슬관절 연골 연화증의 진단에서의 Dual Echo in Steady State (DESS) 영상의 유용성 : 급속 스핀에코 자기공명 영상과 비교 (Usefulness of Dual-Echo in Steady State (DESS) Image in Chondromalacia of Knee Joint: Comparison of DESS and Turbo Spin-Echo MR Images)

  • 목적: 급속 스핀 에코 MR영상과 Dual Echo in Steady State(DESS) 영상을 비교하여 슬관절 연골 연화증의 병소 발견에 있어서 DESS 영상의 유용성을 알아 보고자 하였다. 대상 및 방법: 1997년 1월부터 1998년 7월까지 관절경 수술과 자기 공명 영상을 시해안 환자중 수술 소견에서 슬관절의 연골 연화가 있었던 26명(남:여=14:12, 평균 46.3세)을 대상으로 하였다. 1.5 T 자기 공명 장치를 사용하였고, 급속 스핀 에코 양자 밀도와 T2 강조영상의 게측 게수로써 TR/TE 3000-4200/16-96msec, FOV $140-160{\times}150-160$, matrix size $180{\times}256$, 절편 두께 4.0mm, 절편 간격은 0.5mm로 하였으며, DESS 영상의 계측계수로써 TR/TE 25.4/9.0msec, filp angle $35-35^{\circ}$, FOV $150-160{\times}150-160mm$, matrix size $192{\times}256$, 효과적 절편 두께 1.5mm로 하였으며, DESS 영상은 시상단면을, 양자 밀도 강조 영상과 T2 강조영상은 시상 단면과 관상 단면을 얻어 비교하였다. 영상의 분석은 2명의 방사선과 의사가 합의하여 후향적으로 분석하였고, 슬개-대퇴 구획, 슬관절의 내측, 외측 구획으로 나누었으며, 자기 공명 영상과 관절경 소견의 등급은 0: 정상, I: 연화 또는 수포, II: 표층 균열, III: 심층 균열, IV: 전층 균열 또는 골노출로 나누었고, 각각의 민감도, 특이도, 그리고 정확도를 구하였다. 결과: 관절경 소견상 I등급 7예, II등급 21예, III등급 6예, IV등급은 18예 였으며, 급속 스핀 에코 자기공명 영상에서의 관절경 소견과 구획과 등급이 일치했던 경우는 등급별로 0%, 14%, 0%, 61%로 나타났으며, DESS 영상에서는 각각 0%, 33%, 50%, 67% 이었다(p=0.001). 또한 각 부위에서 등급을 고려하지 않은 경우, 고식적 자기공명 영상에서의 민감도, 특이도, 그리고 정확도는 59.6%, 88.6%, 78.8% 였으며, DESS 영상에서는 73.1%, 88.4%, 82.2%로 나타났다(p=0.007). 결론: 슬관절의 연골 연화증의 진단에 있어서 DESS 영상이 급속 스핀 에코 자기공명영상보다 높은 발견율과 민감도를 보였다. 그러므로 급속 스핀 에코 MR 영상과 더불어 DESS 영상이 슬관절 연골연화즈의 진단에 보다 더 도움을 줄 것으로 생각한다.

이중대역(PCS/IMT-2000) 마이크로스트립 패치 안테나 설계 및 구조에 따른 SAR 분석 (SAR Analysis and Design the Microstrip Patch Antenna on Dual-Band(PCS/IMT-2000))

  • 본 논문에서는 PCS/IMT-2000 이동통신 전화기에 의해 인체 두부에 유기되는 SAR의 1 g 및 10 g 평균 첨두 값을 비교 분석하였다. 평면형 구조인 single-semi-disc MPA와 double-semi-disc MPA를 설계하고, 안테나 설계 파라미터에 따른 공진 특성을 분석하였다. 그 결과 중심주파수 2.0 ㎓에서 대역폭이 15.5 %, 31 %로 나타났다. 그리고 제안한 안테나와 동일한 공진 주파수를 갖는 모노폴 안테나를 설계하여 SAR 값을 비교ㆍ분석하였다. 폴더형 전화기에 부착된 double-semi-disc MPA에 의한 1 g 및 10 g 평균 첨두 SAR 값은 각각 0.811 W/kg과 0.507 W/kg으로 모노폴 안테나의 1.206 W/kg과 0.755 W/kg에 비해 약32.7 % 가량 낮은 것으로 나타났다. 평면형 구조의 안테나는 등방성 방사패턴을 나타내는 모노폴 안테나에 비해 인체 외부 방향으로 지향성 방사 패턴을 나타내어 인체 두부에 유기되는 SAR이 낮은 값으로 나타났다.

