Development of Drying Systems for Accurate Measurement of Particulate Matter by means of Optical Particle Measuring Instruments (광산란 계측기의 미세먼지 측정 정확도 향상을 위한 수분제거 전처리 기술 개발)

  • IIn this study, we have developed drying systems for reducing the error by humidity on measuring particulate matter (PM) in the ambient air with optical particle measuring instruments. Two types of drying systems were designed: drying systems using heating and dilution methods. In addition, 3 types of drying systems using a heating method were designed: Type A (1 hole), B (3 holes) and C (7 holes). After making them, the laboratory and field tests were carried out to evaluate the developed drying systems. As a result, it was shown that the PM concentrations obtained by PM monitoring devices with drying systems agree well with that of the reference devices. Therefore, it could be concluded that the drying systems can be applied to PM monitoring devices for real-time monitoring of the ambient aerosols.

  • Benzoic acid, methyl paraben, and butyl paraben are preservatives that have been used in pharmaceutical, cosmetic, and food products. However, as their toxicities for human have been reported, many nations and organizations including Korea have established a regulation limit for thier usage of these preservatives in food products. The present study developed the isotope dilution liquid chromatography mass spectrometry method for accurate determination of three target preseratives in soysauce. In this study, the isotope dilution liquid chromatography mass spectrometry method was developed for accurate determination of three target preservatives in soy sauce. LC separation was optimized considering the pKa of benzoic acid which is lower than those of methyl and butyl parabens. A C18 column was used with 5 mM ammonium acetate and methanol as mobile phases. Mass spectrometry was operated in negative mode and selected reaction monitoring mode (SRM). Soy sauce sample was cleaned-up with C18 SPE cartridge for removing matrix inferences and color material. Optimized conditions and the method were validated with soy sauce reference materials for the analysis of food preservatives from Health Science Authority in Singapore. The measured values of benzoic acid, methyl and butyl paraben agreed well with reference values within their uncertainties.

  • A new method of calculating sensitivity coefficients of core characteristics relative to infinite-dilution cross sections has been developed. Conventional sensitivity coefficients are evaluated for the changes of effective cross sections which are dependent on individual models of core and cell. Therefore a correction has been derived to the conventional sensitivity coefficients based on the perturbation theory. The accuracy of the present method has been verified by comparing numerical results of sensitivity coefficients with a reference Monte-Carlo method.

  • To analyze co-planar PCBs in food, the isotopic dilution method by high resolution gas chromatography/high resolution mass spectrometry was applied. Among 209 PCB congeners, 12 congeners (#77, #81, #126, #169, #105, #114, #118, #123, #156, #157, #167 and #189) were chosen as target compounds that were toxic congeners re-assessed by WHO in 1998. Milk and milk products including cheese and butter were collected as food samples. Samples were homogenized, spiked with the known amount of the standard mixture and extracted. After extraction, extracts were cleaned up by sulfuric acid impregnated silica gel, purified on silica gel and alumina column chromatography and then analyzed by HRGC/HRMS. As results, the overall recoveries were ranged from 83% to 106% and the limit of detection was about 0.1 pg/g at signal/noise>3. Levels of targets in the selected food samples were 0.001~0.107 pgWHO-TEQ/g.

  • This basic study is focused on the physically removal method of carbon dioxide from the decomposition of ammonium carbarmate to prevent the recombination of ammonium salts. A basic visual experimental set-up was designed and constructed to observe the recombination phenomena from the proper composition of ammonia gas, carbon dioxide gas, and compressed air dilution gas. To quantify the recombination phenomena, a simple device was designed to measure the weight change under severe cases for three different tube sizes. The temperature and pressure in the visual tube and the volumetric flow rates of the nitrogen dilution gas were studied and the conditions to avoid recombination were analyzed according to mean free path theory. Diffusivity values based on the Chapman-Enskog theory were calculated from the experimental data. These value may serve as an index for the prevention of recombination.

Determination of Dibutyltin in Sediments Using Isotope Dilution Liquid Chromatography-Inductively Coupled Plasma Mass Spectrometry

  • A method is described for the determination of dibutyltin (DBT) in sediment by isotope dilution using liquid chromatography inductively-coupled plasma/mass spectrometry (LC-ICP/MS). To achieve the highest accuracy and precision, special attentions are paid in optimization and evaluation of overall processes of the analysis including extraction of analytes, characterization of the standards used for calibration and LC-ICP/MS conditions. An approach for characterization of natural abundance DBT standard has been developed by combining inductively-coupled plasma/optical emission spectrometry (ICP/OES) and LC-ICP/MS for the total Sn assay and the analysis of Sn species present as impurities, respectively. An excellent LC condition for separation of organotin species was found, which is suitable for simultaneous DBT and tributyltin (TBT) analysis as well as impurity analysis of DBT standards. Microwave extraction condition was also optimized for high efficiency while preventing species transformation. The present method determines the amount contents of DBT in sediments with expanded uncertainty of less than 5% and its result shows high degree of equivalence with reference values of an international inter-comparison and a certified reference material (CRM) within stated uncertainties.

