Effect of Pine Silage Feeding on Nutrient Digestibility, Feed Conversion and Carcass Traits of Korean Native Cattle (잣나무 生枝葉사일리지 급여가 한우의 영양소 소화율, 사료요구율 및 도체성적에 미치는 영향)

  • The wastes (the needle leaves and branches) produced from thinning the forest were fermented under an anaerobic condition (pine silage) to utilize as a forage source of ruminants. An in situ trial was conducted with two ruminally fistulated Korean native cows by 4 replicates (2 bags per cow), and in vivo digestibility of pine silage was estimated with five Korean native steers by 5 replicates in incomplete double turn-over design. In order to investigate feed efficiency and carcass traits, forty eight Korean native bulls were assigned to four treatments (0%, 25%, 50%, 75% of pine silage) with a completely randomized design in 12 pens accommodating 4 animals per pen. The amounts of concentrate and roughage allowed to experimental animals were in the range of 2.5% and 0.6% of body weight, respectively. Animals had freely accessed to mineral block and water in stanchion barn.An in situ crude protein digestibility of the roughage sources when suspended for 48 hrs in the rumen was higher for feeding the pine silage than the rice straw, whereas NDF digestibility was vice versa. No differences between the treatments were observed in in vivo digestibilities of dry matter, crude protein and NDF. Daily body weight gain was significantly (P<0.01) higher for the pine silage substitutions compared with the rice straw feeding except for the 25% silage feeding. The pine silage feeding did not affect feed efficiency.In carcass traits, marbling score tended to be higher for the 50% pine silage treatment than the other treatments while back fat thickness tended to be the thinnest for the rice straw feeding. From these results, it may be recommended to substitute pine silage, as a forage source, for rice straw by 50 percent.

Extraction Characteristics of Saponin and Acidic Polysaccharide Based on the Red Ginseng Particle Size (홍삼의 입자크기에 따른 사포닌 및 산성다당체의 추출 특성)

  • Effect of pulverization on total solid, crude saponin, and acidic polysaccharide contents of dried red ginseng main root were tested. Several particle size samples, including red ginseng main root (non pulverized), $10{\sim}40$ mesh powder, $40{\sim}100$ mesh powder, and >100 mesh powder were used in the extraction. The sequential solvent extraction method (1st: 70% EtOH at $70^{\circ}C$ for 12 hr, 2nd: 70% EtOH at $70^{\circ}C$ for 12 hr, 3rd: water at $70^{\circ}C$ for 12 hr) was applied to extract the saponins and acidic polysaccharide. Extraction yield of total solid of pulverized red ginseng ($10{\sim}40$ mesh size) was increased to 20% compared with that of non-pulverized. Especially, the crude saponin content of pulverized red ginseng ($10{\sim}40$ mesh size) showed an increase of 47% over non-pulverized. No difference in the component ratio was observed by pulverization, when the individual ginsenosides were quantified by HPLC. Also, extraction yield of acidic polysaccharide of pulverized red ginseng ($10{\sim}40$ mesh size) was increased 57% compared with that of non-pulverized. The results suggested that pulverization might be useful for increasing the extraction yield of red ginseng components.

Effect of the Plants Mixture and Garlic Composition on Serum Lipid Level of Hypercholesterolemic Rats (식물류 혼합물과 마늘의 복합 조성이 고콜레스테롤혈증 흰쥐의 혈청 지질에 미치는 영향)

  • This study was designed to investigate the effects of 4 kinds of plant water extract mixture and garlic extract (PMC) administration on serum lipid metabolism in hypercholestrolemic rats. The normal group was administered a cholesterol free diet, the control group a 1% cholesterol diet, and each experimental group was given a diet of 1% cholesterol, 1% plant mixture and 0.3, 0.5, 0.7% garlic extract (PMC-I, PMC-II, PMC-III), respectively. Each diet was administered orally to SD-male rats for 4 weeks. Total cholesterol content decreased by about 20% with administration of PMC. Triglyceride content also decreased from 9.3 to 15.0% compared to the control group, and phospholipid was similar to triglyceride. There was no significant difference in HDL-cholesterol content between the control and experimental groups. LDL-cholesterol content of the normal group was 9.4 times lower than the control group and its content was significantly lower in the PMC-II ($68.45{\pm}12.83\;mg/dl$) and PMC-III ($66.35{\pm}5.18\;mg/dl$) groups than the PMC-I group. VLDL-cholesterol content of the PMC-II and III groups were similar to the normal group. Atherogenic index (AI) and cardiac risk factor (CRF) were significantly lower in the PMC group. Blood glucose content was the lowest in the PMC-II ($189.37{\pm}12.02\;mg/dl$) group among all groups tested. Total protein content was $9.56{\pm}0.87{\sim}10.05{\pm}2.69\;mg/dl$ in the PMC-I~III groups and was significantly higher than the normal group. CPT activity did not show a significant difference among the experimental groups, while COT activity was effective only in the PMC-I group. Serum TBARS content in the PMC-III group was lower than in the normal group. Serum antioxidant activity by DPPH radical scavenging was $83.75{\pm}2.32%$ in the PMC-III group, which was significantly higher than the control group.

