Characteristics of Haploid Plants Derived from Interspecific Cross between Cytoplasmic Male-sterile Tobacco $F_1$(Nicotiana tabacum) and Nicotiana africana (연초(Nicotiana tabacum L.) 새포질 웅성불임 $F_1$과 Nicotiana africana의 종간 교배에 의한 반수체 식물의 특성)

  • This study was conducted to investigate the utility and agronomic characteristics and use of cytoplasmic. male-sterile (CMS) haploid plants derived from interspecific cross between (male-sterile NC82$\times$burley21) F$_1$ plant (Nicotiana tabacum L.) and Nicotiana africana. Abundant seeds of high germinability were obtained when Nicotiana tabacum (cytoplasmic male-sterile F$_1$ plants) is pollinated by Nicotiana africana. Most of seedlings died at the cotyledonary stage. The remaining seedlings are viable F$_1$ hybrids or maternal haploids that can be easily distinguished. Number of interspecific Fl hybrids and matermal haploids per capsule obtained from the interspecific cross between cytoplasmic male-sterile tobacco F$_1$ plants and N.africana yielded 2.2 and 0.5 plants, respectively. Out of 149 CMS haploid plants obtained from interspecific cross, 102 plants showed green type while the others were yellow type for leaf and stem. This results agreed with the genetic ratios expected among haploid plants from the F$_1$ hybrids heterozygous for two recessive genes of yellow color of burley tobacco plant. Out of 83 CMS haploid plants inoculated with TMV, 48 plants showed resistant, while the others was susceptible. It agreed with the expected genetic ratios for a single dominant gene of TMV resistance. CMS haploid plant will be useful as a source material for breeding of CMS doubled haploid lines

Changes of Chloroplast Number per Guard Cell pairs of Leaves by Ploidy Level in Nicotiana tabacum L. cv. BY-4 (담배 식물체[Nicotiana tabacum L. cv. BY-4]의 배수성에 따른 공변세포의 엽록체 수 변화)

  • We evaluated a possibility of the use of chloroplast number per guard cell pairs as a measure for ploidy level in the different ploidy levels of tobacco plant (Nicotiana tabacum L. cv. BY-4) . The guard-cell chloroplast numbers of leaves of haploid plant were a half of wild-type plant. Furthermore, the number of chloroplast per guard cell pairs of the leaves of doubled-haploid plant increased in two times compared with that of haploid plant. In addition, the chloroplast number was not changed in the F$_1$ progenies. The change of the chloroplast number by leaf stage was not observed. The results indicate that there is a strong relationship between ploidy level (2x and 4x) and chloroplast number per guard cell pairs. This relationship was also, observed in both in vitro and pot cultured plants. It was determined that the measurement of chloroplast number in guard cells of leaf epidermis is simple to use and less labour intensive, and hence can be considered a practical alternative to the chromosome counting methods or flow cytometry in the tobacco plant.

Development of Clubroot Resistant Doubled-Haploid Inbred Lines in Kimchi Cabbage (Chinese Cabbage) (Brassica rapa L.)

  • Kimchi cabbage (Chinese cabbage), radish and Cabbage are major Brassicaceae vegetables in Korea. Especially, we can easily develop whole plant from one microspore in Kimchi cabbage. To develop clubroot resistant doubled-haploid (DH) inbred lines, we pollinated a clubroot resistant turnip of 'IT 033820' with a Kimchi cabbage (Chinese cabbage) inbred of 'BP 079'. More than 85 DH inbred lines were developed from this combination. We screened about 400 materials including these DH inbred lines, commercial cultivars and breeding materials during 3 years using hydroponic system after inoculating single spore isolation race 4(SSI-04) inoculate. One inbred line derived from this combination selected as clubroot resistant and registered as 'Wonkyo20036ho'. We inoculated 26 DH inbred lines derived from 'Zoong-baek 2ho' using SSI-4, the percent of resistant plants varied from 0 to 83%. However the horticultural traits of highly resistant DH inbred line was poor. Thus we selected one DH line showing 77% resistant with yellow inner leaf and maid good head, was registered as 'Wonkyo20034ho'. Another DH inbred line derived from Korean variety of 'Wol-dong' showing 86% resistant was registered as 'Wonkyo20037ho'. Other DH inbred lines were derived from Chinese cultivar of 'Choon-hi-go-hang-wang' and 'Hwang-shim-zo48' showed 80 and 71% resistant, respectively, was also selected for registration. Even though DH inbred lines derived from turnip showed highly resistant to SSI-04 and provincial inoculate, they showed poor characteristics in horticultural traits. However, commercial seed companies showed interesting for adapting these DH inbred lines in commercial breeding.

