Dermatan sulfate (DS) was isolated from eel skin (Anguilla japonica) bv actinase and endonuclease digeslions followed by ${\beta}$-elimination reaction and DEAE-Sephacel chromatography. DS was a major glycosaminoglycan in eel skin with 88% of the total uronic acid. The content of IdoA2S$\alpha$1longrightarrow4GalNAc4S sequence in eel skin. which is known to be a binding site to heparin cofactor II. was two times higher than that of dermatan sulfate from porcine skin. The anti-lla activity of eel skin dermatan sulfate mediated through heparin cofactor ll(NCL) was 25 units/mg. whereas DS from porcine skin shows 23.2 units/mg. The average molecular weight was determined as 14 kDa by gel chromatography on a TSKgel G3000SWXL column. Based on H1 NMR spectroscopy. we suggest that 3-sulfated and/or 2.3-sulfated ldoA residues are present in the chain.

This experiment was carried out in order to evaluate the immunomodulatory activity of levamisole (LMS) in 5. fgrjn challenged eels with different treatment regimens: 7-day LMS treatment before the challenge, 7-day LMS treatment started simultaneously with the challenge, 14-day treatment before and after the challenge. The antibacterial effect was activated in all treated groups, with the best being obtained in the simultaneously treated group. LMS stimulated the defense mechanisms of the eel as demonstrated by increase in the level of total protein, albumin, trypsin inhibitor capacity, lysozyme activity, antibody titers antibacterial activity and survival rate. These results suggest that antibacterial effects of LMS was achieved by not only non-specific immune response but also specific one in eel.
This study aimed to develop eel dipping sauce adding bokbunja(Rub coreanus Miquel) vinegar and investigated the physicochemical and quality properties of eel dipping sauce for optimization. To accomplish the objective, eel dipping sauce was created by adding bokbunja vinegar. The findings are as follows. Among the composition, moisture and crude fiber content increased as more bokbunja vinegar was added. As for crude protein and crude ash, however, their content decreased as more bokbunja vinegar was added. In terms of crude fat, there was no difference between the control group and addition group. As for pH, the content decreased as more bokbunja vinegar was added. Also. the following properties declined as more bokbunja vinegar was added: titratable acidity; sugar content; salinity; viscosity. In terms of chromaticity, however, the brightness(L), redness(a), and yellowness(b) all increased. The overall acceptability was as follows: 0% < 5% < 10% < 20% < 15% Therefore, the proper content of bokbunja vinegar to add in eel dipping sauce is approximately 15~20%.
Genomic DNAs were extracted from the muscle of twenty-one specimens of three eel species collected in Anguilla japonica (AJ), Muraenesox cinereus (MC) and Conger myriaster (CM) from the Yellow Sea, respectively. In the present study, 7 oligonucleotides primers generated 191 specific loci in the AJ species, 226 in the (MC) species and 181 in the CM species, respectively. The primer BION-02 generated the most loci (a total of 83), with an average of 11.86 in the AJ species. The specific loci generated by oligonucleotides primers exhibited inter-individual-specific characteristics, thus revealing DNA polymorphisms. With regard to average bandsharing value (BS) results, individuals from Conger myriaster species (0.808) exhibited higher bandsharing values than did individuals from Muraenesox cinereus species (0.729) (P<0.05). The longest genetic distance (0.430) displaying significant molecular difference was also between individual no. 01 within Anguilla japonica eel species and individual no. 04 within Anguilla japonica species. In this study, the dendrogram resulted from reliable seven oligonucleotides primers, indicating three genetic clusters composed of group I (ANGUILLA 01~ANGUILLA 07), group II (MURAENESOX 08~MURAENESOX 14) and group III (CONGER 15~CONGER 21). The existence of species differentiation and DNA polymorphisms among three eel species were detected by PCR analysis. As mentioned above, a dendrogram revealed close relationships between individual identities within three eel species. High levels of a significant genetic distance among three eel species showed this PCR approach is one of the most suitable tools for individuals and/or species biological DNA studies.

The changes in osmolality and the gene expression profiles of prolactin (PRL) and growth hormone (GH) with rapid changes in salinity were compared in the eel (Anguilla japonica), crucian carp (Carassius carassius), and masu salmon (Oncorhynchus masou). Fish stocked in freshwater (FW) were abruptly transferred to experimental tanks containing FW, 50% seawater (50% SW), or 100% SW (SW). Blood samples and pituitary glands were collected 2 and 24 hrs after the exposure. No mortality was observed in SW eel (n=6), whereas all of the crucian carp (n=6) and two masu salmon (n=6) exposed to SW died after land 24 hrs, respectively. The PRL mRNA levels of the eel and masu salmon decreased in 50% SW and SW compared to those of the fish kept in FW after 24 hrs, whereas the PRL levels of crucian carp were higher in 50% SW than in FW. Unlike the PRL mRNA levels, the GH mRNA levels of the eel did not differ significantly among three different salinities, while the GH mRNA levels of crucian carp and masu salmon increased significantly in 50% SW and SW after 24 hrs. The serum osmolalities increased marginally in the eel and masu salmon in 50% SW at 24 hrs (19% and 9%, respectively), whereas those of crucian carp increased abruptly in 50% SW (50% increase). These results suggest that the synthesis of PRL and GH is important in relation to the osmoregulatory system with environmental changes in salinity.

