The Extraction of Lignin and Production of Vanillin from Rice Straw (볏짚으로 부터의 리그닌 추출 및 바닐린 생성)

  • Lignin was extracted from the rice straw by using the solvent mixture of buthyl alcohol and distilled water. And the experiment of vanillin production from extracted lignin was performed with the oxidation catalysts; CuO, Cu(OH)2 and CuSO4.5H2O. The optimum conditions of lignin extraction are the reaction temperature 12$0^{\circ}C$ and the mixture of 250mL buthyloloohol, 250mL, distilled water and 25g rice straw in the presence of 2.5g p-toluenesulfonic acid. The yield of vanillin from extracted lignin increased linearly with the increase of reaction temperature. And it increased with the order of Cu(OH)<$_2$ CuO$_4\cdot \;5H_2$Oas oxidation catalysts. The maximum yield of vanillin was 9% in the presence of 2.5%(w/v) CuSO$_4\cdot \;5H_2$O under the following conditions: temperature, 18$0^{\circ}C$; pressure, 13atm; pH 4.0 and reaction time, two hours.

Effects of Shigyungbanha-Tang on the Lipopolysaccharide-Induced Acute Lung Injury in Mice (시경반하탕(柴梗半夏湯)이 LPS로 유발된 급성 폐손상에 대한 영향)

  • This study was performed to investigate the effects of Shigyungbanha-tang(SGT) on the lipopolysaccharide(LPS) induced acute lung injury(ALI) in mice. 1 and 24 h before LPS intratracheal instillation, control group was taken distilled water orally. Treated groups was taken each concentrate SGT(2.5 g/kg, 6.7 g/kg) by orally as same times. Normal group was not instilled with LPS and was taken distilled water. 24 h after LPS intratracheal instillation, lung histology was performed in inflated-fixed lungs in 3 mice of each groups. The other mice of each groups, bronchoalveolar lavege fluids(BALF) was obtained to measure proinflammatory cytokines(TNF-$\alpha$, IL-$1{\beta}$, IL-6) and blood sample was obtained to measure white blood cell(WBC). In vitro, the effect of SGT($100\;ug/m{\ell}$, $500\;ug/m{\ell}$, $1000\;ug/m{\ell}$) on the release of RANTES, TARC induced by TNF-$\alpha$ and IL-4 in human alveolar epithelial cell(A549) was examined. Histopathologically, SGT prevented LPS-induced lung injury. SGT decreased protein, TNF-$\alpha$, IL-$1{\beta}$ and IL-6 according to concentrations. In vitro, $500\;ug/m{\ell}$, $1000\;ug/m{\ell}$ concentrate SGT suppressed the expression of RANTES and TARC on A549 cells. On the basis of these results, SGT had a markedly anti-inflammatory effect in a clinically relevant model of ALI. Nevertheless, further investigations are required to determine the potential clinical usefulness of SGT in the adjunctive therapy of ALI.

  • The purpose of this study is to evaluate the effect of additional enamel etching with phosphoric acid on the microleakage of the adhesion of self-etching primer system. Class V cavity($4mm{\times}3mm{\times}1.5mm$) preparations with all margins in enamel were prepared on buccal surface of 42 extracted human upper central incisor teeth. Prepared teeth were randomly divided into 3 groups. Group 1:no additional pretreatment with 37% phosphoric acid (NE). Group 2:additional pretreatment with 37% phosphoric acid for 10 seconds (E10s). Group 3:additional pretreatment with 37% phosphoric acid for 20 seconds (E20s). The adhesives(Clearfil SE $Bond^{\circledR}$, Kuraray, Osaka, Japan) and composite resins(Clearfil $AP-X^{\circledR}$, Osaka, Kuraray, Japan) were applied following the manufacturer's instructions. All the specimens were finished with the polishing disc(3M dental product, St Paul, MN, USA), thermocycled for 500 cycles between $5^{\circ}C$ and $55^{\circ}C$ and resected apical 3-mm root. 0.028 stainless steel wire was inserted apically into the pulp chamber of each tooth and sealed into position with sticky wax. Surrounding tooth surface was covered with a nail varnish 2 times except areas 1mm far from all the margins. After drying for one day, soaked the samples in the distilled water. Microleakage was assessed by electrochemical method(System 6514, $Electrometer^{\circledR}$), Keithley, USA) in the distilled water.   • The stability comparison tests on purified bee venom (PBV) and bee venom melittin (BVM) in different conditions of temperature, solvent, and concentration were studied. High purity BVM (98.2 %) was separated from PBV by prep-HPLC (column, C4) and used to stability tests in aqueous phase. The stability of the PBV has been increased in the saline solution, while BVM was reduced. In distilled water, the stability of PBV has been reduced, while BVM showed an increasing result. As a result, the appropriate conditions for maintaining the long term stability of BVM were found to be the low temperature (4 ℃), distilled water, and concentration (1.0 mg/mL).

