담수식물 근계로부터 분리된 Pseudomonas cepacia KH410 균주에 대한 납, 구리, 카드뮴의 영향 (Effects of Lead, Copper and Cadmium on Pseudomonas cepacia KH410 Isolated from Freshwater Plant Root)

  • 담수식물 수초의 근계 에 부착하는 미생물의 자정력을 탐색하기 위하여 분리된 세균인 Pseudomonas cepacia KH410 을 대상으로 중금속인 납과 구리, 카드뮴이온에 대한 영향을 조사하였다. 이 균의 최적 배지로 Nutrient기본배지를 사용하였고 최대 균체 생산을 위하여 yeast extract, soytone을 각각 1%씩 첨가하였고, NaCl은 0.5%, 초기 pH는 7.0 그리고 배양은 $28^{\circ}C$에서 24 시간 진탕 배양하였다. 이러한 배양 조건하에서 얻어진 최대 균체는 2.72 g DCW/ι-medium이었다. 각각의 100 mg/ι중금속 첨가에 따른 균체 생산은 납은 1.98 g/ι, 구리는 1.58g/ι, 카드뮴은 0.2 g/ι로 세가지 중금속 첨가에 따른 균체 생산이 낮아졌으며 중금속별로 균체 생산량에 차이를 나타내었다. 균의 생육에 영향을 미치는 각각의 중금속 농도를 비교하여 본 결과 최저생육저지농도 (MIC)가 납은 1.3 mM, 구리는 0.8 mM, 카드윰은 0.4 mM로써 카드윰이 균체에 미치는 영향이 가장 높게 나타났다. 동일 농도인 100 mg/ι용액 내에서의 세가지 중금속에 대한 균체의 변화는 균체끼리의 응집 현상이 광학현미경하에서 1 일에서 4 일 사이에 관찰되었고 전자현미경하에서는 각각의 중금속을 10분 노출시킨 후부터 세포벽 쪽으로 이온들이 이동되는 것이 관찰되었고 2 시간 후에는 세포외피, 원형질분리 및 세포내부로 침적된 양상이 각각 다르게 나타났으며 균체의 손상은 카드뮴이 가장 높았고 그 정도는 카드뮴>구리>납의 순이었다.

전분이용성 효모 Sporobolomyces holsaticus의 균체성분 분석에 대하여 (Analysis of Cellular Components of Starch-Utilizing Yeast Sporobolomyces holsaticus)

  • 미생물을 이용한 식량자원개발의 일환으로 분리배양한 전분이용성 효모, Sporobolomyces holsaticus FRI Y-5의 균체성분에 대하여 검토하였다. 효모균체의 일반조 성분으로서 총 탄수화물이 33.08%, 조단백질이 45.63%, 조지방이 20.01%, 회분이 3.04%였으며, 그 밖에 색소수출물이 4.46%였다. 완전세포(Whole cell)의 가용성단백질함량은 cold and hot NaOH extraction 시 40.89%였으며, 이때 조단백질과 가용성단백질 함량으로부터 핵산함량이 7.6%로 추정되었다. 균체단백질의 아미노산 분석결과, 표준단백질에 비하여 필수아미노산 중 threonine, valine, isoleucine이 약간 적었으며 그때 제한아미노산인 valine의 단백가는 89.4였다. 균체지방의 지방산 조성은 $C_{16:0}$이 6.5%였고, $C_{18:0}$이 2.4%, $C_{18:1}$이 81.9%, $C_{18:2}$이 3.2%, $C_{18:3}$이 6.0%였으며, 이 때 $C_{18:1}$이 특이하게 많았다. 또한 이 효모균체는 미량원소로서 Fe나 Ca에 비하여 Mg과 K을 많이 함유하고 있었으며, 각각 365.33mg/100g, 282.75mg/100g이었고, 비타민 중 $Vit.\;B_2$는 3.7mg/100g이나 niacin은 검출되지 않았다. 색소수출물의 흡광도 실험결과, carotenoid계 색소가 확인되었으며 자외선부분에서 태양광에 파괴되지 않는 물질을 발견하였다.

