곤충 유전자를 이용한 항세균성 펩타이드 생산 및 응용에 관한 기초 연구로서 비병원성 세균(Escherichia coli K12)으로 면역 반응을 유도한 누에로부터 발현량이 증가하는 유전자 중 Hyalophora cecropia의 attacin과 cDNA 상동성을 나타내는 BmInc6 클론을 분리하고 그 특성을 조사하였다. BmInc6 cDNA의 전체염기서열을 분석한 결과 그 크기는 852 bp이고, 35번째 염기에서 변역이 개시되어 679 bp 위치에서 종결되는 open reading frame을 가지며 812번째 위치에 잠정 전사 종결 신호의 존재가 확인되었다(GenBank, AF005384). BmInc6에 의해 coding되는 아미노산은 214개이며, hydropathy 분석 결과 친수성을 나타내는 단백질이었다. 그리고 BmInc6 유전자에 의해 연역되는 펩타이드를 nuecin으로 명명하였다. Nuecin 유전자를 baculovirus 발현 백터계를 이용하여 곤충세포주에서 발현시킨 결과 전사체의 크기는 약 950 bp였고, 세포내 벌현 펩타이드의 분자량은 약 23 kDa이었다. 세포내에서 발현된 nuecin 전구체로 추정되는 23 kDa 펩타이드는 세포외로 분비되는 과정에서 약 3 kDa의 signal 펩타이드가 제거됨으로서 약 20 kDa의 성숙 nuecin으로 된다는 사실을 단백질 전기영동상으로 확인하였다. Nuecin 단백질의 항세균 활성을 수종의 그람 음성 및 양성 세균에 대해 검정한 결과, 특히 E. coli와 Bacillus subtilis에 높은 활성을 나타내었으며, attacin이 한정된 그람 음성 세균에만 항세균 활성을 나타내는데 비해 nuecin은 그람 음성은 물론 그람 양성에도 항세균 활성을 나타내어, 보다 넓은 항세균 스펙트럼을 가지는 것을 알 수 있었다.
스마트 교육 환경과 4차 산업 혁명 시대를 맞이하여 편리한 기능을 갖는 다양한 테크놀로지들을 활용하는 새로운 차원의 디지털 수학 교과서가 필요하게 되었다. 한국의 경우 초 중등 수학 교육에서는 여러 다양한 시도가 있었으나 대학 수학교육의 경우 디지털 수학 교과서 관련 연구는 미비하였다. 본 논문에서는 선형대수학을 중심으로 디지털 콘텐츠와 대화형 실습실을 활용하는 디지털 교과서를 소개한다. 본 교과서는 본 연구진이 직접 개발하여 누구나 http://matrix.skku.ac.kr/LA-K에서 다운로드 받을 수 있도록 제공하였으며, 기존의 종이 교과서(서책형)를 단순히 pdf 형태의 파일로 변환하여 애니메이션이나 참고자료 등을 추가한 수준에서 벗어나 전자책, 웹 콘텐츠, 강의 동영상, 대화형 실습실을 포함한다. 본 선형대수학 디지털 교과서는 학생들이 어떠한 모바일 기기에서든 시간과 장소의 제약 없이 자유롭게 사용할 수 있으며, 계산, 코딩 및 타이핑 과정에서 절약된 시간을 수학 개념을 더 깊이 이해하는데 사용할 수 있다. 코드를 포함한 대화형 실습실 및 동영상 강의를 탑재한 최초의 수학 디지털 교과서로 평가되는 본 연구의 결과물은 차세대 디지털 교과서의 주요 모델 중 하나가 될 것으로 판단된다.
통계적 신호 전송에 관한 위너의 MMSE와 정보이론 관점에서 처음으로 증명한 샤논의 상호 정보량을 비교함으로서 둘 사이의 관련성을 연구하였다. 위너가 신호 전송에서 보려했던 것은 잡음이 존재하는 채널에서 복원하려는 신호의 전송 품질의 근본적 극한(limit)를 계산해내려는 것이라 해석할 수 있다. 반면, 샤논은 엔트로피 개념을 적용하여 상호 정보에 대한 불확실성의 차를 최대화로 하는 전송 정보량의 근본적 상한(upper bound)의 극한(limits)을 계산을 계산하려는 것이라 해석할 수 있다. 본 논문의 관심은 샤논의 점 대 점 채널 용량의 근본적 극한을 계산하는 샤논의 공식을 유도함에 있어 위너의 최소 평균 자승 오차(MMSE) 컴바이너에 의한 최적 위너 필터를 사용했을 때 위너의 MMSE와 샤논의 상호 정보량이 미적분관계로 연결되어 있음을 확인하는 것이며, 위너 필터의 MMSE 와 샤논의 상호 정보량이 만나는 점에서 대역 효율성의 상한과 에너지 효율성의 하한을 계산하였다. mod-${\Lambda}$ AWGN 채널 모델에 의한 적절한 성형 격자 ${\Lambda}$의 선택과 ${\alpha}$의 MMSE 추정에 의해 격자형 부호 방식이 샤논의 원래 채널 용량 극한에 미치게 됨을 확인하였다.

