  • Dry hams and sausages we re processed manually under the environmental conditions in Seoul, Korea, by addition with $NaNO_2$ - equivalent of 264ppm and 185ppm, respectively. The residual nitrites levels throughout the processing and storage periods were 32-67 ppm which are below the maximum legal allowance of 70ppm . The nitrites showed the highest levels at 2- weeks processing of dry sausages and at 5-weeks processing of dry hams, and the levels were gradually decreased thereafter. The residual nitrites in sausages dried in intestinal cases were lower and dropped below 10ppm after 3-weeks processing. The dry hams processed in summer also contained lower levels of nitrites, below 10ppm after 4-weeks processing. The levels of residual nitrates, however, showed highest when that of nitrites were lowest and were increased gradually with the duration of storage.

  • Supercritical carbondioxide is very effective in removing oils from a variety of seed matrices, devoid of any appreciable amount of phospholipid content. However, the limited solubility of phosphalipids in supercritical carbondioxide leaves behind a potentially valuable by-product in spent seed matrix. Any phospholipid extraction process from the spent matrix must maintain the structure and the quality of phospholipids and must be compatible with the end use of the seed protein meal an animal feed or for human consumption. An initial supercritical carbondioxide extraction of soybean flakes was performed at 32 MPa and $80^{\circ}C$ to extract the oils, leaving the phospholipids in the deflatted soybean flakes, A second step was performed on the defatted soybean flake using $X_{eth}$=0.10, Varying the pressure from 175 MPa to 70.6 MPa and temperature from $60^{\circ}C$ to $80^{\circ}C$. For all supercritical carbon dioxide/ethanol mixture extractions, a fraction rich in phospholipids was obtained. The fractions extracted from defatted soybean flakes were dried and them redissolved in chloroform before HPLC-ELSD analysis. Quantitative and qualitative analysis of phospholipids on soybean seeds, defatted soybean flake, and different extracted phospholipid fractions was carried out, to ascertain the effect of extraction pressure and temperature.

  • Background: This study was conducted to compare the physicochemical properties of 12 different cultivars of Rehmannia glutinosa, a plant used in Korean traditional herbal medicine. Method and Results: We investigated the following 12 cultivars of R. glutinosa; Jihwang1, Goryeo, Daegyung, Gogang, Togang, Dagang, Wongang, Yeongang, Hwanggang, Dahwang, Segang and Chunggang. We measured water content, chromaticity, hardness, active components levels, and antioxidant activity. Our results showed that Togang has the lowest water content and the highest yield. The catalpol content was the highest in Togang (40.50 mg/g) and the lowest in Chunggang (22.17 mg/g). The aucubin content was the highest in Hwanggang (0.28 mg/g). Chunggang had the lowest aucubin content, but the highest level of verbascoside. Total flavonoid and total polyphenol were the highest in Daegyung and Chunggang, respectively. There was a strong correlation between the total flavonoid and total polyphenol (r = 0.89) content. Conclusions: There were significant differences in physicochemical properties among 12 cultivars. These properties will affect suitability for processing and final product quality. We therefore suggest the selection of appropriate cultivars for different uses.

  • Objectives : The extract of Sagunja-tang has been traditionally used for restorative treatment of constitutional weakness, vascular and immune disorder, and nervous disease in Oriental country. This study investigated the regulatory effects of Sagunja-tang on the expression, production, and activity of immune mediators. Methods : In this study, the extract of Sagunja-tang was prepared by extracting with distilled water at 100$^{\circ}C$ for 2.5h. The extract was freeze-dried following filtration through 0.45${{\mu}m}$ filter. The extract was dissolved in Hank's balanced salt solution (HBSS) and filtered again through 0.45${{\mu}m}$ filter before use. The level of nitrite, an oxidative product of nitric oxide(NO) was measured in the culture medium by the Griess reaction. The levels of prostaglandin E2(PGE2), Th1 cytokines (IFN-${\gamma}$, IL-2) and Th2 cytokines(IL-4, IL-5, IL-13) were measured by enzyme-linked immunosorbent assay and the protein levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression were determined by Western blot analysis. Also examined the effects of the extract on T-cell proliferation and cytotoxic activity of natural killer cells. Results : In this investigation, Production levels of Th2 cytokines (IL-4, IL-5, IL-13) was inhibited in a dose dependent manner by treatment with the extract. I also found that the extract increased T-cell proliferation and cytotoxic activity of natural killer cells in a dose-dependent manner.Conculsions : These results suggest that the water extract of Sagunja-tang may be useful for a therapeutic drug against a sickly constitution and immune diseases, probably by regulating the production of immune mediators.

  • Sophora flavescens Ait. (Fabaceae) is a medicinal plant widely founded in Northeast Asia, and its dried root (Kushen) has been used as a traditional Chinese herbal medicine. The therapeutic effects of Kushen in micturition disorder was not investigated comprehensively yet. In the present study, we examined and compared the efficacy of three batches of Kushen extract using different ethanol content through an in vitro cell-based assay. Among them, we chose the batch with the highest efficacy and augmented the volume of extract for industrial purpose. The bulk extract was examined in its efficacy in the in vitro cell-based assay, and the therapeutic effects through an in vivo behavioral assay of OAB rats. The main components of the extracts were analyzed by liquid chromatography. Cytotoxicity of the extracts was investigated by MTT assay. The overall efficacy of the extract was as much as, or more than, kurarinone, a potent BKCa channel activator. Thus, the extract was a potent relaxant of urinary smooth muscle by upregulating the activity of BKCa channel. The Kushen extract could be explored as an alternative medicine against overactive bladder patients indicating severe dysfunction of BKCa channel.

