  • Microwave digestion is a preferred pretreatment method for agricultural samples because of its quick chemical reaction and minimum loss of analytes. In this research, a feedback temperature controller was developed to control the temperature inside a vessel for the microwave-assisted digestion system. An existing industrial microwave oven was fitted with the temperature controller for controlling inside temperature of the vessel. Four control methods, On/Off, proportional (P), proportional integral (PI), and proportional integral derivative (PID) were used and compared. Experimental results showed that PID control produced best temperature control performance. The PID controller could maintain the temperature of water sample and rice sample in the digestion system with error range of $-2.5{\sim}3.3^{\circ}C$ and $-1.9{\sim}0.5^{\circ}C$ at set temperature of $170^{\circ}C$, respectively.

  • This study was aimed at investigating a filtration bed system for the production of liquid fertilizer using wastewater generated after anaerobic digestion of animal manure. Slurry, Compost, & Biofiltration-Methane System, designed by Korean National Institute of Animal Science, is now a standard method to recycle anaerobic digestion wastewater. However, currently provided protocol to run the system needs continuous upgrades as the system is relatively new and can be more cost-effective if few adjustments are made. In this study, we tried to enhance the capability of the system to process anaerobic digestion wastewater by replacing the bed materials with the cheaper one, and determining the time point of operation from which filtrates can be utilized for fertilization.

  • The purpose of this study was to evaluate the influences of digestion temperature on the extraction of heavy metals from soil using the standard method established by Korean Ministry of Environment (KMES). A total of 7 heavy metals (As, Cd, Cr, Cu, Ni, Pb and Zn) in soil samples were extracted at varying digestion temperatures [(66 ± 2.0)℃, (73 ± 1.9)℃, (80 ± 1.3)℃, (85 ± 1.7)℃, (92 ± 2.0)℃, (98 ± 1.7)℃]. As, Cd, Cu, Pb and Zn concentrations remained relatively constant over the temperature range, but Ni and Cr concentrations greatly varied with the digestion temperature. The extent of variation in extraction efficiencies as compared to the concentration obtained at 66℃ was in following order; Ni (7.09% ~ 35.42%) > Cr (4.79% ~ 25.40%) > Zn (3.99% ~ 17.52%) > Cu (2.22% ~ 19.34%) > As (3.54% ~ 8.26%) > Cd (-5.08% ~ 1.08%) > Pb (-4.71% ~ -1.70%). The accuracy for certified reference materials at the digestion temperature of 80 and 85℃ was 98.7% ~ 105.8%. Therefore, digestion temperature of 80℃ ~ 85℃ is suggested to obtain reliable and reproducible data when the standard method by Korean Ministry of Environment is employed to analyze multiple heavy metal components in soil samples.

  • This study was conducted to evaluate two sample digestion procedures and instrumental determination parameters for analysis of lead in bone. Amputated human legs were treated by acid digestion or microwave dissolution prior to spectrometric analysis. Inductively coupled plasma mass spectrometry (ICP-MS) and graphite furnace atomic absorption spectrometry (GF-AAS) were used for determining bone lead levels. Recovery efficiencies using standard reference material from acid digestion measured by ICP-MS were in good agreement with those of the certified value, but in cases of acid digestion by GF-AAS and microwave digestion by both two methods, recovery underestimated and overestimated, respectively. For the bone samples, the lead concentrations obtained by ICP-MS after acid digestionwere in good agreement with those by GF-AAS (correlation coefficient = 0.983), but GF-AAS gave systematically higher values than ICP-MS. While a good agreement between two analytical methods after microwave digestion was also obtained (correlation coefficient = 0.950), bone lead concentrations from microwave were relatively higher than those from acid digestion. In conclusion, the use of the simple nitric acid digestion procedure at an ambient temperature coupled to ICP-MS seems to be efficient for the determination of lead in bone in consideration for both the convenience and validity.

