The Establishing Test Method of Bactericidal Activity and the Evaluating of Korean Disinfectants/Sanitizers Efficacy (살균소독력 시험법 확립 및 살균소독력 평가)

  • Bacterial suspension test was used to establish Standardization Test Method to investigate bactericidal activity of disinfectant/sanitizer product. Using acceptable verification methodology, test substance showing 5 log or higher reduction in viable count against Escherichia coli ATCC 10536 and Staphylococcus aureus ATCC 6538, representing Gram-negative and -positive bacteria, respectively, under test conditions for $5\;min{\pm}10\;sec\;at\;20{\pm}1^{\circ}C$ was considered to have sanitizing capability. All disinfectant/sanitizer products tested under manufacturer's recommended in-use condition gave good reduction values against major food-poisoning bacteria. This standardized method was valuable for evaluating efficacy of disinfectants/sanitizers and could be used as Standardization Test Method for assessing bactericidal activity

Virucidal Efficacy against Avian Influenza Virus of a Disinfectant Spray Containing Grapefruit Seed Extracts, Citric Acid, Malic Acid and Benzalkonium Chloride (자몽종자추출물, 구연산, 사과산 그리고 염화벤잘코늄을 주성분으로 하는 스프레이형 소독제의 조류인플루엔자바이러스에 대한 살바이러스 효과)

  • Objectives: This study evaluated the virucidal efficacy against avian influenza virus (AIV) of a disinfectant spray containing 0.25% grapefruit seed extract, 0.2% citric acid, 0.0625% malic acid and 0.0125% benzalkonium chloride. Methods: The disinfectant spray was diluted several times with hard water (HW) and organic matter (OM). Two point five mL of each diluent was added into each test tube, and 2.5 mL of AIV suspension was inserted into each test tube. After 30 minutes of virus-disinfectant contact reaction at $4^{\circ}C$, 2.5 mL of 10% inactivated fetal bovine serum was added into each test tube to neutralize the sanitizer efficacy. The neutralized solutions were serial 10-fold dilutions with phosphate buffer solution, and 0.2 mL of the diluents was injected into the allantoic cavity of five ten-day-old-chickens per dilution time. After incubation of the embryos for five days, the viability of the AIV was examined by hemagglutination titer. The valid dilution of the disinfectant spray was estimated according to the dilution time that the virus titer was inactivated more than $10^4$ 50% egg-infective dose (EID50)/mL compared with pathogen control. Results: In HW and OM conditions, the valid dilutions of the disinfectant spray against AIV were seven- and three-fold dilutions, respectively. The AIV titer of the pathogen control was more than 6.1 log10EID50/mL, and there was no embryonic toxicity. Conclusion: The present study showed that this disinfectant spray has effective virucidal activity against AIV.

Evaluation of Effectiveness of Sanitizers and Disinfectants used in Domestic Food Processing Plants (국내 식품가공공장 현장에서의 살균소독제 유효성 평가)

  • Effects of sanitizers and disinfectants on microorganisms in food-processing plants were investigated. Chlorine and hydrogen peroxide were most effective, showing approximately 2-3 log reductions, among products tested, whereas alcohol showed less than 1 log reduction. Before using sanitizer and disinfectant in food processing plants, aerobic plate, coliforms, and E. coli counts were about $10^1-10^5,\;10^1-10^2$, and below $10\;CFU/cm^2$, respectively. After use of sanitizer, APC decreased to $10^1-10^2$. The result to confirm sanitizing effectiveness for 2 months using showed that Alcohol compound, QAC, hydrogen peroxide compound were effective against Staphylococcus aureus ATCC 6538 and E. coli ATCC 10536 at recommended usage concentrations, whereas effectiveness of chlorine compound decreased from 8 to 4 log 2 weeks after opening of product. Although sanitizers and disinfectants approved by law showed 5 log reduction in vitro for S. aureus ATCC 6538 and E, coli ATCC 10536, sanitizing effectiveness in food processing plants was very low, and effectiveness decreased once used product was stored.

