Antioxidative and Nitrite-scavenging Activities of Edible Mushrooms (버섯류의 항산화성 및 아질산염 소거작용)

  • This study was conducted to investigate the functional characteristics of diethylether and butanol extracts from Ganoderma lucidum, Agaricus bisporus and Lentinus edodes. Electron donating abilities of diethylether and butanol extracts from Ganoderma luculum were 95.07% and 97.75%, respectively. Electron donating abilities of butanol extracts from Agaricus bisporus was 94.33%, and that of Lentinus edodes was 96.09%. Antioxidative activities of diethylether and butanol extracts from Canoderma lucidum were higher than those of BHA. All extracts of Agaricus bisporus and Lentinus edodes showed lower antioxidative activity than that of BHA. Nitrite-scavenging abilities of diethylether and butanol extracts from Ganoderma lucidum were 68.34% and 44.44%, respectively. Nitrite-scavenging abilities of butanol extracts from Agaricus bisporus were 43.39%, and those of Lentinus edodes were 68.23%.

Comparative analysis of total sugar and sugar alcohol contents of the fruiting bodies in edible and medicinal mushrooms (식용 및 약용버섯류의 단당류, 이당류, 당알코올 함량 비교 분석)

  • We analyzed saccharide by dividing and comparing Monosaccharide, Disaccharide and sugar Alcohol. At first, Glucose had outstanding contained quantity of ASI 7114 with 81.11 g/l even comparing with other mushrooms for medical use and edibility. And 119.98 g/l of Fructose was observed at Hericium erinaceum that was more contained quantity than Flammulina velutipes and Lentinus edodes. But, the most contained quantity observed in Ganoderma lucidum was ASI 7015 with 15.70 g/l that was the level of 1/8 approximately against Hericium erinaceum. Ribose was found at low level generally that was hardly contained. Xylose was also observed low level. ASI 7004 was detected at 0.96g/l that was the most content with imperceptible difference by comparing with other mushrooms for medical use and edibility. Next, 35.21 g/l of Trehalose, disaccharide was observed at Agaricus bisporus that was around 11 times of content than ASI 3.09 g/l that was the most content of Ganoderma lucidum. For ${\alpha}$-Lactose, Sparassis crispa has the most amount of 3.38 g/l that was around 12.5 times of ASI 7060 0.27 g/l that was the most content of Ganoderma lucidum. For Glycerol, sugar alcohol, 64.74 g/l was observed at Pleurotus eryngii. We knew it was around 8 times of ASI 7004 8.61 g/l that was the most content of Ganoderma lucidum. 0.72 g/l of Solbitol was observed at Flammulina velutipes. We knew it was around 2times of ASI 7003 0.31 g/l that was the most content of Ganoderma lucidum. Moreover most of Ganoderma lucidum didn't contain Solbitol. 2.96 g/l of Mannitol was observed at Agaricus bisporus. that was the most content among other mushrooms. Also Mannitol was contained in Lentinus edodes and leurotus cornucopiae only. Even Ganoderma lucidum didn't have Mannitol. At last, as a result of myo-Inosito content analysis, it was seemed not to be involved in any of mushrooms.

Cultivation of Sparassis crispa on Several kinds of Medium Density and Particle Size of Sawdust-based Medium Made of Larix kaempferi (낙엽송 톱밥배지 밀도 및 입자크기에 따른 꽃송이버섯의 재배특성)

  • Cauliflower mushroom ($Sparassis$ $crispa$) is being cultivated as a functional mushroom since the mushroom contains larger amount of ${\beta}$-glucan than other edible or medicinal mushrooms. In this study, as a fundamental study for the cultivation process of cauliflower mushroom, we assayed the mycelial growth and the productivity of the mushroom cultivated on the sawdust-based medium made of larch ($Larix$ $kaempferi$) by 4 kinds of medium densities and 6 kinds of particle sizes. Low densities of sawdust-based media (0.68~0.72 g/$cm^3$) showed high variations in mycelial growth at early stage on sawdust-based medium. The coefficient of variance for the mycelial growth decreased from higher than 40% at the 3rd week to lower than 10% at the 7th week. High density of sawdust-based medium (0.80 g/$cm^3$) showed relatively lower variation in the mycelial growth at early stage with less than 30% of coefficient of variance, the high density of sawdust-based medium was thought to be quite stable compared to the lower densities of sawdust-based medium. From the viewpoint of mushroom productivity especially for the goods (excluding bottom of fruiting body), 0.76 g/$cm^3$ was better than any other densities; the return rates of fruiting body from each medium were 12.2, 13.6, 13.1, and 12.0% for 0.80, 0.76, 0.72, and 0.68 g/$cm^3$, respectively. By the way, it took about 40 days for harvesting from primordium formation with the sawdust-based medium excluded the particles less than 1 mm, while it took about 70 days with the medium including the particles less than 1 mm. The yield from the sawdust-based medium with the particles less than 1 mm was also quite less than any other sawdust-based medium especially for the goods. Therefore, we recommend that the sawdust-based medium with larch for the cultivation of cauliflower mushroom be prepared as 0.76 g/$cm^3$ in medium density with excluding the particles less than 1 mm.

