Estimation of deep reservoir temperature of thermal groundwaters in Bugok and Magumsan areas, South Korea

  • In this study, hydrochemical studies of thermal waters in the Bugok and Magumsan areas showing geothermal anomalies were carried, and the applicability of ion seothermometers and multiple mineral equilibrium approach was examined to estimate their potential deep reservoir temperatures. Typical thermal waters of the two areas are clearly grouped into two major types, according to water chemistry: Na-Cl type (group A) and Na-SO4 type (group D). Compared to group A, group B and C waters show some modifications in chemistry. Group E waters show the modified chemistry from group D. Geothermal waters from the two areas showed some different chemical characteristics. The thermal waters of group A and B in Magumsan area are typically neutral to alkaline (pH=6.7 to 8.1) and Cl-rich (up to 446.1 mg/L), while the waters of group D and E in Bugok area are alkaline (pH=7.6 to 10.0) and SO$_4$-rich (up to 188.0 mg/L). The group A (Na-Cl type) and group D (Na-SO$_4$ type) waters correspond to mature or partially immature water, whereas the other types are immature water. The genesis of geothermal waters are considered as follows: group A and B waters were formed by seawater infiltration into reservoir rocks along faults and fracture zones and possibly affected by fossil connate waters in lithologic units through which deep hot waters circulate; on the other hand, group D and E waters were formed by the oxidation of sulfide minerals (mainly pyrite) in surrounding sedimentary rocks and/or hydrothermal veins occurring along restricted fracture channels and were possibly affected by the input and subsequent oxidation of S-bearing gases (e.g. H2S) from deep thermal reservoir (probably, cooling pluton). The application of quartz, Na-K, K-Mg geothermometers to the chemistry of representative group A and D waters yielded a reasonable temperature estimate (99-147$^{\circ}C$ and 90-142$^{\circ}C$) for deep geothermal reservoir. Aqueous liquid-rich fluid inclusions in fracture calcites obtained from drillcores in Bugok area have an average homogenization temperature of 128$^{\circ}C$, which corresponds to the results from ion geothermometers. The multiple mineral equilibrium approach yielded a similar temperature estimate (105-135$^{\circ}C$ and 100-14$0^{\circ}C$). We consider that deep reservoir temperatures of thermal waters in the Magumsan and Bugok areas can be estimated by the chemistry of typical Na-Cl and Na-SO$_4$ type waters and possibly approach 105-135$^{\circ}C$ and 100-14$0^{\circ}C$.

  • The thermal-hydraulic effects of removing the RCC guide thimble plugs are evaluated for Westinghouse type PWR plants as a part of feasibility study: core outlet loss coefficient, thimble bypass flow, and best estimate flow. It is resulted that the best estimate thimble bypass flow increases about by 2% and the best estimate flow increase approximately by 1.2%. The resulting DNBR penalties can be covered within the current DNBR margin. Accident analyses are also investigated and the dropped rod transient is shown to be limiting and relatively sensitive to bypass flow variation.

Biological activity of Euonymus alatus (Thunb.) Sieb. wing extracts (화살나무 날개 추출물의 생리활성)

  • Euonymus alatus (Thunb.) Sieb., also known as the arrow tree in Korea, is a plant in East Asia used in traditional medicine and food. In particular, the wings of E. alatus are rich in phenolic compounds. This study evaluated the antioxidant, α-glucosidase inhibition, and anti-cancer activities of E. alatus wing extracts. The radical and hydrogen peroxide scavenging acitvities and reducing the power of 1,000 ㎍/mL E. alatus wing extracts, were similar to those of the positive control (0.1% BHT, 0.1% α-tocopherol). In addition, ethanol and methanol extract at 250 ㎍/mL showed 95.70 and 94.99% of α-glucosidase inhibition activity, respectively. The ethanol extract of E. alatus wings had the highest total polyphenol and flavonoid contents (867.8 mg% and 521.7 mg%, respectively). The E. alatus wing extracts significantly decreased the cell viability of LNCaP human prostate cancer cells (p<0.001), MDA-MB-231 human breast cancer cells (p<0.001), and HT-29 human colon cancer cells (p<0.001) in a dose-dependent manner. However, there was no significant effect on B16 mouse melanoma cells. Notably, the ethanol extracts showed higher cancer cell growth inhibitory activity in LNCaP and HT-29 cells than the other extracts. These results suggest that E. alatus wing extracts could have significant clinical applications, and our results can be used as basic data for future functional food material development.