  • Inductively coupled plasma-mass spectrometry (ICP-MS), one of the most commonly used instruments for metal analysis, was used to determine total uranium in soil. The method was named as "Modified ASTM C1345-96". When comparing with ASTM C1345-96, digestion time (2~3 days) was shorten to 7 hours and the treatment in furnace was eliminated. In analyses of 26 field soil samples, there was a significant difference in the average concentration of total uranium between modified ASTM C1345-96 and ASTM C1345-96 (F : 6.22 > Fc : 4.03, significance level : 0.05, n=26). The average concentration of modified ASTM C1345-96 was 1.8 times larger than that of ASTM C1345-96. In addition, modified ASTM C1345-96 was compared with other acid digestion methods for soil including ISO 11466, Modified ISO 11466, US EPA-3051, US EPA-3051A and US EPA-3052 using a certificated reference material (SRM 2711a, NIST) and field soil samples with different level of organic matter content (1.6%, 5.8%, 10.6%). Modified ASTM C1345-96 showed the best accuracy of 93.01% for SRM 2711a. Also, modified ASTM C1345-96 showed the higher extraction rates than other digestion methods by 11~45%.

  • Eight Holstein steers ($216{\pm}48kg$ body weight) fitted with ruminal and duodenal cannulas were used to evaluate effects of wheat straw processing (ground vs pelleted) at two straw inclusion rates (7% and 14%; dry matter basis) in dry rolled or steam-flaked corn-based finishing diets on characteristics of digestion. The experimental design was a split plot consisting of two simultaneous $4{\times}4$ Latin squares. Increasing straw level reduced ruminal (p<0.01) and total tract (p = 0.03) organic matter (OM) digestion. As expected, increasing wheat straw level from 7% to 14% decreased (p<0.05) ruminal and total tract digestion of OM. Digestion of neutral detergent fiber (NDF) and starch, per se, were not affected (p>0.10) by wheat straw level. Likewise, straw level did not influence ruminal acetate and propionate molar proportions or estimated methane production (p>0.10). Pelleting straw did not affect ($p{\geq}0.48$) ruminal digestion of OM, NDF, and starch, or microbial efficiency. Ruminal feed N digestion was greater (7.4%; p = 0.02) for ground than for pelleted wheat straw diets. Although ruminal starch digestion was not affected by straw processing, post-ruminal (p<0.01), and total-tract starch (p = 0.05) digestion were greater for ground than for pelleted wheat straw diets, resulting in a tendency for increased post-ruminal (p = 0.06) and total tract (p = 0.07) OM digestion. Pelleting wheat straw decreased (p<0.01) ruminal pH, although ruminal volatile fatty acids (VFA) concentration and estimated methane were not affected ($p{\geq}0.27$). Ruminal digestion of OM and starch, and post-ruminal and total tract digestion of OM, starch and N were greater (p<0.01) for steam-flaked than for dry rolled corn-based diets. Ruminal NDF digestion was greater (p = 0.02) for dry rolled than for steam-flaked corn, although total tract NDF digestion was unaffected (p = 0.94). Ruminal microbial efficiency and ruminal degradation of feed N were not affected (p>0.14) by corn processing. However, microbial N flow to the small intestine and ruminal N efficiency (non-ammonia N flow to the small intestine/N intake) were greater (p<0.01) for steam-flaked than for dry rolled corn-based diets. Ruminal pH and total VFA concentration were not affected ($p{\geq}0.16$) by corn processing method. Compared with dry rolled corn, steam-flaked corn-based diets resulted in decreased acetate:propionate molar ratio (p = 0.02). It is concluded that at 7% or 14% straw inclusion rate, changes in physical characteristics of wheat straw brought about by pelleting negatively impact OM digestion of both steam-flaked and dry-rolled corn-based finishing diets. This effect is due to decreased post-ruminal starch digestion. Replacement of ground straw with pelleted straw also may decrease ruminal pH.

  • Purpose - The purpose of this study is to promote sales of the renewable energy industry and to advance into the Southeast Asian market. Research design, data, and methodology - This study is to develop a highly efficient food waste treatment system for Southeast Asian renewable energy industry. The radiation treatment method was applied for this purpose. Results - To investigate effects of ionization on removal of non-degradable organic matter, the results from gamma irradiation and co-digestion process was compared to those from a co-digestion process. Based on the BMP test results, food wastes were oxidized by hydroxyl radicals, and the specific methane yield was 366 mL CH4/g VS. Methane composition was 82%. A WAS/food wastes co-digestion was developed for the treated of non-degradable organic matter in food wastes. The average efficiency of non-degradable organic matter were 92.2% using the food waste co-digestion. Conclusions - Performance of gamma irradiation and co-digestion process was superior to that of a co-digestion process (10-20%). This implies that food wastes can be high efficient co-digested by the gamma irradiation. It is believed that it will be possible to enter the Southeast Asian energy industry as a strategic technology in the overseas energy recovery industry.