Physiological and Genetic Characteristics of Cultivated Mushroom, Hypsizygus marmoreus

  • A edible mushroom, Hypsizygus marmoreus is commercially cultivated in Northeast Asia. Japan's annual production is 110,000ton or more. Since 2002, cultivation is expanded in Korea. To investigate the morphological, cultural and microscopic characteristics of Hypsizygus marmoreus, 109 isolates were collected from Korea and other countries. Clamp connection, chlamydospore and arthrospore were present in all tested isolates of H. marmoreus except HYM-002 and HYM-004. Also pilealtrama, gilltrama, basidia, basidiospore and cystidia of fruiting body were no difference among the isolates in the present investigation. Morphological characteristics of fruiting body was that color of pileus was brown and white, irregular as marble, the average size 12~22mm and stipes was $46{\sim}91{\times}6{\sim}10mm$. Isolates HYM-031, HYM-047 and HYM-109 formed grayish-brown pileus with a faint pattern. Molecular analysis with RAPD and ITS rDNA sequence analysis were also performed to check the genetic relationships among H. marmoreus isolates. Based on the RAPD analysis using the URP-PCR, all isolates of H. marmoreus were clustered into large 3 groups but more than 90% showed high similarity. In addition, morphological and geographical differences have been classified as an independent cluster. The brown and white strains enclosed in same cluster. So genetically no significance difference was observed between these two strains. ITS gene sequences of 16 selected isolates which were 640 bp long, were aligned and compared. The similarity in ITS sequence was 94.8 to 99.1% among tested isolates and the H. marmoreus isolates in GeneBank. In conclusion the tested isolates were H. marmoreus. Morphological and molecular observations proved that all tested isolates were belonging to H. marmoreus. For the stable artificial cultivation, composition of optimum media, mature period and light condition were established. Optimal formula of artificial cultivation medium was Douglas sawdust: corn cob: soybean meal: wheat bran = 40:30:15:15. In addition, 7% rice bran and 3% yellow sucrose was the most effective composition for spawn's liquid medium. For the maturation of the isolates was favorable for growing for 20 to 30 days at $25^{\circ}C$ and the LED lights in mixture of white and blue was good for growth period. For effective growth, the temperature, humidity and aeration control in every step was important.

Purification and Characterization of a Laccase from the Edible Wild Mushroom Tricholoma mongolicum

  • A novel laccase from Tricholoma mongolicum was purified by using a procedure that entailed ion-exchange chromatographies on DEAE-cellulose, CM-cellulose, and Q-Sepharose, and FPLC-gel filtration on Superdex 75. The purified enzyme was obtained with a specific activity of 1,480 U/mg-protein and a final yield of 15%. It was found to be a monomeric protein with a molecular mass of 66 kDa as estimated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Its N-terminal amino acid sequence was GIGPVADLYVGNRIL, similar to some but also different to other mushroom laccases. The optimum pH and temperature for the purified enzyme were pH 2 to pH 3 and $30^{\circ}C$, respectively. It displayed a low $K_m$ toward 2,7-azinobis (3-ethylbenzothiazolone-6-sulfonic acid) diammonium salt (ABTS) and high $k_{cat}/K_m$ values. The purified laccase oxidized a wide range of lignin-related phenols, but exerted maximal activity on ABTS. It was significantly inhibited by $Hg^{2+}$ ions, and remarkably stimulated by $Cu^{2+}$ ions. It inhibited HIV-1 reverse transcriptase and proliferation of hepatoma HepG2 cells and breast cancer MCF7 cells with an $IC_{50}$ of 0.65 ${\mu}M$, 1.4 ${\mu}M$, and 4.2 ${\mu}M$, respectively, indicating that it is also an antipathogenic protein.

Characterization of intraspecific hybrids by di-mon crossing in Pleurotus eryngii (Di-Mon 교잡법에 의한 큰느타리버섯 교잡주의 특성)

  • Pleurotus eryngii is not edible and medicinal mushrooms indigenous to Korea. To improvement of strain suitable to the geographic setting of Korea, we are mated with 22 dikaryons and 47 monokaryons isolated from Pleurotus eryngii ASI 2547 by Di-Mon mating. 19 strains forming fruit body obtained from clamped 253 bred strains. 7 excellent strains are selected from 19 bred strains by various morphological features of fruit body. Among the selected 7 strains, H6 strain were identified into ASI 2547-like recombinant hybrids with URP uniprimer by RAPD analysis. This suggested that Di-Mon crossing is one of rapid and easy breeding method for strain improvement with molecular techniques.