전달 임피던스를 이용한 볼트 접합부 구조 건전성 모니터링 기법 (Bolt-joint Structural Health Monitoring Technique Using Transfer Impedance)

  • 본 연구에서는 볼트 접합부 건전성 모니터링을 위하여 이중(dual) 압전재료 기법인 전달 임피던스 기법(transfer impedance technique)을 이용하여 볼트 이완을 탐지할 수 있는 기법을 연구한다. 전달 임피던스 기법에서는 하나의 압전재료가 탄성파를 가진하고 다른 압전재료가 이를 센싱함으로서 결함 등에 의한 탄성파 전파의 변화를 감지한다. 한편, 단일 압전재료 기법을 이용하기 위해서는 고가의 임피던스 분석기(impedance analyser)를 이용해야 하지만, 전달 임피던스 기법은 임피던스 분석기를 대체하여 일반적인 함수발생기(function generator)와 디지털 멀티미터(digital multimeter)를 이용한 결함탐지가 가능하기 때문에 저비용의 결함탐지를 수행할 수 있다. 고력볼트 접합부로 구성된 강판 프레임 실험체를 제작하고, 볼트의 조임력을 단계적으로 감소시키며 실험을 수행하였다. 전달 임피던스 기법을 이용하여 손상지수를 구하였고 이를 이용하여 결함 유무를 합리적으로 판정할 수 있었으며, 볼트 이완의 정도가 커질수록 손상지수가 증가함을 알 수 있었다. 동일한 실험체에 대하여 임피던스 분석기를 이용한 단일 압전재료 기법에 대한 실험을 수행하여 결과를 비교하였는데, 결함정도가 커질수록 손상지수가 모두 증가하는 유사한 경향을 보였다. 즉, 고가의 임피던스 분석기를 배제하고 저비용으로 볼트 접합부의 결함상태 추정이 가능한 것으로 판단된다. 향후 결함위치 및 정도를 판정할 수 있는 연구가 추가된다면 볼트 접합부 건전성 모니터링에 효과적으로 활용될 수 있을 것으로 판단된다.

Effects of exploration and molecular mechanism of CsV on eNOS and vascular endothelial functions

  • This study aimed to investigate the effects and potential mechanisms of Chikusetsusaponin V (CsV) on endothelial nitric oxide synthase (eNOS) and vascular endothelial cell functions. Different concentrations of CsV were added to animal models, bovine aorta endothelial cells (BAECs) and human umbilical vein endothelial cells (HUVECs) cultured in vitro. qPCR, Western blotting (WB), and B ultrasound were performed to explore the effects of CsV on mouse endothelial cell functions, vascular stiffness and cellular eNOS mRNA, protein expression and NO release. Bioinformatics analysis, network pharmacology, molecular docking and protein mass spectrometry analysis were conducted to jointly predict the upstream transcription factors of eNOS. Furthermore, pulldown and ChIP and dual luciferase assays were employed for subsequent verification. At the presence or absence of CsV stimulation, either overexpression or knockdown of purine rich element binding protein A (PURA) was conducted, and PCR assay was employed to detect PURA and eNOS mRNA expressions, Western blot was used to detect PURA and eNOS protein expressions, cell NO release and serum NO levels. Tube formation experiment was conducted to detect the tube forming capability of HUVECs cells. The animal vasodilation function test detected the vasodilation functions. Ultrasonic detection was performed to determine the mouse aortic arch pulse wave velocity to identify aortic stiffness. CsV stimulus on bovine aortic cells revealed that CsV could upregulate eNOS protein levels in vascular endothelial cells in a concentration and time dependent manner. The expression levels of eNOS mRNA and phosphorylation sites Ser1177, Ser633 and Thr495 increased significantly after CsV stimulation. Meanwhile, CsV could also enhance the tube forming capability of HUVECs cells. Following the mice were gavaged using CsV, the eNOS protein level of mouse aortic endothelial cells was upregulated in a concentration- and time-dependent manner, and serum NO release and vasodilation ability were simultaneously elevated whereas arterial stiffness was alleviated. The pulldown, ChIP and dual luciferase assays demonstrated that PURA could bind to the eNOS promoter and facilitate the transcription of eNOS. Under the conditions of presence or absence of CsV stimulation, overexpression or knockdown of PURA indicated that the effect of CsV on vascular endothelial function and eNOS was weakened following PURA gene silence, whereas overexpression of PURA gene could enhance the effect of CsV upregulating eNOS expression. CsV could promote NO release from endothelial cells by upregulating the expression of PURA/eNOS pathway, improve endothelial cell functions, enhance vasodilation capability, and alleviate vessel stiffness. The present study plays a role in offering a theoretical basis for the development and application of CsV in vascular function improvement, and it also provides a more comprehensive understanding of the pharmacodynamics of CsV.