  • The Ag-Pd-Cu alloys containing a small amount of Au is commonly used for dental purposes, because this alloy cheaper than Au-base alloys for clinical use. However, the most important characteristic of this alloy is age-hardenability, which is not exhibited by other Ag-base dental alloys. The specimens used were Ag-30Pd-10Cu ternary alloy and Au addition alloy. These alloys were melted and casted by induction electric furnace and centrifugal casting machine in Ar atmosphere. These specimens were solution treated for 2hr at $800^{\circ}C$ and were then quenched into iced water, and aged at 350-$550^{\circ}C$ Age-hardening characteristic of the small Au-containing Ag-Pd-Cu dental alloys were investigated by means of hardness testing, X-ray diffraction and electron microscope observations, electrical resistance, differential scanning calorimetric, energy dispersed spectra and electron probe microanalysis. Principal results are as follows ; Maximum hardening occured in two co-phases of ${\alpha}_2$ + PdCu In stage II, decomposition of the $\alpha$ solid solution to a PdCu ordered phase($L1_o$ type) and an Ag-rich ${\alpha}_2$ phase occurred and a discontinuous precipitation occurred at the grain boundary. From the electron microscope study, it was concluded that the cause of age-hardening in this alloy is the precipitation of the PdCu redered phase, which has AuCu I type face-centered tetragonal structure. Precipitation procedure was ${\alpha}{\to}{\alpha}_1+PdCu{\to}{\alpha}_2+PdCu$ at Pd/Cu = 3 Pd element of Ag-Pd-Cu alloy is more effective dental alloy on anti-corrosion and is suitable to isothermal ageing at $450^{\circ}C$.

Sprouting Characteristics and Herbicidal Responses of Purple Nutsedge (향부자 괴경의 출아특성과 제초제에 대한 반응)

  • To establish an efficient herbicide screening method for purple nutsedge(Cyperus rotundus) control, its sprouting characteristics, tuber production and responses on several herbicides were investigated under greenhouse condition. The tubers stored at $4^{\circ}C$ after sterilization with the diluted prochloraz(Spotac) solution showed higher sprouting than the non-sterilized did. The harvested tubers were not dormant, and the sterilized tubers which stored at low temperature had a sprouting capability of about 80% after 6 months. If the fresh weight of purple nutsedge tubers was decreased to below 48%, they could not sprout. However, the tubers soaked in water and then stored at low temperature could sprout by 88% even 6 months later. Sprouting and initial growth of tuber were much better at $35^{\circ}C$-day/$25^{\circ}C$-night than at 30/$20^{\circ}C$ or 25/$15^{\circ}C$. The half-sected tubers, which were prepared by setting the intact tuber of above 1.2g latitudinally, were shown similar initial growth to the intact but those sected crucifically were not. These results suggest that the half-sected tuber itself can be used as a material on herbicide screening. About 1000 tubers could be harvested when 10 tubers planted in a pot($56{\times}35{\times}16cm$) filled with the artificial soil were cultivated in greenhouse of $35^{\circ}C$-day/$25^{\circ}C$-night for 3 months(April-July, 1993). Chlorimuron, Bentazon and Norflurazon were selected as the standards for the screening because of providing relatively effective control on purple nutsedge in both soil-surface and foliar spray treatment.

Preparation and Characterization of Bamboo-based Activated Carbon by Phosphoric Acid and Steam Activation (인산 및 수증기 활성화에 의한 대나무 활성탄 제조 및 특성 연구)