Platelet-Agglutinating Protein p37 from a Patient with Thrombotic Thrombocytopenic Purpura Has Characteristics Similar to Prethrombin 2

  • Thrombotic thrombocytopenic purpura (TTP) is characterized by widespread platelet thrombi in arterioles and capillaries. Unusually large or multimeric von Willebrand factor, as well as one or ore platelet-agglutinating factors, have been implicated in the pathogenesis of TTP. But, the actual mechanisms of platelet agglutination have not been satisfactorily explained. Recent studies suggested the 37-kDa platelet-agglutinating protein (PAP) p37 to be partially responsible for the formation of platelet thrombi in patients with TTP. We studied mobility in SDS-PAGE, the sequence of N-terminal amino acid residues, DNA and antigenic characteristics of PAP p37, which might be related to the pathogenesis of TTP. PAP p37 was purified from the plasma of a 31-year-old male Korean patient with acute TTP. The findings are as follows: (1) We compared PAP p37 with thrombin through the use of SDS-PAGE, either with or without $\beta$-mercaptoethanol. PAP p37 did not appear to be cleaved between the A- and B-chains of prethrombin 2. However, thrombin did cleave between those of prethrombin 2, but linked with disulfide bridge. (2) N-terminal 21 amino acid sequence of PAP p37 was T-F-G-S-G-E-A-D-X-G-L-R-P-L-F-E-K-K-S-L-E. It appeared to be identical to that of 285-305 amino acid residues of human prothrombin (prethrombin 2). (3) No prothrombin gene DNA mutation was revealed. (4). The antigenicity of PAP p37 was similar to thrombin, which was a result of the competitive binding against the anti-thrombin antibody. With these results, we conclude that PAP p37 has similar characteristics to prethrombin2.

Changing Patterns of Communicable Diseases in Korea (우리나라 전염성 질환의 변화 양상)

  • Before twentieth centuries and during early twentieth centuries, communicable diseases were the major cause of morbidity and mortality in Korea. But reliable data are not available. After 1975, the overall morbidity and mortality from communicable diseases, rapidly declined. Recently many new pathogenic microbes were recognized: L. monocytogenes, Hantaan virus, Y. pseudotuberculosis, P. multocida, L. pneumophilia, Human immunodeficiency virus (HIV), G. seoi, H. capsulatum, C. burnetii, V. cholerae O139, C. parvum, F. tularensis, E. coli O157:H7, B. burgdorferi, S. Typhimurium DT104, Rotavirus, hepatitis C virus and so on. Since the first HIV infection recognized in 1985, the reported cases of infection and deaths from HIV/AIDS have been steady increased each year. Legionnaire's disease, E. coli O157:H7 colitis, listeriosis and crytosporidiasis have been occurring just sporadically among immunocompromized cases. Many re-emerging communicable diseases were occurred in Korea: leptospirosis, malaria, endemic typhus, cholera, tsutsugamushi disease, salmonellosis, hepatitis A, shigellosis, mumps, measles, acute hemorrhagic conjunctivitis, brucellosis and so on. Leptospirosis and tsutsugamushi diseases have been noticed as major public health problems since 1980s. The malaria that had been virtually disappeared for a decade has reappeared from 1993 with striking increase of patients in recent 3-4 years. The distributions of salmonella and shigella serotypes have been changed a lot in recent few decades. Furthermore rapid emergence of antibiotic-resistant bacterial strains induces more difficult and complex problems in control of communicable diseases. We must recognize on the importance of environment and ecosystem conservation and careful prescription of anti-microbial agent in order to prevent communicable diseases.

Effect of Heat Treatment on the Color Change of Blue-Stained Pinus densiflora Boards (열처리에 의한 청변균 변색 소나무 판재의 재색 변화)

  • Red pine is a popular species for making a cutting board in Korea, but easily sap stained. Heat treatment could improve its quality by darkening and equalizing the color of sap stained wood. The color change of sap stained red pine boards was investigated by heat treatment at $190^{\circ}C$. It was observed that the color of heat treated boards got darker and it made the color of sap stain vanished. A colorimeter was used to measure color indexes. It was revealed that the values of the lightness ($L^*$) and the yellowness ($b^*$) decreased as heat treatment repeated while the values of the redness ($a^*$) increased. The average of the color difference (${\Delta}E^*$) between the control and 1st heat treated boards was 16.1, which could be expressed as 'Extremely different' while that between the 1st and 2nd heat treated boards was 8.3, which could be expressed as 'Considerably different'. The fact that heat treatment equalized the color of boards was confirmed by a statistical analysis.