  • 최근 지능화 고도화 되고 있는 사이버위협으로부터 기업의 정보자산을 안전하게 보호하기 위해서는 기술적인 분야뿐만 아니라 관리적, 환경적 분야 등 전방위적 대응체제를 구축하여야 한다. 본 연구에서는 보안적으로 안전한 망 설계를 위해 물리적 망분리와 논리적 망분리등 보안 망 이론에 대한 사례를 분석하여 기업 환경에 적합하고 상시적 대응 및 관리가 가능한 정보보호 관리 모델을 제안한다. 특히, 제안 모델은 기존 망에서의 개선사항을 도출하고, 개선사항이 적용된 망을 설계하기 위해서 중앙 관리성을 향상시킨 실시간 보안 대응 능력, 보안 위협 발생 시 선제 탐지 및 능동적 대처, 중요 장비 이중활르 통한 고가용성, 고성능, 고신뢰성 확보, 개별 네트워크의 보안 정책 통합 관리, 개별 네트워크의 망 분리로 보안성 향성 등의 기능을 적용하였다.

복분자의 유방암 세포 사멸 및 사멸 유전자 발현 유도 효과 (Inducing Effects of Rubus coreanus on Cell Death and Apoptotic Gene Expressions in Human Breast Cancer Cells)

  • We investigated the inducing effects of Rubus coreanus extract (RCE) on apoptosis and its related gene expressions in human breast cancer cells. MDA-MB-231 cells were cultured in the presence of 0, 200, 300, and $400{\mu}g/mL$ RCE for 24h. MTT assay demonstrated that relative cell viability measured a decrease in a dose-dependent manner (p<0.05). This dependency was also found in the increasing levels of cell death by a dual staining with Hoechst 33322 and propidium iodide (p<0.05). These close associations was also observed by different stages of apoptotic processes, as shown by an Apoptosis Detection Kit. To determine whether the alterations in such cell activities obtained above cause the induction of apoptotic genes, PT-PCR was performed expressions of both Bcl-2 and Bax mRNAs. The Bcl-2/Bax ratio which is an important indicator of apoptosis, was found to have significantly decreased dose dependence (p<0.05). Western blot analysis also demonstrated that Caspase-3 significantly increases in a dose-dependent manner (p<0.05) in addition to similar alterations of other proteins examined. Taken these results together, the ethanolic RCE used induces a reduction in cell viability along with increased membrane permeability. This leads to a precautious apoptotic process and, subsequently, cell death through the apoptotic pathway involving Bax and Caspase-3 in human breast cancer MDA-MB-231 cells.

Screening and Evaluation of Streptomyces Species as a Potential Biocontrol Agent against a Wood Decay Fungus, Gloeophyllum trabeum

  • Two-hundred and fifty-five strains of actinomycetes isolated from soil samples were screened for their antagonistic activities against four well-known wood decay fungi (WDF), including a brown rot fungus, Gloeophyllum trabeum and three white rot fungi Donkioporia expansa, Trametes versicolor, and Schizophyllum commune. A dual culture assay using culture media supplemented with heated or unheated culture filtrates of selected bacterial strains was used for the detection of their antimicrobial activity against four WDF. It was shown that Streptomyces atratus, S. tsukiyonensis, and Streptomyces sp. greatly inhibited the mycelial growth of the WDF tested compared with the control. To evaluate the biocontrol efficacy of S. atratus, S. tsukiyonensis, and Streptomyces sp., wood blocks of Pinus densiflora inoculated with three selected Streptomyces isolates were tested for weight loss, compression strength (perpendicular or parallel to the grain), bending strength, and chemical component changes. Of these three isolates used, Streptomyces sp. exhibited higher inhibitory activity against WDF, especially G. trabeum, as observed in mechanical and chemical change analyses. Scanning electron microscopy showed that cell walls of the wood block treated with Streptomyces strains were thicker and collapsed to a lesser extent than those of the non-treated control. Taken together, our findings indicate that Streptomyces sp. exhibits the potential to be used as a biocontrol agent for wood decay brown rot fungus that causes severe damage to coniferous woods.

