This study was conducted to detect the toxoplasma-specific DNA in peripheral blood collected from cats experimentally infected with Toxoplasma gondii (RH strain) and from domiciled cats by B1 gene-base polymerise chain reaction(PCR). The sensitivity of oligonucleotide primer, T-1 & T-2, designed from toxoplasma B1 gene amplification method was compared with parasite detection by mouse inoculation(MI). And also, latex agglutination test(LAT) and indirect fluorescent antibody test(IFAT) were conducted to detect the fluctuation of serum antibodies compared with the detection of toxoplasma by PCR and MI. Toxoplasma B1 gene PCR was shown consistently high sensitivity and the results obtained by PCR agreed completely with those from MI. All blood samples collected before infection with T gondii gave negative results by PCR and MI. Also, toxoplasma Bl gene PCR was not cross reaction with Neospora caninum DNA and normal cat leucocyte as controls. The toxoplasma-specific DNA was detected by PCR in blood of 5 cats experimentally infected with T gondii 6 days after infection and the detection of this specific-DNA was long lasted in blood for 64 days after infection. The detection of toxoplasma-specific DNA by PCR could be identified as few as 10 tachyzoites and the isolation of T gondii by MI could be isolated as few as 1 tachyzoite from tenfold serial dilution of T gondii with normal cat blood, respectively. In healthy domiciled cats, the toxoplasma-specific DNA and the parasite were detected and isolated in blood from 3 of 56(5.3%) cats by both PCR and MI, respectively. In the results of antibody test from the total 56 heads of healthy domiciled cats, the positive rates are 15(26.7%) by LAT and 19(33.9%) by IFAT. These results suggest that PCR detection of toxoplasma can be applied as a sensitive and specific diagnostic and research tool.
Objectives: The aim of this in vitro study was to evaluate the biocompatibility of newly proposed root-end filling materials, Biodentine, Micro-Mega mineral trioxide aggregate (MM-MTA), polymethylmethacrylate (PMMA) bone cement, and Smart Dentin Replacement (SDR), in comparison with contemporary root-end filling materials, intermediate restorative material (IRM), Dyract compomer, ProRoot MTA (PMTA), and Vitrebond, using human periodontal ligament (hPDL) fibroblasts. Materials and Methods: Ten discs from each material were fabricated in sterile Teflon molds and 24-hour eluates were obtained from each root-end filling material in cell culture media after 1- or 3-day setting. hPDL fibroblasts were plated at a density of $5{\times}10^3/well$, and were incubated for 24 hours with 1:1, 1:2, 1:4, and 1:8 dilutions of eluates. Cell viability was evaluated by XTT assay. Data was statistically analysed. Apoptotic/necrotic activity of PDL cells exposed to material eluates was established by flow cytometry. Results: The Vitrebond and IRM were significantly more cytotoxic than the other root-end filling materials (p < 0.05). Those cells exposed to the Biodentine and Dyract compomer eluates showed the highest survival rates (p < 0.05), while the PMTA, MM-MTA, SDR, and PMMA groups exhibited similar cell viabilities. Three-day samples were more cytotoxic than 1-day samples (p < 0.05). Eluates from the cements at 1:1 dilution were significantly more cytotoxic (p < 0.05). Vitrebond induced cell necrosis as indicated by flow cytometry. Conclusions: This in vitro study demonstrated that Biodentine and Compomer were more biocompatible than the other root-end filling materials. Vitrebond eluate caused necrotic cell death.
This study was carried out to investigate the factors affecting fertilization in vitro of follicular oocytes with frozen-thawed spermatozoa in Korean Native Cattle. The bovine ovaries were obtained at a slaughter house and the follicular oocytes were recovered by aspirating the follicular fluid from the visible follicles of 3~6mm. The bovine oocytes were matured in vitro for 20~24 hours in TCM-199 containing FCS and hormones. The matured oocytes were fertilized in vitro using Percoll-separated frozen-thawed spermatozoa in BO solution. The effects of dilution and fertilization media, capacitating method, concentration of inseminated sperm and time after insemination of fertilization, were observed. The results obtained are summarized as follows : 1. The fertilization rate of frozen-thawed sperm inseminated in BO solution with caffeine and heparin together(56.4%) was higher than that of sperm inseminated in BO solution with either caffeine(10.5%) or heparin(8.9%) and without both caffeine and heparin(0%)(P<0.05). 2. The fertilization rate(56.3%) of frozen-thawed sperm inseminated in BO solution with both caffeine and heparin without preincubation was higher than that of sperm preincubated(2.9%)(P<0.05). 3. The fertilization with high concentration of frozen-thawed sperm(1.4~1.8$\times$107cells/ml) in BO solution containing caffeine and heparin resulted in higher fertilization rate, 76.7%, than the low concentration of sperm(0.8~1.0$\times$107cells/ml), 32.7%(P<0.01). 4. When the oocytes were inseminated with frozen-thawed sperm in BO solution containing caffeine and heparin without preincubation, fertilization rate increased by time and the rates were 5.9, 46.0 and 59.4% at 8, 16 and 24 hours, respectively.

