장 평활근의 수축성에 대한 berberine의 효과 (Effect of berberine on intestinal contractility)

  • Berberine $(10^{-7}-10^5M)$ increased the contractility dose-dependently in isolated rabbit ileal and jejunal segments. Atropine and hemicholinium abolished this response but not mecamylamine. Berberine$(10^{-8}-10^5M)$ enhanced the transmurally-stimulated(80 V, 0.5 ms, 0.05 Hz) twitch response in the isolated guinea-pig ileal segments. Atropine and hemicholinium also abolished this response but not mecamylamine. Effect of KCI, carbachol and histamine were not affected by pretreatment with berberine$(10^{-5}M)$. The results of our study suggest that berberine increases the intestinal contractility by increasing a small amount of acetylcholine release from the postganglionic parasympathetic nerve terminal but not by a direct activation of muscarinic receptors.

황기 부탄올 분획물이 생쥐의 체액성 면역기능에 미치는 영향 (Effects of the Butanol Fraction of Astragali Radix on the Humoral Immune Function in Mice)

  • Effect of the butanol fraction of Astragali Radix (BFAR) on the humoral immune response were investigated in ICR mice. Mice were divided into 4 groups and BFAR at doses of 5,25 and 125 mg/kg were administered orally to mice daily for 3 weeks, and the normal animals were given vehicle. The results of this study are summarized as follows; the relative weight of spleen was markedly increased by BFAR treatment, compared with that in normal mice. However, the body weight gain and the relative weight of liver were not affected. Splenic plaque forming cells and hemagglutination titers to sheep red blood cells, and the secondary IgG antibody response to bovine serum albumin were also dose-dependently enhanced by BFAR treatment. In these mice, BFAR did not increase serum alanine aminotransferase total protein, sect albumin and albumin/globulin ratio when compared with those in normal mice. Thus, these findings indicate that BFAR significantly enhances humoral immune response to antigen in concentrations that do not affected liver function.

가토(家兎)에서 Norepinephrine, Phenylephrine 및 Clonidine의 승압반응(昇壓反應)에 대한 Diltiazem의 영향(影響) (Effects of Diltiazem on Norepinephrine-, Phenylephrine- and Clonidine-induced Pressor Response in Rabbits)

  • To examine the selectivity of diltiazem, used in the cardiovascular diseases, on alpha-1 and alpha-2 adrenoceptor-induced pressor responses, effect of diltiazem on alpha-adrenocepter agonist-induced pressor responses were investigated in urethane-anesthetized rabbits and spinal rabbits. The results are summarized as follows: 1. Intravenous diltiazem(10, 30, 100, 300, $1000{\mu}g/kg$) produced dose-dependent depressor response in rabbits. 2. Pressor responses to intravenous norepinephrine($10{\mu}g/kg$) and phenylephrine ($30{\mu}g/kg$) were inhibited by pretreatment with intravenous diltiazem in rabbits and no difference was noted between the degree of both inhibitions of the pressor response by diltiazem. 3. Presser responses to intravenous norepinephrine ($3{\mu}g/kg$), phenylephrine ($20{\mu}g/kg$) and clonidine ($300{\mu}g/kg$) were inhibited by pretreatment with intravenous diltiazem in spinal rabbits. No difference was noted between the inhibition of norepinephrine-induced pressor response and that of phenylephrine-induced pressor response by diltiazem. The inhibition of clonidine-induced pressor response by diltiazem was slightly prominent than that of norepinephrine- or phenylephrine-induced pressor response. These results suggest that diltiazem significantly inhibits both pressor responses mediated by alpha-1 and alpha-2 adrenoceptors.

횐쥐 대동맥의 수축반응과 열충격단백질에 대한 비소의 영향 (Effect of Arsenic on Heat Shock Protein and Vascular Contractility of Rat Aorta)

