Effects of dietary protein level on growth performance and nitrogen excretion of dairy heifers

  • Objective: Protein supplementation is costly and can result in excess nitrogen (N) excretion. The objective of this study was to evaluate the effects of feeding different levels of dietary protein on average daily gain, body size, rumen fermentation, and nitrogen excretion of 8 to 10 month-old Holstein heifers. Methods: Thirty-six Holstein heifers were divided into 12 blocks according to age ($273{\pm}6.2d$) and were randomly assigned to diets containing a low (10.2% dry matter [DM]), medium (11.9% DM), or high (13.5% DM) level of dietary crude protein (CP). All diets contained approximately 70% roughage and 30% concentrate with similar dietary metabolizable energy (ME) content (2.47 Mcal/kg). Results: Dry matter intake did not differ among the treatments, and average daily gain increased with the increasing dietary protein, 0.79, 0.95, 0.97 kg/d for low, medium, and high group, respectively. Body height increased linearly with increasing dietary CP but no other significant differences in body dimensions were found among the treatments. The increased ratios of dietary CP improved the rate of rear teat length growth remarkably (p<0.05). There was no difference in rumen pH or ruminal major volatile fatty acid (acetate, propionate, and butyrate) concentration among the 3 diets, but rumen ammonia-N concentration increased with the higher dietary CP (p<0.05). Increasing N intake led to increased total N excretion; urinary N excretion was significantly increased (p<0.05) but fecal N excretion was similar among the treatments. Conclusion: These data suggest that the diet containing 11.9% CP (ME 2.47 Mcal/kg) could meet the maintenance and growth requirements of 9 to 11 month-old Holstein heifers gaining approximately 0.9 kg/d.

식이 단백질과 cysteine 수준이 흰쥐의 Cadmium 중독에 미치는 영향 (Effect of Dietary Protein and Cysteine Levels on Cadmium Toxicity in Rats)

  • This study was performed to investigate the effect of dietary protein and cysteine levels on cadmium toxicity in rats. Seventy-two male rats of Sprague-Dawley strain weighting 171$\pm$3g were blocked into 12 groups according to body weight, and were raised for 30 days. cadmium chloride was given at levels of 0 or 400ppm, protein at levels of 7, 15 and 40%, and cysteine was added(total dietary cysteine contents : 0.45%) to diet or not. The results are summarized as follow. Food intake, weight gain, food were lower than those of cadmium free group. But, these were increased with increasing dietary protein level and cysteine addition. Fecal cadmium excretion was remarkably increased in high protein (40%) groups. Thus, cadmium retention rates were decreased in high protein groups. Metallothionein concentrations in liver and kidney were increased in cysteine addition, and cadmium administration. Especially, these were remarkably increased in cadmium and cysteine added groups. Urinary calcium excretion was increased with cadmium administration, but urinary protein excretion and creatinine clearance were not changed in these animal. In conclusion, food intake, weight gain and organ weights were decreased with administration. Cadmium toxicity was alleviated by increasing fecal cadmium excretion, while cysteine addition increased metallothionein concentrations in liver and kidney. From these results, it was shown that cadmium toxicity was alliviated by synergistic effect of high protein level and cysteine addition.

식이 단백질의 종류와 함황아미노산 함량이 성장기 쥐의 골밀도에 미치는 영향 (The Effect of Dietary Protein Source and Sulfur Amino acid Content on bone Metabolism in Growing Rats)

  • This study was performed to evaluate the effect of dietary protein source and sulfur amino acid content on bone metabolism in ra. Thirty male rats (body weight 145$\pm$2g) were divided into three groups. The rats in the first group were fed on casein 20% diet as animal protein source and those in the second group were fed on soy 20% diet as plant protein source. Sulfur amino acid ratio of these group was 1.07:1. The rats in the third group were fed on soy 20% diet and the sulfur amino acid were supplemented with the amount contained as much in the soy 20% diet. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks, The total body, spine, femur bone mineral density and bone mineral content were measured using Dual Energy X-ray Absorptiometry Calcium, phosphate, pyridinoline, creatinine in urine and calcium, phosphate, alkaline phosphatase, osteocalcin in serum were measured. During the experimental period, plant protein (soy protein) group had a lower urinary Ca excretion, urine pyridinoline & crosslinks value and had a higher Ca efficiency in total bone and femur bone mineral density than animal protein (casein) group. There were no significant differences in serum calcium, phosphate, alkaline phosphatase and osteocalcin among the three groups of the rats. The findings from this study demonstrated that plant protein (soy protein) is beneficial of bone mineral density because it had a higher Ca efficiency in total bone and femur bone mineral density than animal protein (casein). However, the supplementation of sulfur amino acid on soy results were consistent with prior studies that dietary sulfur amino acid load had a negative effect on calcium balance. The rats fed sulfur amino acid supplementation diet increased urinary calcium excretion and decreased calcium efficiency for total and femur mineral density.   • 본 연구에서는 식이단백질 수준이 성장기 흰쥐의 신장 발달과 기능에 미치는 영향을 조사하기 위해 평균체중이 97.5$\pm$1.9g인 숫컷 흰쥐를 5%, 15% 그리고 50% casein 식이로 6주간 사육하였다. 체중증가량은 단백질 수준이 높을수록 높았으며 특히 단백질수준이 신장무게에 미친영향이 간, 비장, 흉선 등이 다른 장기에 비해 컸다. 신장내 총 DNA와 RNA 함량에서는 유의적인 차이가 없었다. 총 protein과 단위 g당 protein 함량은 식이내 단백질수준이 높을 수록 증가했다. 사구체여과율(GFR)은 식이단백질 수준이 높을수록 높았으나 신혈류(RBF)는 각 군 사이에 유의적인 차이를 보이지 않았다. 뇨질소배설량은 단백질 수준에 비례하여 유의적인 차이를 보였고 노칼슘배설량은 5%군이 낮은 경향을 보였다. 본 연구 결과로 성장기의 흰쥐에서 고단백식이는 신장의 무게증가를 촉진하며 사구체여과율을 증가시킴을 알 수 있다.