재조합 효모의 유가배양에서 Exoinulinase생산을 위한 Promoter의 선별 (Selection of Constitutive Promoter for Exoinulinase Production in Fed-Batch Culture of Recombinant Yeast)

  • Klyyveromyces marxianus exoinulinase를 Saccharomyces cerevisiae에서 구성적으로 과발현 생산하기 위해, 구성적 promoter인 GAPDH, ADH1, PGK 및 ENOI promoters 하류에 exoinulinase 유전자 (INUI)의 ORF를 in frame으로 연결한 각각의 plasmi에 YIGP, pADHI,-INU, pPGK-INU 및 pENO-INU 를 구축하였다. 이들 각 plasmid를함유한 형질전환주 4종을 포도당 농도 5% 배지에서 회분배양한 결과 균체증식은 promoter에 따라 큰 차이를 보이지 않았지만 exoinulinase 발현수준과 plasmid 안정성은 사용한 promoter 에 크게 좌우되었다. 즉 exoinulinase 발현수준은 GAPDH PGK ADH1 ENO1 promoter 각각 1.70, 1.67 1.29, 0.80 unit/ml 였으며 plasmid 안정성은 GAPDH promoter 계의 55%를 제외하고 모두 80%이상으로 높게 나타났다. 이상의 plasmid 안정성과 exoinulinae 발현수준을 고려하여 ADH1 및 PGK 발현계를 선정하여 유가배양하였다 Yeast extract와 포도당을 간헐적으로 공급한 유가배양 결과, 두 발현계에서 약 30 g-DCW/1의 균체농도를 얻었지만, ADHI promoter 계에서는 3.70 unit/ml 의 최대 exoinulinase 활성과 96%의 plasmid 안정성을 보여TRh 반면에 PGK promoter 계는 각각 2.70 unit/ml/와 80%를 나타내었다. 따라서 plasmid 안정성과 긴 배양시간을 고려할 때 비선택적 영양배지를 사용하는 고농도세포 유가배양에서 ADH1 promoter가 exoinulinase 의 구성적 과발현, 생산에 더 적합할 것으로 사료된다.

Production and Characterization of Multi-Polysaccharide Degrading Enzymes from Aspergillus aculeatus BCC199 for Saccharification of Agricultural Residues

  • Enzymatic hydrolysis of lignocellulosic biomass into fermentable sugars is a key step in the conversion of agricultural by-products to biofuels and value-added chemicals. Utilization of a robust microorganism for on-site production of biomass-degrading enzymes has gained increasing interest as an economical approach for supplying enzymes to biorefinery processes. In this study, production of multi-polysaccharide-degrading enzymes from Aspergillus aculeatus BCC199 by solid-state fermentation was improved through the statistical design approach. Among the operational parameters, yeast extract and soybean meal as well as the nonionic surfactant Tween 20 and initial pH were found as key parameters for maximizing production of cellulolytic and hemicellulolytic enzymes. Under the optimized condition, the production of FPase, endoglucanase, ${\beta}$-glucosidase, xylanase, and ${\beta}$-xylosidase was achieved at 23, 663, 88, 1,633, and 90 units/g of dry substrate, respectively. The multi-enzyme extract was highly efficient in the saccharification of alkaline-pretreated rice straw, corn cob, and corn stover. In comparison with commercial cellulase preparations, the BCC199 enzyme mixture was able to produce remarkable yields of glucose and xylose, as it contained higher relative activities of ${\beta}$-glucosidase and core hemicellulases (xylanase and ${\beta}$-xylosidase). These results suggested that the crude enzyme extract from A. aculeatus BCC199 possesses balanced cellulolytic and xylanolytic activities required for the efficient saccharification of lignocellulosic biomass feedstocks, and supplementation of external ${\beta}$-glucosidase or xylanase was dispensable. The work thus demonstrates the high potential of A. aculeatus BCC199 as a promising producer of lignocellulose-degrading enzymes for the biomass conversion industry.

항균성 물질을 생산하는 Lactobacillus amylovorus IMC-1의 배양학적 특성 (Cultural Characteristics of Lactobacillus amylovorus IMC-1 Producing Antibacterial Substance)

