Rheological Properties of Konjac Glucomannan Dispersons (구약감자 Glucomannan 현탁액의 유동특성)

  • Konjac(Amorphophallus konjac) glucomannan dispersions were prepared from konjac flour, pretreated konjac flour and purified glucomannan. Konjac glucomannan dispersions showed non-Newtonian fluid behavior without yield stress and higher shear stress at fixed shear rate than the dispersions of gum xanthan, gum carrageenan and sodium alginate. While temperature increased, shear stress at fixed shear rate of konjac glucomannan dispersion steadily decreased. The apparent viscosity of konjac glucomannan dispersion was in its maximum at pH 7, whereas decreased on the outskirts of pH 7. The change in apparent viscosity was not found up to 1% sodium chloride addition in case of konjac glucomannan dispersion. However, the apparent viscosity of konjac glucomannan dispersion decreased up to sugar addition of 10%, afterwards increased.

Dissolution Characteristics of Biphenyl Dimethyl Dicarboxylate from Solid Dispersions and Permeation through Rabbit Deuodenal Mucosa (고체분산체로부터 비페닐디메칠디카르복실레이트의 용출 특성 및 토끼의 십이지장 점막 투과)

  • To increase the dissolution rate of practically insoluble biphenyl dimethyl dicarboxylate (DDB), various solid dispersions were prepared with water soluble carriers, such as povidone (PVP K-30), poloxamer 407, sodium deoxycholate (SDC) and polyethylene glycol (PEG) 6000, at drug to carrier ratios of 1:3, 1:5 and 1:10 (w/w) by solvent or fusion method. Dissolution test was performed by the paddle method. The dissolution rate of DDB tablets (25 mg) on market was found to be very low (11.44, 9.02 and 6.42% at pH 1.2, 4.0 and 6.5 after 120 min, respectively). However, dissolution rates of DDB from various solid dispersions were very fast and reached supersaturation within 10 min. DDB-PEG 6000 solid dispersion appeared to be better in enhancing the in vitro dissolution rate than others. Furthermore, the incorporation of DDB and phosphatidylcholine (PC) into ${\beta}-cyclodextrin$ at ratios of 1:2:20, 1:5:20 and 1:10:20 resulted in a 4.9-, 11.2- and 19.6-fold increase in DDB dissolution after 120 min as compared with the pure drug, respectively. This might be attributed to the formation of lipid vesicles which entrapped a certain concentration of DDB during dissolution. On the other hand, the permeation of DDB through rabbit duodenal mucosa was examined using some enhancers such as SDC, sod. glycocholate (SGC) and glycyrrhizic acid ammonium salt (GAA). Only trace amounts of DDB were found to permeate through deuodenal mucosa in the absence of enhancer. SDC was found to markedly decrease the permeation flux of DDB, however, SGC and GAA (5 mM) enhanced the flux of DDB 1.6 and 2.4 times higher as compared with no additive, respectively.

  • Solid dispersion, defined as the dispersion of one or more active ingredient in a carrier or matrix at solid state, is an efficient strategy for improving dissolution of poorly water-soluble drugs for enhancement of their bioavailability. Compared to other conventional formulations such as tablets or capsules, solid dispersion which can be prepared by various methods has many advantages. However, despite numerous studies which have been carried out, limitations for commercializing these products remain to be solved. For example, during the manufacturing process or storage, amorphous form of solid dispersion can be converted into crystalline form. That is, the dissolution rate of solid dispersion would continuously decrease during storage, resulting in a product of no value. To resolve these problems, studies have been conducted on the effects of excipients. In fact, modification of the solid dispersions to overcome these disadvantages has progressed from the first generation to the recent third generation products. In this review, an overview on solid dispersions in general will be given with emphasis on the various manufacturing processes which include the use of polymers and on the stabilization strategies which include methods to prevent crystallization.