Effects of Plant Growth Regulators on Haploid Production of Wheat (밀 반수체 생산에서 식물생장조절제의 영향)

  • The introduction of doubled haploid (DH) approach into breeding programs has reduced the times and population sizes required for the production of pure lines. We carried out the experiment for development on effective method of producing haploid in wheat. Emasculated spikelets of wheat were pollinated with maize pollen and cultured in the solution containing 40 g/L sucrose and 2,4-D, NAA, 2,4,5-T and dicamba 24 h after pollination, and then incubated until embryo rescue. Fourteen to sixteen days after pollination, the embryos are excised and cultured in half-strength MS basal medium supplemented with 20 g/L sucrose and 1 mg/L NAA. The type of plant growth regulators was found to be most significant in production of haploid plants. The application of synthetic auxins to pollinated florets, stimulates haploid embryo development to a stage where the embryos can be rescued onto nutrient media. The percentage of seeded florets was significantly affected by 100 mg/L 2,4-D, 150 mg/L 2,4,5-T and 50 mg/L dicamba. The percentage of embryos formed was significantly increased by treatment with 2,4-D and 2,4,5-T at 100 mg/L, and dicamba at 50 mg/L, but the treatments with 150 mg/L 2,4-D inhibited embryo development and plant regeneration. The optimum application time of plant growth regulators was 24 hrs after pollination.

QTL Analysis of Protein Content in Double-haploid Lines of Rice

  • The objective of this study was to characterize the main-effect QTLs, epistatic QTLs and QTL-by-environment interactions (QE), which are involved in the control of protein content. A population of 120 doubled haploid (DH) lines derived from a cross between 'Samgang' and 'Nagdong', was planted and determined for protein content over three years. Based on the population and a genetic linkage map of 172 markers, QTL analysis was conducted by WinQTLcart 2.5 and QTLMAPPER. Three main-effect QTLs affecting protein content of brown rice were detected from 2004 to 2006 on chromosomes 1 and 11. The qPC11.2 was repeatedly detected across two years. Seven pairs of epistatic loci were identified on eight chromosomes for protein content and collectively explained 39.15% of phenotype variation. These results suggest that epistatic effects might be an even more important component of the genetic basis for protein content and that the segregation of the DH lines for protein content could be largely explained by a few main-effect QTLs and many epistatic loci.

Inheritance of Herbicide (glufosinate) Resistance in Transgenic Rice Plant through Anther Culture

  • Haploid system by anther culture allows the development of homozygous lines when doubled. The response of anther culture to Basta (glufosinate) resistance was investigated on transgenic plants (cv. Anjungbyeo) in order to identify inheritance of bar gene associated with Basta. Most of the regenerated transgenic plants were sterile, and only a few plants produced viable seeds ($A_1$) in the greenhouse. The bar gene was analysis by PCR in basta resistant transgenic plant ($TA_0$). The transgenic seeds ($A_1$) were significantly germinated in Basta solution compared with non-transformed seeds. As a result of anther culture, in regenerated haploid plants, segregation ratio was 1:1 in five of eight cross combinations. In diploid plants, segregation ratio was 1:1 in seven of eight cross combinations. Although there was some differences in the cross combinations, most of the combinations had 1:1 segregation ratio which supports the theory. The difference may be a result of the small sample size or the difference of anther culture response caused by genotypic difference. Hence, when many cross combinations were anther-cultured the results would support the theory.

