Comparison of Total Phenol, Flavonoid Contents, and Antioxidant Activities of Orostachys japonicus A. Berger Extracts (와송(Orostachys japonicus A. Berger) 추출물의 총 페놀, 플라보노이드 함량 및 항산화 활성 비교)

  • The purpose of this study was to measure the bioactivity and antioxidant activity of Orostachys japonicus A. Berger. Orostachys japonicus A. Berger have been known to contain functional materials such as kaempferol, hydroquinone, methyl gallate, quercetin, gallic acid etc. To identify the main functional materials of Orostachys japonicus A. Berger, the contents of flavonoid and phenol were measured. We extracted Orostachys japonicus A. Berger powder from four solvents such as chloroform:methanol (CM, 2:1, v/v), distilled water (DW), 70% methanol, 70% ethanol. After that, this study determined tannin, total phenol, flavonoid content, DPPH radical scavenging activity, ABTS radical scavenging activity, ferric reducing antioxidant power and reducing power of Orostachys japonicus A. Berger extracts and as results of comparing each extract. respectively. From the above results shows that antioxidant activity and bioactivity of Orostachys japonicus A. Berger extracts was higher in the order of 70% ethanol, 70% methanol, DW and CM (p<0.05). The results showed that antioxidant activity of Orostachys japonicus A. Berger extracts supposed to affect by the total phenol and flavonoid contents.

Evaluation of biological activity for Dangyuja (Citrus grandis) leaves and investigation of optimal concentrations extracted by alternative ethanol concentrations (에탄올 농도별 당유자 잎의 최적추출조건 및 생리활성 평가)

  • TheCitrus grandis Osbeck is a special product in the Jeju island. The product has been as a remedy for liver damage and hang over. This study demonstrates how to investigate and compare the antioxidant, phenol content, tyrosinase and ${\alpha}$-glucosidase inhibitory activity, antimicrobial, and alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activity with the C. grandis leaves extracted in different ethanol concentrations. From the yield, a 20% ethanol extract demonstrated the highest results among the other extracts. The distilled water extract showed the most abundant in a total phenol content and highest ABTS radical scavenging activity and reducing power assay. In the DPPH radical scavenging activity, ${\alpha}$-glucosidase and tyrosinase inhibitory assay (used ${\text\tiny{L}}$-tyrosine as substrate), the 80% ethanol extract exhibited a higher value than other extracts. The 60% ethanol extract showed prominent activities in the tyrosinase inhibitory (used ${\text\tiny{L}}$-dopa as substrate), ADH and ALDH activity assay. In the minimum inhibitory concentration (MIC) assay, 60% and 80% ethanol extracts inhibited the bacterial growth almost similarly. Moreover, the gram-positive bacteria was more restrained than the gram-negative bacteria. The resultsrevealed that the distilled water and 80% ethanol extract showed a relatively higher antioxidant activity compared to other extracts. The 60 ~ 80% ethanol extracts demonstrated potential tyrosinase, ${\alpha}$-glucosidase inhibitory, antimicrobial, ADH and ALDH activities. Therefore, the C. grandis is suggested to be considered as a functional material for various proposes.

Effect of Insulin-like Action and Insulin Sensitizing on 3T3-Ll Adipocytes from Coicis Semen (의이인이 3T3-Ll Adipocytes에서 인슐린성 작용과 인슐린 민감성에 미치는 영향)

  • Objectives : In this study, water extracts from Coix lachryma-jobi Linne var. mayuen Stapf. were investigated for their effects on insulin-like action and glucose uptake in 3T3-Ll cells. Methods : We examined the effects of insulin-like action on the differentiation of 3T3-Ll fibroblasts. The Coicis Semen was treated with hot water and the extract was freeze-dried. The hot water extract was chromatographed on nonionic polymer resin (Amberlite XAD-4, Sigma) with distilled water (Fr. 1), 20% (Fr. 2), 40% (Fr. 3), 60% (Fr. 4), 80% (Fr. 5), and 100% EtOH (Fr. 6), successively. Results : Total extract of Coicis Semen was fractionated into 0 to 100% MeOH with Amberlite XDA-4 column. Treatment of cells with $10{\;}\mu\textrm{g}/ml$ of total extracts of Coicis Semen significantly increased the differentiation (p<0.05). At $1{\;}\mu\textrm{g}/ml$ level of insulin, the differentiation was accelerated (p<0.01). The effect of extracts plus insulin on the differentiation was greater than that of insulin alone. In 3T3-Ll adipocytes, glucose uptake was higher by 2.7 times with 5 uM of total extract in low dosage of insulin (3 ng/ml) than without total extract. 5 and 50 uM of water and 40% MeOH fractions increased glucose uptake by 3.5 times in 3T3-Ll adipocytes (p<0.00l). Conclusions : Coicis Semen contains compounds which playa role of insulin-like action and insulin sensitizer.

