Isoforms of Glucose 6-Phosphate Dehydrogenase in Deinococcus radiophilus

  • Glucose 6-phosphate dehydrogenase (G6PDH, EC 1.1.1.49) in Deinococcus radiophilus, an extraordinarily UV-resistant bacterium, was investigated to gain insight into its resistance as it was shown to be involved in a scavenging system of superoxide $(O_2^{-1})$ and peroxide $(O_2^{-2})$ generated by UV and oxidative stresses. D. radiophilus possesses two G6PDH isoforms: G6PDH-1 and G6PDH-2, both showing dual coenzyme specificity for NAD and NADP. Both enzymes were detected throughout the growth phase; however, the substantial increase in G6PDH-1 observed at stationary phase or as the results of external oxidative stress indicates that this enzyme is inducible under stressful environmental conditions. The G6PDH-1 and G6PDH-2 were purified 122- and 44-fold (using NADP as cofactor), respectively. The purified G6PDH-1 and G6PDH-2 had the specific activity of 2,890 and 1,033 U/mg protein (using NADP as cofactor) and 3,078 and 1,076 U/mg protein (using NAD as cofactor), respectively. The isoforms also evidenced distinct structures; G6PDH-1 was a tetramer of 35 kDa subunits, whereas G6PDH-2 was a dimer of 60kDa subunits. The pIs of G6PDH-1 and G6PDH-2 were 6.4 and 5.7, respectively. Both G6PDH-1 and G6PDH-2 were inhibited by both ATP and oleic acid, but G6PDH-1 was found to be more susceptible to oleic acid than G6PDH-2. The profound inhibition of both enzymes by ${\beta}-naphthoquinone-4-sulfonic$ acid suggests the involvement of lysine at their active sites. $Cu^{2+}$ was a potent inhibitor to G6PDH-2, but a lesser degree to G6PDH-1. Both G6PDH-1 and G6PDH-2 showed an optimum activity at pH 8.0 and $30^{\circ}C$.

Multichannel Anticollision Protocol for Improving Tag Collection Performance in Active RFID Systems (능동형 RFID 시스템에서 태그 수집 성능 향상을 위한 다중채널 기반 충돌방지 프로토콜)

  • Tag collection is one of the major functions in RFID systems. In this paper, we propose a multichannel slotted-aloha anticollision protocol to mitigate the tag collision problem and improve tag collection performance in active RFID systems. In the proposed anticollision protocol, while an RFID reader transmits commands to multiple RFID tags via a common channel, it receives multiple tag responses over multiple channels simultaneously. We also implemented an active RFID system supporting the proposed anticollision protocol. In experiments with the implemented reader and 60 tags, the proposed dual-channel slotted-aloha anticollision protocol showed a large improvement in tag collection performance compared with the single-channel slotted-aloha anticollision protocol. With 60 tags, the average time spent on tag collection using the dual-channel anticollision protocol was 600.543 ms, which was 46.3% of the 1289.721 ms required using the single-channel anticollision protocol.

  • The next decade promises drastic improvements and additions to global navigation satellite systems (GNSS). Plans for GPS modernization include a civilian code measurement on the L2 frequency and a new L5 signal at 1176.45 MHz. Current speculations indicate that a fully operational constellation with these improvements could be available by 2013. Simultaneously, the Galileo Joint Undertaking is in the development and validation stages of introducing a parallel GNSS called Galileo. Galileo will also transmit freely available satellite navigation signals on three frequencies and is scheduled to be fully operational as early as 2008. In other words, a dual system receiver (e.g., GPS+GALILEO) for general users can access six civil frequencies transmitted by at least fifty eights navigation satellites in space. The advent of GALILEO and the modernization of GPS raise a lot of attention to the study of the compatibility and interoperability of the two systems. A number of performance analyses have been conducted in a global scale with respect to availability, reliability, accuracy and integrity in different simulated scenarios (such as open sky and urban canyons) for the two systems individually and when integrated. Therefore, the scope of this article aims at providing the technical benefits analysis for Taiwan specifically in terms of the performance indices mentioned above in a local scale, especially in typical urban canyon scenarios. The conclusions gained by this study will be applied by the Land Survey Bureau of Taiwanese as the guideline for developing future GNSS tracking facilities and dual GNSS processing module for precise surveying applications in static and kinematic modes.

A Comparison of Fault Tolerant Ethernet Implementation Approaches (고장 극복 (Fault Tolerant) Ethernet 구현 방안의 비교 분석)

  • Reliability is absolutely necessary in most recent mission critical systems which usually utilize the Ethernet based computer networks. A typical way to increase system reliability is to equip the fault tolerant Ethernet. In this paper we try to formulate the fault tolerant dual Ethernet concept and presents a comparison of its implementation approaches. Two types of dual Ethernet configuration are analyzed; the divided and the connected. Then the characteristics of three current implementation approaches which are the hardware based, the software based, and the recently proposed hybrid approach are compared. The results show that the hardware based or the hybrid approaches can be a better solution for the real time mission critical systems. Also for the systems which require the use of Commercial-Off-The-Shelf (COTS) hardware for fault tolerant Ethernet the possible choice is the software based or the hybrid approach.