Antimicrobial Activity and Bactericidal Activity of Caesalpinia sappan L. Extract (소목 추출물의 항균력 및 살균소독력)

  • Antimicrobial activity and bactericidal activity of Caesalpinia sappan L. extracts were investigated against five food-borne pathogens, E. coli, S. aureus, S. typhimurium, B. cereus and L. monocytogenes. Methanol extract of Caesalpinia sappan L. revealed antimicrobial activities against five pathogens. In particular, by paper disc diffusion the highest activity was shown against L. monocytogenes. Antimicrobial activities of methanol extracts showed the most potent activities, but hexane fraction had no activity. Fractions of ethyl acetate and butanol turned out to have higher antimicrobial activities against Gram(+) bacteria than Gram(-) bacteria. The minimum inhibitory concentration against five food-borne pathogens was 1.563 mg/ml on Gram(+) bacteria and 3.125 mg/ml on Gram(-) bacteria. The result of antimicrobial activity in a shaking flask method showed that bacterial growth rate fell by more than 99.999% at 3.125 mg/ml of methanol extract. The highest rate of viable reduction (99.998%) was shown at 0.781 mg/ml of methanol extract against L. monocytogenes. After five minutes of reaction between test strains and methanol extracts, the growth rates of five kinds of bacteria were reduced by more than 99.999% at a concentration of 100 mg/ml. Therefore, it is suggested that methanol extracts of Caesalpinia sappan L. can be developed as a natural sanitizer or disinfectant.

Assessment of Both Standard and Isolated Vibrio parahaemolyticus on Efficacy of Commercial Sanitizers and Disinfectants (Vibrio parahaemolyticus 표준 및 식품분리 균주에 대한 살균소독제 유효성분별 감수성 평가)

  • This study evaluated the bactericidal effect of 10 sanitizers and disinfectants such as ethanol (75 and 95%), iodine (15 and 25 ppm), chlorine (100 and 200 ppm), quaternary ammonium, acid, hydrogen peroxide, and peroxide acetic acid against V. parahaemolyticus. Ten strains of V. parahaemolyticus isolated from Korean foods and 4 strains of standard V. parahaemolyticus were compared for efficacies of various sanitizers and disinfectants by EN 1276 method based on quantitative suspension test. Ethanol (75 and 95%), 25 ppm of iodine, 100 ppm of quaternary ammonium, 145 ppm of hydrogen peroxide and acid showed more than $5log_{10}CFU/mL$ reduction in both clean and dirty conditions. Tests result of chlorine (100 ppm) showed more than $5log_{10}CFU/mL$ reduction in clean condition. Iodine (15 ppm) showed more than $5log_{10}CFU/mL$ reduction except 4 isolated and 1 standard V. parahaemolyticus in clean condition. iodine (15 ppm) also showed under $5log_{10}CFU/mL$ reduction $(0.93{\sim}3.73log_{10}CFU/mL)$ in dirty condition. Eleven hundred ppm of hydrogen peroxide was evaluated as weak sanitizer and disinfectant due to their $0.99{\sim}4.79log_{10}CFU/mL$ reduction on both clean and dirty conditions. Consequently, ethanol, iodine (25 ppm), chlorine (200 ppm), quaternary ammonium, acid and peroxide acetic acid were thought to be effective sanitizer and disinfectant against V. parahaemolyticus.

  • Conformity to legal permission standard of $5\;log_{10}$ CFU/mL reduction of foodborne pathogens or spoilage bacteria such as Escherichia coli, Staphylococcus aureus, Salmonella typhimurium, Pseudomonas aruginosa, and Enterococcus hirae was examined in 11 domestic commercial sanitizers and disinfectant. One chlorine compound, two iodophor compounds, two peroxide compounds, and three quaternary ammonium compounds (QACs) met advised standard concentration (100%), showing $7\;log_{10}$ CFU/mL reduction, and met legal standard by $5-6\;log_{10}$ CFU/mL reduction at 75% of advised standard concentration. At 10% dilution, one chlorine compound, one iodophor compound, two peroxide compounds, and two QACs satisfied legal standard.