The minimum inhibitory concentration (MIC) of eight antibiotics against Edwardsiella tarda 49 strains isolated from olive flounder (Paralichthys olivaceus) was determined by the broth microdilution method. E. tarda showed 38.8% and 61.2% resistance to ampicillin and amoxicillin. Both resistance rates of E. tarda were 4.1% against ciprofloxacin and norfloxacin. The $MIC_{50}$ values of oxytetracycline, amoxicillin and oxolinic acid were $64{\mu}g/ml$, $32{\mu}g/ml$ and $4{\mu}g/ml$. The $MIC_{50}$ value to ciprofloxacin was $0.25{\mu}g/ml$ which was lowest among eight antibiotics tested.
Turbulence produced on roadways is one of the major factors determining the dilution rates at the initial stage of traffic emissions of air pollutants and, thus, the distribution of air pollutants near the roadways. Field experiments were conducted on Gyeongbu Highway, one of the busiest highways in Korea, for 4~7 days in winter, spring, and summer. Two three-dimensional ultrasonic anemometers were installed on both sides of the highway to estimate turbulence intensities (vertical wind fluctuation and kinetic turbulence energy) induced by the roadway. Roadway-induced turbulence consists of three components: structural road-induced turbulence (S-RIT), thermal road-induced turbulence (T-RIT), and vehicle-induced turbulence (VIT). The contribution of T-RIT to the total RIT was insignificant (less than 10%), and the majority of RIT was S-RIT (by the highway embankment) and VIT. In this study, we propose the empirical relationships of VIT as a function of traffic density and wind speed under free-flow traffic conditions. Although this empirical relationship appears to underestimate the VIT, it can be applied to the air quality models easily because the relationship is simple and only needs readily obtainable input variables (wind speed and traffic information).

In order to evaluate the present status of hook-worm infection in the rural areas in Korea, stool examinations were undertaken in several selected rural villages in each year of 1970 and 1977. The methods employed were cellophane thick smear technique and brine floatation technique in 3,454 specimens collected from 7 different rural villages for the prevalence rate of hookworm infection and Stoll's dilution egg counting technique for the intensity of hookworm infection. The incidences of Ancylostoma duodenale and Necator americanus infections were determind by polyethylene tube coproculture and by the evacuated adult worms after treatment with anthelmintics to the patients who were infected by hookworms. The results are as follows : 1) In 1970, 323(18.6%) out of 1,737 specimens collected from 4 different rural areas were positive for hook worm infection and the ranges of prevalence rates in those suveyed areas were shown as 6.9 to 38.2 per cent. On the other hand, 144(8.2%) out of 1,717 specimens collected from 3 different rural areas in 1977 were shown as positive for hookworm infection and 7.5 to 10.6 per cent of prvalence rates were shown in the surveyed areas. 2) In the age group under 10 years, there was no infection of hookworm in the studied areas in 1977, however in 1970 the prevalence of the hookworm infection was shown as very low in the age group of 5~9. On the whole, the hookworm prevalence steadily increased from the age of 15 in the studied areas in 1970, however in 1977 the prevalence increased from the age of 20 to the older age groups. 3) There was not observed much differences in the intensity of hookworm infection among the studied areas in 1970 and 1977. The mean EPG in each studied areas were shown under 1,000 EPG and mean numbers of evacuated worms per infected persons were counted as 9.4 to 15. 1. 4) Both A.duodenale and N.americanus have been found in all studied areas but the incidences and st-ate of distribution of two species of hookworn were shown as different in each localities.
This study was carried out in order to evaluate the growth promotion effects on tomato crops and to assess the mineral nutrient concentrations of farm-made liquid fertilizers used in organic cultivation in South Korea. We hope that this study will help to develop of a standardized manufacturing technique for these organic liquid fertilizers. We collected 62 farm-made liquid fertilizers made from various raw materials including fish, seaweed, food scraps, plant and crop by-products, and other materials. Two groups of tomato seedlings were treated at different times, one at 20 days and the other at 40 days after sowing. We used both foliar and soil applications. These seedlings were treated using liquid fertilizers at various dilution rates (x1000, x500 and x100). When foliar application was used, seedlings after 20 days had a 20-30% increase in shoot fresh weight with 47-48 fertilizers and seedlings after 40 days had a 20-30% increase in shoot fresh weight with 17-32 fertilizers. When soil application was used, seedlings after 20 days had the same increase in shoot fresh weight with 30-31 fertilizers and seedlings after 40 days also saw the same increase with 6-7 fertilizers. Therefore, our studies showed that application of liquid fertilizers to seedlings 20 days after sowing was most effective and that foliar treatment was more effective than soil treatments. We also observed that the higher the concentrations of fertilizer, particularly when applied twice rather than just once, the higher the rates of growth, which promoted shoot fresh weight more than plant height. Our results imply that mineral nutrients in liquid fertilizers seem to be the probable cause for the growth promotion observed in this study. However, more study is required to determine exactly which mineral nutrients are most effective.

