Typing of Extended-Spectrum ${\beta}$-Lactamase (ESBL) Producing Enterobacteriaceae Isolated from Slaughterhouse in Pusan, Korea (부산 도축장에서 분리된 광범위 베타 락탐 분해효소(Extended-Spectrum ${\beta}$-Lactamase, ESBL)생성 장내세균의 형별분류)

  • The emergence of extended spectrum ${\beta}$-lactamase producing bacteria is causing very serious problems in Korea. Although there have been many reports about these bacteria Isolated from patients and clinical specimens, there is no report of extended spectrum ${\beta}$-lactamase producing organisms isolated from natural environment in Korea. This study was conducted to investigate the biological characteristics and extended spectrum ${\beta}$-lactamase types of eighteen strains of extended spectrum ${\beta}$-lactamase producing Klebsiella pneumoniae and Escherichia coli isolated from a slaughterhouse in Pusan in Korea during 2002 to 2004. Extended spectrum ${\beta}$-lactamases were identified by double-disk synergy test, conjugation, isoelectric focusing values and gene sequencing. Eight strains of Klebsiella pneumoniae and two strains of Eschericha coli were isolated kom pigs and transferred extended spectrum ${\beta}$-lactamase genes to recipient Escherichia coli J53 (sodium azide resistant and ceftazidime senstive) strain by conjugation. The conjugants of extended spectrum ${\beta}$-lactamase genes were alignments and translated to amino acids by BCM and NCBI blast. Eight conjugants of Klebsiella pneumonae were typed TEM-52, and two strains of Escherichia coli, SHV-12, but CMY-1 type were not detected in this study.

A Study on Soot Formation in Premixed Constant-Volume Combustion at High Pressures (高壓下의 定積 豫混合氣燃燒에 있어서 煤煙생成에 關한 硏究)

  • The effect of pressure on soot formation in premixed propane-air combustion is investigated at high pressures over the pressure range of 1 to 5 MPa by using a specially designed constant volume combustion bomb. The combustiom chamber of disk type with eight spark plugs located on the circumference at an interval of 45deg is 100mm in diameter by 14mm thick. The end gases are compressed to high pressures by the eight converging flames. The soot volume fraction in the chamber center during the final stage of combustion at the highest pressure is measured by the in-situ laser extinction technique, and the burnt gas temperature during the same period is measured by the two-color method. It is found that the soot yield rises with 50 to 100% for the respective equivalence ratio range of 1.9-2.2 at an interval of 0.1 when the combustion pressure is increased from 1 to 5 MPa, and that the turbulent flames decrease in the soot yield as compared with the laminar flames because the burnt gas temperatures increase with the drop of heat loss.

  • As the demand for slim laptops requires low-height optical disc drives, vibration problems of optical disc drives are of great concern. Additionally, with the decrease of a track width and a depth of focus in high density drives, studies on vibration resonance between mechanical parts become more important. From the vibration point of view, the performance of optical disc drives is closely related with the relative displacement between a disc and an objective lens which is controlled by servo mechanism. In other words, to read and write data properly, the relative displacement between an optical disc and an objective lens should be within a certain limit. The relative displacement is dependent on not only an anti-vibration mechanism design but also servo control capability. Good servo controls can make compensation for poor mechanisms, and vice versa. In a usual development process, robustness of the anti-vibration mechanism is always verified with the servo control of an objective lens. Engineers partially modify servo gain margin in case of a data reading error. This modification cannot correct the data reading error occasionally and the mechanism should be redesigned more robustly. Therefore it is necessary to verify a mechanism with respect to the possible servo gain plot. In this study we propose the experimental verification method for anti-vibration mechanism with respect to the existing servo gain plot. Thismethod verifies axial vibration characteristics of optical disc drives on the basis of transmissibility. Using this method, we verified our mechanism and modified the mechanism for better anti-vibration characteristics.

