Cloning of Transgenic Rabbit Embryos Expressing Green Fluorescent Protein Gene by nuclear Transplantation (Green Fluorescent Protein 발현 토끼 수정란의 핵이식에 의한 복제)

  • The principal objective of this study was to clone transgenic embryos in order to improve the efficiency of transgenic animal production by the combination of microinjection and nuclear transplantation techniques. Mature female New Zealand White rabbits were superovulated by eCG and hCG treatments, fllowed by natural mating. Zygotes were collected from the oviducts at 18∼22 h after hCG injection by flushing with D-PBS containing 5% fetal calf serum(FCS). Two to three picoliters of green fluorescent protein(GFP) gene wa microinjected into male pronucleus. The foreign gene-injected zygotes were cultured in TCM-199 or RD medium containing 10% FCS with a monolayer of rabbit oviductal epithelial cells in a 5% CO2 incubator. The morulae expressing GFP gene were selected and their blastomeres were separated for the use of nuclear donor. Following nuclear transplantation of fluorescence-positive morula stage blastomeres, 13 (21.3%) out of 61 fused oocytes developed to blastocyst stage and all of the cloned blastocysts expressed GFP. The results indicate that the screening of transgene in rabbit embryos by GFP detection could be a promisible method for the preselection of transgenic embryos. Also the cloning of preselected transgenic embryos by nuclear transplantatin could be efficiently applied to the multiple production of transgenic animals.

Anti-inflammatory effects of Hataedock with Coptidis Rhizoma and Glycyrrhiza Uralensis on Allergic Rhinitis through Regulating IL-4 Activation (알레르기성 비염에서 황련-감초 하태독법의 IL-4활성 조절을 통한 항염증효과)

  • The aim of this study is to evaluate the anti-inflammatory effect of Hataedock treatment using Coptidis Rhizome and Glycyrrhiza Uralensis (CG) mixed extract in allergic rhinitis induced NC/Nga mice. We divided NC/Nga mice into 3 groups as follows; allergic rhinitis-induced group after CG Hataedock treatment (CGT, n=10), no treatment group (Ctrl), allergic rhinitis elicited group (ARE). To induce allergic rhinitis, NC/Nga mice of 3 weeks age were sensitized on 7, 8 and 9week by Ovalbumin (OVA) antigen in intranasal space. Hataedock using CG extract was administered on week 3 in allergic rhinitis-induced group (CGT) after Hataedock treatment. To identify distribution of Interlukin (IL)-4, Cluster of differentiation 40 (CD40), high-affinity IgE receptor ($Fc{\varepsilon}RI$), substance P, Matrix metallopeptidase 9 (MMP-9), Nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$) p65, Inducible nitric oxide synthase (iNOS) and Cycloxygenase-2 (COX-2), we used histological examination. CGT significantly inhibited IL-4 and CD40 response compared with ARE. The reduction of Th2 cytokine expression decreased inflammatory mediators such as $Fc{\varepsilon}RI$, substance P, MMP-9, $NF-{\kappa}B$ p65, iNOS and COX-2. Such immunological improvement induced reduction of respiratory epithelial damage and mucin secretion in goblet cell. These results indicate that Hataedock treatment suppresses allergic rhinitis through modulating of Th2 responses and diminishing various inflammatory mediators in nasal mucosal tissue. It might have potential applications for prevention and treatment of allergic rhinitis.

Evaluation of the effects of whitening mouth rinses combined with conventional tooth bleaching treatments

  • Objectives: The aim of the present study was to evaluate the effect of whitening mouth rinses alone and in combination with conventional whitening treatments on color, microhardness, and surface roughness changes in enamel specimens. Materials and Methods: A total of 108 enamel specimens were collected from human third molars and divided into 9 groups (n = 12): 38% hydrogen peroxide (HP), 10% carbamide peroxide (CP), 38% HP + Listerine Whitening (LW), 10% CP + LW, 38% HP + Colgate Plax Whitening (CPW), 10% CP + CPW, LW, CPW, and the control group (CG). The initial color of the specimens was measured, followed by microhardness and roughness tests. Next, the samples were bleached, and their color, microhardness, and roughness were assessed. Data were analyzed through 2-way analysis of variance (ANOVA; microhardness and roughness) and 1-way ANOVA (color change), followed by the Tukey post hoc test. The Dunnett test was used to compare the roughness and microhardness data of the CG to those of the treated groups. Results: Statistically significant color change was observed in all groups compared to the CG. All groups, except the LW group, showed statistically significant decreases in microhardness. Roughness showed a statistically significant increase after the treatments, except for the 38% HP group. Conclusions: Whitening mouth rinses led to a whitening effect when they were used after conventional treatments; however, this process caused major changes on the surface of the enamel specimens.

