가족치료와 가족코칭의 융합적 접근 (Convergence Approach of Family Therapy and Family Coaching)

  • 본 연구는 2000년대 이후 전 세계적으로 개인 한 명의 내담자를 두고 상담과 코칭의 융합적 적용에 대한 중요성이 학계와 임상을 중심으로 급속히 진전됨에 따라 가족치료와 가족코칭의 융합적 접근을 시도해 보았다. 이를 위해 국내·외 선행 문헌 연구를 바탕으로 가족치료의 주요 이론 가운데 코칭과 접목하여 임상 만족도와 효과성을 높일 수 있는 코칭 기술을 제시하였다. 또한 가족치료가 가족코칭과 융합적 적용이 가능한지, 가족치료의 주요 이론들이 임상현장에서 코칭과 접목하여 활용될 때 어떤 유익이 있는지를 살펴보았다. 이를 위해 이중구속과 의사소통 코칭, 가짜 친밀성과 진실한 라포, 부부 균열과 갈등 중재, 분화와 객관적 관조를 비교·분석해 보았다. 본 연구를 기반으로 가족 치료뿐만 아니라 상담 및 심리치료 현장에서 코칭과의 융합적 접근을 시도함으로써 새로운 임상 치료모델과 가족코칭의 학문적 정립을 구축해 나가는 데 기여하기를 바란다.

Differentiation of Dopaminergic Neurons from Mesenchymal-Like Stem Cells Derived from Human Umbilical Cord Vein

  • One of the most extensively studied populations of multipotent adult stem cells are mesenchymal stem cells (MSCs). MSCs derived from the human umbilical cord vein (HUC-MSCs) are morphologically and immunophenotypically similar to MSCs isolated from bone marrow. HUC-MSCs are multipotent stem cells, differ from hematopoietic stem cells and can be differentiated into neural cells. Since neural tissue has limited intrinsic capacity of repair after injury, the identification of alternate sources of neural stem cells has broad clinical potential. We isolated mesenchymal-like stem cells from the human umbilical cord vein, and studied transdifferentiation-promoting conditions in neural cells. Dopaminergic neuronal differentiation of HUC-MSCs was also studied. Neural differentiation was induced by adding bFGF, EGF, dimethyl sulfoxide (DMSO) and butylated hydroxyanisole (BHA) in N2 medium and N2 supplement. The immunoreactive cells for $\beta$-tubulin III, a neuron-specific marker, GFAP, an astrocyte marker, or Gal-C, an oligodendrocyte marker, were found. HUC-MSCs treated with bFGF, SHH and FGF8 were differentiated into dopaminergic neurons that were immunopositive for tyrosine hydroxylase (TH) antibody. HUC-MSCs treated with DMSO and BHA rapidly showed the morphology of multipolar neurons. Both immunocytochemistry and RT-PCR analysis indicated that the expression of a number of neural markers including NeuroD1, $\beta$-tubulin III, GFAP and nestin was markedly elevated during this acute differentiation. While the stem cell markers such as SCF, C-kit, and Stat-3 were not expressed after neural differentiation, we confirmed the differentiation of dopaminergic neurons by TH/$\beta$-tubulin III positive cells. In conclusion, HUC-MSCs can be differentiated into dopaminergic neurons and these findings suggest that HUC-MSCs are alternative cell source of therapeutic treatment for neurodegenerative diseases.

  • Various stem cells or progenitor cells are being used to treat cardiovascular disease in ischemic heart disease, stem ceil therapy is expected to regenerate damaged myocardium. To evaluate effects of stem cell treatment, the method to image stem cell location, distribution and differentiation is necessary. Optical imaging, MRI, nuclear imaging methods have been used for tracking stem cells. The methods and proglems of each imaging technique are reviewed.

저신장 소아를 위한 성장호르몬 치료 (Short Stature and Growth Hormone Therapy)

  • Normal growth and development is of prime concern during childhood. The treatment of children with growth hormone deficiency has been revolutionized by growth hormone therapy. An improved height outcome with a final height within the target height range has been achieved. However, close follow-up with regular clinical and laboratory monitoring is essential for achieving the desirable height outcome. The theoretical unlimited supply of growth hormone has led to its wide spread use in a variety of disorders other than a growth hormone deficiency. Initially used in children with Turner syndrome, growth hormone is now used to treat chronic renal failure, an idiopathic short stature and intrauterine growth restrictions in addition to a wide array of newly emerging indications. This review summarizes the basics for a proper growth assessment, the differentiation of normal and abnormal growth causes of a short stature, and the indications for growth hormone treatment.