  • In order to examine if arsenic, one of environmental stresses, contributes to hypertension as one of cardiovas cular pathological factors, this study was perfarmed in vivo and in vitro, using intacted or pithed rats and aorta ring preparation, respectively. And also the relationship between expression of heat shock protein (HSP) 90 and vasoactives-induced contractile response was elucidated. To measure blood pressure, the carotid arterial pressure was recorded on physiograph(Grass Co. 79E) connected to strain gauge. On the other hand, contractile response of vascular ring preparation isolated from rat was determined in organ bath and was recorded on physiograph connected to isometric transducer. And HSP was detacted by Western blotting whole cell Iysis. Preganglionic nerve stimulation was increased by 26.0% in arterial pressure of rat treated with arsenic. Vascular contractile response was monitored and HSP were measured by Western blotting of whole Iysis prepared from samples exposed with 0, 0.5, 1, 2 and 4 mM of arsenic for 8 hours. The dose-vascular responses of potassium chloride were augmented by increasing dose of arsenic in the strips exposed to arsenic for 8 hours, and were not augmented for 1, 3, 5 hours. And the response of relaxation of rat aorta induced by histamine was not influenced by arsenic stress. The increase of HSP 90 expression in rat aorta was pronounced at 8 hours after 4 mM of arsenic treatment, but HSP 60 expression was not. Arsenic stress not only increased the expression of HSP 90 in the rat aorta, but also augmented contractions to potassium chloride. These results indicated that arsenic stress was sufficient to induce heat shock protein 90, resulting in increased vascular contractility in rat aorta.

폐암으로 유발된 무기폐에 대한 방사선 치료의 효과 (Radiation Effect on Airway Obstruction from Lung Cancer)

  • 1986년 4월부터 1988년 12월까지 영남대학병원 치료 방사선과에 폐암으로 확진후 등록된 환자중 기관지정 검사상 지도 폐쇄의 소견이 있으며 흉부 X-선 사진상 현저한 폐음영이 있었던 환자 21명에 대한 방사선 치료 성적은 다음과 같다. 1. 71.4%의 환자(15/21)에서 임상 증상의 호전을 보였다. 2. 소세포성 암환자는 30GY/10 fractions의 선량에서도 100%의 증상 호전 및 흉부 X-선상 소견의 호전을 보였다. 3. 비소세포성 암환자에서 50GY이상 조사군 이 45GY이하 조사군 보다 우수한 성적을 보였다. 4. 병변의 위치와 무기폐의 호전정도는 무관하였다.

Development and Performance of a Hand-Held CZT Detector for In-Situ Measurements at the Emergency Response

  • Background: A hand-held detector for an emergency response was developed for nuclide identification and to estimate the information of the ambient dose rate in the scene of an accident as well as the radioactivity of the contaminants. Materials and Methods: To achieve this, the most suitable sensor was first selected as a cadmium zinc telluride (CZT) semiconductor and the signal processing unit from a sensor and the signal discrimination and storage unit were successfully manufactured on a printed circuit board. Results and Discussion: The performance of the developed signal processing unit was then evaluated to have an energy resolution of about 14 keV at 662 keV. The system control unit was also designed to operate the CZT detector, monitor the detector, battery, and interface status, and check and transmit the measured results of the ambient dose rate and radioactivity. In addition, a collimator, which can control the inner radius, and the airborne dust sampler, which consists of an air filter and charcoal filter, were developed and mounted to the developed CZT detector for the quick and efficient response of a nuclear accident. Conclusion: The hand-held CZT detector was developed to make the in-situ gamma-ray spectrometry and its performance was checked to have a good energy resolution. In addition, the collimator and the airborne dust sampler were developed and mounted to the developed CZT detector for a quick and efficient response to a nuclear accident.

악성 폐쇄성 황달 환자에 있어서 간문에 대한 방사선치료의 역할 (The Role of Porta Hepatis Irradiation in Relieving Malignant Obstructive Jaundice)

  • 1984년부터 1988년까지 인제대학교 의과대학 치료방사선과에서 전이성 암에의해 발생한 악성 폐쇄성 황달환자 13명을 대상으로 간문을 포함하는 국소방사선치료를 시행하였다. 총 3명의 환자중 추적가능한 7명은 표준 방사선 조사야에 2.6주에서 6주 동안에 걸쳐 $2600\~5480$cGy의 방사선 조사를 받았으며 이중 5명은 완전관해, 1명은 부분 관해를 보여주었다. 전체 13명의 환자에서의 평균 생존기간은 3개월 이었으며 7명의 추적가능한 환자에서는 10.5개월이었다. 완전 관해를 보인 환자 5명중 2명은 황달의 재발없이 1년 이상의 생존기간을 보여주었다. 따라서 간문의 전이성 암으로 인해 발생한 악성 폐쇄성 황달 환자에 있어서 간문을 포함하는 조사야의 국소적 방사선 치료는 황달 해소와 증상완화에 많은 도움을 줄 수 있는 것으로 생각된다.