Biocompatibility and bioactive potential of the NeoMTA Plus endodontic bioceramic-based sealer

  • Objectives: This study evaluated the biocompatibility and bioactive potential of NeoMTA Plus mixed as a root canal sealer in comparison with MTA Fillapex. Materials and Methods: Polyethylene tubes filled with NeoMTA Plus (n = 20), MTA Fillapex (n = 20), or nothing (control group, CG; n = 20) were inserted into the connective tissue in the dorsal subcutaneous layer of rats. After 7, 15, 30 and 60 days, the specimens were processed for paraffin embedding. The capsule thickness, collagen content, and number of inflammatory cells (ICs) and interleukin-6 (IL-6) immunolabeled cells were measured. von Kossa-positive structures were evaluated and unstained sections were analyzed under polarized light. Two-way analysis of variance was performed, followed by the post hoc Tukey test (p ≤ 0.05). Results: At 7 days, the capsules around NeoMTA Plus and MTA Fillapex had more ICs and IL-6-immunostained cells than the CG. However, at 60 days, there was no significant difference in the IC number between NeoMTA Plus and the CG (p = 0.1137) or the MTA Fillapex group (p = 0.4062), although a greater number of IL-6-immunostained cells was observed in the MTA Fillapex group (p = 0.0353). From 7 to 60 days, the capsule thickness of the NeoMTA Plus and MTA Fillapex specimens significantly decreased, concomitantly with an increase in the collagen content. The capsules around root canal sealers showed positivity to the von Kossa stain and birefringent structures. Conclusions: The NeoMTA Plus root canal sealer is biocompatible and exhibits bioactive potential.

The influence of sodium hypochlorite concentration on the fibrin structure of human blood clots and transforming growth factor-beta 1 release: an ex vivo study

  • Objective: This study investigated the effects of various concentrations of sodium hypochlorite (NaOCl) on human whole-blood clotting kinetics, the structure of the blood clots formed, and transforming growth factor (TGF)-β1 release. Materials and Methods: Human whole blood was collected from 5 healthy volunteers and divided into 4 groups: CG (control, 0.5 mL of blood), BN0.5 (0.5 mL of blood with 0.5 mL of 0.5% NaOCl), BN3 (0.5 mL of blood with 0.5 mL of 3% NaOCl), and BN5.25 (0.5 mL of blood with 0.5 mL of 5.25% NaOCl). The effects of NaOCl on clotting kinetics, structure of fibrin and cells, and release of TGF-β1 were assessed using thromboelastography (TEG), scanning electron microscopy (SEM), and enzyme-linked immunosobent assay, respectively. Statistical analysis was conducted using the Kruskal Wallis and Mann-Whitney U tests, followed by the post hoc Dunn test. A p value < 0.05 indicated statistical significance. Results: The blood samples in BN0.5 and BN3 did not clot, whereas the TEG of BN5.25 showed altered clot formation. Samples from the CG and BN3 groups could only be processed with SEM, which showed that the latter lacked fibrin formation and branching of fibers, as well as clumping of red blood cells with surface roughening and distortion. TGF-β1 release was significantly highest in BN3 when all groups were compared to CG (p < 0.05). Conclusions: Each concentration of NaOCl affected the release of TGF-β1 from blood clots and altered the clotting mechanism of blood by affecting clotting kinetics and cell structure.

Design and Implementation of eBook Annotation Ontology Based on Non-First Normal Form (Non-First Normal Form에 입각한 eBook Annotation 온톨로지의 설계와 구현)

  • 본 연구에서는 온라인 다중 사용자 환경의 eBook 어노테이션 시스템 개발에서 데이터를 의미 기반으로 관리하고, 데이터에 대하여 상호 공통적인 이해를 표현하며, 그리고 데이터에 대한 무결성 검사 등을 지원하기 위해서 eBook 어노테이션 온톨로지를 구축하였다. eBook 어노테이션 테이터에 대한 상호 공통적인 이해의 표현을 위해서 한국 전자책 문서 표준인 EBKS(Electronic Book of Korea Standard)를 기반으로 구축 하였으며 구축된 온톨로지는 Conceptual Graph(CG)를 사용하여 표현하였다. 의미 기반의 처리를 위해서 본 온톨로지에서는 다국어(Multilingua) 관계를 고려하였으며 또한 오노테이션 데이터 생성 시 중요도를 표현하기 위해서 중요성 axiom을 고려했고, $NF^2$(Non-First Normal Form)에 입각하여 온톨로지를 설계함으로서 어노테이션 데이터의 검색에 활용도를 높였다. 제안된 온톨로지는 어노테이션 데이터의 재사용성을 높일 수 있고 의미 정보를 활용함으로써 eLearning, cyberclass과 같은 다중 사용자 환경에서 효과적인 협업을 가능하게 한다. 본 연구에서는 구현한 eBook annotation 시스템은 구축한 온톨로지를 사용함으로써 의미 기반의 데이터 관리가 가능하다. 또한 어노테이션 생성 시 온톨로지 구조를 모르더라도 어노테이션을 생성할 수 있는 인터페이스를 구현하였다.

