Correlation for Human in Vitro Fertilization with the Zona-Free Hamster Penetration Assay (인간의 체외수정과 투명대제거 햄스터난자의 침투능에 관한 연구)

  • We compared fertilizing potential measurements by the zona-free hamster egg penetration assay with the in vitro fertilization and embryo transfer program was evaevulated for their ability to fertilize zona free hamster egg. Spermatozoa from 12 presumeably fertile donors and from the male partners of 56 infertile couples were evaluated for their ability to fertilizing potentials. Penertration rates of fertile donors were $36.2{\pm}27.7%$ ; Fertilization rates of infertile couples between with normal semen parameters and with abnormal semen parameters were $28.7{\pm}19.1$, $5.7{\pm}8.9%$, respectively. Sperm motility of couples with penetration rates between on 15-30% and on 30> were $54.1{\pm}4.6$, $55.5{\pm}8.3%$ respectively. Hamster penetration rates of couples participating in an in vitro fertilization and embryo transfer program was $38.9{\pm}29.9%$. But in one case, a positive fertility assessment was obtained in the absence of fertilization of the wife's eggs attributable to egg immaturity. This method may have potential value as a diagnostic tool in evaluation human sperm fertilization capacity which avoids the ethical and logistical problems associated with fertilizing of human eggs in vitro.

Relationship between Differential Expression of Estrogen Receptor and Follicle Stimulating Hormone Receptor Genes in Ovary and Heterosis of Egg Number Traits in Chickens

  • In order to understand the molecular mechanism of heterosis of reproduction traits in chickens, we used the quantitative real-time reverse transcriptional polymerase chain reaction (Quantitative real-time RT-PCR) technique to investigate the differential expression of estrogen receptor (ESR) and follicle stimulating hormone receptor (FSHR) genes in 32-week-old ovaries of inbred chickens and their hybrid offspring in $4{\times}4$ diallel crosses, which involved White Plymouth Rock (E), CAU Brown (D), Silkies (C) and White Leghorn (A). We found that there were significant differences in mRNA expression of ESR and FSHR genes not only between hybrids and their parental lines (p<0.01), but also among different crosses (p<0.01). Furthermore, positive correlations between differential expression of both ESR and FHSR in hybrids and heterosis percentages of 32-week-old and 42-week-old egg number traits were significant at p<0.05. Our results suggested that differential expression of ESR and FSHR genes in the ovaries of inbred chickens and their hybrids could play roles in the formation of heterosis of egg number traits to some extent.

Analysis of Microbial Contamination in Commercial Saengshik Products (유통 생식제품의 미생물 오염 분석)

  • This study was performed to assess the presence of contaminated microorganisms of Escherichia coli, Clostridium perfringens, and Bacillus cereus in the 112 commercial Saengshik products. E. coli was not detected in all the samples, but C. perfringens was detected in 11 products (9.8%). The number of the bacteria was less than 100 CFU/g, which was satisfactory to KFDA microbiological requirement. B. cereus was detected less than $10^2{\sim}10^3$ CFU/g in 7 products and $10^3{\sim}10^4$CFU/g in 13 products out of 25 products. Those detected bacteria from tryptose sulphite cycloserine agar and mannitol egg yolk polymyxin agar showed the typical characteristics of Gram positive and contained lecithinase, which can decompose egg-yolks layers in the biochemical test. Therefore, much more attention must be applied to satisfy the B. cereus requirement for Saengshik products sold in the market.

  • A total of 860 cellotape anal swab specimens from 326 school children of urban area, 243 of rural farm village and 291 of rural fishing village was submitted for Enterobius vermicularis egg detection from September through November, 1985. Each specimen was collected in the morning. Among the total 860 children's submitting specimens, 316 (36.7%) were positive for E. vermicularis eggs. Of the 326 children in the urban area, E. vermicularis eggs were detected in the 45 (13.8%). This detection rate was lower significantly as compared with 143 (55.6%) of 243 children in the farm village or 136 (46.7%) of 291 children in the fishing village. The egg detection rate in the male children was lower remarkably than that in the female children.

The Influence of Microinjection of Foreign Gene into the Pronucleus of Fertilized Egg on the Preimplantation Development, Cell Number and Diameter of Rabbit Embryos

  • The aim of this in vitro study was to test the effect of microinjection (Mi) of foreign gene into the rabbit egg pronucleus and epidermal growth factor (EGF) addition on the blastocyst rate, the cell number and the diameter of embryos, and to determine possible relationships between embryo cell number and embryo diameter. Blastocyst rate was significantly decreased in gene- Mi (G-Mi/E0) group (63.1%) comparing to intact ones (83.5%, $p_1$<0.05). The addition of EGF at 20ng/ml (G-Mi/E20) or 200 ng/ml (GMi/ E200) to gene-Mi embryos did not affect blastocyst rate (65.6 and 55.2% resp.). As a control for Mi, the eggs were microinjected with the same volume of phosphate-buffered solution (PBS-Mi) instead of the gene construct solution. Cell numbers and embryo diameters were measured from embryo images obtained on confocal laser scanning microscope. Bonferroni-modified LSD test showed that the embryo cell number in PBS-Mi group was significantly lower ($p_1$<0.05) and in gene-Mi group was tended to decrease compared with intact embryos. Embryo diameter was not different among experimental groups. No effect of EGF given at any doses both on the cell number and embryo diameter was found. A positive correlation between cell number and embryo diameter was observed in all groups of embryos. Since embryo diameter was not changed under the influence of Mi or EGF addition in this study, this seems to be more conservative characteristics of the embryo morphology. These results suggest that the pronuclear microinjection compromises developmental potential of embryos, decreasing blastocyst rate and embryo cell number, whilst embryo diameter is not affected. No effects of EGF on studied parameters were confirmed. Declined quality of Mi-derived embryos is caused by the microinjection procedure itself, rather than by the gene construct used.