  • Hypochlorous acid(HOCl) is a chemical that is a safe sanitizer and disinfectant approved by the Food and Drug Administration as a food additive, exhibiting strong sterilizing power with low effective chlorine concentration of pH 5.0-6.5 and effective chlorine concentration 10-80 ppm. To apply to fishery industries, we develope the HOCl ice for store or delivery of fishery products. However when HOCl is being frozen, the contained HOCl are expelled out from the ice due to the molecular structures of ice; there is no space to contain HOCl inside. To increase chlorine containing amount in ice, we develop the alginate particles containing HOCl which is bio comparable since alginate is a natural polymer extracted from the brown algae and it is widely used for drug delivery and containing substances, etc. We produce HOCl with water as base solution suppressing osmotic flow from fishery products, and mix it with the developed alginate particles and made HOCl-alginate ice and checked the remaining amount of HOCl. We measure the change of pH and chlorine concentration optimizing the best concentration of alginate particles. Finally, we produce the alginate particle HOCl ices with respect to the alginate's optimal concentration.

Optimization of disposable paper-based test strips for hypochlorous acid detection

  • The Covid-19 pandemic has increased demand for chlorine-based sanitizing solutions, most of which contain hypochlorous acid (HOCl) as an active agent. Free chlorine (HOCl) in these sanitizers is crucial for their efficacy. Disposable test strips are affordable and convenient tools for determining various qualitative and quantitative parameters. In this study, disposable opto-chemical test strips were developed by physically immobilizing 3,3',5,5'-tetramethylbenzidine (TMB) and o-dianisidine (o-D) reagents on chromatography and filter paper-based test strips for the visualization and detection of free chlorine in the form of HOCl. The reagents undergo a rapid color change upon reaction with chlorine through a redox reaction. The paper-based test strips showed rapid color change within a minute and a low sample volume requirement (1 ml). This portable, disposable paper-based test strip is a simple and cost-effective way to rapidly detect the presence of HOCl sanitizers for home and field applications. Both TMB and o-D successfully detected chlorine. Chromatography paper proved to be the more efficient option among the two papers used as substrates for the reagents (TMB and o-D). It exhibited high retention capacity and high performance in terms of color transformation when reacting with HOCl, even after two months of storage.

  • Bactericidal efficacies of various sanitizers and disinfectants against 10 Bacillus cereus strains isolated from Korean foods and 8 standard B. cereus strains were investigated. The sanitizing capabilities of ethanol, iodine, chloride, quaternary ammonium, hydrogen peroxide, and peroxide acetic acid were investigated using the EN 1276 method based on quantitative suspension testing. The resistance against sanitizers and disinfectants was higher for wild-type than standard strains, and the bactericidal activities decreased in dirty conditions. Ethanol, chlorine, and iodine at the maximum level allowed under Korean food sanitation laws showed a great effectiveness against B. cereus. Hydrogen peroxide at 1,100 ppm showed the lowest bactericidal activity against B. cereus. These results indicate that the legally allowed maximum concentrations of sanitizers and disinfectants in Korea do not reduce all B. cereus strains by at least $5\;{\log}_{10}\;CFU/mL$.

In Vitro and In Vivo Inhibitory Effects of Gaseous Chlorine Dioxide against Fusarium oxysporum f. sp. batatas Isolated from Stored Sweetpotato: Study II

  • Chlorine dioxide ($ClO_2$) has been widely used as an effective disinfectant to control fungal contamination during postharvest crop storage. In this study, Fusarium oxysporum f. sp. batatas SP-f6 from the black rot symptom of sweetpotato was isolated and identified using phylogenetic analysis of elongation factor 1-${\alpha}$ gene; we further examined the in vitro and in vivo inhibitory activities of $ClO_2$ gas against the fungus. In the in vitro medium tests, fungal population was significantly inhibited upon increasing the concentration and exposure time. In in vivo tests, spore suspensions were drop-inoculated onto sweetpotato slices, followed by treatment using various $ClO_2$ concentrations and treatment times to assess fungus-induced disease development in the slices. Lesion diameters decreased at the tested $ClO_2$ concentrations over time. When sweetpotato roots were dip-inoculated in spore suspensions prior to treatment with 20 and 40 ppm of $ClO_2$ for 0-60 min, fungal populations significantly decreased at the tested concentrations for 30-60 min. Taken together, these results showed that $ClO_2$ gas can effectively inhibit fungal growth and disease development caused by F. oxysporum f. sp. batatas on sweetpotato. Therefore, $ClO_2$ gas may be used as a sanitizer to control this fungus during postharvest storage of sweetpotato.
