Lespedeza species are mainly used for wildlife food and cover and for erosion control. The germination of these species can be enhanced after a fire occurrence in forest, which is known as fire-activated seeds to germinate. While the heat treatment could break seed dormancy of Lespedeza, its germination rate was quite low. We investigated that chemical scarification could promote germination of L. tomentosa. Seeds were soaked in 100% sulfuric acid (H2SO4) for 0, 1, 3, 6, 12, 24, 48, 96, 192, and 384 min, and then washed in distilled water for 24 h. Very few seeds were germinated in control (H2SO4 for 0 min). More than 90% of seeds were germinated in H2SO4 for 24, 48, and 92 min. However, some damage was observed in roots and cotyledons of seedling dipped in H2SO4 for a long time. To search the optimal soaking time in H2SO4 without defects, seeds scarified in H2SO4 for 30, 60, 90, 120, 150, 180, and 300 min were sown the commercial soil medium. Seeds treated with H2SO4 for 90 min and 150 min emerged by about 92% and 84%, respectively. Therefore, H2SO4 treatment could break the seed dormancy of Lespedeza species, and especially in case of L. tomentosa the optimal treatment time in sulfuric acid was one to two hours. Germination of L. tomentosa began promptly following the scarification and was completed within about one month, indicating that seeds has no physiological dormancy, just has physical dormancy.
It was reported that esthetic composite resin restoration reinforces the strength of remaining tooth structure with preserving the natural tooth structure. However, it is unknown how much the strength would be recovered. The purpose of this study was to compare the fracture resistance of three types of undermined cavity filled with composite resin with that of non-cavitated natural tooth. Forty sound upper molars were allocated randomly into four groups of 10 teeth. After flattening occlusal enamel, undermined cavities were prepared in thirty teeth to make three types of specimens with various thickness of occlusal structure (Group $1{\sim}3$). All the cavity have the 5 mm width mesiodistally and 7 mm depth bucco-lingually. Another natural 10 teeth (Group 4) were used as a control group. Teeth in group 1 have remaining occlusal structure about 1 mm thickness, which was composed of mainly enamel and small amount of dentin. In Group 2, remained thickness was about 1.5 mm, including 0.5 mm thickness dentin. In Group 3, thickness was about 2.0 mm, including 1 mm thickness dentin. Every effort was made to keep the remaining dentin thickness about 0.5 mm from the pulp space in cavitated groups. All the thickness was evaluated with radiographic Length Analyzer program. After acid etching with 37% phosphoric acid, one-bottle adhesive (Single $Bond^{TM}$, 3M/ESPE, USA) was applied following the manufacturer's recommendation and cavities were incrementally filled with hybrid composite resin (Filtek $Z-250^{TM}$, 3M/ESPE, USA). Teeth were stored in distilled water for one day at room temperature, after then, they were finished and polished with Sof-Lex system. All specimens were embedded in acrylic resin and static load was applied to the specimens with a 3 mm diameter stainless steel rod in an Universal testing machine and cross-head speed was 1 mm/min. Maximum load in case of fracture was recorded for each specimen. The data were statistically analyzed using one-way analysis of variance (ANOVA) and a Tukey test at the 95% confidence level. The results were as follows: 1. Fracture resistance of the undermined cavity filled with composite resin was about 75% of the natural tooth. 2. No significant difference in fracture loads of composite resin restoration was found among the three types of cavitated groups. Within the limits of this study, it can be concluded the fracture resistance of the undermined cavity filled with composite resin was lower than that of natural teeth, however remaining tooth structure may be supported and saved by the reinforcement with adhesive restoration, even if that portion consists of mainly enamel and a little dentin structure.

