크롬친화세포의 배양방법

  • 목적 :크롬친화세포는 카테콜아민 (CA)을 분비하는 새포로서 무신 수질내에 주로 존재한다. 따라서 부신피질의 영향을 배제하여 여러가지 자율신경계 작용 약물의 약리작용을 연구하는데 중요한 조직이다. 그러므로, 부신으로부터 크롬친화세포를 분리하여 배양하는 방법을 습득하여 자율신경계 작용약물연구에 이용코자 히스타민을 이용하여 CA 분비작용을 연구하였다. 방법: 도살장에서 소를 즉사시킨 후 좌우양측 부신을 분리하여 collagenase digestion으로 부신수질로부터 분리하고 Percoll gradient에 의해서 chromaffine cell을 정제하였다. 이렇게 정제한 크롬친화세포는 Dulbecco's modified Eagle medium에 10% fetal calf serum과 함께 culture dish에다 넣어 5% $CO_2$, incubator에서 유지시켜 주면서 실험을 시행하였다. 배양세포는 분리후 일주일 이내에 사용하였다. Catecholamine측정은 electrochemical detector를 연결하여 HPLC로 측정하였다.

Long Bone Fractures in Raptors: 28 cases (2004-2007)

  • Medical records from the Veterinary Medical Teaching Hospital of the University of Missouri-Columbia from 2004 to 2007 were available for 28 raptors that underwent long bone fracture repair. There were 14 owls, 10 hawks, 2 vultures, 1 eagle, and 1 falcon. Mean body weight was 780 g (ranged from 150 to 1400 g) for 14 owls; 650 g (ranged from 150 to 1270 g) for 10 hawks; 1760 g (ranged from 1520 to 2000 g) for 2 vultures; 5000 g for 1 eagle; and 130 g for 1 falcon. Of all 28 fracture cases, 11 cases (39%) and 1 case (3%) were related to hit-by-car and shooting respectively. Physical examination revealed dehydration in 18 raptors (64%) and lethargy in 12 raptors (42%). Forty one long bone fractures were included in 28 cases. The radiographs revealed 13 ulnar fractures (32%), 12 humeral fractures (30%), 10 radial fractures (25%), 4 tibiotarsal fractures (9%), 1 femoral fracture (2%), and 1 fibular fracture (2%). External skeletal fixation using polymethylmethacrylate (PMMA) combined with intramedullary fixation was used in 19 long bone fractures (46%). Intramedullary fixation using intramedullary Kirschner pin was used in 16 long bone fractures (39%). No surgical treatment was performed in 6 long bone fractures (15%). This study reported that many of raptors presented dehydration and lethargy when admitted for treatment. Therefore, proper hydration and nutrition are critical pre-surgical requirements. In addition, combination of internal fixation and external skeletal fixation using PMMA might be better option to treat raptors with comminuted fracture that results from mostly trauma of hit-by-car.

홍화 추출물이 생쥐 골수 유래 중간엽 줄기세포의 지방분화에 미치는 영향 (Effects of Carthamus Tinctorius Extract on Adipogenic Differentiation of Mouse Bone Marrow-Derived Mesenchymal Stromal Stem Cells)

  • Objective: This study investigated the effect of purified Carthamus tinctorius (C. tinctorius) extracted with a hot water and ethanol method on adipogenic differentiation of mouse bone marrow-derived mesenchymal stromal stem cells (mBMSCs). Methods: The C. tinctorius was extracted using hot water and ethanol. The samples were concentrated by a rotary evaporator and were then dried using a freeze-dryer. The mBMSCs were cultured and maintained in a minimum essential medium eagle alpha (${\alpha}-MEM$) supplemented with 10% FBS and 1% antibiotic antimycotic solution. To induce adipogenic differentiation, the cells were treated with Dulbecco's modified eagle's medium-low glucose (DMEM-LG) containing 1 mg/mL insulin, 1 mM dexamethasone, and 0.5 mM 3-isobutyl-1-methylxanthine. To evaluate the adipogenic differentiation ability, oil-red O staining was performed after adipogenic differentiation for 21 days. The mRNA expression and protein level of adipogenic-related genes were quantified by quantitative real-time PCR and western blotting, respectively. Results: In the results of the MTT assay, no concentrations of C. tinctorius extracts showed toxicity on mBMSCs, so we fixed the treatment concentration of the extract at 100 ng/mL. In oil-red O staining, the water-C. tinctorius extract treatment significantly decreased adipogenic differentiation compared with the control and ethanol extract groups. The water-C. tinctorius extract group in particular showed reduced mRNA and protein expression of Peroxisome proliferator-activated receptor gamma ($Ppar{\gamma}$) and CCAAT/enhancer-binding protein alpha ($C/ebp{\alpha}$), which are adipogenic-related transcription factors. Conclusion: These data suggest that extract of C. tinctorius decreased the adipogenic differentiation of mBMSCs, while only water-C. tinctorius extract had an effect on different adipogenesis in mBMSCs. The C. tinctorius will be a useful therapeutic reagent for the prevention of obesity-related diseases such as diabetes, hyperlipidemia, coronary artery disease, and osteoporosis.

