Selection and Characteristics of Bacteriocin-Producing Microorganism to Utilize in Anti-Bacterial Rice Brain Protein Film Production (항균성 미강 단백질 필름 개발을 위한 Bacteriocin 생성균주의 선별 및 특성)

  • This study was conducted to select the bacteriocin-producing microoreanism cultivated in the rice bran culture and to characterize the produced bacteriocin for the further purpose of economical and anti-bacterial rice bran protein film. Pseudomonas putida 21025 was cultivated from rice bran and identified as a producer of a bacteriocin which showed bactericidal activity against Pseudomonas aeruginosa 9027. Bacteriocin produced by Pseudomonas putida 21025 showed a broad spectrum of activity against spoilage and soil bacteria. The activity of the bacteriocin produced by Pseudomonas putida 21025 decreased after 1 hr of staying at the temperature of $50^{\circ}C$, and with the presence of some organic solvents, except hexane and ethanol. However, the bacteriocin activity was stable throughout the pH ranges of 6-9 for 2 hrs, at the temperature lower than $50^{\circ}C$, and with the presence of ethanol for 3 hrs. The bacteriocin was partially purified by 50% ammonium sulfate precipitation followed by subsequent dialysis. Direct detection of the partially purified bacteriocin on SDS-PAGE suggested that it had an apparent molecular mass of about 21.6 kDa.

An Analysis of Optimal Sequences for the Detection of Wake-up Signal in Disaster-preventing Broadcast (재난방송용 대기모드 해제신호 검출을 위한 최적 부호 성능 분석)

  • Recently, the need for disaster-preventing broadcast has increased gradually to cope with natural disaster like earthquake and tsunami causing enormous losses of both life and property. In disaster-preventing broadcast system, the wake-up signal is used to alert user terminal and switch the current state of channel to the emergency channel, which is for the fast and efficient delivery of emergency information. In this paper, we propose the detection method of wake-up signal for disaster-preventing broadcast systems. The wake-up signals for disaster-preventing broadcast should have a good auto-correlation property in low power and narrow-band conditions that does not affect the existing digital television (DTV) system. The suitability of the m-sequence and complementary code (CC) is analyzed for wake-up signals according to signal to noise ratio. A wake-up signal is proposed by combining the direct sequence spread spectrum (DSSS) technique and pseudo noise (PN) sequences such as Barker and Walsh-Hadamard codes. By using the proposed method, a higher detecting performance can be achieved by the spreading gain compared to the single long m-sequence and the Golay code.

Growth-inhibitory Effects of Citrus Oils and Synthetic Agricultural Fungicides on Molds Isolated from Putrefied Citrus Fruits (감귤 부패 사상균에 대한 감귤 정유와 시판 합성 농약의 항균 효과)

  • Growth-inhibitory effects of citrus oils and agricultural fungicides, which were on the market, on several molds isolated from putrefied citrus fruits were investigated. When fungicidal activities of 11 kinds of synthetic agrochemicals against 6 species of molds, Alternaria alternata, Rhizopus sp., Botrytis cinerea, Monilia candida, Penicillium italicum and Penicillium digitatum, were investigated, agrochemicals containing mancozeb or fluazinam as an effective component had the broadest fungicidal spectrum. Agrochemicals containing iprodione, benomyl, azoxystrobin or thiophanate were less effective on the molds and those containing conazole derivative were intermediately effective. Resistance of the molds to the agrochemicals were species- and agrochemical-dependent. Among those molds tested, Penicillium italicum and Alternaria alternata showed relatively higher level of survival in the presence of synthetic fungicides. On the other hand, when the molds were exposed to citrus oils by direct contact, no cell could survive regardless of the species. The fungicidal activity of citrus oils was also confirmed by paper disk method and microscopic observation. These results suggested that citrus oils had broad killing activity against molds. Therefore, it would be necessary to design method for the application of citrus oils in order to improve post-harvest storage of citrus fruits.