  • Communication antennas used at sea have been developed remarkably. However, the speed of this development is insufficient when compared with user demands. Therefore, we developed an antenna for UWB/Bluetooth that operates on 3 [GHz] and 5.72 [GHz] bands in order to use the high-speed communication network system which improved antenna miniaturization, gain and radiation patterns. To improve bandwidth, a microstrip patch antenna was selected and CST Microwave Studio 2014 program was used. Through the program, we calculated the slot width, length, transmission line width, etc. using a theoretical formula at each step. These figures were checked through simulation to see if they meet production standards. UWB for high-speed wireless communication for short-range communication at sea, Bluetooth for exchanging information at a short distance by connecting each device, and corresponding technology can be easily utilized.

  • A design of an elliptic curve cryptography (ECC) processor that supports both pseudo-random curves and Koblitz curves over $GF(2^m)$ defined by the NIST standard is described in this paper. A finite field arithmetic circuit based on a word-based Montgomery multiplier was designed to support five key lengths using a datapath of fixed size, as well as to achieve a lightweight hardware implementation. In addition, Lopez-Dahab's coordinate system was adopted to remove the finite field division operation. The ECC processor was implemented in the FPGA verification platform and the hardware operation was verified by Elliptic Curve Diffie-Hellman (ECDH) key exchange protocol operation. The ECC processor that was synthesized with a 180-nm CMOS cell library occupied 10,674 gate equivalents (GEs) and a dual-port RAM of 9 kbits, and the maximum clock frequency was estimated at 154 MHz. The scalar multiplication operation over the 223-bit pseudo-random elliptic curve takes 1,112,221 clock cycles and has a throughput of 32.3 kbps.

  • In this paper, we propose MSAG-SCS-MMA-I adaptive blind equalization with double step size with very small residual ISI and MSE at steady-state while significantly improving the convergence speed of the traditional SCS-MMA-I algorithm in 256-QAM system. And we evaluate the equalization performance for this algorithm. Different step sizes according to the absolute value of decision-directed error instead of a fixed step-size are applied to the tap update equation of MSAG-SCS-MMA-I, which is controlled by binary flags of '1' or '0' obtained from SCS-MMA-I and decision-directed algorithms. This makes for excellent equalization performance. As a result of computer simulation, we confirmed that the proposed algorithm has more better performance than the MMA, SCS-MMA-I, and MSAG-SCS-MMA-I algorithms in terms of the performance index such as residual ISI, MSE, and MD.

Cyclodextrins' effect on the enatioseparation of some PPIs and capillary electrophoresis method development for determining rabeprazole enantiomers

  • Over the past decades, chiral switch of the proton pump inhibitors (PPIs) has been received widespread attention in therapeutic advantages as well as pharmaceutical analysis. In present study, the influence of cyclodextrins (CDs) on the chiral separation of four common PPIs (lansoprazole, omeprazole, pantoprazole, and rabeprazole) was investigated. The results demonstrated that capillary electrophoresis (CE) with dual CDs as a chiral selector system is a possible and promising method for the enantioseparation of these PPIs. Rabeprazole, which is the most challenging and acid-labile candidate among four PPIs, was selected for further development of the technique. To optimize CE condition, the effects of capillary parameters and background electrolytes on the enantioseparation were investigated. Finally, the best chiral separation was acheived by using sulfobutyl ether-${\beta}$-CD, and ${\gamma}$-CD as dual chiral selectors. The developed CE method not only provided the effective chiral separation but also showed the good stability of rabeprazole. The proposed method was successfully validated according to the International Conference on Harmonization guideline and effectively applied to determine rabeprazole enantiomers in commercial rabeprazole tablets, with recoveries ranging from 97.17 % to 103.29 % of the label content.

Strength analysis of the driving shift gears for a 67 kW class agricultural tractor according to tire type

  • The purpose of this study was to measure the engine torque and rotational speed of a 67 kW class agricultural tractor according to tire type during plow tillage and to analyze the gear strength of the driving shift for the tractor. A field test was performed under the condition with a single tire (Test A) and dual tires (Test B) to increase the ground width of the rear tires. A load monitoring system was developed, and the engine torque and rotational speed were measured using controller area network (CAN) communication. The engine torque and rotational speed during plow tillage were calculated as the equivalent torque and speed using Palmgren Miner's rule. As a result, the equivalent torque and speed in Test A and Test B were 181.0 Nm and 1,913 rpm and 206.1 Nm and 2,130 rpm, respectively. As the ground width of the rear tire was increased, the bending stress in Test B was about 9.9 to 10.5% higher than that of the Test A, and the contact stress was about 4.6 to 4.9% higher than that of the Test A. Under all conditions, the safety factor for the bending and contact stress was 1 or more. Thus, the driving shift gears for the dual tire type are considered safe.

Dual Band Microstrip Antenna for Design Wimax/LTE 5G for Ship Radio Communication (선박 무선통신을 위한 Wimax/LTE 5G 용 이중대역 마이크로스트립 안테나 설계)

  • In this paper, we designed a microstrip patch antenna that can be applied to the Wimax/LTE 5G system among wireless media usable in coastal ships. The substrate of the proposed antenna is FR-4 (er=4.3), the size is 22 mm × 30 mm, and it can be used in the 3.5 GHz and 5.8 GHz bands of Wimax/LTE 5G by constructing a simple structure using a microstrip patch antenna. CST Microwave Studio 2014 was used for simulation, and the gain of the simulation result is 2.41dB at 2.4 GHz and 3.96 dB at 3.5 GHz. S-Parameter also showed a result of less than -10 dB (VSWR 2:1) in the desired frequency band, and designed a small variable and a miniaturized antenna so that the antenna can be used in mobile phones or electronic devices.