  • Background: Ginsenosides with less sugar moieties may exhibit the better adsorptive capacity and more pharmacological activities. Methods: An efficient method for the separation of four minor saponins, including gypenoside XVII, notoginsenoside Fe, ginsenoside Rd2, and notoginsenoside Fd, from Panax notoginseng leaves (PNL) was established using biotransformation, macroporous resins, and subsequent preparative high-performance liquid chromatography. Results: The dried PNL powder was immersed in the distilled water at $50^{\circ}C$ for 30 min for converting the major saponins, ginsenosides Rb1, Rc, Rb2, and Rb3, to minor saponins, gypenoside XVII, notoginsenoside Fe, ginsenoside Rd2, and notoginsenoside Fd, respectively, by the enzymes present in PNL. The adsorption characteristics of these minor saponins on five types of macroporous resins, D-101, DA-201, DM-301, X-5, and S-8, were evaluated and compared. Among them, D-101 was selected due to the best adsorption and desorption properties. Under the optimized conditions, the fraction containing the four target saponins was separated by D-101 resin. Subsequently, the target minor saponins were individually separated and purified by preparative high-performance liquid chromatography with a reversed-phase column. Conclusion: Our study provides a simple and efficient method for the preparation of these four minor saponins from PNL, which will be potential for industrial applications.

  • Objective: This study aimed to characterize the effect of pre-treated black garlic (BG) extracts addition into retorted Korean ginseng chicken soup (Samgyetang) on the fatty acid composition and flavour-related indexes. Methods: Four different treatments; Samgyetang made with a 5% (w/w) addition of garlic (G), fresh BG (FBG), oven-dried BG (DBG), or encapsulated BG (EBG) extracts were developed and compared to negative control (NC) without any extract addition. Prepared samples were cooked via retorting at 121.1℃, 1.5 kgf/cm2 for 1 h. Results: The BG treated samples were higher in C18:3n3 and C18:2n6 fatty acids, with thrombogenic index was 18% to 20% lower than the NC. EBG yielded the highest umami-related nucleotides (5'-guanosine monophosphate and 5'-inosine monophosphate) and modified some free amino acid (alyne, phenylalanine and leucine) thus possessed the highest equivalent umami concentration among samples. Some individual aldehydes (pentanal, hexanal, and heptanal) were lower, while furans and volatile sulfur compounds were higher than the NC and G treatment group, indicating a potential suppression of unpleasant flavour alongwith the intensificiation of favourable flavour from the addition of BG extracts into retorted Samgyetang. Conclusion: Taken together, the synergistic results of this study indicate that incorportating suitable pre-treatment of BG extract could be of critical importance for the development of the retorted Samgyetang with improved flavour and functionalities.

  • We previously reported that dried Lysimachia christinae whole plant extract exerted significant neuroprotective activity. In this study, we tried to isolate neuroprotective compounds of L. christinae. We evaluated the neuroprotective activity of the four fractions (hexane, chloroform, ethyl acetate, and n-butanol fractions) of methanol extract. Among them, ethyl acetate and n-butanol fractions showed most potent neuroprotective activity against glutamate excitotoxicity. Nine compounds were isolated from ethyl acetate and n-butanol fractions of L. christinae extract and identified as cynaroside (1), (3,4,5,6-tetrahydroxytetrahydro-2H-pyran-2-yl)methyl-3-hydroxy-2-octyldopentaconta-23,33-dienoate (2), androst-16-ene-3,6-diol (3), 2-hydroxy-24-propoxytetracos-4-enoic acid (4), 2-hydroxy-24-methoxytetracos-4-enoic acid (5), 12-(stearoyloxy)octadec-9-enoic acid (6), β-sitosterol (7), (E)-4-(3,4-dimethoxyphenyl)but-3-en-1-yl palmitate (8) and (1S,2S,3R,4R)-4-(((2S,3R,4R,5R,6S)-2-(((2R,3R,4S,5R,6R)-2-(3,4-dimethoxyphenethoxy)-3,5-dihydroxy-6-(hydroxymethyl) tetrahydro-2H-pyran-4-yl)oxy)-4,5-dihydroxy-6-methyltetrahydro-2H-pyran-3-yl)oxy)cyclohexane-1,2,3-triol (9) by spectroscopic data such as UV, IR, NMR, Mass spectroscopy. Their neuroprotective activity was evaluated by MTT assay. Cynaroside (1) and androst-16-ene-3,6-diol (3) had significant neuroprotective activity against glutamate-injured HT22 cells. The neuroprotective efficacy of cynaroside (1) and androst-16-ene-3,6-diol (3) was related to their anti-oxidative activity.