연어 세포주의 전염성 췌장괴사 바이러스의 감염 증상에 관한 연구 (A Study on infection symptom of infectious pancreatic necrosis virus(IPNV) in chinook salmon embryo cell line)

  • CHSE-214 무지개송어 세포주는 10% fetal bovine serum과 2mM-glutamine이 첨가된 Eagle's 기본배지에서 성장하였다. CHSE-214 세포주의 최적온도는 $20^{\circ}C$였다. IPN바이러스는 CHSE-214 세포주에서 복제되었으며, 세포변성효과를 나타내었다. 또한 IPN 바이러스의 감염시간에 따른 감염증상을 역위상차 현미경으로 관찰하였다. 감염 6-12시간 후 세포는 정상세포와 비슷하였다. 감염 18-24시간 후 세포는 일부가 정상 세포보다 더 둥근형태로 되었고, 일부세포는 세포배양용 플라스크 바닥에서 떨어져 부유상태로 되었다. 감염 30시간 후 세포는 더더욱 비정상적인 형태로 되었다. 감염 48~68시간 후 감염된 세포는 파열되었고, 아주 높은 세포 병변 효과를 나타내었다.

한국해군의 전략커뮤니케이션 (SC : Strategic Communication) 적용 방안 (Ways to apply the Strategic Communications to the ROK Navy)

  • The aim of this study is to find ways to apply the strategic communications to the Republic of Korea Navy. It may be a little bit late for the ROK Navy to accommodate the SC in these days because the adoption and implementation of the SC by the Combined Forces Command has already been begun. It was in 2007 when the SC was in place in the CFC. ROK's Ministry of National Defense and Joint Chiefs of Staff have also mulled over the SC and begun to apply it in part in the annual exercises such as Key Resolve command post drills and Foal Eagle field training exercise, etc. For the ROK Navy, in addition to those exercises, it is faced with further areas like North Korean maritime provocations and other maritime incidents where its version of SC is needed. As noted, the SC is not intended to deter or defend directly those provocations and incidents, but aims to create conditions favorable to the achievement of the navy's strategic objectives. The ROK Navy has to establish a SC planning center and implementing organizations within the Headquarters to be consistent with its above organizations such as MND, JCS, and CFC that have already applied the SC in part or in entirety. SC center and other related organizations need to be under the control of VCNO and the center needs to be located and administered by the policy division in N-5 at the HQs. The vision of the navy's SC is the winning without combats and the least damages in time of war. In other for the navy to reach the vision, the strategies to be executed are early establishment of SC implementing organizations, forming consensus over the need for the SC within the navy, strengthening core competencies to apply the SC, acquiring the SC experts and making doctrines on the SC. The SC, in addition, in the navy has to be planned and implemented in not only peace time and crisis time but also war time.

Sol-gel 법으로 제작한 Ga-doped ZnO 박막의 도핑 농도와 열처리 온도가 전기적 및 광학적 특성에 미치는 효과 (Effects of Doping Concentrations and Annealing Temperatures on the Electrical and Optical Properties of Ga-doped ZnO Thin Films by Sol-gel Method)

  • Sol-gel 법으로 Eagle 2000 유리 기판 위에 Ga 도핑 농도와 열처리 온도에 따른 GZO 박막을 제작하여, 전기적 및 광학적 특성을 조사하였다. 1 mol% Ga 이 도핑되고 $600^{\circ}C$에서 열처리한 GZO 박막에서 가장 우수한 (002) 배향성이 관찰되었다. Hall 측정 결과, Ga 도핑 농도가 증가함에 따라 segregation 효과로 인한 캐리어 농도의 감소와 비저항 값의 증가가 관찰되었다. 1 mol% Ga 이 도핑되고 $600^{\circ}C$에서 열처리한 GZO 박막에서 가장 큰 캐리어 농도($9.13{\times}10^{18}cm^{-3}$)와 가장 낮은 비저항 ($0.87{\Omega}cm$) 값을 나타내었다. 모든 박막은 가시광 영역에서 약 80 % 이상의 투과율을 보였으며, Ga 농도가 1 에서 4 mol% 로 증가함에 따라 에너지 밴드 갭이 좁아지는 Burstein-Moss 효과가 관찰되었다.