  • Contaminated liver fluke egg in the environment has led to the high prevalence of human opisthorchiasis associated with cholangiocarcinoma in Southeast Asia. To find the effective lessening methods of Opisthorchis viverrini eggs in the contaminated environment, we investigated the temperature conditions for killing of these trematode eggs in vitro. Numerous O. viverrini eggs were obtained in the proximal part of uteri of adult worms from experimental hamsters. Mature eggs with miracidium were allocated by experimental groups (2 control: positive and negative and 4 treatment: 50, 60, 70, and $80^{\circ}C$) with 0.85% saline, and treated by the experimental plan. Eggs in each experimental groups were observed under the confocal microscope after stain with Propidium Iodide (PI) to evaluate the effect of temperatures. Eggs in 70 and $80^{\circ}C$ groups were all killed after over 10 min heated. Majority of eggs in $60^{\circ}C$ (10, 15, and 30 min heated), 70 and $80^{\circ}C$ (5 min heated) groups were inactivated. However in $50^{\circ}C$ group, below half of eggs were to be killed in all time lapse (10, 15 and 30 min). In order to prevent O. viverrini infection and cholangiocarcinoma, direct treatment of sewage by heating at 70 or $80^{\circ}C$ at least 10 min is essential. Therefore, treatment of O. viverrini eggs at a high temperature is a potential method for controlling egg contamination in sewage.

  • Increase in daily protein consumption per capita from 1975(85.1 g) to 2001(88.4 g) was 3.3 g. This trend was relatively slower than the case of Japan where daily protein consumption was 84.7 g in 1975 and 90.3 g in 2001. Animal-related protein in 2003 was 45.7 g in which 61% was originated from meat, milk and egg whereas 39% was composed of fish and its relevance. The trend of protein consumption fairly come up with the ideal ratio of 5:5 between animal-originated protein and plant-originated protein, following the base case of Japan. The effect of animal protein on human health can vary depending on one's viewpoint and its controversy is still a subject of debate. For reason, two faces of positive and negative effects on human health coexists. However, there is no doubt that positive effect is far more than negative one. It is not important whether or not animal protein is more beneficial for human health. However, it is more important how human balance between two proteins.

High endemicity of Metagonimus yokogawai infection among residents of Samchok-shi, Kangwon-do

  • A small-scale epidemiological survey was undertaken during 1997-1998 on the residents along the Osib-chon (Stream), Samchok-shi (City), Kangwon-do (Province), to evaluate the status of Metagonimus yokogawai infection. A total of 165 fecal samples was collected and examined by cellophane thick smear and formalin-ether sedimentation techniques. The egg Positive rate of M. yokogawai was 29.7%, showing a remarkable difference between males (46.6%) and females (16.3%). To obtain the adult flukes of M. yokogawai, 11 egg positive persons were treated with praziquantel and purged with magnesium sulfate. A total of 242,119 adult flukes (average 22,010 per person, 367 119,650 in range) was collected from diarrheic stools, all of which were identified as M. yokogawai. The results show that M. yokogawai is still highly endemic in this area.

Diagnostic Value of the Intradermal Test for the Infection with Clonorchis sinensis (간흡충증 진단을 위한 피내반응검사의 의의)

  • The intradermal(ID) test has been widely used in Korea and several reports about the results of the ID test are known. We examined the egg of Clonorchis sinensis(C. s.) by ID test in 443, stool's egg-counting technique in 79 and direct smear(cellophane thick smear technique) in 1304 subjects. The results are as follows.: 1. The positive rate of C. s. was 3.8% out of 1304 persons. 2. The sensitivity of ID test was 82.1% out of 39 persons and, the specificity was 64.6% out of 404 persons. 3. The false positive of ID test 35.4% out of 404 persons and, the false negative was 17.9% out of 39 persons. Intradermal test is a rapid, sensitive and useful supplementary diagnostic tool for the detection of Clonorchiasis infection and must be used as screening test with direct smear of stool but cross reaction with other helminth infections and moderate false reaction are the main disadvantages in its practical application.

Prevalence of Gymnophalloides seoi infection in coastal villages of Haenam-gun and Yeongam-gun, Republic of Korea

  • One coastal village in Haenam-gun and two in Yeongam-gun, Jeollanam-do were surveyed for intestinal parasite infections by fecal examination. The egg positive rates of Gymnophalloides seoi were high, $24.1\%$ (14/58) in Haenam-gun and $9.3\%$ (11/118) in Yeongam-gun. The egg positive rates of heterophyids, including Heterophyes nocens, and of Clonorchis sinensis were $10.3\%\;and\;6.9\%$ in Haenam-gun, and $14.4\%\;and\;8.5\%$ in Yeongam-gun, respectively. After praziquantel treatment and purgation, a total of 37,761 fluke specimens were recovered from 17 patients; 11 in Haenam-gun and 6 in Yeongam-gun. Gymnophalloides seoi was the most commonly recovered species, with 37,489 specimens in total (2,205 per person). Other recovered flukes included Heterophyes nocens, Stictodora fuscata, Heterophyopsis continua, Pygidiopsis sUmma, and undetermined species. These results indicate that the areas surveyed are new endemic foci of G. seoi.