Development of Black Shank Resistant Burley Tobacco Germplasm, KB 104 and KB 106, and Their Characteristics (버어리종 역병저항성 계통 KB 104와 KB 106의 육성경과 및 특성)

  • Black shank(Phytophthora parasitira roar. nicotianae) resistant burley tobacco (Nicotiana tabacum L.) germplasms, KB 104 and KB 106, were developed by Korea Ginseng & Tobacco Research Institute. KB 104 was developed from the single cross of Burley 21$\times$Newton 77, using a modified pedigree method. KB 104 was highly resistant to black shank, and its agronomic characteristics and chemical contents were comparable to those of Burley 21, and value per 10a was slightly higher than Burley 21, KB 106 is a maternally derived doubled haploid made by N. africana method from the single cross of Burley 21$\times$ Va 509. KB 106 was also highly resistant to black shank, had two more harvestable leaves per plant and flowered three days later than Burley 21 did. Total alkaloid and nicotine contents of KB 106 were significantly lower than those of Burley 21. But its nornicotine content was higher than Burley 21 5. Production of doubled haploid population derived from the microspore culture of rapeseed (Brassica napus L.) F1 generation and analysis of fatty acid composition (유채 잡종 1세대의 소포자 배양에 의한 배가반수체 집단 선발 및 지방산 조성 분석)

  • Brassica napus, an oil crop that produces rapeseed oil, is an allotetraploid (AACC, 2n = 38) produced by natural hybridization between B. rapa and B. oleracea. In this study, microspore was cultured using the F1 developed from a cross between 'EMS26' line with high oleic acid content and 'J8634-B-30' lines. The flower bud size showing the nuclear development at the late uninucleate and binucleate stage with high embryogenesis rate was 2.6 ~ 3.5 mm. Microspores were cultured using only this size and after then most microspore embryo developed into secondary embryos and then regeneration plants obtained from the developed multilobe. The analysis of the ploidy of the plants revealed that 66.7% and 27.8% of the total lines were tetraploids and octoploids, respectively. The sizes of stomatal cells in tetraploids, octoploids, and diploids were 25.5, 35.6, and 19.9 ㎛, respectively, indicating that ploidy level was positively correlated with cell size. Furthermore, 62 tetraploid doubled haploid (DH) lines were selected. The average oleic acid (C18:1) and linolenic acid (C18:3) concentrations of DH were 72.3% and 6.2%, respectively. Oleic acid and linolenic acid concentrations exceeded the two parental values in 5 and 14 DH lines, respectively, suggesting that these two fatty acids had transgressive segregation. Therefore, the DH population can be utilized for the biosynthesis of unsaturated fatty acids in rapeseed and related genes. It can also be used as a breeding material for varieties with high oleic acid concentrations.

Genetic Mapping of QTLs that Control Grain Characteristics in Rice (Oryza sativa L.) (벼의 낱알 특성에 관여하는 양적형질유전자좌 분석)

  • We performed a molecular marker-based analysis of quantitative trait loci (QTLs) for traits that determine the quality of the appearance of grains, using 120 doubled-haploid (DH) lines developed by another culture from the F1 cross between ‘Cheongcheong’ (Oryza sativa L. ssp. Indica) and ‘Nagdong’ (Oryza sativa L. ssp. Japonica). The traits studied included length, width, and thickness of the grains, as well as length-to-width ratio and 1,000-grain weight. The objective of this study was to determine the genetic control of these traits in order to formulate a strategy for improving the appearance of this hybrid. Within the DH population, five traits exhibited wide variation, with mean values occurring within the range of the two parents. Three QTLs were identified for grain length on chromosomes 2, 5, and 7. Three QTLs were mapped for grain width on chromosome 2: qGW2-1, qGW2-2, and qGW2-3. Six chromosomes were identified for the grain length-to-width ratio; four of these were on chromosome 2, whereas the other two were on chromosomes 7 and 12. One QTL influencing 1,000-grain weight was identified and located on chromosome 8. The results presented in the present study should facilitate rice-breeding, especially for improved hybrid-rice quality.