Errors of Antibiotic Susceptibility Testing from Automated and Manual Systems in Clinical Isolates of Acinetobacter baumannii

  • Acinetobacter baumannii is an aerobic, gram-negative and glucose-non-fermenting bacterium, which has emerged as a serious opportunistic pathogen. Many clinical microbiology laboratories use the Vitek 2 system for the routine antimicrobial susceptibility testing process, including testing on A. baumannii isolates. However, in case of amikacin, it is now recommended to perform additional antimicrobial susceptibility testing for A. baumannii strains due to the relatively lower minimum inhibitory concentration (MIC) in the Vitek 2 system compared to conventional reference methods. In our study, we assessed MIC for amikacin susceptibility testing of A. baumannii isolates in the Vitek 2 system, the agar dilution, Etest, and disk diffusion method. We collected 40 gentamicin-resistant, A. baumannii strains (amikacin MIC by Vitek 2:${\leq}2{\mu}g/mL$, 2 isolates; $4{\mu}g/mL$, 34 isolates; $8{\mu}g/mL$, 4 isolates) from a University hospital and compared the Vitek 2 system to other reference methods for testing susceptibility to amikacin. The Vitek 2 system showed major errors in all of the 40 isolates, yielding a low MIC. The results of our study strongly suggested that the Vitek 2 system was not a reliable method to test the MICs of gentamicin; ranging from ${\geq}16{\mu}g/mL$ for amikacin susceptibility. Other tests, such as agar dilution, Etest, or disk diffusion methods, should be paralleled to determine the MIC of amikacin in A. baumannii.

A short study of uncertainty for post column isotope dilution method in HPLC-ICP/MS (HPLC-ICP/MS에서 후 컬럼 동위원소 희석법의 기초적인 불확도 연구)

  • A short study for the uncertainty of post column isotope dilution method has been performed for the analysis of Selenomethionine in HPLC-ICP/MS. Major error sources studied were concentration and the flow rate of Se isotope solution, atomic weights of Se in spike and sample, and isotope ratio measured for the spiked sample. Uncertainties were obtained for each factor and the contribution for the total concentration uncertainty was 54.4% and 0.61%, 0.0072% and 0.018%, and 45.0%, respectively. The biggest contribution factor was concentration of the spike solution and the second was the isotopic ratio measured for the spiked sample solution. The mass flow rate of spike and atomic weights did not show much contribution. The calculated total uncertainty was $1.46ng{\cdot}g^{-1}$ for the standard SeMet ($126.30ng{\cdot}g^{-1}$). The experimental result was $127.09{\pm}1.46ng{\cdot}g^{-1}$ and the relative uncertainty was 1.20%.

Standardization of an enzyme-linked immunosorbent assay for detection of antibody to avian reticuloendotheliosis virus (세망내피증 바이러스 항체검출을 위한 ELISA 표준화)

  • Enzyme-linked immunosorbent assay (ELISA) for detection of antibodies to reticuloendotheliosis virus (REV) at single serum dilution was standardized. REV HI, one of the Korean field isolates, was inoculated into chicken embryo fibroblast (CEF) cells and was harvested from the culture fluids and cells after 10 to 12 days. Viruses were purified by centrifugation at the $107,000{\times}g$ for 12 hours on 20, 30, 45% (W/V) sucrose gradient. Virus specific fraction was collected and used as ELISA antigen. To standardize ELISA, the optimal concentration of coating antigen ($1{\mu}g/well$) and conjugate (1/1000) was determined by corrected OD (OD value of positive serum-OD value of negative serum) and P/N ratio (OD value of positive serum/OD value of negative serum). To calculate ELISA titer by measuring absorbance at 1/400 single serum dilution, serum titrations were carried out for various sample sera together with standard positive and negative sera. The observed titers of serum samples were plotted against sample/positive (s/p) ratios at 1/400 serum dilution. From the above data, the ELISA titers could be calculated by the equation of $log_{10}$ ELISA titer = 2.2763 ($log_{10}$ s/p) + 3.482 (r = 0.93). For evaluating the sensitivity, the standardized method were compared with conventional agar gel immunodiffusion (AGID) test method using serum samples collected from REV infected field chicken flocks. Fifty seven of 60 samples (95%) were positive for REV by ELISA, whereas only 11 (18.3%) samples were positive by AGID test. This results suggested that the ELISA tests developed in this study could be used for detection of antibodies to REV with high sensitivity.

Reduced sulfur gas loss in the bag sampling procedure - Comparison by calibration slope values (악취황 성분의 채취와 샘플백 내부의 시료 유실율 비교: 검량선의 기울기를 이용한 평가)

  • In this work, sample loss of reduced sulfur compounds (RSC) was investigated with respect to bag sampling techniques. For comparison purpose, calibration slopes were obtained by analyzing standards prepared by 'within syringe dilution' (WSD) method against those made either by 'Tedlar bag dilution' (TBD) or by 'polyester aluminum bag dilution' (PBD) method. The results indicated that the recovery rate of TBD showed the mean values of about 87%, while those of PBD about 77%. Despite the fact that sample loss is inevitable, precise sampling of reduced sulfur compounds may still be possible, if one acknowledges and be prepared for such loss rates of bag sampling methods.