Nutritional Analyses and Antioxidant Activity of Apple Pomace (사과 부산물의 영양성분 분석 및 항산화 효과)

  • To enhance the applications of apple pomace, which is a by-product of apple, this study analyzed the nutritional components, ursolic acid content, and antioxidant activity of different solvent (distilled water, fermented alcohol, and methanol) extracts. The samples included hot air-dried and freeze-dried apple pomace. The moisture, protein, fat, ash, and total dietary fiber contents of hot air-dried apple pomace were 3.2%, 3.9%, 2.4%, 2.0%, and 28.5%, respectively, and those of freeze-dried apple pomace were 8.2%, 3.4%, 2.4%, 1.8%, and 33.0%, respectively. Ursolic acid was not detected in the distilled water extract of either sample. However, in hot air-dried apple pomace, the methanol extract was 1,753.32 ㎍/ml, and the fermented alcohol extract was 1,532.94 ㎍/ml. In freeze-dried apple pomace, the methanol extract was 1,407.04 ㎍/ml, and the fermented alcohol extract was 1,221.81 ㎍/ml. The total polyphenol and flavonoid contents were 306.7 ㎍/ml and 950.1 ㎍/ml, respectively in methanol extracts of hot air-dried apple pomace and 277.6 ㎍/ml and 925.0 ㎍/ml, respectively in methanol extracts of freeze-dried apple pomace. 2, 2'-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities of hot air-dried apple pomace were 73.3% in methanol extract and 59.4% in fermented alcohol extract, and those of freeze-dried apple pomace were 76.1% in methanol extract and 66.0% in fermented alcohol extract. Both samples had the lowest antioxidant activity in distilled water extracts. Similar to DPPH radical scavenging activity, both samples showed increasing 2,2'-azino-bis(3-ethylbenzothiazoline-6- sulfonic acid) (ABTS) radical scavenging activity in the order of methanol, fermented alcohol, and distilled water. All samples had stronger reducing power than ascorbic acid (311.5 ㎍/ ml) as a positive control.

Antioxidant and Antibacterial Activity of Extracts from Brassica juncea czerniak et coss., Celosia cristata L., and Beta vulgaris L. (맨드라미, 비트, 홍갓 색소 추출물의 항산화 및 항균 효과)

  • We sought to study the qualities and scientific benefits of Dongchimi, a traditional Korean food. We compared and analyzed ingredients used for the appearance and storability of dongchimi - honggot (Brassica juncea czerniak et coss), cockscomb (Celosia cristata L.), and beet Beta vulgaris L.). We specifically examined the antioxidative and antibacterial activity of pigments from extracts of these ingredients. Distilled water ($H_2O$) and 1% citric acid were used to safely extract pigments. The antioxidative activity of the pigments was then measured for total phenolic compounds, SOD (Super Oxide Dismutase), and EDA (Electron Donation Ability) by DPPH. The antibacterial activity of was also assessed by a Paper disc solution. Our results show that the pigments had sufficient antioxidative activity and had antibacterial properties against Gram positive and negative bacteria. In particular, Cockscomb (used for enhancing color) contained the highest amount of polyphenol compounds and had the most efficient antioxidative activity.

Optimal Growth Condition of Pleurotus ostreatus Cultured in the Foodwastes Extracts (음식폐기물 추출물을 이용한 느타리버섯균의 최적성장조건)

  • For the purpose of the methodological development to convert the food wastes into resources, we have attempted to culture the mushroom hypha, Pleurotus ostreatus. The food-wastes were mixed with distilled water, and the mixture was autoclaved to produce fluid, which was centrifugated and used as the growth media. Concentrations of the food wastes extracts were prepared with 10, 20, 30, 40, and 50%(W/V), and the initial pH were set variously with 4, 5, 6, and 7. These were cultured for 9 days at the temperature of $25^{\circ}C$ and the rotation rate of 120 rpm. The result is that the fluid form of the mushroom hypha have been grown best at the concentration of 30% and the optimal pH was 5 and 6.