Studies on extraction and color characteristics of purple sweet potato (PSP) pigment were performed to provide the basic information for the utilization of PSP as a new source of natural food colorant. PSP pigment was extracted well with the polar solvents such as distilled water, ethanol, and methanol. but hardly extracted with the non-polar solvents. Among the tested solvents, 20% ethanol solution containing 0.1% citric acid was found to be the most efficient for extraction of the pigment from PSP. PSP contained high amount of pigment not only in the epidermis but also in the flesh of the potato. The PSP pigment was heat stable even under pretreatments such as autoclaving and blanching of the potato before extraction. The optimum temperature of the extraction for the PSP Pigment was decided to be $30^{\circ}C$ by considering the stability and the rate of extraction. The pigment was markedly influenced by the change of pH. The color of the pigment solution was red at the pH range of $1.0{\sim}3.0$, became blue at $7.0{\sim}8.0$, then turned green at $9.0{\sim}10.0$. A characteristic batho-chromic shift of the pigment solution was observed as the pH of the solution increased.

The mechanisms of aflatoxin $B_1(AFB_1)$ degradation by ammonia and alkaline pH during the storage of Korean soy sauces were studied. In the 0.05%, 0.1% and 0.5% of ammonia solutions, almost all of $AFB_1$(96-100%) was degraded after 2 to 24 hrs of incubation at $30^{\circ}C$. Increased levels of ammonia in both home made soy sauce(HMSS) and commercial soy sauce(CSS) caused slow increases in pH. The pH change was higher in CSS than in HMSS. The degradations of $AFB_1$ were not observed in the samples of HMSS, CSS, distilled water and 20 % of NaCl solution during the storage, however, when the pHs of the samples were adjusted to 10, the toxin was completely removed in all samples. $AFB_1$ was stable at pH 5 and 7 in bath buffer solutions and buffer solutions+0.2% ammonia, however, $AFB_1$ was degraded completely at pHs more than 9. $AFB_1$ was not degraded even at high concentrations of ammonia(0.2-1.0%) when the pH was maintained at 7 in the buffer solution. It indicated that ammonia content in the system was not important but the higher pH was the reason to degrade $AFB_1$. When the pHs of HMSS, CSS, buffersolution and buffer solution + 0.2% ammonia were adjusted to 5 and then reacted with $AFB_1$ for 5 days, the toxin was stable in all samples. However, when the pHs of the samples were adjusted to 7, about 60-70 % of $AFB_1$ was degraded in HMSS and CSS after 5 days of incubation during which the pH was not changed, but $AFB_1$ in the buffer solution and buffer solution + 0.2% ammonia was not degraded at all in the same conditions.
This in vitro study evaluated the influence of a flowable composite resin on the tensile bond strength of resin to enamel and dentin treated with Er:YAG laser and diamond bur. 96 Buccal enamel and mid-coronal dentin were laser-irradiated using an Er:YAG laser and treated with diamond bur. Each groups(48) were divided two small groups depends on acid-etching procedure. Light-cure flowable resin(Metafil Flo) and self-cure resin(Clearfil FII New Bond) were used in this study. After surface etching with 37% phosphoric acid and the application of an adhesive system, specimens were prepared with a hybrid composite resin. After 24hours storage in distilled water at 37$^{\circ}C$, all samples were submitted to the tensile bond strength evaluation, using a universal testing machine(Z020, Zwick, Germany). The obtained results were as follows: 1. TBS of acid-etching group were higher than those of non-etching group in both enamel and dentin treated with Er:YAG laser and diamond bur. Laser 'conditioning' was clearly less effective than acid-etching. Moreover, acid etching lased enamel and dentin significantly improved the microTBS of M-Flo. 2. In enamel, TBS of laser-irradiated group were lower than those of bur-prepared group. However, in flowable resin subgroup, there were not differed those between two groups in dentin. 3. In laser-treated group, TBS of flowable composite resin were higher than those of self-curing resin in dentin, however, there was no difference in enamel. From this study, we can conclude that the self- and light-cure composite resin bonded significantly less effective to lased than to bur-cut enamel and dentin, and that acid-etch procedure remains mandatory even after laser ablation. We suggest that Er:YAG laser was useful for preparing dentin cavity with flowable resin filling.