Effects of Ibuprofen on Bioavailability of Ciprofloxacin (이부프로펜이 씨프로플록사신의 생체이용률에 미치는 영향)

  • The effects of ibuprofen on bioavailability of ciprofloxacin were studied in rabbits. Animals were divided into three groups ; group A received 60 mg/kg of ciprofloxacin; group B and C received 60 mg/kg of ciprofloxacin with 60 and 240 mg/kg of ibuprofen, respectively. Ciprofloxacin and ibuprofen were given by single oral administration. Serum concentrations of ciprofloxacin were measured by high performance liquid chromatography with UV detector and pharmacokinetic parameters were calculated. Area under the serum concentrations versus time curve (${\pm}\;S.E.$) of ciprofloxacin were decreased in group B and C compared with group A ($12.26\;{\pm}\;0.94$ and $12.57\;{\pm}\;0.94$ vs. $15.71\;{\pm}\;1.06\;{\mu}g{\cdot}hr/ml$, p<0.05), whereas total clearances were increased ($1.81\;{\pm}\;0.13$ and $1.76\;{\pm}\;0.12$ vs. $1.40\;{\pm}\;0.09\;l/hr/kg$, p<0.05). No significant differances in these parameters were observed between group B and C. Relative bio-availability of group B and C to group A were 78 and 80%, respectively. These results suggest that the coadministration of ibuprofen with ciprofloxacin may reduce the bioavailability of ciprofloxacin.

Char Oxidation Characteristics of High Ash Coal in Drop Tube Furnace (고회분탄의 촤 산화 반응 특성 연구)

  • The char oxidation characteristics of high ash coal were experimentally investigated at several temperatures (from 900 to $1300^{\circ}C$) for 4 types of coals (Gunvor, Glencore, Noble, and ECM) under atmospheric pressure in a drop tube furnace (DTF). The char reaction rate was calculated from the exhaust gas concentrations (CO and $CO_2$) using FT-IR, and the particle temperature was measured using the two-color method. In addition, the activation energy and pre-exponential factor for high ash coal char were calculated based on the Arrhenius equation. The results show that as the ash content increases, the particle temperature and area reactivity decreases. This is because in high ash coal, the large heat capacity of the ash, ash vaporization, and relatively low fixed carbon content of ash suppress combustibility during char oxidation. As a result, the higher ash content of coal leads to high activation energy.

Expression and Characterization of Trehalose Biosynthetic Modules in the Adjacent Locus of the Salbostatin Gene Cluster

  • The pseudodisaccharide salbostatin, which consists of valienamine linked to 2-amino-1,5-anhydro-2-deoxyglucitol, is a strong trehalase inhibitor. From our Streptomyces albus ATCC 21838 genomic library, we identified thirty-two ORFs in a 37-kb gene cluster. Twenty-one genes are supposed to be a complete set of modules responsible for the salbostatin biosynthesis. Through sequence analysis of the gene cluster, some of the upstream gene products (SalB, SalC, SalD, SalE, and SalF) revealed functional resemblance with trehalose biosynthetic enzymes. On the basis of this rationale, we isolated the five genes (salB, salC, salD, salE, and salF) from the S. albus ATCC 21838 and cloned them into the expression vector pWHM3. We demonstrated the noticeable expression and accumulation of trehalose, using only the five upstream biosynthetic gene cluster of salbostatin, in the transformed Streptomyces lividans TK24. Finally, 490 mg/l trehalose was produced by fermentation of the transformant with sucrosedepleted R2YE media.

  • Spectroscopic diagnostics on reactive plasmas was carried out in a facing target sputtering unit with BaO +12Fe composite targets and 50% $O_2+$ Ar sputter gas. Spectra of rective plasmas were composed of peaks which were assigned to be Ba, B$a^+$, Fe, FeO, F$e^+$, Ar, $Ar^+$, O, $O^+$. As detecting positions in plasmas were far away from targets, the relative peak intensities of the ions and neutral species were decreased, but the relative intensities of the former decreased faster than those of the latter.