Effects of different levels of dietary crude protein on the physiological response, reproductive performance, blood profiles, milk composition and odor emission in gestating sows

  • Objective: This study was conducted to evaluate the effects of crude protein (CP) levels on the physiological response, reproductive performance, blood profiles, milk composition and odor emission in gestating sows. Methods: Seventy-two multiparous sows (Yorkshire×Landrace) of average body weight (BW), backfat thickness, and parity were assigned to one of six treatments with 10 or 11 sows per treatment in a completely randomized design. Experimental diets with different CP levels were as follows: i) CP11, corn-soybean-based diet containing 11% CP; ii) CP12, corn-soybean-based diet containing 12% CP; iii) CP13, corn-soybean-based diet containing 13% CP; iv) CP14, corn-soybean-based diet containing 14% CP; v) CP15, corn-soybean-based diet containing 15% CP; and vi) CP16: corn-soybean-based diet containing 16% CP. Results: There was no significant difference in the performance of sow or piglet growth when sows were fed different dietary protein levels. Milk fat (linear, p = 0.05) and total solids (linear, p = 0.04) decreased as dietary CP levels increased. Increasing dietary CP levels in the gestation diet caused a significant increase in creatinine at days 35 and 110 of gestation (linear, p = 0.01; linear, p = 0.01). The total protein in sows also increased as dietary CP levels increased during the gestation period and 24 hours postpartum (linear, p = 0.01; linear, p = 0.01). During the whole experimental period, an increase in urea in sows was observed when sows were fed increasing levels of dietary CP (linear, p = 0.01), and increasing blood urea nitrogen (BUN) concentrations were observed as well. In the blood parameters of piglets, there were linear improvements in creatinine (linear, p = 0.01), total protein (linear, p = 0.01), urea (linear, p = 0.01), and BUN (linear, p = 0.01) with increasing levels of dietary CP as measured 24 hours postpartum. At two measurement points (days 35 and 110) of gestation, the odor gas concentration, including amine, ammonia, and hydrogen sulfide, increased linearly when sows fed diets with increasing levels of dietary CP (linear, p = 0.01). Moreover, as dietary CP levels increased to 16%, the odor gas concentration was increased with a quadratic response (quadratic, p = 0.01). Conclusion: Reducing dietary CP levels from 16% to 11% in a gestating diet did not exert detrimental effects on sow body condition or piglet performance. Moreover, a low protein diet (11% CP) may improve dietary protein utilization and metabolism to reduce odor gas emissions in manure and urine in gestating sows.

Effects of Dietary Protein Sources on Growth and Body Composition in Olive Flounder (Paralichthys olivaceus)

  • A 6-week feeding trial was conducted to investigate the effects of six different dietary animal protein sources on growth and body composition of olive flounder, Paralichthys olivaceus in recirculating system. White fish meal (WFM), flounder muscle (FLM), carp muscle (CM), blood meal (BM), squid liver powder (SLP) and casein (CA) were used as the main animal protein sources in the six experimental diets. Fish averaging $2.9\pm0.03g$ $(mean\pm SD)$ were distributed to each aquarium as a group of 15 fish and were fed one of the six experimental diets to each treatment of triplicate groups. After 6-week of the feeding trial, fish fed white fish meal (WFM) and flounder muscle (FLM) diets showed a significant higher weight gain $(WG\%)$ (P<0.05) than those of fish fed the CM, BM, SLP and CA diets. Fish fed BM diet showed the lowest WG among all the dietary treatments. Feed conversion ratio (FCR) and protein efficiency ratio (PER) showed the similar trend as WG. Hematocrit and hemoglobin were not affected by the dietary treatments. Fish fed the FLM and CM diets showed significant higher survival rate than those of fish fed BM diets, and there was no significant difference in survival of fish fed WFM, FLM, CM, SLP and CA diets. These results indicated that WFM and FLM are the best dietary protein sources tested in olive flounder.

