Method development for efficacy testing of veterinary disinfectants using bacteriophage MS2 (Bacteriophage MS2를 이용한 소독제 효력시험 확립에 관한 연구)

  • In virucidal efficacy testing, the chemical inactivation cannot be determined for all viruses due to the difficulties or the inability to culture sufficiently or the risk of exposure to the viruses. Therefore, disinfectants against these viruses could be evaluated by different methods and surrogate viruses are used as alternative. In this study we developed a method for efficacy testing of veterinary disinfectants using one of the candidate surrogate viruses, bacteriophage MS2, as part of the research on the selection of surrogate viruses for efficiency of efficacy testing of veterinary disinfectants. This method is based on the Animal and Plant Quarantine Agency (APQA) guidelines for efficacy testing of veterinary disinfectants. Bacteriophage and disinfectant are reacted in suspension in accordance with the APQA guidelines and then a newly established double agar layer method is applied for the efficacy test. The double agar layer method is summarized as follows: 1) The bottom agar with 1.5% agar is boiled and cooled before poured into petri dishes at volume of 20 mL, and dried under biological safety cabinet. 2) The top agar with 0.7% agar is boiled and kept at 50℃ before E. coli culture was seeded. 3) The serially diluted bacteriophage MS2-disinfectant mixtures 0.05 mL and E. coli host 0.01 mL (OD600 0.2~0.3) are mixed with 5 mL of top agar and incubate them at 50℃ for 5 min for reaction. 4) The resulting mixture is poured over top of a bottom agar plate and rocked sufficiently to ensure that the top agar covers the entire surface of the bottom agar. 5) The double agar layer is then placed under biological safety cabinet to allow the agar layer to solidify and subsequently incubated at 37℃ for 24 hr. 6) Following incubation, the plates may be inspected for plaques and record results.

Survey on Practical use of Sanitizers and Disinfectants on Food Utensils in Institutional Foodservice (단체급식소의 기구등의 살균소독제 사용실태 조사)

  • This study surveyed on the actual conditions of using sanitizers and disinfectants for improvements of sanitization on food utensils at 105 school and 20 industry foodservice operations. The questionnaire which was administered to 125 foodservices was used as a mail or visitation method. The answers of asking "Perception on temporary authorization system of sanitizers and disinfectants on food utensils" were 75% in contract managed school foodservices, 81.8% in self operated school foodservices, and 50% in industry. Main factors to choose sanitizers were sterilizing power (38.6%, 28.6%, 38.9%) and safety (32.6%, 46.1%, 33.3%) at every foodservices. Keeping ratio of sanitizers and disinfectants guidelines in contract managed school, self operated school and industry foodservices were 64.8%, 52% and 73.7%, respectively. If easy and practical guideline is developed, most foodservices replied to use if for disinfection of foodservices. Most of the foodservices were not only knowing sanitizers and disinfectants but also possessing a guideline. However, they didn't perform disinfection according to the guideline due to its complexity. Consequently, we suggest that it is necessary to provide an easy and practical "sanitizers and disinfectants guideline" and useful information.

Application of 'Sponge Model' with Disinfectants for the Inhibition of Listeria monocytogenes (Listeria monocytogenes의 증식억제를 위한 살균제 'Sponge model'의 응용)