Bond strength of resin cement to $CO_2$ and Er:YAG laser-treated zirconia ceramic

  • Objectives: It is difficult to achieve adhesion between resin cement and zirconia ceramics using routine surface preparation methods. The aim of this study was to evaluate the effects of $CO_2$ and Er:YAG laser treatment on the bond strength of resin cement to zirconia ceramics. Materials and Methods: In this in-vitro study 45 zirconia disks (6 mm in diameter and 2 mm in thickness) were assigned to 3 groups (n = 15). In control group (CNT) no laser treatment was used. In groups COL and EYL, $CO_2$ and Er:YAG lasers were used for pretreatment of zirconia surface, respectively. Composite resin disks were cemented on zirconia disk using dual-curing resin cement. Shear bond strength tests were performed at a crosshead speed of 0.5 mm/min after 24 hr distilled water storage. Data were analyzed by one-way ANOVA and post hoc Tukey's HSD tests. Results: The means and standard deviations of shear bond strength values in the EYL, COL and CNT groups were $8.65{\pm}1.75$, $12.12{\pm}3.02$, and $5.97{\pm}1.14MPa$, respectively. Data showed that application of $CO_2$ and Er:YAG lasers resulted in a significant higher shear bond strength of resin cement to zirconia ceramics (p < 0.0001). The highest bond strength was recorded in the COL group (p < 0.0001). In the CNT group all the failures were adhesive. However, in the laser groups, 80% of the failures were of the adhesive type. Conclusions: Pretreatment of zirconia ceramic via $CO_2$ and Er:YAG laser improves the bond strength of resin cement to zirconia ceramic, with higher bond strength values in the $CO_2$ laser treated samples.

ASYMMETRIC LIGHT CURVES OF BLACK HOLE BINARIES AND THE DOPPLER BEAMING EFFECT (블랙흘 쌍성계의 비대칭 광도 곡선과 DOPPLER BEANING 효과)

  • Black hole binary candidates are known to be composed of a black hole with 10 $M_{\odot}$ and a K or M type companion. Because the companion is believed to fill the Roche lobe that is very aspherical, the light curves of black hole binaries are characterized by an ellipsoidal variation. It has been known that the ellipsoidal light curves exhibit asymmetric maximum brightness at the orbital phases 0.25 and 0.75, which has been attributed to star spots or the hot impact points of the accretion flow on to the accretion disk around the black hole. In this paper, it is pointed out that the special relativistic beaming effect contributes to the asymmetry of several percent often observed in the light curves. The typical orbital velocity 400 km $s^{-1}$ observed in black hole binaries may induce the temperature difference $\DeltaT/T$ ~ 1/400 of the late type companion star in the observer`s rest frame, because of the special relativistic Doppler beaming effect. This difference in temperature can result in several per cent of brightness sensitively dependent on the wavelength band, which is comparable to what has been observed in most black hole binary candidates. Considering the significant contribution of the special relativistic Doppler beaming effect, we conclude that the estimation of the sizes and temperatures of the star spots or the hot impact point needs serious revision.

Synthesis of Mg2Ni by mechanical alloying and its electrochemical characteristics for Ni-MH secondary battery (Ni-MH 2차 전지용 Mg2Ni의 기계적 합금화법에 의한 제조 및 전기화학적 특성)

  • The $Mg_2Ni$ hydrogen storage alloys which have much higher theoretical discharge capacity than $AB_5$ and $AB_2$ type alloys were synthesized by mechanical alloying with some additives and subjected to the electrochemical measurements. Two different processes were employed to the synthesis of $Mg_2Ni$ alloys with using the high energy ball mill SPEX 8000. One was only ball milling, 12 hrs, the Mg and Ni powders for 12 hrs with additives such as $AB_5$, Ni, Co and Cu powders. In the other process the Mg and Ni powders were ball milled for 1 hr first and then heat treated at $300{\sim}400^{\circ}C$ for 1 hr to get $Mg_2Ni$ alloy, and finally the $Mg_2Ni$ alloy powders were ball milled with the additives for 12 hrs. The alloy powders prepared were compacted at room temperature under $7.64tons/cm^2$ into disk type electrodes for the electrochemical measurements. The experimntal results showed that the electrodes prepared with the heat treated alloy powders had a higher discharge capacities than those without heat treatment. The addition of Ni caused an increase of the discharge capacity and the addition of Co improved the cycling characteristics. The electrode prepared by ball milling of $Mg_2Ni$ and 10wt% Ni powders has showed the highest discharge capacity, 546mAh/g.alloy, which was 55% of the theoretical capacity.