This study was carried out to examine the effects of vitamin E on chronic gastric ulcer induced by alcohol treatment in rats. Chronic gastric ulcer model was established by oral administration of 70% ethanol at one time and supply of 15% ethanol for additional 7 days. Male Sprague-Dawley rats, approximately 200 g, were fasted for 24 hours and orally gavaged with 1 mL of 70% ethanol for the induction of acute ulcer. A supply of 15% ethanol dissolved in distilled water for 7 days were followed to maintain chronic gastric ulcer. Acute ulcer group was sacrificed at 3 hours after oral administration of 1 mL of 70% ethanol. Chronic groups were divided into three groups according to vitamin E levels; low-vitamin E (LVE, 0 mg/mL oil/day), normalvitamin E (NVE, 1 mg/mL oil/day) and high-vitamin E (HVE, 10 mg/mL oil/day). These groups were fed vitamin E free diets which were made of vitamin E free vitamin mix followed AIN-93M pattern for 7 days. Histological findings of congestion, hemorrhage and necrosis in gastric tissue were shown severely in acute ulcer group and LVE group of chronic ulcer groups. The concentration of gastrin in serum was significantly higher in LVE group. The content of histamine in stomach was lower in acute ulcer group but there was no significant difference among the chronic groups regardless of vitamin E levels. Content of malondialdehyde (MDA) in gastric tissue was higher in HVE group and activities of antioxidant enzyme, glutathione peroxidase (GPx) and catalase, were lower in HVE group. Myeloperoxidase (MPO) activities as a marker of neutrophils infiltration was significantly higher in LVE group. These results suggested that vitamin E supplementation has positive effects on healing of alcohol-induced chronic gastric ulcer through alleviation of gastric tissue injuries and reduction of the MPO activity in gastric tissue and gastrin in serum.
Exogenous factor and nutrients affecting for conidial germination of Colletetrichum dematium f. sp. capsicum causing red pepper anthracnose were studied by slide germination test. Optimum temperature of conidial germination was at $28^{\circ}C$, ranging 15 to $35^{\circ}C$. Optimum pH was at 5.5, ranging 4.5 to 8.0, and more than 90% of relative humidity (RH) was optimum. Poor conidial germination of the fungus was observed on sterile distilled water, but potato sucrose broth (PSB), red pepper fruit broth (RPFB), green pepper fruit broth (GPFB) and pepper leaf broth (PLB) furnished a satisfactory nutrients for conidial germination. Exogenous supply of carbon and nitrogen sources were essential for conidial germination, while potassium, phosphorous and sulfur were not evident as that for carbon and nitrogen. Soluble starch was the most suitable as a carbon source for conidial germination and followed by D-glucose, D-galactose and lactose in that order. Maximum germination was attained in the $1{\times}10^4$ conidia per ml. Germination was decreased with increment of conidial concentration, and in the density of $5{\times}10^4$ conida per ml, germination was nearly supipressed. It suggested existing a self-inhibitor. Non-washed conidia germinated more than washed conidia, and conidial germination was also gradually decreased by increasing conidial density.
It is well known that most of the beverages on sale contain acid. Among these beverages, one of the most typically purchased products is Coca-Cola ($Coke^{(R)}$), which affects the demineralization of the tooth enamel surface. However other beverages also affect the erosion of the tooth enamel surface. Therefore, the aim of this study was to observe the influence that acid containing beverages have upon the tooth enamel surface. In this study, 30 enamel specimens were obtained from extracted maxillary third molars without cracks. The study measured the pH concentrations and the Ca and P concentration levels of the acidic beverages. The control without exposure was kept in distilled water, and each enamel specimen was stored for 30-min, 60-min, and 120-min. The erosion effect of the enamel surface was observed by SEM. The results obtained from this study can be summarized as follows: 1. The pH concentration of acidic beverages, without milk, was 3.105 on average. The beverages eroded the enamel surface and the erosion degree was determined by the pH concentration of beverages. 2. The erosion degree significantly increased by as the exposure time increased. The difference in the erosion degree between the 30-min group and the 60-min and the 120-min group was significant, but the erosion degree of the 120-min group was significantly less than that of the initial group. 3. When compared with the beverages containing similar pH concentrations, the demineralization degree of the beverages containing higher concentrations of Ca and P was lower.