  • 발효식품의 제조에 유용한 starter 개발 및 probiotics로서 이용 가능성을 타진하기 위하여 항균성 물질을 생산하는 Lactobacillus amylovorus IMC-1 균주를 이용한 skim milk medium에서의 산생성, 단백질 분해활성, lactose 분해활성 및 유사구균과 혼합발효 그리고 병원성 세균에 대한 살균력 등을 검토하였다. L. amylovorus IMC-1은 10% skim milk에 yeast extract를 첨가하여 12시간 배양하였을 때 산도 0.8%이었고, 72시간째에는 2.7%로 우수한 산 생성력을 보였다. IMC-1 균주의 ${\beta}-galactosidase$ 활성은 균체 1 mg당 약 $39{\mu}M/minute$로 매우 강한 반면, $phospho-{\beta}-galactosidase$의 활성은 매우 낮았다. 또한 이 균주는 10% skim milk에서 배양 12시간째 $6\;{\mu}g/mL$, 72시간째에 $69\;{\mu}g/mL$의 유리 tyrosine을 생산하여 단백질 분해활성이 낮았다. 전기영동상의 band pattern으로부터 casein중 ${\alpha}-casein$을 주로 이용하며, ${\beta}-casein$은 거의 이용하지 못함을 알 수 있었다. 10% skim milk에서 IMC-1 균주 및 St. thermophilus NIAI 510의 단독 또는 혼합배양을 실시한 결과, 혼합배양, IMC-1 균주 및 St. thermophilus 순으로 산 생성력이 높았으며, 유산구균 단독배양 및 혼합배양에서는 시간이 경과함에 따라 치즈 향기가 증가하는 것을 확인할 수 있었다. 또한 10% skim milk medium에서 혼합배양에 의한 최적의 유산발효 시간은 산 생성력 및 공존 균수 등을 고려할 때, yeast extract 0.1% 첨가시에는 16시간, 0.5% 첨가시에는 12시간 배양하는 것이었다. IMC-1이 생산한 항균성 물질은 Escherichia coli O157을 배양 24시간째 2 log정도 감소시켰고, 48시간 배양 후에는 피검균이 검출되지 않았다. 또한 Shigella flexneri는 배양 4시간째 2 log이상 감소하였고, 6시간 배양 후에는 검출되지 않아 항균성 물질은 강한 살균작용을 나타냄을 알 수 있었다.

Effects of Dietary Synbiotics from Anaerobic Microflora on Growth Performance, Noxious Gas Emission and Fecal Pathogenic Bacteria Population in Weaning Pigs

  • Synbiotics is the term used for a mixture of probiotics (live microbial feed additives that beneficially affects the host animal) and prebiotics (non-digestible food ingredients that beneficially affect the organism). This study investigated the effect of probiotics from anaerobic microflora with prebiotics on growth performance, nutrient digestibility, noxious gas emission and fecal microbial population in weaning pigs. 150 pigs with an initial BW of 6.80${\pm}$0.32 kg (20 d of age) were randomly assigned to 5 dietary treatments as follows: i) US, basal diet+0.15% antibiotics (0.05% oxytetracycline 200 and 0.10% tiamulin 38 g), ii) BS, basal diet+0.2% synbiotics (probiotics from bacteria), iii) YS, basal diet+0.2% synbiotics (probiotics from yeast), iv) MS, basal diet+0.2% synbiotics (probiotics from mold), v) CS, basal diet+0.2% synbiotics (from compounds of bacteria, yeast and mold). The probiotics were contained in $10^{9}$ cfu/ml, $10^{5}$ cfu/ml and $10^{3}$ tfu/ml of bacteria, yeast and molds, respectively. The same prebiotics (mannan oligosaccharide, lactose, sodium acetate and ammonium citrate) was used for all the synbiotics. Pigs were housed individually for a 16-day experimental period. Growth performance showed no significant difference between antibiotic treatments and synbiotics-added treatments. The BS treatment showed higher (p<0.05) dry matter (DM) and nitrogen digestibility while ether extract and crude fiber digestibility were not affected by the dietary treatment. Also, the BS treatment decreased (p<0.05) fecal ammonia and amine gas emissions. Hydrogen sulfide concentration was also decreased (p<0.05) in BS, YS and MS treatments compared to other treatments. Moreover, all the synbioticsadded treatments increased fecal acetic acid concentration while the CS treatment had lower propionic acid concentration than the US treatment (p<0.05) gas emissions but decreased in fecal propionate gas emissions. Total fecal bacteria and Escherichia coli populations did not differ significantly among the treatments, while the Shigella counts were decreased (p<0.05) in synbiotics-included treatment. Fecal bacteria population was higher in the YS treatment than other treatments (p<0.05). The BS treatment had higher yeast concentration than YS, MS and CS treatments, while US treatment had higher mold concentrations than MS treatment (p<0.05). Therefore, the results of the present study suggest that synbiotics are as effective as antibiotics on growth performance, nutrient digestibility and fecal microflora composition in weaning pigs. Additionally, synbiotics from anaerobic microflora can decrease fecal noxious gas emission and synbiotics can substitute for antibiotics in weaning pigs.