In here, we investigated the quantitative analysis method of indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) with liquid chromatography-mass/mass spectrometry (LC-MS/MS) or gas chromatography-MS. Two ways of clean-up process were investigated for LC-MS/MS instrumental analysis of IAA, but both a simple dilution and hydrophile-lipophile balance (HLB) solid phase extraction (SPE) were not met the optimal recovery rates for quantitative analysis. On the other hand, the clean-up method for GC-MS was finally optimized through HLB-SPE from 250-folds diluted sample and methylation with trimethylsilyl chloride in methanol for 4 h. The limit of detection for methyl ester of IAA and IBA were both 1.4 mg/L, and recovery rates showed 93-107% from the concentrated liquid fertilizer.

In order to keep the competitiveness of LPG fuel for transportation fuel, the difference in fuel consumption with gasoline and cost for an aftertreatment system should be reduced with continuous development of technology for LPG engine. In the present study, spray-guided type direct injection combustion system, whose configuration is composed of direct injector in the vicinity of spark plug, was employed to realize stable lean combustion. A certain level of nitrogen oxides($NO_x$) emits due to a locally rich mixture regions in the stratified mixture. With the application of EGR system for the reduction of $NO_x$, 15% of $NO_x$ reduction was achieved whereas fuel consumption and hydrocarbon emission increased. By the application of EGR, the combustion speed reduced especially appeared at initial flame development period and peak heat release rates and increasing rates for heat release rate decreased as EGR rate increased due to the dilution effect of intake air.
An epidemiological survey on Metagonimus yokogawai infection was performed in the eastern coast area of Kangwon province. Formalin-ether concentration technique and Stoll's dilution egg counting method were employed to figure out the prevalence and intensity of M. yokogawai infection and eating habits of inhabitants with raw freshwater fish. The results are summarized as follows: 1. The prevalence rate of M. yokogawai infection was 6.6% among a total of 2, 357 examinees; 8.6% (115 out of 1, 345) in male and 4.0% (40 out of 1, 012) in female. 2. The prevalence rates were different by the streams where the examinees were residing: 17.8% along the area of Ohsip-cheon (stream), 11.8% along Maeup-cheon, 1.7% along Hwasang-cheon, 1.3% along Yeongok-cheon, 0.9% along Namdae-cheon, 0.9% along Sa-cheon and 0% in the other streams (Yongchon-cheon, Kangnung Namdae-cheon and Jusoo.cheon) in decreasing order. 3. By social strata, the positive rate was 8.0% (122 out of 1, 521) in general inhabitants and 3.9% (33 out of 836) in school children. 4. By the intensity of infection by means of EPG counts in feces; the light infection (less than 400/EPG) was 74.1%, moderate infection (401-1, 000/EPG) 17.3% and heavy infection (more than 1, 001/EPG) 8.6%. 5. Twelves out of 50 sweetfish (Plecoglossus altivelis) (24%) caught from the streams in eastern coast of Kangwon-Do were infected with metacercariae of M. yokogawai, and the number of larvae detected were 70.7 per 6sh in average. The metacercarial infection rates in fish were also different by the streams; 100% in Ohsip-cheon of Samcheok-Gun, 14.3% in Sa-cheon, 16.7% in Yeongokcheon of Myeongju-Gun and 0% in Namdae-cheon of Yangyang-Gun, respectively. 6. Six hundred ninety live (695) out of 1, 396 inhabitants and school children (49.8%) were experienced in eating raw freshwater fishes (Plecoglossus altivezis, Triboledon hakonensis, Coreoperca sp., Morose sp., Carassius sp. & Cyprinus sp. and Zacco platypus). The data were assayed with the questionnaire collected from the examinees. In summarizing the above results, it is known that the Ohsip-cheon area of Samcheok-Gun is highly infected region, and the other areas of Kangnung Namdae-cheon, Yeongok-cheon and Yangyang Namdae-cheon are very lowly infected or non-infected regions for M yokogawai infection.