상급종합병원 암센터에서 Nivolumab 사용평가와 치료성과에 미치는 영향인자 (Evaluation of Nivolumab Use and Factors related to Treatment Outcomes in a Cancer Center of a Top Tier General Hospital)

  • Background: We strived to evaluate the status of nivolumab use and associated factors on the clinical efficacy of the drug. Methods: The study was retrospectively conducted in patients who had been administered nivolumab at least once at the cancer center of Seoul National University Hospital from June 2015 to April 2017. Data were collected from electronic medical records. A medication-use evaluation was performed based on the American Society of Health-System Pharmacists mediation-use guidelines. Results: Sixty-six of the 74 patients (89.2%) showed indications approved for nivolumab use by the Korean Ministry of Food and Drug Safety (MFDS; n=55) or the US Food and Drug Administration (FDA; n=11). Approximately 73.0% of the patients were administered the approved dose of 3 mg/kg but 25.7% were administered an unapproved fixed dose of 100 mg. The overall response rate was 21.7%, and the response rate of non-small cell lung cancer patients, who accounted for the largest number of indications, was 18.8%. Adverse reactions were found in 90.1% of the patients and were mostly mild (86%). The expression of programmed death-ligand 1 (PD-L1) was analyzed as a factor affecting treatment response (p=0.028, odds ratio [OR]=11.331). Conclusion: PD-L1 expression was found to affect treatment response. However, caution is required while using an unapproved dosage and in the absence of monitoring for effectiveness and safety. Therefore, an effective protocol or instruction manual for the proper use of nivolumab should be considered.

사료성분에 의한 닭의 혈중 Cholecystokinin 농도의 상승 (Enhanced Release of Cholecystokinin by Dietary Components in Chicks)

  • 닭의 Cholecystokinin (CCK) 방출에 미치는 사료성분에 대한 효과에 대해서 stomach tube에 의한 사료투여에 의해, 특수 CCK-8 antibody 를 이용해서 그 혈중 CCK의 농도를 측정했다. 실험 1에서, 무단백질사료에 대두단백질 및 대두단백질과 같은 조성의 아미노산혼합물을 첨가해서 투여한 결과 무단백질사료에 비교해서 유의하게 높은 증가를 보였으며, 아미노산혼합물사료투여가 단백질사료투여보다 빠른 증가가 관찰되었다. 무단백질사료에 아미노산을 단독으로 첨가하여 투여한 결과. phenylalanine 첨가 사료에서 Plasma중의 CCK농도가 가장 높게 나타나 아미노산혼합물과 같은 수준으로 나타났으며. 뒤이어 arginine이, valine에서 가장 낮은 분필반응이 관찰되었다. 실험 2와 3에서, 단백질사료에 SBTI를 첨가하여 투여한 결과, SBTI첨가 사료에 의해 Plasma CCK 농도는 투여 후 급격히 증가하였으며, 그 반응은 1,000mg첨가가 100mg/kg첨가보다 높은 dose response가 관찰되었다. SBTI첨가 사료에 의한 crop emptying에는 처리군과 차이가 나타나지 않아, SBTI가 단독으로 plasma CCK농도를 상승시킨다고 생각된다.

Enhancing immune responses to inactivated foot-and-mouth virus vaccine by a polysaccharide adjuvant of aqueous extracts from Artemisia rupestris L.

  • Background: New-generation adjuvants for foot-and-mouth disease virus (FMDV) vaccines can improve the efficacy of existing vaccines. Chinese medicinal herb polysaccharide possesses better promoting effects. Objectives: In this study, the aqueous extract from Artemisia rupestris L. (AEAR), an immunoregulatory crude polysaccharide, was utilized as the adjuvant of inactivated FMDV vaccine to explore their immune regulation roles. Methods: The mice in each group were subcutaneously injected with different vaccine formulations containing inactivated FMDV antigen adjuvanted with three doses (low, medium, and high) of AEAR or AEAR with ISA-206 adjuvant for 2 times respectively in 1 and 14 days. The variations of antibody level, lymphocyte count, and cytokine secretion in 14 to 42 days after first vaccination were monitored. Then cytotoxic T lymphocyte (CTL) response and antibody duration were measured after the second vaccination. Results: AEAR significantly induced FMDV-specific antibody titers and lymphocyte activation. AEAR at a medium dose stimulated Th1/Th2-type response through interleukin-4 and interferon-γ secreted by CD4+ T cells. Effective T lymphocyte counts were significantly elevated by AEAR. Importantly, the efficient CTL response was remarkably provoked by AEAR. Furthermore, AEAR at a low dose and ISA-206 adjuvant also synergistically promoted immune responses more significantly in immunized mice than those injected with only ISA-206 adjuvant and the stable antibody duration without body weight loss was 6 months. Conclusions: These findings suggested that AEAR had potential utility as a polysaccharide adjuvant for FMDV vaccines.