백서(白鼠) 중격측좌핵에서 Haloperidol로 유발된 세포외 도파민 농도 변화에 대한 (-)-3-PPP 전처치 효과 (Effect of Pretreatment of (-)-3-PPP on the Haloperidol-Induced Extracellular Dopamine Concentrations in the Nucleus Accumbens of Rats)

  • Objectives : To investigate the effects of (-)-3-PPP(0.5, 2, and 10mg/kg, s.c.) and haloperidol(0.1, 0.5, and 2mg/kg, s.c.) on the extracellular dopamine concentrations, and the effect of pretreatment with (-)-3-PPP(2mg/kg) on the haloperidol(2mg/kg)-induced extracellular dopamine concentrations in the nucleus accumbens(NAS) of free moving rats. Methods : Dopamine levels in dialysate were determined with high pressure liquid chromatography(HPLC) with electrochemical detection(ECD). Results : (1)(-)-3-PPP had dual actions depending on the doses: at 2mg/kg, it decreased and at 10mg/kg, increased extracellular dopamine concentrations ; (2) haloperidol at all doses increased dopamine levels with higher dose having a greater increase; and (3) pretreatment of (-)-3-PPP reduced the increase in dopamine levels elicited by acute treatment with haloperidol. Conclusions : These findings suggest that pretreatment of (-)-3-PPP in low dose could accelerate the onset of therapeutic effect of haloperidol by diminishing the haloperidol-induced dopamine release in the limbic system.

Centromere Repeat DNA Originated from Brassica rapa is Detected in the Centromere Region of Raphanus sativus Chromosomes

  • Fluorescence in situ hybridization (FISH) is a powerful tool for the detection of DNA sequences in the specific region of the chromosomes. As well as for the integrated physical mapping, FISH karyotype analysis has to be preceded. Karyotype of Raphanus sativus 'Wonkyo 10039' was analyzed by a dual-color FISH technique; using various repetitive DNA probes, including 5S rDNA, 45S rDNA, and centromere retrotransposon. The length of the somatic metaphase chromosome ranged from 1.35 to $2.06{\mu}m$ with a total length of $15.29{\mu}m$. The chromosome complements comprised of eight pairs of metacentrics and one pair of submetacentric. Bleached DAPI Band analysis revealed a heterochromatin region, covering 28.6% to 50.4% each chromosomes. 5S and 45S rDNA sequences were located on two and three pairs of chromosomes, respectively. The centromere retrotransposon of Brassica (CRB) is a major component in Brassica related species that has been maintained as a common centromere component. CRB signals were detected on the centromere and pericentromeric region of R. sativus 'Wonkyo 10039' and three basic Brassica species (B. rapa, B. nigra, and B. oleracea). These results will provide a valuable background for physical mapping and elucidation of the evolutionary relationship among the Brassica related species.

Microcantilever biosensor: sensing platform, surface characterization and multiscale modeling

  • The microcantilever (MCL) sensor is one of the most promising platforms for next-generation label-free biosensing applications. It outperforms conventional label-free detection methods in terms of portability and parallelization. In this paper, an overview of recent advances in our understanding of the coupling between biomolecular interactions and MCL responses is given. A dual compact optical MCL sensing platform was built to enable biosensing experiments both in gas-phase environments and in solutions. The thermal bimorph effect was found to be an effective nanomanipulator for the MCL platform calibration. The study of the alkanethiol self-assembly monolayer (SAM) chain length effect revealed that 1-octanethiol ($C_8H_{17}SH$) induced a larger deflection than that from 1-dodecanethiol ($C_{12}H_{25}SH$) in solutions. Using the clinically relevant biomarker C-reactive protein (CRP), we revealed that the analytical sensitivity of the MCL reached a diagnostic level of $1{\sim}500{\mu}g/ml$ within a 7% coefficient of variation. Using grazing incident x-ray diffractometer (GIXRD) analysis, we found that the gold surface was dominated by the (111) crystalline plane. Moreover, using X-ray photoelectron spectroscopy (XPS) analysis, we confirmed that the Au-S covalent bonds occurred in SAM adsorption whereas CRP molecular bindings occurred in protein analysis. First principles density functional theory (DFT) simulations were also used to examine biomolecular adsorption mechanisms. Multiscale modeling was then developed to connect the interactions at the molecular level with the MCL mechanical response. The alkanethiol SAM chain length effect in air was successfully predicted using the multiscale scheme.