This study was to separate wild yeast from the Houttuynia Cordata Thunb(HC) extracts fermented in traditional way and investigate the characteristics of fermentation from the HC extracts. As a result, the longer the cultivation time, the more the contents of alcohol in $15^{\circ}Brix$ sugar solutions increased. When it reached to 90 hours since it cultivated, it ranked HCE 12%, HCD 11.2%, and HCA 10.5% in order. As for HCF, HCG, HCB, and HCC), this study has shown that the contents of alcohol were from 7.5% through 8.5%. As a result of selecting germ strains like HCE and HCD with the highest alcohol-genicity in the sugar solutions as separated yeast from HC and of comparing and reviewing the existing Saccharomyces germ strains and fermentation power in the medium of HC(Juice extraction of HC : Distilled water(1:1), this study has found out that 'pH' decreased from $4.09{\sim}4.12$ before fermentation through $3.57{\sim}3.78$ after fermentation. And, the sugar concentration decreased from $15.00{\sim}15.19^{\circ}Brix$ through $7.75{\sim}9.57^{\circ}Brix$ before fermentation. Also, the acid value increased from $0.138{\sim}0.210%$ before fermentation through $0.282{\sim}0.45%$ after fermentation. In addition, as the contents of alcohol became $3.6{\sim}4.6%$ after fermentation, isolates HCD and HCE from HC had higher value rather than ones of DJ97, YJK, R12, and RCY separated from persimmons, apples, and grapes. The result value of color was minimum $4.75{\pm}1.44$ and maximum $6.85{\pm}1.63$, and HCE marked the highest record among the items of sensory evaluation. The overall acceptability was in normal level like minimum $3.95{\pm}1.17$ and maximum $5.00{\pm}1.41$. It is considered as it could lower sensory evaluation because the acceptability of flavor was not satisfied. After all, the significance(p<0.05) among the germ strains was not recognized in aspects of color, flagrance, flavor, and overall acceptability.
This study was conducted to investigate the effects of various heat treatments on the microbial reduction and germination of red radish seeds for the development of effective and economical sterilization methods of improving microbial safety without reducting the germination rate. Hydrothermal treatment was conducted at 60, 65, 70, 75, 80, and $90^{\circ}C$ for 30 and 60 seconds, and dry heat treatment was performed at 70, 80, 90, and $100^{\circ}C$ for 5 minutes. In the seeds that underwent the hydrothermal treatment, time had little effect on the microbial reduction. There was no significant microbial reduction over time. However, there was significant microbial reduction as temperatures increased (p<0.001). The total plate count (TPC) was reduced by more than 3 logs, and Listeria monocytogenes was not detected at temperatures above $70^{\circ}C$. In the seeds that were subjected to the dry heat treatment, the TPC and the population of the L. monocytogenes were significantly reduced as the temperatures increased (p<0.001). After treatment at $100^{\circ}C$ for 5 minutes, the TPC and the L. monocytogenes were reduced by 3 logs. As with the microbial reduction, time had little effect on the germination. There were no significant changes in the germination after the hydrothermal treatment over time; but at the temperatures above $75^{\circ}C$, the germination rate significantly decreased as the temperature increased (p<0.001). When the seeds were soaked after the hydrothermal treatment, their germination was stimulated. The dry heat treatment at temperatures of $80^{\circ}C$ and higher significantly decreased the germination rate as the temperature increased (p<0.001). Dry heat treatment before the germination of the seeds soaked in distilled water for three hours significantly decreased the germination at temperatures greater than $90^{\circ}C$ (p<0.05). This study showed that appropriate heat treatments can increase the microbiological safety and germination of red radish seeds.