Reaction Characteristics of Combined Steam and Carbon Dioxide Reforming of Methane Reaction Using Pd-Ni-YSZ Catalyst (Pd-Ni-YSZ 촉매를 이용한 수증기-이산화탄소 복합개질 반응 특성)

  • In this study, the reaction characteristics of combined steam and carbon dioxide reforming of methane (CSCRM) reaction using Pd-Ni-YSZ catalyst were investigated according to types of catalysts and gas compositions. Catalysts were prepared in the form of powder and porous disk. The injected gases were supplied at different ratios of $CH_4/CO_2/H_2O$. As a result, the conversion of $CH_4$ and $CO_2$ was improved as a result of using the porous disc type catalyst as compared with that of the powder type catalyst. When the $CH_4/CO_2/H_2O$ ratio of the feed gas was 1 : 0.5 : 0.5, the $H_2/CO$ ratio was adjusted close to 2. However, after 6 hours of the reaction, $CH_4$ conversion was partially reduced by the carbon deposition and the pressure drop increased from 0.1 to 0.8. This issue was then solved by optimizing the water content. As a result, it was confirmed that the durability was secured by preventing the carbon deposition when the gas was supplied at a $CH_4/CO_2/H_2O$ ratio of 1 : 0.5 : 1, and the conversion rate was maintained at a relatively high level.

Clonal Dissemination of Multidrug Resistant Acinetobacter baumannii Isolates Harboring blaOXA-23 at One University Hospital in Daejeon, Korea (대전지역 소재 대학병원에 blaOXA-23 유전자를 가지고 있는 다제내성 Acinetobacter baumannii의 확산)

  • Acinetobacter species isolates are important opportunistic pathogens and commonly implicated in nosocomial infections. The therapeutic options for treatment of the bacterial infections are limited because the bacteria isolates are usually multidrug resistant (MDR). In the current study, we investigated various carbapenemase genes in 68 Acinetobacter species isolates. Antimicrobial susceptibilities were tested using the disk diffusion method. Screening of carbapenemase genes was performed via multiplex PCR. In addition, PCR and DNA sequencing were used to identify the carbapenemase genes. Repetitive extragenic palindromic-PCR (REP-PCR) was also performed to assess the clonality of isolates. In our study, A. baumannii isolates were highly resistant to all agents tested while all non-A. baumannii isolates were susceptible to all agents tested, with the exception of aztreonam and cefotaxime. All 51 A. baumannii isolates contained the $bla_{OXA-51}$ gene and 37 (72.5%) isolates also harbored the $bla_{OXA-23}$ gene. In addition, 39 MDR A. baumannii isolates were identified in our study and 37 isolates contained the $bla_{OXA-23}$ gene. The 37 MDR strains harboring $bla_{OXA-23}$ showed type I (n=22) or type II (n=15) banding patterns on their REP-PCR profiles. Our results suggest clonal relation and horizontal spreading of MDR A. baumannii isolates containing the $bla_{OXA-23}$ gene at the hospital located in Daejeon. Continuous investigation of antimicrobial resistant determinants and monitoring emergence and dissemination of MDR isolates is required to prevent and control infection and colonization of MDR A. baumannii isolates.

AN IN-VITRO WEAR STUDY OF DENTAL PORCELAINS AND HUMAN ENAMEL (치과용도재에 의한 법랑질 마모에 관한 연구)