All living organisms use numerous signal-transduction pathways to sense and respond to their environments and thereby survive and proliferate in a range of biological niches. Molecular dissection of these signalling networks has increased our understanding of these communication processes and provides a platform for therapeutic intervention when these pathways malfunction in disease states, including infection. Owing to the expanding availability of sequenced genomes, a wealth of genetic and molecular tools and the conservation of signalling networks, members of the fungal kingdom serve as excellent model systems for more complex, multicellular organisms. Here, we employed Cryptococcus neoformans as a model system to understand how fungal-signalling circuits operate at the molecular level to sense and respond to a plethora of environmental stresses, including osmoticshock, UV, high temperature, oxidative stress and toxic drugs/metabolites. The stress-activated p38/Hog1 MAPK pathway is structurally conserved in many organisms as diverse as yeast and mammals, but its regulation is uniquely specialized in a majority of clinical Cryptococcus neoformans serotype A and D strains to control differentiation and virulence factor regulation. C. neoformans Hog1 MAPK is controlled by Pbs2 MAPK kinase (MAPKK). The Pbs2-Hog1 MAPK cascade is controlled by the fungal "two-component" system that is composed of a response regulator, Ssk1, and multiple sensor kinases, including two-component.like (Tco) 1 and Tco2. Tco1 and Tco2 play shared and distinct roles in stress responses and drug sensitivity through the Hog1 MAPK system. Furthermore, each sensor kinase mediates unique cellular functions for virulence and morphological differentiation. We also identified and characterized the Ssk2 MAPKKK upstream of the MAPKK Pbs2 and the MAPK Hog1 in C. neoformans. The SSK2 gene was identified as a potential component responsible for differential Hog1 regulation between the serotype D sibling f1 strains B3501 and B3502 through comparative analysis of their meiotic map with the meiotic segregation of Hog1-dependent sensitivity to the fungicide fludioxonil. Ssk2 is the only polymorphic component in the Hog1 MAPK module, including two coding sequence changes between the SSK2 alleles in B3501 and B3502 strains. To further support this finding, the SSK2 allele exchange completely swapped Hog1-related phenotypes between B3501 and B3502 strains. In the serotype A strain H99, disruption of the SSK2 gene dramatically enhanced capsule biosynthesis and mating efficiency, similar to pbs2 and hog1 mutations. Furthermore, ssk2, pbs2, and hog1 mutants are all hypersensitive to a variety of stresses and completely resistant to fludioxonil. Taken together, these findings indicate that Ssk2 is the critical interface protein connecting the two-component system and the Pbs2-Hog1 pathway in C. neoformans.
Objective: Oxidative stress (OS) is a pathological process arising from the excessive production of free radicals in the body. It has the potential to alter animal gene expression and cause damage to the jejunum. However, there have been few reports of changes in the expression of long noncoding RNAs (lncRNAs) in the jejunum in piglets under OS. The purpose of this research was to examine how lncRNAs in piglet jejunum change under OS. Methods: The abdominal cavities of piglets were injected with diquat (DQ) to produce OS. Raw reads were downloaded from the SRA database. RNA-seq was utilized to study the expression of lncRNAs in piglets under OS. Additionally, six randomly selected lncRNAs were verified using quantitative real-time polymerase chain reaction (qRT-PCR) to examine the mechanism of oxidative damage. Results: A total of 79 lncRNAs were differentially expressed (DE) in the treatment group compared to the negative control group. The target genes of DE lncRNAs were enriched in gene ontology (GO) terms and Kyoto encyclopedia of genes and genomes (KEGG) signaling pathways. Chemical carcinogenesis-reactive oxygen species, the Foxo signaling pathway, colorectal cancer, and the AMPK signaling pathway were all linked to OS. Conclusion: Our results demonstrated that DQ-induced OS causes differential expression of lncRNAs, laying the groundwork for future research into the processes involved in the jejunum's response to OS.
일반적으로 누에나방의 평균 생존시수는 암컷과 수컷이 각각 8일과 5일로 알려져 있다. 그러나 J037 품종의 성충 수명은 상당히 길며, 반대로 Daizo(sdi) 품종은 상당히 짧은 성충 수명 특성을 갖고 있다. 이러한 누에 품종 간 성충 수명의 차이는 유전적 차이에 따른 현상으로 추정되어 왔다. 본 연구에서는 장단명누에 품종의 발현유전자를 분석함으로써 누에 수명 관련 유전자를 탐색하여 그 특성을 분석하고자 하였다. 우선, 장명 수나방 mRNA를 사용하여 cDNA 유전자은행을 제작하고, 제작된 유전자은행으로부터 2,688개 클론을 무작위 선발한 후 장단명 수나방 cDNA를 각각의 탐침으로 차별화선별을 수행하였다. 본 연구에서 차별화 발현하는 193개의 클론을 선발하여 EST 상동성 분석을 통해 최종적으로 154개의 J037 수나방 수명 관련 독립유전자를 선발할 수 있었는데, 장명 탐침에서 과발현하는 124개와 단명 탐침에서 과발현하는 30개로 구성된다. 154개 독립유전자 중 가장 많은 발현빈도수를 보인 유전자는 cytochrome oxidase subunit-1으로서 모두 9회로 확인되었고, 현재까지 기능에 관해 알려진 바 없는 1-50번 독립유전자가 5회로 두 번째로 높은 발현빈도를 보였다. 154개 선발 독립유전자를 기능별로 분류한 결과, unclassified protein군에 가장 많은 24%의 독립유전자가 포함되어 있었다. 본 연구에서 두 번째로 발현빈도가 높은 클론(ID;1-50)의 염기서열 및 아미노산 서열을 분석하였는데, 1-50번 유전자는 전체 1,523 bp 염기로 구성되어 있으며, 723개 염기쌍이 240개의 아미노산을 coding 하고 있었다. 본 연구에서 선발된 주요 수명 관련 유전자는 국제유전자은행 EST database에 등록하였다.