This research was conducted to investigate pollutant load unit factor from aquacultural farms. Pollutant load unit factors were investigated on the 13 kinds of fish type, i.e., Trout, Leather Carp, Eel, Carp, Cartfish, Freshwater Eel, Crucian Carp, Colored Carp, Sturgeon, Marsh Snail, Sweetfish, Pond Snail, Eriocheir Sinensis. Water qualities in aquacultural farms were investigated wide range of values by fish type and pollutant items. High BOD and COD values were observed at the Leather Carp (Ponded water system), Cartfish (Ponded water system) and Freshwater Eel (Recirculating System). TOC and DOC values were relatively high at the Freshwater Eel (Recirculating System) and Eriocheir Sinensis (Extensive). Eel (Ponded water system) and Freshwater Eel (Recirculating System) produced high concentrated nutrient pollutants, i.e., T-N, T-P. Pollutant load unit factors are dependent on fish type, aquacultural type, water quality items, etc. If some fishes have similar basic unit values, those could be applied for TMDLs as a group of fish having same pollutant load unit. The water quality concentrations of post treatment facilities' (mainly reservoir tank) effluent were higher than those of fish raising bath because of extracting pollutant from sediment in the reservoir tank. Therefore, it needs to the management and regulations about post treatment facilities.

A seasonal variations of chemical components and gel forming ability in wild common carp (Cyprinus car-pio) and conger eel (Astroconger myriaster) caught around Pusan, Korea monthly for one year (from May,1984 to April, 1985) were investigated. During a year variation range in the moisture and crude lipid contents were found in conger eel meat showing 61.0-76.6% and 5.4-20.8%, while crude protein, ash and carbohydrate were no appreciable differences . Ail of the moisture of chemical components were nearly constant in wild common carp for 12 months. The contents of moisture were less, while crude lipid were more in conger eel meat from September to February than those of the other months. Qualities of the fish jelly were the best for that months in conger eel. Fish jellies were good for 12 months in wild common carp. Gel forming ability was related to the content of chemical components and fishing season in conger eel.

In the present study, we investigated the effects of recombinant eel gonadotropins (rec-GTHs) on maturation induction in immature ovarian culture in vitro and sex steroid hormones in vivo in the Japanese eel Anguilla japonica. To study the in vitro effects of rec-GTHs on estradiol-17β (E2) production in immature ovarian tissues, ovarian tissues were incubated with different doses of rec-follicle-stimulating hormone (rec-FSH) or rec-luteinizing hormone (rec-LH). The results revealed that the E2 levels in the rec-FSH (0.1, 0.5, or 1 ㎍/mL)- and rec-LH (0.1 or 0.5 ㎍/mL)-treated groups were significantly higher than those in the female eels from the control group. Furthermore, to investigate the in vivo effects of rec-GTHs on the gonadosomatic index (GSI) and plasma sex steroid hormone levels, the eels were injected intraperitoneally with eel's ringer (control), salmon pituitary extract (SPE; for female eels), human chorionic gonadotropin (hCG; for male eels), rec-FSH, rec-LH, and rec-FSH + rec-LH once a week. The results revealed that except for the SPE and the hCG groups, none of the groups exhibited a significant difference in GSI values. However, in vivo plasma E2 levels increased at the end of 4 weeks after rec-FSH treatment in female eels. Based on these results, it is suggested that rec-GTHs may have a positive effect on sexual maturation in female eels; however, further studies on complementary rec-protein production systems and additional glycosylation of rec-hormones are needed to elucidate hormone bioactivity in vivo and in vitro.
The present study aimed to investigate the mechanism of cell surface receptor loss by two constitutively activating mutants (designated L469R, and D590Y) and two inactivating mutants (D417N and Y558F) of the luteinizing hormone receptor (LHR) in the Japanese eel Anguilla japonica, known to naturally occur in human LHR transmembrane domains. We investigated cell surface receptor loss using an enzyme-linked immunosorbent assay in HEK 293 cells. The expression level of wild-type eel LHR was considered to be 100%, and the expression levels of L469R and D417N were 97% and 101%, respectively, whereas the expression levels of D590Y and Y558F slightly increased to approximately 110% and 106%, respectively. The constitutively activating mutants L469R and D590Y exhibited a decrease in cell surface loss in a manner similar to that of wild-type eel LHR. The rates of loss of cell surface agonist-receptor complexes were observed to be very rapid (2.6-6.2 min) in both the wild-type eel LHR and activating mutants. However, cell surface receptor loss in the cells expressing inactivating mutants D417N and Y558F was slightly observed in the cells expressing inactivating mutants D417N and Y558F, despite treatment with a high concentration of agonist. These results provide important information on LHR function in fish and the regulation of mutations of highly conserved amino acids in glycoprotein hormone receptors.