Clinicoradiologic evaluation of styloid process calcification

  • Purpose: This study was performed to investigate the prevalence, morphology, and calcification pattern of the elongated styloid process in the Mathura population and its relation to gender, age, and mandibular movements. Materials and Methods: The study analyzed digital panoramic radiographs of 2,706 adults. The elongated styloid process was classified with the radiographic appearance based on the morphology and calcification pattern. The limits of mandibular protrusion were evaluated for each subject. The data were analyzed by using a Student's t-test and chi-squared test with significance set at p=0.05. Results: Bilateral elongation having an "elongated" type styloid process with a "partially mineralized" pattern was the most frequent type of styloid process. No correlation was found between styloid process type and calcification pattern on the one hand and gender on the other, although elongated styloid was more prevalent in older and male populations (p<0.05). Further styloid process elongation showed no effect on mandibular protrusive movement (p>0.05). Conclusion: Dentists should recognize the existence of morphological variation in elongated styloid process or Eagle syndrome apparent on panoramic radiographs. We found higher prevalence of elongated styloid process in the population of the Mathura region when compared with other Indian populations. The calcification of the styloid process was more common in the older age group with no correlation to gender, mandibular movement and site. "Type I" with a "partially calcified" styloid process was observed more frequently in the population studied.

정전분무 열분해법에 의한 나노분말의 제조 및 하이드록시 아파타이트 형성능력 평가 (Preparation of electrostatic spray pyrolysis derived nano powder and hydroxyapatite forming ability)

  • 본 연구에서는 새로운 나노 분말 제조방법 중의 하나인 정전분무 열분해법을 이용하여 칼슘 포스페이트 나노분말을 제조하였다. 정전 분무된 분말은 공기 중에서 $400^{\circ}C$로 30분간 열처리하여 고상화하였다. 결정화된 분말의 하이드록시 아파타이트 형성능력을 평가하기 위하여 Eagle's minimum essential medium solution(MEM)을 사용하였으며, MEM 용액에 침전된 후의 분말의 특성평가를 위하여 X-선 회절 분석법, 전계 방사 주사형 전자 현미경, 에너지 분산 X-선 분광계 및 퓨리에 변환 적외선 분광계를 사용하여 분석을 행하였다. 비정질 구조를 가진 나노 분말은 MEM 용액에 15일 침전 후, 분말의 표면에 유도된 하이드록시 아파타이트 결정을 확인할 수 있었다.

편도와 접근을 통한 측방인두간극 이물제거의 치험 증례 (REMOVAL OF FOREIGN BODY IN THE LATERAL PHARYNGEAL SPACE VIA TRANSTONSILLAR APPROACH)

  • Lateral pharyngeal space is one of potential fascial planes of head and neck, that may become involved by various pathological processes, such as infection, inflammation and neoplasm. The calcified stylohyoid ligament with styloid process is also located in this space, so this space is more acquainted with Eagle's syndrome in oral and maxillofacial field. During the mandibular transbuccal fixation procedures of 29-year old female patient who had right condylar neck and left parasymphysis fracture, we had lost one 10.0 mm miniscrew. After confirming the location of the lost miniscrew from different angled plain skull radiographies, we tried to find it in the lateral pharyngeal space via transtonsillar approach at the time of plate removal operation. This case report is aimed to share our valuable experience of the effective approach way to the lateral pharyngeal space, which has many advantages, such as short operative time, minimal bleeding, fast post-operative recovery, and less morbidity. The related literature is also reviewed.

고주파 마그네트론 스퍼터링법에 의해 제작된 ITZO (indium tin zinc oxide) 박막의 전기적 및 광학적 특성 (Electrical and Optical Properties of ITZO Thin Films Deposited by RF Magnetron Sputtering)

  • 본 연구에서는 RF magnetron sputtering 법으로 상온에서 공정압력 (1~7 mTorr) 을 변화시켜가며 유리기판(Eagle 2000) 위에 ITZO ($In_2O_3$ : $SnO_2$ : ZnO = 90wt.%: 5wt.%: 5wt.%) 박막을 제작하여, 구조적 특성과 광학적 및 전기적 특성을 조사하였다. XRD 와 FESEM 측정을 통해, 공정압력에 무관하게 모든 ITZO 박막이 부드러운 표면의 비정질 구조를 가지고 있음을 확인할 수 있었다. 공정압력 3mTorr 에서 증착한 ITZO 박막이 비저항 $3.08{\times}10^{-4}{\Omega}{\cdot}cm$, 가시광 영역에서 평균 투과도 81 % 와 재료평가지수 $10.52{\times}10^{-3}{\Omega}^{-1}$ 의 가장 우수한 전기적 및 광학적 특성을 나타내었다.