EFFECTS OF DIETARY CELLULOSE AND PROTEIN LEVELS ON GROWTH PERFORMANCE, ENERGY AND NITROGEN UTILIZATION, LIPID CONTENTS AND DEVELOPMENT OF INTERNAL ORGANS IN GROWING CHICKS

  • In order to investigate the effects of dietary cellulose and protein levels on chick performance, four semi-purified diets were formulated so as to contain cellulose at levels of 5% (LC) and 20% (HC) in combination with 10% (LP) and 20% (HP) protein, and fed ad libitum to 1-week-old White Leghorn male chicks for 3 weeks. There were no significant differences in feed intake, body weight gain and feed efficiency between the LC-HP and HC-HP groups. All parameters were lower in the LP groups; the HC-LP group consumed very small amount of feed and lost body weight during the experiment. The retention rates of DM, ash, nitrogen and energy were higher in the HP than the LP groups. The triglyceride concentration of carcass was lower in the HC-LP group and that of liver was higher in the LC-LP group. The carcass total cholesterol level was higher in the HC-HP group. The relative weight of most digestive organs was higher in the HP group irrespective of the cellulose level. In conclusion, the chick performance was primarily influenced by dietary protein level, and when the chicks were fed inadequate levels of protein, the low cellulose level gave a better performance than the high cellulose level.

  • Dietary protein is the most expensive component in flounder feed because of the high protein requirement of this species. The non-protein energy sources may also influence the protein utilization of fish. The inclusion of adequate levels of non-protein energy sources in diets can minimize use of protein as an energy source. Protein sparing effect by fat and carbohydrate has been studied in other fish. The Requirement of Ruminal Degradable Protein for Non-Structural Carbohydrate-Fermenting Microbes and Its Reaction with Dilution Rate in Continuous Culture

  • A continuous culture study was conducted to determine the impact of ruminal degradable soy protein (S-RDP) level and dilution rate (D) on growth of ruminal non-structural carbohydrate-fermenting microbes. Corn starch, urea and isolated soy protein (ISP) were used to formulate three diets with S-RDP levels of 0, 35 and 70% of total dietary CP. Two Ds were 0.03 and $0.06h^{-1}$ of the fermenter volume in a single-effluent continuous culture system. As S-RDP levels increased, digestibilities of dietary dry matter (DM), organic matter (OM) and crude protein (CP) linearly (p=0.001) decreased, whereas digestion of dietary starch linearly (p=0.001) increased. Increasing D from 0.03 to $0.06h^{-1}$ resulted in decreased digestibilities of dietary DM and OM, but had no effect on digestibilities of dietary starch (p=0.77) and CP (p=0.103). Fermenter pH, the concentration of volatile fatty acids (VFA) and daily VFA production were unaffected (p=0.159-0.517) by S-RDP levels. Molar percentages of acetate, propionate and butyrate were greatly affected by S-RDP levels (p=0.016-0.091), but unaffected by D (p=0.331-0.442). With increasing S-RDP levels and D, daily bacterial counts, daily microbial N production (DMNP) and microbial efficiency (MOEFF; grams of microbial N produced per kilogram of OM truly digested) were enhanced (p=0.001). The increased microbial efficiency with increasing S-RDP levels is probably the result of peptides or amino acids that served as a stimulus for optimal protein synthesis. The quantity of ruminal degradable protein from soy proteins required for optimum protein synthesis of non-structural carbohydrate-fermenting microbes appears to be equivalent to 9.5% of dietary fermented OM.

  • 단백질의 수준과 종류, 지방의 수준에 따라서 나타나는 실험동물의 성장, 장기와 지방조직을 관찰하였으며 mitogen에 의한 lympocyte의 생산능력 및 PFC에 의한 면역능력을 관찰한 결과는 다음과 같았다. 1) 실험1의 결과, 체중증가량과 장기 무게는 지방수준에 관계없이 6%의 casein을 섭취한 군이 15%나 30% casein군에 비하여 낮았다. 그러나 15%군과 30%군 사이에는 유의적인 차이가 없었다. 2) 전체 실험을 통하여 본 연구에서는 단백질의 종류나 수준에 따른 면역능력의 차이는 나타나지 않았다. 그러나 단백질의 수준이나 질에 관계없이 고지방식이(30%)를 섭취하였을 때가 저지방식이를 섭취하였을 때에 비하여 면역능력이 낮은 경향을 보였다.
