Dietary Value of Three Benthic Diatom Species on Haliotis discus hannai Larvae (북방전복 Haliotis discus hannai 유생에 대한 3종 부착 규조류의 먹이효율)

  • Although the method of seedling production of Haliotis discus hannai is well known, the optimum benthic diatom species as a live food at early larval stage are not fully developed. In this study three Pennales diatom species, Caloneis schroederi, Rhaphoneis sp., and Cocconeis californica were examined on settlement, metamorphosis, survival, and growth of Haliotis discus hannai larvae. The larvae fed Raphoneis sp. or C. californica showed high settlement rate with 80-82% within 48 hrs, which was significantly higher than those fed C. schroederi or mixed diets with three diatom species. The larvae fed the former microalgal species also showed higher metamorphosis rate with 32-34% than the latter species with 10-12% within 4 days. With regard to survival and growth of the larvae, single diet with Rhaphoneis sp. or C. californica had better dietary value than the mixed diets for the early larvae of H. discus hannai.

STUDIES ON THE PROPAGATION OF TOP SHELL-I Spawning and early development of the top shell, Turbo cornutus SOLANDER (소라 Turbo cornutus SOLANDER의 증식에 관한 연구-I 소라의 산란과 초기발생)

  • Fertilization and early development of turbo cornutus was studied based on the samples which were collected in Yeosu area. Particular emphasis was paid on induction of artificial spawing, fertilization rate, preembryonic development, the growth of the early larva and larval survival to various salinity. Among the various methods for induction of artificial spawning which have been tested for the present study, drying by exposure to air is the. most efficient, and percentage fertilization rate was $83.8-96.4\%$. The diameter of fertilized eggs was $0.182{\pm}0.0028mm$; and the diameter of egg membrane was $0.245{\pm}0.093mm$. Under the temperature range of $20.6-25.4^{\circ}C$ the larvae hatched out after 11:05-11:15 hours of fertilization. After 3.0-3.5 days of fertilization the planktonic larvae begand to settle, and the settlement terminated within 5 days. During the period of 150 days of early culturing the diameter growth of shell(M) and the diameter of shell aperture(A) was formulated as follows: $$1972\;M=0.33e^{0.02070D}$$ $$A=0.19e^{0.02282D}$$ $$1973\;M=0.32e^{0.02282D}$$ $$A=0.16e^{0.02596D}$$ During the same period of early culturing the relative growth of shell diameter and the diameter of shell aperture was formulated as follows : 1972 A=0.6478 S-0.1575 1973 A=0.5897 S-0.0515 After 11 days of larval hatching $0.02-0.18\%$ of planktonic larvae settled. After 150 days of settlement the survival rate of the early shells was $7.4-21.6\%$. Under the temperature range of $21.0-22.7^{\circ}C$ the optimum salinity range for the development of egg and the planktonic larvae was $30-35\%_{\circ}$.

Host range, Life cycle and Natural enemies of Mulberry scale (Pseudaulacaspis pentagona) on Prunus mume in south Korea (매실나무에 기생하는 뽕나무깍지벌레 (Pseudaulacaspis pentagona)의 기주범위, 생활사 및 천적에 관한 조사)

  • Studies were conducted to investigate hosts, life cycle, population dynamics and natural enemies of mulberry scale, Pseudaulacaspis pentagona Ta. & Tozz., from 1987 to 1988 in southern region of Korea. The host plants were 22 families and 74 species. Host plants belonged to genus Prunus in general were severely damaged due to mulberry scale. Mulberry scale was most likely to have three generations a year with first occurrence of larval stage from mid-May to late June, second occurrence from mid-July to mid-August and third occurrence from early September to late October including 3 peaks in early and mid-June, late July and mid-September. In case of each stage occurrence, eggs were peaked on 10th day, larvae on 25th day and adults on 7th day from beginning of occurrence, respectively. Natural enemies were observed as parasitoids of 3 species such as Aphytis diaspidis, Archenomus orientalis and Apterencyrtus mocrophagus and as predators of 3 species such as Chilocorus Kuwanae, Chilocorus rubidus and Hormonia axyridis. Shoot growth was affected by mulberry scale and Lepra to dwindle as much as half of normal growth.