  • Dental porcelain is one of the materials of choice for restoration where esthetics is of concern, but has a considerable potentials of wear. The wear of enamel is variable when opposed by different porcelain systems and surface conditions, and the exposed dentine and opaque porcelain due to clinical failure is expected to have high potentials of wear. The purpose of this study was to investigate the wear effects of self-glazed, polished incisal porcelain. polished dentine and opaque porcelain against human enamel in the laboratory by use of a pin-on-disk type wear tester. 4 types of dental porcelain($Vita-{\Omega}$, Ceramco-II, Vintage powder $Vita-{\alpha}$ of In-Ceram system) and type IV gold alloy as cotrol group were used for test specimens. Intact buccal cusps of maxillary premolar were used for enamel specimens, and the cusp converged to a point and was devoid of visible abrasion, caries, decalcification. The upper part was the cusp of a maxillary premolar and the lower part was a porcelain specimen. The enamel wear was deter-mined by weighing the cusp before and after each test. Surface profilometer was used to quantitate wear of the porcelain specimens. Vicker's hardness tester was used to evaluate the surface hardness of test specimens. The SEM was used to evaluate the wear surfaces. The results were as follows : 1 Self-glazed porcelain produced more enamel wear than polished porcelain, especially the enamel wear of $Vita-{\alpha}$ self-glazed porcelain was 3.2 times more than that of other groups. 2. Opaque porcelain produced least porcelain wear, $Vita-{\alpha}$ self-glazed porcelain produce greatest porcelain wear, but there was no statistically significant difference between the groups(p>0.05). 3. The enamel wear of dentine porcelain was 3.8 times more than that of polished inisal porcelain(p<0.05), and the enamel wear of opaque porcelain was 1.9 times more than that of polished inisal porcelain, but there was no statistically significant difference between the groups(p>0.05) 4. Overglazed porcelain produced less enamel wear than self-glazed porcelain, and more enamel wear than polished porcelain, but there was no statistically significant difference between the groups(P>0.05). 5. The hardness number of $Vita-{\Omega}$ dentine and Ceramco-II opaque porcelain was larger, but that of Vintage dentine and $Vita-{\alpha}$ self-glazed porcelain was similar to other groups. 6. Examination of SEM photographs revealed that overglazed porcelain had smoother surface than self-glazed porcelain, and self-glazed porcelain had smoother surface than polished porcelain. Much polishing scratches and larger porosities were observed on the opaque porcelain specimen, and much polishing scratchess and small porosities were observed on the dentine porcelain specimen.

Current Status of the KMTNet Active Nuclei Variability Survey (KANVaS)

  • Multi-wavelength variability is a staple of active galactic nuclei (AGN). Optical variability probes the nature of the central engine of AGN at smaller linear scales than conventional imaging and spectroscopic techniques. Previous studies have shown that optical variability is more prevalent at longer timescales and at shorter wavelengths. Intra-night variability can be explained through the damped random walk model but small samples and inhomogeneous data have made constraining this model hard. To understand the properties and physical mechanism of intra-night optical variability, we are performing the KMTNet Active Nuclei Variability Survey (KANVaS). Using KMTNet, we aim to study the intra-night variability of ~1000 AGN at a magnitude depth of ~19mag in R band over a total area of ${\sim}24deg^2$ on the sky. Test data in the COSMOS, XMM-LSS, and S82-2 fields was obtained over 4, 6, and 8 nights respectively during 2015, in B, V, R, and I bands. Each night was composed of 5-13 epoch with ~30 min cadence and 80-120 sec exposure times. As a pilot study, we analyzed data in the COSMOS field where we reach a magnitude depth of ~19.5 in R band (at S/N~100) with seeing varying between 1.5-2.0 arcsec. We used the Chandra-COSMOS catalog to identify 166 AGNs among 549 AGNs at B<23. We performed differential photometry between the selected AGN and nearby stars, achieving photometric uncertainty ~0.01mag. We employ various standard time-series analysis tools to identify variable AGN, including the chi-square test. Preliminarily results indicate that intra-night variability is found for ~17%, 17%, 8% and 7% of all X-ray selected AGN in the B, V, R, and I band, respectively. The majority of the identified variable AGN are classified as Type 1 AGN, with only a handful of Type 2 AGN showing evidence for variability. The work done so far confirms there are more variable AGN at shorter wavelengths and that intra-night variability most likely originates in the accretion disk of these objects. We will briefly discuss the quality of the data, challenges we encountered, solutions we employed for this work, and our updated future plans.
