방사능탐지용 CAYS 함침 이중구조 폴리설폰막의 형상 및 특성에 제막공정의 습도가 미치는 영향 (Vapor Exposure Effect of a Casting Solution on the Embedding and Radioactive Detection of CAYS in Double-layered Polysulffne Film)

  • 방사능 오염도 측정에 사용하기 위한 이중구조 고분자막이 폴리설폰과 세륨활성화된 이트륨실리케이트(CAYS)를 이용하여 제조되었다. 제조된 막은 순수 고밀도 고분자 지지층과 이에 제막된 고분자 용액의 상전환 공정에 의해 고형화된 CAYS 함침 활성층의 이중구조로 구성된다. 제막공정에서 대기방치 공정이 생략되었을 때 CAYS를 포함하는 활성층은 전형적인 비대칭 구조를 지니며, CAYS 입자들이 고분자 구조 사이에 박혀있는 형상을 지닌다. 제막공정에서 대기에 방치하는 시간이 증가할수록 막의 형상은 스폰지 구조를 띠며 CAYS는 고분자 구조로부터 분리되어 막 내부에 셀 같은 공간에 밀집되어 존재함을 보였다. 한편, 두 충 사의 계면형상은 고분자 고형화 과정에서의 상전환 속도와 밀접한 관련되었으며, 대기방치 시간의 증가에 따라 계면의 구분이 뚜렷하게 나타나지 않았다. 방사능 탐지 특성에서 스폰지 구조를 지니는 막의 고분자 구조는 방사성핵종이 통과할 수 없는 밀집된 형상을 지니면서 탐지효율의 감소를 초래하는 것으로 나타났다.

Mechanism of guanine-specific DNA damage by UVA and its role in photocarcinogenesis and photoaging

  • Solar UV light is a well-known carcinogen. UVA radiation is probably carcinogenic to humans. In addition, recent investigations point to the importance of UVA irradiation in the photoaging. We investigated the mechanism of sequence- specific DNA damage using $\^$32/P-Iabeled DNA fragments in relation to carcinogenesis and aging. Furthermore, we investigated whether UVA accelerates the telomere shortening in human WI-38 fibroblasts. The exposure of double- stranded DNA fragments to 365 nm light in the presence of endogenous sensitizers produced sequence-specific cleavage at the 5' site of 5'-GG-3' and 5'-GGG-3' sequences. In addition, HPLC analysis revealed that sensitizers plus 365 nm light increased the 8-oxodG content of double-stranded DNA. We discuss the mechanisms of guanine-specific DNA damagecaused by excited photosensitizers in relation to carcinogenesis and aging.

  • Three-dimensional natural convection from a discrete flush-mounted circular heat source on the bottom of a cubic enclosure was studied by using a holographic interferometric tomography. The heat source was located at the off-center of the bottom plate so that three-dimensional temperature field can be achieved. A set of multidirectional holographic interferogram was recorded by employing a double-reference beam, double-exposure holographic technique in order to eventually reconstruct the temperature fields. The recorded interferometric data appear good enough to be further processed to extract optical pathlength data from them and finally reconstruct the temperature fields. A complete analysis of the temperature fields including the field reconstructions and comparison with thermocouple measurements is underway and will be reported shortly.

  • In this study, a practical holographic interferometric tomography system, which is instantaneous and non-contact for measuring three dimensional flow field, was developed. The system consists of holographic recording/reconstruction system, fringe analysis code and computational tomography code and it is developed with Gill environment for general users. The developed system was applied to three-dimensional natural convection from a discrete flush-mounted circular heat source on the bottom of a cubic enclosure. The heat source was located at the off-center of the bottom plate so that three-dimensional temperature field can be achieved. A set of multi-directional holographic interferograms was recorded by employing a double-reference beam, double-exposure holographic technique. Optical pathlength data were extracted from the recorded interferometric data and finally three dimensional temperature field inside the cube was reconstructed.

Electron Spin Resonance and Electron Nuclear Double Resonance Studies on the Photoinduced Charge Separation of N-Methylphenothiazine in Phenyltriethoxysilane, Vinyltriethoxysilane and Methyltriethoxysilane Gel Matrices

  • The photoproduced cation radical of N-methylphenothiazine doped in the different kind of matrices of phenyltriethoxysilane (PhiTEOS), vinyltriethoxysilane (VTEOS), and methyloiethoxysilane (METOS) was comparatively studied with electron spin resonance (ESR) and electron nuclear double resonance (ENDOR). The photoinduced charge separation efficiency was determined by integration of ESR spectra which correspond to the amount of photoproduced cation radical in the matrices. This was correlatively studied with the polarity and pore size of the gel matrices. The polarity of the matrices was comparatively determined by measuring λ$\sub$max/ values of PC$_1$ in the different matrices. The relative pore size among the matrices was determined by measuring relative proton matrix ENDOR line widths of the photoproduced cation radical of PCI. The decay kinetic constants of the cation radical of PCI in the different matrices was relatively studied with fitting the biexponential decay curves after exposure into the ambient condition. This is correlatively interpreted with the polarity and pore size of the matrices.