Traditional Knowledge on Wild Mushrooms in the Surrounding Villages Hallyeo-Haesang National Park (한려해상국립공원 인근 지역에서의 야생버섯에 대한 전통 지식)

  • Traditional knowledge on wild mushrooms in Gyeongnam province near of Hallyeo-Haesang National Park was investigated. It was interviewed from 145 of people in age of 50th's to 90th's living in 53 villages. Total 19 mushrooms were used for food in which Ramaria botrytis was the most frequently used and followed Macrolepiota procera and Lentinula edodes. L. edodes was uniquely both medicinal and edible. Chlorophyllum neomastoidea, poisonous mushroom, was used as an insect repellent. The most various mushrooms were found in Irun, Geoje where 10 species of mushrooms were used and followed Jingyo, Hadong where 8 species were used. Among the interviewed people, the number of female was more than double compared to that of male and most of the interviewed people were 70th's and 80th's in age.

Genes Expressed During Fruiting Body Formation of Agrocybe cylindracea

  • Agrocybe cylindracea, an edible mushroom belonging to Bolbitiaceae, Agaricales, is widely used as invaluable medicinal material in the oriental countries. This study was initiated to find the genes expressed during the fruiting body formation of A. cylindracea. The cDNAs expressed differentially during fruiting body morphogenesis of A. cylindracea were isolated through subtractive hybridization between vegetative mycelia and fruiting bodies. The cDNAs expressed in the fruiting body morphogenesis of A. cylindracea were cloned and twenty genes were identified. Eleven were homologous to genes of known functions, three were homologous to genes in other organism without any function known. Six were completely novel genes specific to A. cylindracea so far examined. Some genes with known functions were a pleurotolysin, a self-assembling poreforming cytolysins; Aa-Pril and Pir2p, specifically induced genes during fruiting initiation of other mushroom, Agrocybe aegerita; an amino acid permease; a cytochrome P450; a MADS-box gene; a peptidylprolyl isomerase; and a serine proteinase. For other clones, no clear function was annotated so far. We believe the first report of the differentially expressed genes in fruiting process of A. cylindracea will be great helps for further research.

The Effects of Flammulina velutipes Water Extract on the Activation of Spleen Cell and Macrophage in Mice (팽이버섯 열수추출물이 마우스 비장세포와 대식세포의 증식 및 활성에 미치는 효과)

  • Flammulina velutipes is an edible mushroom and contains a lot of fiber, vitamin $B_1$, $B_2$, niacin and folic acid. This study was conducted to explore the effects of the Flammulina velutipes mushroom on immune cells and immunity. Th1 cytokine productions as $IFN-{\gamma}$, $TNF-{\alpha}$, and IL-2 were measured in an activated macrophage by Flammulina velutipes water extract in seven concentrations (0, 5, 10, 50, 100, 250, 500, and $1,000{\mu}g/mL$). Also, the splenocyte proliferation index was measured at 48 hours after treatment of the Flammulina velutipes water extract in seven concentrations or mitogen, LPS and ConA. The $IFN-{\gamma}$ and $TNF-{\alpha}$ productions were increased by treatment of the Flammulina velutipes water extract. The $TNF-{\alpha}$ production was significantly higher in the $50{\sim}1,000{\mu}g/mL$ Flammulina velutipes water extract treated macrophages. The $IFN-{\gamma}$ production of macrophages treated with the Flammulina velutipes water extract increased significantly in all groups, and the highest $1000{\mu}g/mL$ concentration. The splenocyte proliferation index was enhanced when the $10{\sim}1,000{\mu}g/mL$ Flammulina velutipes water extracts were treated compared to the control. These primary results suggest that Flammulina velutipes may enhance the immune function by activation of the macrophage and spleen cell.

Identification and Functional Analysis of Mating Type Loci in the Pleurotus eryngii

  • Pleurotus eryngii has recently become a major cultivated mushroom; it uses tetrapolar heterothallism as a part of its reproductive process. Sexual development progresses only when the A and B mating types are compatible. Such mating incompatibility occasionally limits the efficiency of breeding programs in which crossing within loci-shared strains or backcrossing strategies are employed. Therefore, understanding the mating system in edible mushroom fungi will help provide a short cut in the development of new strains. We isolated and identified pheromone and receptor genes in the B3 locus of P. eryngii and performed a functional analysis of the genes in the mating process by transformation. A genomic DNA library was constructed to map the entire mating-type locus. The B3 locus was found to contain four pheromone precursor genes and four receptor genes. Remarkably, receptor PESTE3.3.1 has just 34 amino acid residues in its C-terminal cytoplasmic region; therefore, it seems likely to be a receptor-like gene. Real-time quantitative RT-PCR (real-time qRT-PCR) revealed that most pheromone and receptor genes showed significantly higher expression in monokaryotic cells than dikaryotic cells. The pheromone genes PEphb3.1 and PEphb3.3 and the receptor gene PESTE3.3.1 were transformed into P5 (A3B4). The transformants were mated with a tester strain (A4B4), and the progeny showed clamp connections and a normal fruiting body, which indicates the proposed role of these genes in mating and fruiting processes. This result also confirms that PESTE3.3.1 is a receptor gene. In this study, we identified pheromone and receptor genes in the B3 locus of P. eryngii and found that some of those genes appear to play a role in the mating and fruiting processes. These results might help elucidate the mechanism of fruiting differentiation and improve breeding efficiency.