Structure and apoptotic function of p73

  • p73 is a structural and functional homologue of the p53 tumor suppressor protein. Like p53, p73 induces apoptosis and cell cycle arrest and transactivates p53-responsive genes, conferring its tumor suppressive activity. In addition, p73 has unique roles in neuronal development and differentiation. The importance of p73-induced apoptosis lies in its capability to substitute the pro-apoptotic activity of p53 in various human cancer cells in which p53 is mutated or inactive. Despite the great importance of p73-induced apoptosis in cancer therapy, little is known about the molecular basis of p73-induced apoptosis. In this review, we discuss the p73 structures reported to date, detailed structural comparisons between p73 and p53, and current understanding of the transcription-dependent and -independent mechanisms of p73-induced apoptosis.

Comparison of Reprogramming Methods for Generation of Induced-Oligodendrocyte Precursor Cells

  • Direct conversion by trans-differentiation is of growing interest in cell therapy for incurable diseases. The efficiency of cell reprogramming and functionality of converted cells are important considerations in cell transplantation therapy. Here, we compared two representative protocols for the generation of induced-oligodendrocyte progenitor cells (iOPCs) from mouse and rat fibroblasts. Then, we showed that induction of Nkx6.2, Olig2, and Sox10 (NOS) was more effective in mouse fibroblasts and that induction of Olig2, Sox10, and Zfp536 (OSZ) was more effective at reprogramming iOPCs from rat fibroblasts. However, OSZ-iOPCs did not show greater proliferation than NOS-induced cells. Because the efficiency of iOPCs generation appears to differ between cell species depending on transcription factors and culture conditions, it is important to select appropriate methods for efficient reprogramming.

Ornithine Decarboxylase: A Promising and Exploratory Candidate Target for Natural Products in Cancer Chemoprevention

  • Ornithine decarboxylase (ODC), the first enzyme in the polyamine biosynthesis, plays an important role in tumor progression, cell proliferation and differentiation. In recent years, ODC has been the subject of intense study among researchers, as a target for anti-cancer therapy and specific inhibitory agents, have the potential to suppress carcinogenesis and find applications in clinical therapy. In particular, it is suggested that ODC is a promising candidate target for natural products in cancer chemoprevention. Future exploration of ornithine decarboxyalse inhitors present in nature may offer great hope for finding new cancer chemporeventive agents.

탈모 치료에 관한 최신 동향 (The Latest Trends of Treatment for Alopecia)

  • Objective : The purpose of this study is to analyse research trend on alopecia including androgenetic alopecia, alopecia areata, etc.Methods : We searched Pubmed, CNKI and OASIS on the title "alopecia" in recent 5 years.Results : We found 23 studies on Pubmed, 20 on CNKI, and 3 on OASIS about treatments of alopecia. The subjects of them were about 5α-reductase inhibitor, Minoxidil, steroids, immunosuppression, laser therapy, PRP, microneedling, stem cell, herbal medicine, integrative medicine, external treatments and syndrome differentiation.Conclusions : In recent studies in western medicine were about combination therapy or safety and presented validities and superiorities about new treatments. Integrative medicine, external treatment and researches about patients' characteristics were reported in TCM. Korean medicine has advantages of considering whole body and phychological problems.

Highlighted STAT3 as a potential drug target for cancer therapy

  • Signal transducer and activator of transcription 3 (STAT3) is a cytoplasmic transcription factor that regulates cell proliferation, differentiation, apoptosis, angiogenesis, inflammation and immune responses. Aberrant STAT3 activation triggers tumor progression through oncogenic gene expression in numerous human cancers, leading to promote tumor malignancy. On the contrary, STAT3 activation in immune cells cause elevation of immunosuppressive factors. Accumulating evidence suggests that the tumor microenvironment closely interacts with the STAT3 signaling pathway. So, targeting STAT3 may improve tumor progression, and anti-cancer immune response. In this review, we summarized the role of STAT3 in cancer and the tumor microenvironment, and present inhibitors of STAT3 signaling cascades.

Effect of Photobiomodulation on the Mesenchymal Stem Cells

  • Photobiomodulation forms the basis of photomedicine and is defined as the effect of coherent or non-coherent light sources, such as low-level lasers and light-emitting diodes, on cells and tissues. This treatment technique affects cell functions, proliferation, and migration, and plays an important role in tissue regeneration. Mesenchymal stem cells (MSCs) are known to be beneficial for tissue regeneration, and the combination of stem cell therapy and laser therapy appears to positively affect treatment outcomes. In general, a low-power laser has a positive effect on MSCs, thereby facilitating improvements in different disease models. This study elucidates the mechanisms and effects of low-power laser irradiation on the proliferation, migration, and differentiation of various MSCs that have been examined in different studies.