Alcaligenes latus와 Comamonas acidovorans의 균체성장 및 Poly[3-hydroxybutyrate-co-4-hydroxybutyrate] 합성 특성 (Characteristics of Cell Growth and Poly[3-hydroxybutyrate-co-4-hydroxybutyrate] Synthesis by Alcaligenes latus and Comamonas acidovorans)

  • Characteristics of cell growth and poly(3-hydroxybutyrate-co-4-hydroxybutyrate) [P(3HB-co-4HB)] synthesis was investigated through flask and batch cultures of Alcaligenes latus and Comamonas acidovorans. The specific growth rate of C. acidovorans increased with yeast extract concentration and decreased with 1,4-butanediol concentration. Optimum glucose concentration for growth of C. acidovorans was 20 g/L. In one-step flask cultures of C. acidovorans, final dry cell weight and PHA content decreased with the ratio of 1,4-butanediol to glucose, while the 4HB fraction in copolymers gradually increased to 100 $mol\%$ with an initial 1,4-butanediol concentration of 20 g/L as single carbon source. The specific growth rate of A. latus decreased with v-butyrolactone concentration and optimum sucrose concentration for growth was 10 g/L. In batch cultures of A. latus, 4HB fraction increased with initial v-butyrolactone concentration. P(3HB-co-4HB) with 19 $mol\%$ 4HB was obtained when the initial ratio of v-butyloractone (g/L) to sucrose (g/L) was 10 : 10.

Production of Indole-3-Acetic Acid by Enterobacter sp. DMKU-RP206 Using Sweet Whey as a Low-Cost Feed Stock

  • In this study, we investigated Indole-3-acetic acid (IAA) production by a rice phylloplane bacteria, Enterobacter sp. DMKU-RP206, using sweet whey as a feed stock instead of lactose. We succeeded in using sweet whey for Enterobacter sp. DMKU-RP206 to produce 3,963.0 mg IAA/l with the optimal medium containing 1.48% sweet whey, 1.42% yeast extract and 0.88% $\text\tiny{L}$-tryptophan. The medium pH was adjusted to 6 and the culture conditions were shaking at 200 rpm on an orbital shaker at $30^{\circ}C$ for 3 days. We also evaluated the effect of IAA in culture filtrates of Enterobacter sp. DMKU-RP206 on the promotion of jasmine rice growth in a pot experiment. Compared with the negative control (without IAA), the result showed that biosynthetic IAA produced by Enterobacter sp. DMKU-RP206 significantly increased the growth of jasmine rice (Oryza sativa L. cv. KDML105) in terms of length and dry weight of shoot. This work thus reveals the impact of IAA produced by Enterobacter sp. on the promotion of jasmine rice growth.

Inactivation of Salmonella on Eggshells by Chlorine Dioxide Gas

  • Microbiological contamination of eggs should be prevented in the poultry industry, as poultry is one of the major reservoirs of human Salmonella. ClO2 gas has been reported to be an effective disinfectant in various industry fields, particularly the food industry. The aims of this study were to evaluate the antimicrobial effect of chlorine dioxide gas on two strains of Salmonella inoculated onto eggshells under various experimental conditions including concentrations, contact time, humidity, and percentage organic matter. As a result, it was shown that chlorine dioxide gas under wet conditions was more effective in inactivating Salmonella Enteritidis and Salmonella Gallinarum compared to that under dry conditions independently of the presence of organic matter (yeast extract). Under wet conditions, a greater than 4 log reduction in bacterial populations was achieved after 30 min of exposure to ClO2 each at 20 ppm, 40 ppm, and 80 ppm against S. Enteritidis; 40 ppm and 80 ppm against S. Gallinarum. These results suggest that chlorine dioxide gas is an effective agent for controlling Salmonella, the most prevalent contaminant in the egg industry.

Inhibition of Aflatoxin Production of Aspergillus flavus by Lactobacillus casei

  • Lactobacillus casei KC-324 was tested for its ability to inhibit aflatoxin production and mycelial growth of Aspergillus flavus ATCC 15517 in liquid culture. flatoxin $B_{1}$ biosynthesis and mycelial growth were inhibited in both simultaneous culture and individual antagonism assays, suggesting that the inhibitory activity was due to extracellular metabolites. produced in cell-free supernatant fluids of the cultured broth of L. casei KC-324. In cell-free supernatant fluids of all media tested, deMan, Rogosa and Sharpe broth, potato dextrose broth, and Czapek-Dox broth+1% yeast extract showed higher antiaflatoxigenic activity. In these case, fungal growths, however, was not affected as measured by mycelial dry weight. The antiaflatoxigenic metabolites from L. casei KC-324 were produced over wide range of temperatures between $25^{\circ}C$ and $37^{\circ}C$. However, these metabolites were not thermostable since the inhibitory activity of the supernatant was inactivated within 30 minutes at $100^{\circ}C$ and $121^{\circ}C$. The inhibitory activity was not influenced by changing pH of supernatant between 4 and 10. However, the antiaflatoxigenic activity was slightly reduced at pH 10.