MICROLEAKAGE OF COMPOMER IN CLASS II CAVITY (제 2 급 와동에 충전된 Compome의 미세누출에 관한 연구)

  • To evaluate the micro leakage of compomer, 4 materials were divided into 4 groups of 15 cavities each.(Group 1: Z-100, Group 2: Dyarct AP, Group 3: Fuji II LC, Group 4: Compoglass) After the class II cavities were prepared using carbide bur No. 553, all specimen were restored by incremental filling technique. In group 3, Z-100 was filled with a base of a light curing glass-ionomer. After 7 days, all specimens were thermocycled between $5^{\circ}C$ and $55^{\circ}C$ for 500 cycles, followed by placement in 50% silver nitrate dye for 2 hours at $37^{\circ}C$. After rinsed in distilled water, these teeth were immersed in photodeveloping solution and exposed to fluorescent light for 6 hours. Teeth were then washed in distilled water to remove the photodeveloping solution, sectioned mesio-distally and evaluated. The results were as follows : 1. In the cervical portion, there was significant difference between Fuji II LC and other groups(Z-100, Dyract AP, Compoglass), Fuji II LC had the least value.(p<0.05) 2. In the cervical portion, there was not significant difference among Dyract AP, Z-100 and Compoglass. 3. In the occlusal portion, there was not significant difference among Dyract AP, Z-100 and Compoglass. From the results above, In enamel, microleakage of compomer such as Dyract AP and Compoglass resemble to that of composite resin. It is thought that it is due to characteristics of composite resin portion of compomer. But in dentin, microleakage of compomer is higher than that of resin modified glass ionomer cement, it is thought that in compomer, acid-base reaction is not developed with dentin.

Effects of Ojeok-san on the Maturation of Follicles and NGF Expression in Rats with Estradiol Valerate-induced Polycystic Ovaries (오적산(五積散)이 Estradiol Valerate로 유발된 다낭성 난소 모델에서 난포의 성숙 및 NGF발현에 미치는 영향)

  • Objectives: This study was designed to investigate the effect of Ojeok-san(OJS) on the estradiol valerate(EV)-induced polycystic ovaries(PCO) in rats. Methods: PCO was induced by single intramuscular injection with estradiol valerate(EV)(4mg) in female rats. Normal group(n=8) were injected with sesame oil and orally administrated distilled water for sixty days. PCO control group (n=8) were injected with EV and orally administrated distilled water for sixty days. OJS treated group(n=8) were injected with EV and orally administrated OJS for sixty days. Then, we measured weights of body, ovaries, adrenal glands and uterus of rats. The histomorphometrical changes of ovaries were also evaluated. And we observed the NGF and CRF expression by immunohistochemistry. Results: The results were as follows - The weights(mg) of ovaries in OJS treated group($57.4{\pm}9.4$) were significantly increased(p < 0.05) compared with PCO control group($42.3{\pm}8.5$). - The numbers of mature follicles in OJS treated group($9.3{\pm}2.5$) were significantly increased(p < 0.05) compared with PCO control group($6.1{\pm}2.1$). - The expressions of NGF-immunoreactive cells in the ovarian granulosa cells in OJS treated group were weaker than PCO control group. Conclusion: From the these results, we concluded that Ojeok-san(OJS) has inhibitory effect on the development of EV-induced polycystic ovary. And it's effect maybe related with decreased NGF activities in the ovary.

Surface microhardness of three thicknesses of mineral trioxide aggregate in different setting conditions

  • Objectives: This study aimed to compare the surface microhardness of mineral trioxide aggregate (MTA) samples having different thicknesses and exposed to human blood from one side and with or without a moist cotton pellet on the other side. Materials and Methods: Ninety cylindrical molds with three heights of 2, 4, and 6 mm were fabricated. In group 1 (dry condition), molds with heights of 2, 4, and 6 mm (10 molds of each) were filled with ProRoot MTA (Dentsply Tulsa Dental), and the upper surface of the material was not exposed to any additional moisture. In groups 2 and 3, a distilled water- or phosphate-buffered saline (PBS)-moistened cotton pellet was placed on the upper side of MTA, respectively. The lower side of the molds in all the groups was in contact with human blood-wetted foams. After 4 day, the Vickers microhardness of the upper surface of MTA was measured. Results: In the dry condition, the 4 and 6 mm-thick MTA samples showed significantly lower microhardness than the 2 mm-thick samples (p = 0.003 and p = 0.001, respectively). However, when a distilled water- or PBS-moistened cotton pellet was placed over the MTA, no significant difference was found between the surface microhardness of samples having the abovementioned three thicknesses of the material (p = 0.210 and p = 0.112, respectively). Conclusions: It could be concluded that a moist cotton pellet must be placed over the 4 to 6 mm-thick MTA for better hydration of the material. However, this might not be necessary when 2 mm-thick MTA is used.