Preparation and Characterization of Poly(ethylene glycol) Based Pranlukast Solid Dispersion (친수성 Poly(ethylene glycol)을 이용한 프란루카스트 고체분산체의 제조 및 특성 분석)

  • In this study, poly(ethylene glycol) (PEG) was used as a hydrophilic polymer carrier to develop solid dispersion formulations for enhancing solubility and dissolution rate of pranlukast, one of poorly soluble drugs that has been broadly used for the treatment of asthma. PEG based solid dispersions with or without poloxamer were prepared by hot melting and solvent evaporation methods. The resultant solid dispersions were characterized by DSC and powder X-ray measurements, and their morphological properties were observed to be partially changed to amorphous state with reduced crystallinity. Dissolution and solubility tests showed that the solubility and dissolution rate of the solid dispersions were significantly enhanced. The solid dispersion formulation prepared by the hot melting method with a chemical composition of pranlukast:PEG:poloxamer = 1:5:1 demonstrated the most enhanced solubility and dissolution rate. The results suggest that the solid dispersions based on PEG and poloxamer are promising systems for the enhancement of solubility and bioavailability of pranlukast.

  • We studied a low density 8CB liquid crystal-hydrophobic aerosil dispersions imbedded in submicron-size cylindrical pores. The nanosize spherical aerosil particles are adsorbed at the pore wall and hinder the planar anchoring. The adsorption is temperature dependent, and an axial to radial molecular configuration transition occurs within the cylindrical pores.

  • This study describes a simple and effective experimental determination of pump-power dependence of refractive indices and dispersions of erbium-doped fibers (EDFs) of various doping concentrations. Systermatical analysis on light-induced change of the refractive indices and dispersion for a signal wavelength range of 1510 nm-1560 nm with a pump of 980 nm wavelength has been made by measuring the pump-induced phase changes of the signal beam in Mach-Zehnder type interferometer, which has an optically pumped EDF and a reference EDF in each of the two arms, respectively. The measured pump-induced refractive indices match well with the theoritically predicted ones. The results show that pump-induced refractivity and dispersion changes of the EDF increase with the increasing pump power and $Er^{3+}$-doping concentration as expected from theory.

  • The rheological properties of exopolysaccharide, p-KG03, produced by marine microalgae Gyrodinium impudicum strain KG03 had been studied. The intrinsic viscosity of this p-KG03 was calculated to 65.22 and 50.75 $d{\ell}/g$ using Huggins and Kramer equations (xanthan gum 24.41 and 24.03). Aqueous dispersions at p-KG03 concentrations ranging from 0.1 to 1.0 % (w/w) showed marked shear-thinning properties as Power-Law behavior. In aqueous dispersions of p-KG03 1.0 %, consistency index (K) and flow behavior index (n) were 2,172 and 0.52. The apparent viscosity and the influence of shear rate on different conditions as p-KG03 concentrations, pH, NaCl, $CaCl_2$ and temperature in aqueous solutions were measured. And p-KG03 had mixed with aqueous solutions of xanthan gum and gellan gum, and invested the change of mixed aqueous solution behavior.

  • Virtual hosting is a typical service to connect each directory of site and domain name. If traffic amounts may increase at one site present in the server, then it affects traffic amounts of other sites as well (including the sites which have flew requests). To overcome this problem, we suggest a simple feedback-control concept for the system by periodically monitoring the traffic and properly actuating traffic dispersions by investigating the log file. Specifically, large files are to be served in a backup server (to reduce the workload of the main server) by changing their own URL's in html format. In other words, it automatically redistributes the workload by using the URL. Furthermore, we also use the redirecting method by just adding html tags to html header. This method efficiently handles the workload and maintains the capability of the server effectively to the varying workload.

Preparation of Waterborne Polyurethane Dispersion Based on Siloxane Polyal (실록산 폴리올의 도입에 따른 수분산성 폴리우레탄의 제조)

  • Waterborne polyurethane dispersions (WPUD) were prepared by poly(ethylene glycol) adipate as the polyester type, ${\alpha},{\omega}-hydroxyalkyl$ terminated polydimethylsiloxane (PDMS-diol) as the polysiloxane type, hexamethylene diisocyanate, and isophorone diisocyanate, dimethylol propionic acid. The effects of PDMS-diol contents on the particle size, thermal and surface properties of WPUD were investigated. The structures of the synthesized WPUD were confirmed using by FT-IR. The surface, thermal and mechanical properties were investigated by measuring the contact angles, DSC, TGA and UTM. As PDMS-diol contents increased, the particle size, the contact angle, and the elongation was increased, while the tensile strength was decreased. Also the thermal stabilities of the synthesised WPUD were increased as PDMS-diol contents increased.