Fabric dyeing with Laminaria japonica as a marine resources (해양자원을 활용한 직물염색-다시마-)

  • This study focuses on the effects of dyeing conditions and mordanting conditions, changes of colorfastness, antibacterial properties and UV protective of silk fabric dyed with Laminaria japonica extracts. A natural colorant was extracted from Laminaria japonica using distilled water as extractants. According to the results, maximum dye uptake (K/S) were obtained at 100% V/V colorant concentration, $80^{\circ}C$, 60 min and pH 2. Silk fabric was dyed with Laminaria japonica extract at $80^{\circ}C$ for 60 min with pre-treatment or post-treatment of various metal salts as mordants. The Laminaria japonica extracts produced yellow hue on silk fabric in pre-mordanting and manifested green yellow hue mordanted with $CuSO_4$ in post-mordanting method. Mordant $CuSO_4$ for silk fabric was found to give good light fastness (rating 4). UV protection property did not increase significantly upon mordanting.

  • The antifungal activities of 153 extracts from 51 species of Korean mushroom on six phytopathogenic fungi were investigated. The powder of fruit-body of each mushroom was extracted with petroleum ether, 80% ethanol and distilled water. The water extracts of four mushrooms including Amanita virosa showed antifungal activities on Alternaria alternata, Colletotrichum graminicola, Fusarium solani, Idriella bolleyie. The 80% ethanol extracts of seven mushrooms including Boletus auripes showed antibiotic activities against A. alternata, C. gramincola, Cylindrocarpon destructans, F. solani, F. oxysporum cucumerinum, and I. bolleyie. The petroleum ether extracts of six mushrooms including Amanita citrina showed antibiotic activities against A. alternata, C. destructans, and C. graminicola. The 102 extracts of 34 mushrooms including Agaricus arvensis didn't show antibiotic activities.

Antioxidant and Physiological Activities of Different Solvent Extracts from Messerschmidia sibirica (모래지치(Messerschmidia sibirica) 추출물의 항산화성 및 생리활성)

  • Messerschmidia sibirica is a halophyte that grows in high-salinity areas of coastal sand dunes and sandy shores. This study was conducted to investigate the constituents, antioxidant potency, and physiological activities of M. sibirica. Mineral analyses revealed that potassium, calcium, sodium, and magnesium were the most prevalent minerals in M. sibirica leaves and stems. We used 70% ethanol, 80% methanol, and distilled water as solvents to prepare extracts from M. sibirica tissues, with extraction yields of between approximately 19% and 27% being obtained. Among the six types of extract, the leaf ethanol extract (LEE) was characterized by the highest total polyphenol and flavonoid contents, and the antioxidant activities of the LEE were also the highest among the different solvent extracts. In addition, the leaf water extract was shown to have the highest tyrosinase and α-glucosidase inhibitory activities, whereas the leaf methanol extract was found to have the highest elastase inhibitory activity. Notably, all leaf extracts were established to have more than 75% β-secretase (BACE1) inhibitory activity at a final concentration of 0.5 mg/mL. These results indicate that leaf extracts of M. sibirica may have beneficial antioxidant properties, and could thus have potential application as functional supplements in food and pharmaceutical products.

Antioxidant Activity and Antibacterial Activities from Different Parts of Broccoli Extracts under High Temperature (고온에서의 브로콜리 부위별 추출물의 항산화성 및 항균성)

  • The radical scavenging activity of ethanol, acetone and distilled water extracts of broccoli including leaf, flower, flower plus stem, stem, peel and positive control vitamin C, respectively, were investigated. Each sample under assay condition exhibited free radical scavenging activity (FRSA) toward DPPH radical. Among five samples, S (stem) extracts, F (flower) extracts and L (leaf) extracts of broccoli showed the FRSA in the dot-blot assay. Strong FRSA potential of the ethanol extracts of broccoli revealed at pH $2\~6$ of acid regions and at $60\~80^{\circ}C$. The extracts of green tea and Artermisia were found to have effect against Staphylococcus aureus. Whereas, only the aqueous extracts of broccoli F and FS (flower-stem) of the five extracts marked strong antibacterial activity against Bacillus amyloliquefaciens in high temperature.
