of Aquic Fluvaquentic Eutrudepts). Six zeolite rates were 0, 3, 5, 10, 20 and $40Mg\;ha^{-1}$. Experimental design was a completely randomized design. Chinese cabbage was grown three times consecutively. Established plant number of plant and yield as fresh weight (F.W.) were measured and soil samples were taken before and after harvesting. Chinese cabbage yield was $76.9Mg\;ha^{-1}$ at a rate of $20Mg\;zeolite\;ha^{-1}$, $54.3Mg\;ha^{-1}$ at a rate of $5Mg\;zeolite\;ha^{-1}$, and $51.3Mg\;ha^{-1}$ at control (no zeolite), respectively. Second order regression analysis using zeolite rate and yield showed that optimum zeolite application rate was between 24 and $26Mg\;ha^{-1}$. The regression equation explained about 88% of the yield variability. The electrical conductivity (EC) decreased from 3.2 to $1.0dS\;m^{-1}$ for all treatments so that salt accumulation was not a concern. Based on the results, we recommend that optimum zeolite application rate is between 20 and $24Mg\;ha^{-1}$ for Chinese cabbage under greenhouse cultivation.

Usefulness of Electrophysiological Tests in Movement Disorders (이상운동 질환에서의 신경생리적 검사법의 유용성)

  • In clinical neurology various different electrophysiological tests are widely used to demonstrate the unsuspected malfunctioning in the nervous system and to monitor over time the clinical status of patients. In addition clinical neurologists and neurosurgeons take advantage of the intraoperative monitorings to increase the quality of neurosurgical operations in the posterior fossa, in the spinal cord, or in visual pathways. In the field of movement disorders, elecrophysiolgical tests provide neurologists with making accurate differential diagnoses with useful therapeutic stratergies as well as with investigating the pathophysiological machanisms. By using the electromyographic tests it could be possible for us to evaluate the types of blephalospasm, the extent of hemifacial spasm, the level of myoclonus, and the prime muscles of torticollis etc. Sometimes the myographic guidance may be critical for choosing the exact injecting site of botulinum toxin. These several decades various electroencephalographic and evoked potential tests has been utilized in the electrophysiological laboratories to understand the basic pathophysiology of myoclonus, spasticity and other central motor dysfunctions. It could be one of the breakthroughs in the area of behavorial neurology that the brain function can be mapped by the spontaneous or evoked electrical activities of nervous system since the movement related potentials (MRPs) had been studies for several decades. Various reflex tests such as masseter reflex, blink reflex, click evoked vestibulocollic reflex, facial reflex, stretch reflex, flexor reflex, H-reflex, H-reflex recovery curve, vestibular inhibition of H-reflex, reciprocal inhibition, recurrent or Renshaw reflex, Ib inhibition, cutaneous reflex have been also used to understand normal or abnormal physiology in movement disorders. Polysomnography, posturography and gait studies are also applied in clinical neurology in association with with movement disorders which are useful in deciding the treatment regimen.

Understanding of Growth Habits of $VO_2$ Film on Graphene and Their Effects on Metal to Insulator $Transition_2$

  • Growth of metal oxides on graphene may lead to a better understanding of delicate effects of their growth habits on their underlying physics. The vanadium dioxide ($VO_2$) is well known for its metal-to-insulator transition accompanied by a reversible first order structural phase transition at 340 K. This transition makes $VO_2$ a potentially useful material for applications in electrical and optical devices. We report a successful growth of $VO_2$ nanostructures on a graphene substrate via a vapor-solid transport route. As-grown $VO_2$ nanostructures on graphene were systematically characterized by field emission scanning electron microscopy, x-ray diffraction, Raman spectroscopy, FT-IR spectroscopy and high resolution transmission electron microscopy. These results indicate that the strain between $VO_2$ and graphene layers may be easily controlled by the number of underlying graphene layer. We also found that the strain in-between $VO_2$ and graphene layer affected its metal-to-insulator transition characteristics. This study demonstrates a new way for synthesizing $VO_2$ in a desired phase on the transparent conducting graphene substrate and an easy pathway for controlling metal-to-insulator phase transition via strain.