Characterization of Diethyl Phthalate(DEP) Removal using Ozone, UV, and Ozone/UV Combined Processes (오존, UV, 오존/UV 혼합 공정을 이용한 Diethyl Phthalate(DEP)의 제거특성 연구)

  • Three candidate processes(ozone alone, UV alone and ozone/UV combined processes) were evaluated for the removal of diethyl phthalate(DEP). Of the candidates, the ozone/UV process showed the highest removal efficiency of DEP. To elucidate a major oxidant for DEP oxidation in the ozone/UV process, the effects of pH and hydroxyl radical($OH^{\circ}$) scavenger were investigated. As a result, it was found that $OH^{\circ}$ plays a important role for DEP elimination. Meanwhile, the direct reaction between ozone and DEP was negligible. Observing the pseudo first-order rate of DEP removal in ozone alone and ozone/UV processes, the different pattern was obtained from two processes. The ozone/UV process was well plotted following the pseudo first-order. but in the ozone alone process the rate was divided into fast and slow phases. DEP degradation characteristics in ozone alone and ozone/UV was also investigated by observing the HPLC spectrum. We detected unknown compounds that were guessed to DEP byproducts and observed the formation and disappearance of the unknown compounds according to reaction time. Observing of high removal of TOC in ozone/UV combined process, it was found that DEP and DEP byproducts are completely oxidized by ozone/UV combined process.

  • A simulation procedure which can represent time-dependent nuclear characteristics of TRIGA Mark-III reactor is developed. CITATION, a multi-group diffusion-depletion program, has been utilized as calculational tool. The group structure employed in this study consists of 7 groups: -3-fast and 4-thermal-which is conventionally utilized in TRIGA type reactor analysis. Three-dimensional nuclear characteristics are synthesized by combining results from two-dimensional plane calculation and two-dimensional cylinder calculation, since direct three-dimensional approach is not yet possible. An effort ia made to develope a method which can extract effective zone and group dependent bucklings by neutron diffusion theory rather than conventional zone and/or group independent Ducklings by neutron transport theory, since neutron leakage is quite high for small core such as research reactors. It is turned out that the method developed in this study gives satisfactory results. The calculation is performed under assumptions that all control rods are fully withdrawn, that no samples are inserted in the irradiation holes and that the core is located in the center of the reactor pool. Burnup-dependent variation of core excess reactivity, time dependent change of Xe-135 poisoning and reactivity worth of rotary specimen rack are calculated and compared with operation records. Neutron flux and power distribution as well as neutron spectrum in each irradiation .facility are presented.

  • The specificity and sensitivity of oligonucleotide primers were examined for the rapid detection of Salmonella in meat samples. The oligonucleotide primers used in this study were designed with the modification of mdh and invA sequence in the chromosome of Salmonella Typhimurium. Through the subsequent analysis of the specificity and sensitivity of the primers, two types of oligonucleotide primers, SLM1 and SLT4 were selected for the detection of Salmonella genus specific and S. Typhimurium species specific, respectively. The lowest detection limit of each primer was represented as 1 cell per reaction when reacted with a prepared DNA solution. The detection efficiency of the two primers was analysed with beef and pork samples intentionally contaminated with a mixture of Salmonella culture, and three preparation methods -, namely direct reaction after extraction, enrichment after extraction, and DNA extraction after enrichment for PCR reaction, - were also compared. No differences were found in the results according to meat sources and preparation methods.

ANALYSIS OF OPTICAL TRANSMISSION CHARATERISTICS BY VISIBLE LIGHT INTO SHADE GHIDES (가시광선영역에서 shade guide에 따른 광투과도 특성 연구)

  • The purpose of this study was the evaluation of spectral transmission of the commercial three shade guides and the relationship of hue, value, and chroma according to the transmission rate. The spectral transmittance of three shade guides - Vita Lumin Shade Guide, Vintage and Unibond Shade Guide, and Vitapan 3D-Master Shade Guide - were measured. For direct transmission measurements, each shade tabs were placed at the entrance port of the 1mm diameter intergrating sphere. The intensity of the light source passing through the shade tabs to the right angle was continuously recorded for wavelength from 400 to 700 nm. A transmission spectrum and digital data were obtained for each measurement and they were evaluated using Microcal Origin program. The obtained results of this study were as follows : 1. In Vita Lumin Shade Guide, sum of the transmission rate of the shade tabs at the wavelength 400-700nm was decreased in agreement with the arrangement order at each A, B, C, D group, except D4 shade tab. However, there were no relationships between the transmission rate of the shade tabs and the value-oriented. 2. In Vintage and Unibond Shade Guide, sum of the total transmission rate of the shade tabs was decreased in agreement with the arrangement order at each A, B, C, B group. When all shade tabs arranged in value-oriented, transmission rate was accord with the order, except D4 shade tab. 3. When shade tabs of the Vitapan 3D-Master Shade Guide have the same value and hue, sum of their total transmission rate decreased in accordance with the chroma-oriented. When the shade tabs have the same value and chroma, there were no differences from the order of the transmission rate to the various hue type. However, in the 'R' tabs of reddish hue type, the transmission rate increased at the long wavelength range area. In conclusion, we need the quantitative analyzing instruments in transmission determination. Vitapan 3D-Master Shade Guide covers the tooth color space taking into account the parameters of the systematic value, chroma, hue oriented, and the transmission rate relatively accorded with that sequence.