Effect of Neutralization of Red Mud on Arsenic Stabilization in Soils (레드머드 중화 방법에 따른 토양 중 비소의 안정화 특성 평가)

  • Since the amount of red mud, generated from aluminum smelting process as a by-product, has increased worldwide, the recycle and metal resource recovery from the red mud is becoming more important. In this study, in order to recycle the red mud as a soil stabilizer to remediate arsenic contaminated soils, neutralization of red mud was investigated. Red mud was neutralized by washing with distilled water and NaCl, CaCl2, FeCl3, and HCl solutions and heating at 200-800℃, and arsenic stabilization characteristics in soils were evaluated with the neutralized red mud. Although washing with distilled water was not effective in neutralizing red mud, the application of the washed red mud to soils lowered the soil pH compared to the application of untreated red mud. Among NaCl, CaCl2, FeCl3, and HCl solutions, washing with FeCl3 showed the most effective in lowering pH of the red mud from pH 10.73 to pH 4.26. Application of the neutralized red mud in soils resulted in quite different arsenic stabilization efficiency depending on soil samples. In M1 soil, which showed relatively high arsenic stabilization efficiency by untreated red mud, the neutralization of red mud resulted in little effect on arsenic stabilization in soil. On the other hand, in M2 soil, which showed low arsenic stabilization efficiency by untreated red mud, the neutralization of red mud increased arsenic stabilization significantly. Soil characteristics such as clay minerals and pH buffering capacity seemed to affect reactions between red mud and soils, which resulted in different effects of the red mud application on soil pH and arsenic stabilization efficiencies.

Effect of Yeonsan Ogye bioactive peptides on anti-oxidant indexes in rats' liver

  • Purpose: This study investigated the effect of bioactive Yeonsan Ogye peptides (YOPs) intake on changes in the hepatic anti-oxidant indexes in male rats. Methods: Sprague-Dawley male rats were divided into 3 groups and given a casein-based AIN-93G diet and distilled water ad libitum without any added YOPs (control), distilled water with 250 mg of YOPs (Y250), or 500 mg of YOPs (Y500) per kg of body weight for 4 weeks. YOP dose was decided as referred to in the referenced study where toxicity did not occur. The hepatic anti-oxidant indexes were determined using a commercial kit. Statistical analysis was performed using SPSS version 23.0 and are expressed as $mean{\pm}standard$ error of mean. Differences among the groups were evaluated by one-way analysis of variance followed by post hoc Duncan's multiple comparisons test. Results: There were no differences in the body weights, weight gain, food intake, food efficiency ratio, or organ weight, including liver, kidney, spleen, thymus, and epididymal fat, among all of the groups. The hepatic nitric oxide (NO) level in the Y500 group was lower than that in the control and Y250 groups, and the hepatic malondialdehyde (MDA) level was lower in the Y500 group than in the Y250 group. The differences in hepatic superoxide dismutase (SOD) and catalase (CAT) activities were not statistically significant between the groups. From these results we speculated that YOPs may have anti-oxidative abilities to regulate NO and MDA production without affecting SOD and CAT activities. Conclusion: YOPs are presumed to act as anti-oxidants in the animal or human body.

The Effects of Alpiniae Oxyphyllae Fructus on Osteoporosis and Muscle Dystrophy of Male Mice (수컷 생쥐의 골다공증과 근위축에 대한 익지인(益智仁)의 효과)

  • Objective: To investigate the effect of Alpiniae oxyphyllae fructus (AOF) on the alleviation of musculoskeletal disorders caused by aging, we conducted experiments on osteoporosis and muscle atrophy. Methods: The experimental group was classified into a control group, aging-elicited (AE) group and AOF group. The control group comprised 8-week-old Institute of Cancer Research (ICR) mice. The AE and AOF groups were ICR mice at 50 weeks of age. For the AE group, 10 mL of distilled water was administered once a day for 180 days without any treatment. An AOF extract (0.54 g/kg) was dissolved in distilled water and administered to the mice in the AOF group once a day for 180 days. Results: In the experiment on the alleviation of osteoporosis, the distribution of glucosaminoglycan in the bone matrix of the femoral bone was increased in the AOF group; moreover, the osteocalcin (OCN) positive reaction was increased and 8-OHdG positivity was decreased. In addition, AOF positively decreased RANKL, positively increased OPG, and positively decreased MMP-3. Muscle fiber loss in the endomysium following muscle degeneration of the quadriceps was reduced more in the AOF group compared with the AE group, and caspase-3 positive responses were also decreased. In addition, the 8-OHdG and p-lkB positivity in the AOF group decreased compared with the AE group, and the Myo-D positivity increased. Conclusion: We found that increasing bone formation alleviates osteoporosis, and that reducing bone loss alleviates muscle atrophy by reducing muscle loss and increasing muscle development.
