Isolation and Identification of Biofilm-Forming Marine Bacteria on Glass Surfaces in Dae-Ho Dike, Korea

  • Bacterial strains were isolated from biofilms formed on glass slides submerged in seawater in Dae-Ho Dike. Eight strains showing fast attaching ability were selected and identified. Their exopolysaccharide (EPS)-producing ability and EPS properties were characterized. Based on Microlog System, 4 among the 8 strains were identified as Micrococcus luteus and the rest were Bacillus thuringiensis, Bacillus megaterium,, Staphylococcus saprophyticus and Agrobacterium vitis. A, vitis was reidentified as Sulfitobacter pontiacus based on 16S rDNA sequence data. The amount of water-soluble EPS produced by the 8 strains ranged from 0.114 to 1.329 g$.$l$\^$-1/ and the productivity was negatively correlated with the cell biomass. The molecular weight of the produced EPS ranged from 0.38 to 25.19$\times$10$\^$4/ Da. Glucose and galactose were ubiquitous sugar components. Mannose, ribose, and xylose were also major sugar components. The molecular weight and composition of the EPS showed strain-specific variation.

Depositional Environment and Distribution of Heavy Metal off the Shihwa Dam

  • Depositional environment off the Shihwa Dam has been studied to investigate the change of sedimentation process and the pollution. In order to understand how the sediments are distributed, polluted and modified, depositional factors have been analyzed and compared with the previous data. Study area, located off the Shihwa Dam, was surveyed to collect 25 bottom samples and 2 cores in 1996 and echo-sounding in 1997. These sediments were analyzed for the study of the global characteristics of sediment such as grain size and organic matter. Among these samples, the selected twenty surface sediments were analyzed for the comparison with their contents of metallic elements (Al, Mn, Fe, V, Cr, Co, Ni, Cu, Zn, Cd, Pb, As). According to field and lab analysis of sediments, three sedimentological zones have been generally identified around study area; near the dam (sandy Silt), near the dike (Sand) and offshore (silty Sand) zones. Textural parameters show that the content of silt and clay is dominant near the dam excepting the dike zone of LNG Storage Base and offshore (Palmido). The total concentration of Mn, Ni, Fe, Zn and Cd in bulk sediments was increased after the construction of the dam, while the content of Mn and Cr were higher near tidal channel than in the offshore area. Meanwhile, the annual increasing pattern of some heavy metal has appeared in this area. Based on this primary study, modification of the depositional environment may be caused by the construction of the dam and LNG Storage Base. Additionally, environmental evaluation on organic/inorganic factors has been suggested for interpreting environmental changes caused by coastal development in the nearshore such as the Shihwa coastal area.

금강하구둑 어도의 생태적 기능 평가 (Ecological Function Assessment for a Fishways of Geumgang Estuary Dike)

  • The ichthyofauna and passing efficiency of fishways were surveyed in Geumgang estuary from May to October in 2009. The fishes collected were classified into 24 species, 23 genera belonging to 8 families. These 24 species consist of 17 freshwater fish species, 3 migration fish species and 4 peripheral freshwater fish species. Seasonal function of fishways showed the migration from May to June and returning to the reservoir of fishes be washed out to sea when sluice gate open by rainfall in July and August. Most of the fishes catched in fishways moved from middle tide to high tide except for July. Fishes ascended fishways between the middle ebb tide and the ebb tide only July, because small freshwater swim against the fishway for leaching the reservoir through the newly formed flow when the water level at the fishway kept dropping. Small fishes that body length less than 200 mm mainly used gabion type fishways and fishes over 200 mm utilized a cascade type fishways. Based on the total individuals collected, the efficiency of the fishway was 1 % for the anadromous fish and over 80 % for freshwater fish. Therefore, it is concluded that ecological function of the fishway in the Geumgang estuary dike be very important not only for fish migration but also freshwater fishes's returning.