Ontogeny and Characterization of Major Haemolymph Protein(MHP) in Helicoverpa assulta (담배나방 (Helicoverpa assulta)의 발생중 Major Haemolymph Protein(MHP)의 변화 및 특성)

  • A persistent major haemolyruph protein (MHP) was confirmed, and its ontogeny and physicochemical charadedstics were investigated in Helicoverpa assulta. The MHP existed continually during larval-pupal-adult development, and its ontogeny was similar to that of total haemolymph protein concentration during development. Its content increased with larval growth, and kept to high level during pupal-adult development except for temporary decrease at the early pupal and adult stages. The MHP was purified by ammonium sulfate precipitation, gel filtration and ion exchange chromatography. The purified MHP was determined to be hexamer glycolipoprotein (pI 5.9, M.W. 414kDa) consisted of single type subunit (69kDa). Amino acid analysis suggested that the MHP contained a relatively high content of aromatic amino acids (18.27 mole % of tryptophan, 7.47 mole % of tyrosine and 6.51 mole % of phenylalanine) compared to storage proteins from other insects. Immunodiffusion test and electrophoretic analysis of the organ proteins (gut, fat body, and Malphigian tubule) suggested that the major haemolymph protein was present in the fat body.

  • Settlement, recruitment, growth and production of Mactra venerifomis were investigated at Kunsan tidal flat located in the west coast of Korea from lune 1993 to May 1994. High density of planktonic larvae of M. veneriformis was observed between the mid and early July in 1993. The size of the newly settled larvae ranged from 250 to 350um in shell length with mean density of 601$inds.\cdot m^{-3}$. Recruitment of M. veneriformis larvae was occurred mainly in the mid June to mid July 1993. The larvae have no substratum preference during the larval settlement phase. Mortality of larval was affected by environment (substratum, exposure time, density of adult clams). After settlement they grew until November, and stopped growing in winter from December. The estimated turnover rate of M. veneriformis larvae was estimated as high as 4.91 and 4.94. Annual production of the 0-age group was estimated to be 2,900g $m^{-2}\;\cdot\;yr^{-l}$, which was relatively high compared with those of other age groups.

Enriched Rotifer Feeding Efficiency in the Walleye Pollock Theragra chalcogramma Depends on Larval Fatty Acid Composition (명태(Theragra chalcogramma) 자어의 지방산 조성에 따른 영양강화 로티퍼의 먹이효율)

  • The objectives of this study were to confirm the nutritional requirements and improve the survival of the walleye pollock Theragra chalcogramma, a cold seawater fish, by enrichment. We analyzed the fatty acids and amino acids of fertilized pollock eggs before hatching, just-hatched larvae, larvae that had absorbed only the yolk sac, and larvae starved for 2 days after yolk absorption. For the survival improvement experiment, we administered docosahexaenoic acid (DHA), eicosapentaenoic acid (EPA), and DHA-EPA enrichment. Fatty acid decreased DHA and EPA content. On the $30^{th}$ day, body growth was significantly greater in pollock given the EPA and DHA-EPA treatments (P<0.05). Larval survival at 20 and 30 days after hatching (DAH) was greatest under the DHA-EPA treatment (P<0.05). Survival was significantly lower under the EPA treatment at 10 DAH, but then increased to approach that seen under the EPA-DHA treatment (P<0.05). Therefore, we determined that reduced survival in hatchlings of high-mortality pollack could be improved by controlling EPA and DHA content during enrichment. We conclude that cold seawater fish must be given feed that meets their nutritional needs, which can be accomplished using newly manufactured enrichment products for the larvae of cold seawater fish, such as pollock.

Reproduction and Early Life History of Gunnel, Pholis fangi in the Yellow Sea off Korea (한국 서해 흰베도라치 (Pholis fangi)의 생식과 초기 생활사)

  • Larvae of the gunnel Pholis fangi were collected in coastal waters off Daecheon with a bag net from March to June, 1988, and with a ring larva net in February 1989. Maturity and spawning period were analyzed by examination of the gonads of adult fish collected with a bag net from May 1998 through November 1999. In February, the larvae were widely distributed in the outer and inner Cheonsu Bay. From March to April the larvae were present mainly the inner bay; they were absent there in May and found mainly in the outer bay. After June, few gunnel larvae were collected in the study area. This suggests a seaward movement of gunnel from the nursery grounds of the bay to offshore feeding grounds. The otolith of larvae smaller than 10 mm in total length did not show a distinct growth stop. The growth stop is believed to be formed in the early larval stage when the total length is about 10 mm. This period coincides with the time of shoreward migration, suggesting a metabolic change during this period. At a total length of 30 to 40 mm, the shape of the otolith changes from spherical to elongate. Daily growth rate in length was estimated by the Gompertz equation, which is represented as follows: TL = 6.702exp{2.925"1-exp (-0.008 t)"} ($r^2=0.94$, N = 92) Assuming daily deposition of growth increments in the otolith, the time of first growth increment formation was shown to be from December to January. Gonad observations show that Pholis fangi spawns from November to December. So, the hatching time is thought to be about one month.