Study on Biochemical Constituents of Caprine Ovarian Follicular Fluid after Superovulation

  • The experiment was designed on 42 non pregnant Black Bengal goat. Out of which 18 were subjected to a superovulatory treatment comprising of eCG and hCG for embryo transfer study. The remaining 24 goats received no treatment and served as control for parameter studied as well as recipient for embryo transfer studies. Important biochemical constituents such as acid and alkaline phosphatase, total protein and cholesterol and inorganic phosphorus were estimated in the follicular fluid of control and treated group and the values were separately recorded for small medium and large size follicle. The results indicated a significant effect on acid phosphotase activity due to size of follicle. The value increased progressively from small to medium and from medium to large follicles. Alkaline phosphotase activity showed reverse trend. Alkaline phosphotase decreased progressively as size increased. The concentration of inorganic phosphorus did not reveal any significant difference between the control and treatment groups and also between the different size follicles. The concentration of protein decreased significantly from small to medium and from medium to large, although no difference was observed between the control and treatment groups. The concentration of Cholesterol in the follicular fluid indicated a significant increase from small to medium and to large follicle. Here also no difference was observed due to treatment. Similar in the composition of follicular fluid in the respect of above mentioned constituents indicated no of super ovulatory treatment on follicular fluid composition.

Seismic Tomography using Graph Theoretical Ray Tracing

  • Seismic tomography using the graph theoretical method of ray tracing is performed in two synthetic data sets with laterally varying velocity structures. The straight-ray tomography shows so poor results in imaging the laterally varying velocity structure that the ray-traced tomographic techniques should be used. Conventional ray tracing methods have serious drawbacks, i.e. problems of convergence and local minima, when they are applied to seismic tomography. The graph theretical method finds good approximated raypaths in rapidly varying media even in shadow zones, where shooting methods meet with convergence problems. The graph theoretical method ensures the globally minimal traveltime raypath while bending methods often cause local minima problems. Especially, the graph theoretical method is efficient in case that many sources and receivers exist, since it can find the traveltimes and corresponding raypaths to all receivers from a specific source at one time. Moreover, the algorithm of graph theoretical method is easily applicable to the ray tracing in anisotropic media, and even to the three dimensional case. Among the row-active inversion techniques, the conjugate gradient (CG) method is used because of fast convergence and high efficiency. The iterative sequence of the ray tracing by the graph theoretical method and the inversion by the CG method is an efficient and robust algorithm for seismic tomography in laterally varying velocity structures.

Comparison of the trometamol-balanced solution with two other crystalloid solutions for fluid resuscitation of a rat hemorrhagic model

  • Currently, the optimal resuscitation fluid remains debatable. Therefore, in the present study, we designed a trometamol-balanced solution (TBS) for use as a resuscitation fluid for hemorrhagic shock. Hemorrhagic shock was induced in 18 male Wistar-Kyoto rats, which were assigned to normal saline (NS), Ringer's solution (RS), and TBS groups. During the hemorrhagic state, their hemodynamic parameters were recorded using an Abbott i-STAT analyzer with the CG4+ cartridge (for pH, pressure of carbon dioxide, pressure of oxygen, total carbon dioxide, bicarbonate, base excess, oxygen saturation, and lactate), the CG6+ cartridge (for sodium, potassium, chloride, blood glucose, blood urea nitrogen, hematocrit, and hemoglobin), and enzyme-linked immunosorbent assay kits (calcium, magnesium, creatinine, aspartate aminotransferase, alanine aminotransferase, bilirubin, and albumin). Similar trends were found for the parameters of biochemistries, electrolytes, and blood gas, and they revealed no significant changes after blood withdrawal-induced hemorrhagic shock. However, the TBS group showed more effective ability to correct metabolic acidosis than the NS and RS groups. TBS was a feasible and safe resuscitation solution in this study and may be an alternative to NS and RS for resuscitation in hemorrhagic shock patients without liver damage.