  • Bamboo is an evergreen perennial plant, and it is known as one of the most productive and fastest-growing plants in the world. It grows quickly in moderate climates with only moderate water and fertilizer. Traditionally in Asia, bamboo is used for building materials, as a food source, and as versatile raw materials. Bamboo as a biomass feedstock can be transformed to prepare activated carbon using the thermal treatment of pyrolysis. The effect of process variables such as carbonization temperature, activation temperature, activation time, the amount of steam, and the mixing ratio of phosphoric acid and bamboo were systematically investigated to optimize the preparation conditions. Steam activation was proceeded after carbonization with a vapor flow rate of $0.8{\sim}1.8mL-H_2O\;g-char^{-1}\;h^{-1}$ and activation time of 1 ~ 3 h at $700{\sim}900^{\circ}C$. Carbon yield and surface area reached 2.04 ~ 20.59 wt% and $499.17{\sim}1074.04m^2\;g^{-1}$, respectively, with a steam flow rate of $1.4mL-H_2O\;g-char^{-1}\;h^{-1}$ for 2 h. Also, the carbon yield and surface area were 24.67 wt% and $1389.59m^2\;g^{-1}$, respectively, when the bamboo and phosphoric acid were mixed in a 1:1 weight ratio ($700^{\circ}C$, 2 h, $1.4mL-H_2O\;g-char^{-1}\;h^{-1}$). The adsorption of methylene blue into the bamboo activated carbon was studied based on pseudo first order and second order kinetics models. The adsorption kinetics were found to follow the pseudo second order model, which is governed by chemisorption.

Landslide Susceptibility Mapping Using Deep Neural Network and Convolutional Neural Network (Deep Neural Network와 Convolutional Neural Network 모델을 이용한 산사태 취약성 매핑)

  • Landslides are one of the most prevalent natural disasters, threating both humans and property. Also landslides can cause damage at the national level, so effective prediction and prevention are essential. Research to produce a landslide susceptibility map with high accuracy is steadily being conducted, and various models have been applied to landslide susceptibility analysis. Pixel-based machine learning models such as frequency ratio models, logistic regression models, ensembles models, and Artificial Neural Networks have been mainly applied. Recent studies have shown that the kernel-based convolutional neural network (CNN) technique is effective and that the spatial characteristics of input data have a significant effect on the accuracy of landslide susceptibility mapping. For this reason, the purpose of this study is to analyze landslide vulnerability using a pixel-based deep neural network model and a patch-based convolutional neural network model. The research area was set up in Gangwon-do, including Inje, Gangneung, and Pyeongchang, where landslides occurred frequently and damaged. Landslide-related factors include slope, curvature, stream power index (SPI), topographic wetness index (TWI), topographic position index (TPI), timber diameter, timber age, lithology, land use, soil depth, soil parent material, lineament density, fault density, normalized difference vegetation index (NDVI) and normalized difference water index (NDWI) were used. Landslide-related factors were built into a spatial database through data preprocessing, and landslide susceptibility map was predicted using deep neural network (DNN) and CNN models. The model and landslide susceptibility map were verified through average precision (AP) and root mean square errors (RMSE), and as a result of the verification, the patch-based CNN model showed 3.4% improved performance compared to the pixel-based DNN model. The results of this study can be used to predict landslides and are expected to serve as a scientific basis for establishing land use policies and landslide management policies.

A Concentrated Onion Extract Lowers Serum Lipid Levels in Rats Fed a High-Fat Diet (흰쥐에서 양파 농축액의 고지혈 개선 작용)

  • It is known that onions, or bioactive compounds therein, providehealth benefits. The present study was designed to investigate whether a concentrated onion extract lowered blood lipid levels in rats fed a high-fat diet. Initially, male Sprague-Dawley rats were housed singly in an environment in which temperature and light duration were controlled, and had free access to a nutritionally adequate AIN-93G diet and deionized water. After an acclimatization period, rats were weight-matched and assigned to one of the following five groups: 1) a control group, fed the AIN-93G diet mixed with 10% (w/v) lard and 0.7% (w/v) cholesterol to induce hyperlipidemia (control); 2) three experimental groups, fed the AIN-93G diet mixed with a high-fat source plus concentrated onion extract at three different levels (termed the low-dose, medium-dose, and high-dose groups); and, 3) a placebo group, fed the AIN-93G diet with fats plus the same concentrated extract but devoid of onion-derived material. All five groups freely ingested their respective diets over 6 weeks. At 0, 3, and 6 weeks, blood samples were collected from the orbital sinus following overnight food deprivation. At 6 weeks, livers were collected. Both control and experimental groups continually gained body weight throughout the study. No significant differencein body weight gain was observed among groups. However, the serum concentrations of triglycerides, total cholesterol, and non HDL-cholesterol were significantly reduced by ingestion of concentrated onion extract. Also, the hepatic levels of total lipids and total fatty acids, especially C18:1 (oleic acid), were significantly decreased in rats fed a high level of concentrated onion extract, compared with the control and placebo groups. These results provide clear evidence that ingestion of a concentrated onion extract has a profound inhibitory effect on serum lipid levels in rats fed a high-fat diet. Our findings indicate that a concentrated onion extract may be used to alleviate hyperlipidemia by lowering serum cholesterol and triglyceride levels.