  • The antimicrobial effects of two disinfectants commonly used in food industry on Listeria monocytogenes ATCC 15313 were studied. The two disinfectants tested were commercial benzalkonium chloride (BAC) and sodium hypochlorite (NaOCl). Their effects were studied on cells suspended in disinfectants (in vitro) and in the sponge model with the disinfectants (in vivo). When cells were exposed to $0\~0.1\%$ BAC and $0\~150\;ppm$ NaOCL for 20 minutes, BAC and NaOCl concentration more than $0.25\%$ and 100 ppm showed the antimicrobial effects respectively. This organism decreased rapidly during the first $0.5\~1$ minute followed by a slower decrease during the rest of the exposure time. Fifteen ml of cell solution $(about\;10^7\;CFU/ml\;in\;the\;TSB)$ was mixed with 15 ml of disinfectants in the sponge $(6.0{\times}4.0{\times}4.0cm)$, BAC and NaOCl concentration more than $0.1\%$ and 300 ppm showed the antimicrobial effects, and at $0.25\%$ and 800 ppm diminished in cell numbers 3-log cycles during the first 20 minutes. In the case of sponge model, 15 ml of cell solution and 15 ml of disinfectants $(0.25\%\;of\;BAC,\;800\;ppm\;of\;NaOCl)$ were suspended in the sponge during 20 minutes, washing with 200 ml of sterilized distilled water, and this sponge was transfered in the 100 ml TBS, and then incubated at various temperature. The cells were increased about 1-log cycle during 24 hrs at $5\~15^{\circ}C$. And the others temperature, the cells growth was in proporation to storage tepmerature and the cells were about $10^9\;CFU/ml$ after $1\~3$ days incubations.

Impact of Water Quality Parameters on the Disinfection of Total Coliform with Chlorine Dioxide

  • This study investigated the inactivation of the total coliform, an indicator organism in chlorine dioxide, in order to assess the optimal disinfection procedure for drinking water treatment and distribution systems. This research focus on a number of factors, including the dosage of disinfectant, contact time, pH, temperature and DOC. Water samples were taken from the outlet of a settling basin at a conventional surface water treatment system. As the pH increased in the range of pH 6-9, the bactericidal effects of disinfectants decreased. Changes in levels of pH did not significantly after the disinfection effect of chlorine dioxide for total coliform. With an increase in temperature, there was a subsequent increase in the bactericidal effects of disinfectants. Thus, it is evident that a decrease in temperature will higher the CT values required to inactivate coliform for during the winter. DOC addition can also reduce total coliform inactivation. DOC is the most significant variable in total coliform inactivation with chlorine dioxide.

Hygienic Aspects of Campylobacter Enteritis (Campylobacter장염에 의한 식품위생학적인 고찰)

  • Campylobacter jejuni has been recognized as one of the causes of human gastroenteritis. The feces of a variety of reservoir animals contain c. jejuni as commensals in the intestinal tracts, and are fundamental source of contamination. The intestinal organisms contaminate carcasses, equipment tools hands of the processing line workers and air of the processing facility. Once the contamination happens in the slaughterhouses or the meat processing facilities, it is very difficult to keep the carcasses free from the infection of c. jejuni. Various disinfectants are effective in minimizing the number of Campylobacter infections in the processing facilities by washing contaminated carcasses, tools, and hands. Direct contact with infected animals has been incriminated in transmission of infection caused by C. jejuni. Freezing, cooling with dry air and gamma irradiation are an effective way for preserving the meat and eliminating the transmission, but broad and enforced studies are needed for the practical use.

Efficacy of Disinfectants against Cytoplasmic Polyhedrosis Virus and Microspordia of Tasar Silkworm, Antheraea mylitta D.

  • Bleaching powder solution (1 to $5\%$), slaked lime solution (0.1 to $0.5\%$) and formalin (1 and $2\%$) were tested for their efficacy against cytoplasmic polyhedrosis virus and Nosema mylittansis spores to control virosis and pebrine respectively in tasar silkworm, Antheraea mylitta in indoor rearing condition. All the disinfectants tested were found effective in suppressing the infection of virosis and pebrine significantly. Complete inactivation of Antheraea mylitta cytoplasmic polyhedrosis virus (AmCPV) was recorded when treated with $4\%$ bleaching powder, $0.4\%$ slaked lime for 20 min and $2.0\%$ formalin for 30 min. Similarly treatments of $3.0\%$ bleaching powder solution for 20 min and $2.0\%$ formalin for 30 min were found effective in complete inactivation of N. mylittanis spores.