  • This paper proposes an adaptive pilot beacon equipment for mobile communication systems based on direct spread spectrum technology which generates the pilot channel for handoff between base stations by using the information acquired from the downstream wireless signal regarding the overhead channel information. Such an adaptive pilot beacon equipment will enable low power operation since among the wireless signals, only the pilot channel will be generated and transmitted. The pilot channel in the downstream link of the CDMA receiver is used to acquire time and frequency synchronization and this is used to calibrate the offset for the beacon, which implies that time synchronization using GPS is not required and any location where forward receive signal can be received can be used as the installation site. The downstream link pilot signal searching within the CDMA receiver is performed by FPGA and DSP. The FPGA is used to perform the initial synchronization for the pilot searcher and DSP is used to perform the offset correction between beacon clock and base station clock. The CDMA transmitter the adaptive pilot beacon equipment will use the timing offset information in the pilot channel acquired from the CDMA receiver and generate the downstream link pilot signal synchronized to the base station. The intermediate frequency signal is passed through the FIR filter and subsequently upconverted and amplified before being radiated through the antenna.

  • In this paper, Antenna, LNA, Mixer, VCO, and Modulation/Demodulation in Baseband processor which are the RF main components in Wireless LAN system for ultra high-speed communications network are studied. Antenna bandwidth and selective fading due to multipath can be major obstacles in high speed digital communications. To solve this problem, wide band MSA which has loop-structure magnetic antenna characteristics is designed. Distributed mixer using dual-gate GaAs MESFET can achieve over 10dB LO/RF isolation without hybrid, and minimize circuit size. As linear mixing signal is produced, distortions can be decreased at baseband signals. Conversion gain is achieved by mixing and amplification simultaneously. Mixer is designed to have wide band characteristics using distributed amplifier. In VCO design, Oscillator design method by large signal analysis is used to produce stable signal. Modulation/Demodulation system in baseband processor, DS/SS technique which is robust against noise and interference is used to eliminate the effect of multipath propagation. DQPSK modulation technique with M-sequences for wideband PN spreading signals is adopted because of BER characteristic and high speed digital signal transmission.

Genetic and clinical characteristics of Korean patients with neurofibromatosis type 2

  • Purpose: Neurofibromatosis type 2 (NF2) is characterized by multiple tumors, including vestibular schwannoma (VS) and others affecting cranial and peripheral nerves. NF2 is caused by mutation of the NF2 gene. The mutation spectrum of NF2 has not been characterized in Korean patients. In the current study, the clinical and genetic characteristics of Korean NF2 patients were analyzed. Materials and Methods: Twenty-five unrelated Korean families were enrolled according to the Manchester criteria. Genetic analysis was performed by direct sequencing and multiplex ligation-dependent probe amplification methods using genomic DNA from peripheral lymphocytes or tumor tissues. Results: All patients had bilateral/unilateral VS and/or other cranial and peripheral nerve tumors. Two patients were familial cases and the other 24 patients were sporadic. Germline NF2 mutations were detected in peripheral lymphocytes from both familial cases, but only in 26.1% of the 23 sporadic families. Somatic mutations were also found in tumor tissues from two of the sporadic families. These somatic mutations were not found in peripheral lymphocytes. A total of 10 different mutations including 2 novel mutations were found in 40.0% of studied families. Five mutations (50.0%) were located in exon 6 of NF2, the FERM domain coding region. Conclusion: Family history was an important factor in identifying germline NF2 mutations. Further study is required to investigate whether exon 6 is a mutation hotspot in Korean NF2 patients and its correlation to phenotypic severity.