Caffeine Induces the Stress Response and Up-Regulates Heat Shock Proteins in Caenorhabditis elegans

  • Caffeine has both positive and negative effects on physiological functions in a dose-dependent manner. C. elegans has been used as an animal model to investigate the effects of caffeine on development. Caffeine treatment at a high dose (30 mM) showed detrimental effects and caused early larval arrest. We performed a comparative proteomic analysis to investigate the mode of action of high-dose caffeine treatment in C. elegans and found that the stress response proteins, heat shock protein (HSP)-4 (endoplasmic reticulum [ER] chaperone), HSP-6 (mitochondrial chaperone), and HSP-16 (cytosolic chaperone), were induced and their expression was regulated at the transcriptional level. These findings suggest that high-dose caffeine intake causes a strong stress response and activates all three stress-response pathways in the worms, including the ER-, mitochondrial-, and cytosolic pathways. RNA interference of each hsp gene or in triple combination retarded growth. In addition, caffeine treatment stimulated a food-avoidance behavior (aversion phenotype), which was enhanced by RNAi depletion of the hsp-4 gene. Therefore, up-regulation of hsp genes after caffeine treatment appeared to be the major responses to alleviate stress and protect against developmental arrest.

  • In ascidians, a primitive chordate, maternal cytoplasmic factors and inductive interactions are involved in the specification of cell fates in early embryos. The larval structure of ascidians is relatively simple, and the major mesodermal tissues of the tadpole larva are notochord, muscle, and mesenchyme. Formation of muscle cells is a cell-autonomous process, and localized maternal macho-l mRNA specify muscle fate in the posterior marginal zone of the early embryo. In contrast, inductive influence from endoderm precursors plays important roles in the specification of notochord and mesenchyme fates. FGF-Ras-MAPK signaling is involved In the induction of both tissues. The difference in responsiveness of the posterior mesenchyme and anterior notochord precursors to FGF signaling is caused by the presence or absence of intrinsic factors that inherited from the posterior-vegetal egg cytoplasm, respectively. In these inductions, directed signal polarizes the induced cells and promotes asymmetric cell divisions to produce two daughter cells with distinct fates.

COMPARATIVE STUDIES ON THE EARLY EMBRYONIC DEVELOPMENT AND GROWTH OF MERETRIX LUSORIA AND CYCLINA SINENSIS (대합(Meretrix lusoria)과 가무락(Cyclina sinensis)의 초기발생 및 성장에 관한 비교연구)

  • The comparative studies Were conducted with respect to the artificial spawning early embryonic development, metamorphosis and growth of two species Meretrix lusoria and Cyclina sinensis collected from Inchon, Anmyon island and Buan areas from 1969 to 1974. The highest rate of artificial spawning of M. lusoria, which treated with a dilute ammoniun hydroxide(4/100-5/100N)-seawater solutions, was $25.0-33.3\%$, whereas in C. sinensis the rate of spawning was lower than that of M, lusoria under the similar experimental conditions$(12.5-19.0\%)$. However, the rate of artificial spawning of C. sinensis increased $40\%$ by repeated thermal stimulation. The rate of artificial fertilization of M. lusoria and C. sinensis showed highest value from those individuals which were treated with 1/1000N $NH_4OH$ solution. Their fertilized eggs, then, showed a normal development in the 1/1000N $NH_4OH$ solution. In the early embryonic development of M. lusoria and C. sinensis, the appearance of each of polar body, trochophore and D-shaped veliger were observed around 50min. 5-6 hours, and 23 hours after artificial fertilization respectively. The larval shell lengths of M. lusoria reached to $109,5{\pm}0.7\mu,\;144.6{\pm}1.3\mu$ and $208.0{\pm}0.0\mu$ around, 1, 11 and 20 days, after fertilization respectively. The larval shell lengths of C. sinensis reached to $110.5{\pm}0.6\mu,\;147.8{\pm}1.7\mu,\;and\;235.0{\pm}0.0\mu$ around 1, 10 ana 20 days, after fertilization respectively. The correlations of relative growth rate between the shell length(L) and sell height(H) found by the following simple formula from D-shaped veliger to metamorphosing stage. H=0.77L+6.82 for M. lusoria H=0.75L+8.50 for C. sinensis.