A Study on the Degree of Contamination of Air and Materials in a Hospital (병원내 공기오염과 물품의 항균에 대한 상태조사)

  • The incidence of hospital infection has been seriously increased in the general hospital in recent years. This study was performed on hospital air and materials in a General Hospital in Seoul from June to December in 1980. The results were as follows: 1. Air sampling was done in multiple strategic areas by exposing standard petridishes for 5 minutes. There was a significant difference of airborne microbe between places. ($F._{99}$ = 3.2, p < 0.01). 2. The mean colony count was 8.6$\pm$6.2 colonies / plate / 5 minutes. 3. Gram stains of colony in air sampling were Gram (+) cocci 66.5%. Gram (+) rod 18.4%, Gram (-) cocci 1.3%, Gram (-) rod 8.7% Fungus 4.5%. 4. For the evaluation of sterilization of steam sterilizer and ethylene oxide gas sterilizer, biological monitoring were done by commercial spore strip. Positive culture was obtained in 2 out of 41 tests on 3 steam sterilizers, and in 3 out of 13 tests on ethylene oxide gas sterilizer. 5. Product sampling and culture were done for 2 kinds of disinfectants and 30 sets of various operation package or dressing materials. Positive culture was obtained in one disinfectants.

Fast Screening of Harmful Disinfectants in Household Products via Low-Temperature Plasma Ionization-Mass Spectrometry

  • Isothiazolinone derivatives are widely used in consumer products as disinfectants or preservatives, but there are growing concerns about their impact on human health. Therefore, rapid screening of these biocides is very important for proper control and regulation of potentially hazardous substances. To this end, low-temperature plasma (LTP) ionization mass spectrometry (MS) was investigated to demonstrate its potential for direct and selective analysis of isothiazolinones from sprayed aerosol samples. Benzisothiazolinone (BIT) was clearly identified from a commercial fabric deodorant using LTP ionization MS and MS/MS. LTP allowed selective ionization of BIT directly from the simply sprayed aerosol sample and illustrated its potential for fast screening without sample pre-treatments. Selective nature of LTP ionization, on the other hands, implicates use of LTP ionization MS as a general screening method for specific groups of hazardous chemicals in commercial products.

  • It is important to select the correct disinfectants and to use them appropriately in order to prevent the initial spread of highly infectious livestock disease, such as foot-and-mouth disease or highly pathogenic avian influenza. This study describes a smartphone application developed to enable livestock workers to observe information related to disinfectants for the prevention of livestock disease in the domestic market, regardless of time and location, through a Linux-based Android mobile platform. This application (Konkuk-Disinfectant Information Database) provides information on disinfectant names, components, license and use; it was designed to enable the user to share disinfectant information through social media.

Present Status and Problems of Chemical Seed Treatment of Seedborne Diseases (종자소독의 현황과 문제점)

  • A wide variety of pathogens are known io be seedborne, carried either as infectious mycelium internally or as contaminants on the seed coat. When seed is infected with a pathogen, the seed nay be rendered nonviable or it may remain viable but produce weak seedling. In some cases, the Infected seedling nay not be severely weakened, but nay serve as a source of primary inoculum within a community of plants. A recent problem nay be the dissemination of seedborne pathogens occurring as a result of the massive movements of seed, as a part of the 'Green revolution' Disease of great danger to agriculture may be introduced with seed from other parts of world. Seed treatment with organic mercury compounds in liquid form had become popular since about 1955. Organic mercury compounds contributed considerably to the increase in production of many crops and vegetables. In 1975, however, the use of organic mercury compound was forbidden because of doubts regarding their residual mammalian toxicity in agricultural products. Benomyl-thiram mixture, thiophanate methyl-;hiram mixture and TCMB have now been registered as seed disinfectants for the use of rice blast, brown spot and Bakanae disease. Oxathiinsthiram mixture has been registered as seed disinfectant for barley and wheat loose smut and leaf stripe of barley. Agricultural techniques have made such rapid progress that the nursery methods changed from the use of paddy nursery to box nursery designed for machine-transplanting. The spread of rice transplanting machines has caused increase of seedborne diseases. Among seedborne diseases, Bakanae disease has remarkably increased and causes much damage recently. In order to counter this trend, seed disinfectants must also be diversified. First, effective non-selective disinfectants need to be developed, and second, appropriate control methods always need to be prepared in parallel with the development of new techniques for cultivation.