  • This study investigate the hydration heat history with variation of surface insulating material in cold weather concreting. According to the results, the temperature of concrete lowers below zero in 24hours, so early frost damage occurs in the case of exposure and 1 fold bubble sheet, but the lowest temperature keeps above zero, so a adiabatic effect is very favorable in the case of double bubble sheet and 부직포. Compressive strength of core specimen at 7 and 28 days is highest In the case of double bubble sheet and 부직포. But, considering convenience of construction and economical efficiency, it is thought that the most effective surface insulating material is 1 fold bubble sheet +blanket.

Double Labeling of Binding Sites in Cellulosic Substrates Using Endo- and Exoglucanase-Gold Complexes

  • Thin sections of cellulose fibers were incubated with an endo- and an exoglucanase labeled with gold particles of differing sizes. The hydrolytic sites were then visualized under transmission electron microscopy (TEM). The potential interaction between the ${\beta}$-1, 4-glucan substrates and the endo- and the exoglucanases was investigated using cellulosic and lignocellulosic substrates. The simultaneous visualization was very successful in distinguishing preferred substrates for each cellulase in lignocellulosic substrates. When plant lignocellulose was preincubated with endocellulase, density of the gold labeling greatly increased suggesting that preliminary exposure of lignocellulosic material to endocellulase may have enhanced the accessibility of the substrate to endocellulase and exocellulase. This result provided a plausible explanation for the observed endo/exo cellulase co-hydrolysis.

  • Two-dimensionally arrayed nanocolumn lattices were fabricated by using double-exposure laser holographic method. The hexagonal lattice was formed by rotating the sample with 60 degree while the square lattice by 90 degree before the second laser-exposure. The reactive ion etching for a typical time of 30 min using CH$_4$/H$_2$ plasma enhanced the aspect-ratio by more than 1.5 with a slight increase of the bottom width of columns. The etch-damage was observed by photoluminescence (PL) spectroscopy which was removed by the wet chemical etching using HBr/$H_2O$$_2$/$H_2O$ solution, leading into the enhanced PL intensities of the PCs.

  • In this paper, the temperature history and the strength development of concrete corresponded to various bubble sheets layer and curing temperature. Based on the results, In case of the test temperature of -5℃, concrete subject in the exposure condition, result in a frost damage at initial stage by a fall of below zero temperature. In case of the combination of PE film and non woven fabric was after 36 hour, and combination of bubble sheet over double, a tremendous insulating effect of bubble sheet over double is confirmed due to the temperature of concrete fall of below zero temperature after 60 hours. Meanwhile, regarding the -15℃ of temperature, special measure for insulation curing is necessary to secure stability against early frost damage because frost damage was not affected by the lapping thickness of bubble sheet subjected to severe cold weather condition.

Metal Effects of Urban Air Particulates on Cytokine Production and DNA Damage

  • Epidemiologic studies have demonstrated an association between short-term exposure to particulate air pollutants and increased mortality. However the biological mechanism underlying these associations have not been fully established and also the chemical and physical characteristics of the pollutant particles are not well understood. The metal constituents of air pollutant particles and their bioavailability are considered to Play an important role as possible mediators of Particle-induced airway injury and inflammation. Sprague-Dawley rat alveolar macrophage cells (NR8383) were exposed to airborne and acid-leached particulate matter (PM). Titanium oxide and nickel subsulfide were used as negative and positive controls. Particle-induced reactive oxygen species formation in cells was detected using the fluorescent probe 2',7'-dichlorofluorescin diacetate. Expression of TNF-$\alpha$ and IL-6 were measured by enzyme-linked immunosorbent assay, and PM-induced DNA double-strand breaks were determined with $\lambda$DNA/Hind III marker. Metals associated with air pollutant particles mediated intracellular oxidant production in alveolar macrophages, and the cytotoxicity and proinflammatory cytokine production induced by PM were associated with oxidative stress. The oxidants produced by air pollutant particles also are likely to induce DNA double-strand breaks. Our findings in alveolar macrophage cells exposed to PM and acid-leached PM support the hypothesis that metal components in urban air pollutants and their bioavailabilities might play an Important role in the induction of the adverse health effects.