A Study on Coagulation and Ozone Treatment of Nonbiodegradable Substances in Landfill Leachate (생물학적 난분해성물질을 함유한 매립장 침출수의 응집 및 오존처리에 관한 연구)

  • The objectives of this study are summarized with two. The one is to remove of $COD_{Cr}$, color and turbidity with coagulation and ozonation in nonbiodegradable landfill leachate, the other is to compare of water qualities with pre-ozonation and post-ozonation. The results are summarized as follows ; 1) 90 minutes ozonation with $75mgO_3/min$($4.5gO_3/hr$) was conducted at pH 4,7, 10 to remove $COD_{Cr}$. Removal efficiencies were investigated with 48.2%, 52.6%, and 62.3% respectively. As increasing pH, $COD_{Cr}$ removal efficiencies were increased, it was considered that hydroxyl radical($OH{\cdot}$) which strongly oxdize and nonselectively react with organic compounds, was rapidly produced by ozone self-decomposition at high pH. 2) Alum, ferric chloride and ferrous sulfate were used for coagulation as inorganic coagulant. Ferric chloride was investigated with optimal coagulant, and it removed $COD_{Cr}$ about 12.0% at pH5 and dosage of $2,000m{\ell}/{\ell}$. Cation(C-101P), anion(A-601P) and nonion(SC-050) were tested to remove organic pollutants in landfill leachate. Cation(C-101P) was investigated with the most effective organic coagulant, and removal efficiency of $COD_{Cr}$ was 19.8% at pH5 and dosage of $100m{\ell}/{\ell}$. 3) Color and turbidity were removed up to 88.6%, 97.0% at pH10, when contacted 90 minutes with ozone, respectively. These removal efficiencies were much higher than those of $COD_{Cr}$ and $COD_{Mn}$. It was considered that ozone could oxdize the triggering materials of color and turbidity selectively and preferentially. 4) $COD_{Cr}$, color and turbidity were more effectively removed with pre-ozonation than post-ozonation about 8%, 3.5% and 1% respectively. These results were well corresponed with other's studies that pre-ozonation will increase the effect of coagulation.

Hypoglycemic Effects of Pills Made of Mulberry Leaves and Silkworm Powder in Streptozotocin-Induced Diabetic Rats (뽕잎과 누에가루 혼합환의 Streptozotocin유발 당뇨쥐에서의 혈당강하 효과)

  • This study was carried out to examine the effects of pills made of mulberry leaves and silkworm powder on lowering blood glucose level. Experimental animals were Sprague-Dawley male rat weighing 100$\pm$10 g and pills were supplemented with 0.4% (4 g/kg) diet. Experimental groups were assigned to diabetic group (DM group) and pill supplemented groups. Pill supplemented groups were classified 100% mulberry leaves (M group), mixing 25% silkworm powder to mulberry leaves (25SM group), mixing 50% silkworm powder to mulberry leaves (50SM group), mixing 70% silkworm powder to mulberry leaves (75SM group) and 100% silkworm powder (100S group). Experimental diets and water fed ad libitum, and streptozotocin was injected to induce diabetic state after 3rd weeks and sacrificed on the 9th day. The contents of 1-deoxynojirimycin(DNJ) were increased with adding the silkworm powder. The contents of GABA and rutin were increased with adding the mulberry leaves. In vitro, intestinal mucosa $\alpha$-glucosidase inhibitory activities were significantly increased in pills which mixed with silkworm powder by 50%. Blood glucose levels were high in groups which mixed with silkworm powder by 50% compared to DM group. Intestinal mucosa maltase activity in proximal part was significantly reduced in pill supplemented group compared to DM group and pill supplemented groups were no significant difference. Enzyme activity in middle part was no significant difference in experimental groups. Enzyme activity in distal part was decreased in pill supplemented groups, especially in 50SM, 75SM and 100S groups were significantly reduced compared to DM group. Sucrase and lactase activities in pill supplemented groups were significantly reduced at proximal part, and there was no significant difference in middle and distal parts. In conclusion, pills made of mulberry leaves and silkworm powder increased the $\alpha$-glucosidase inhibitory activity in vitro and reduced the blood glucose levels by controlling the disaccharidase activities of intestinal proximal part in STZ-induced diabetic rat. The synergistic effect was the highest when mulberry leaves was mixed with silkworm powder by the ratio of 50 : 50.
