Antioxidative and Hypoglycemic Effects of Silk Fibroin/SericinMixtures in High Fat-Fed Mice

  • The effect of dietary feeding of silk fibroin/sericinmixtureson the antioxidative status and glucose metabolism in high fat-fed mice was investigated. The mice weregiven experimental diets for 6 weeks: normal control (NC),high fat (HF) andhigh fat supplemented with F100 (pure fibroin, HF-F100), F81 (81:19 fibroin-sericin, w/w, HF-F81) or F50 (50:50 fibroin-sericin, w/w, HF-F50). The silk protein-fed mice showed decreased lipid peroxidation, enhancedantioxidant enzymesactivities and lower blood glucose level relative to HF group. The HF-F50 animals exhibited significantly lower insulin level, higher glycogen concentration, enhanced hepatic glucokinaseactivity and reduced glucose-6-phosphate and phosphoenolpyruvatecarboxynaseactivities than the HF ones. The $in$ $vivo$ antioxidant activity and hypoglycemic action tended to increase with increased amount of sericin and decreased fibroin content in the diet. These findings demonstrate that silk protein, particularly sericin, may be beneficial in suppressing high fat diet-induced hyperglycemiaand oxidative stress.

Less Weight Gain in Obese Rats by Feeding Biocellulose

  • This study evaluated the effect of biocellulose and the diet formulation on reducing body weight gain of obese rats induced by high-fat diet for 10 weeks. Thirty male Sprague-Dawley rats were randomly assigned to high-fat diet group (OB-CON), high-fat diet group containing 5% biocellulose (OB-BIO), and high-fat diet group containing 5% dietary formulation (OB-DF). After feeding each diet for additional 10 weeks, body weight gains of OB-BIO and OB-DF groups were lower by 3.3 and 4.8%, respectively as compared with that of OB-CON group. Although not significant, measured values of the perirenal and visceral fat pads of OB-BIO and OB-DF groups were lower than those of the OB-CON group. The weight of interscapular brown adipose tissue did not show significant difference in all groups. The size of adipocyte in rats was lower in both OB-BIO and OB-DF groups. Thus, biocelluose and the diet formulation showed the anti-obesity effect.

Effects of Houttyunia cordata Ethanol Extracts on Serum Lipids and Antioxidant Enzymes in Rats Fed High Fat Diet (고지방 식이에 따른 어성초 추출물 투여가 혈청지질 및 항산화 효소 활성에 미치는 영향)

  • Effects of Houttuynia cordata ethanol extracts on lipid metabolism and antioxidant enzymes in Sprague Dawley male rats were investigated. High fat used in the diet mixture included 10% of lard, 1% of cholesterol and 0.25% of sodium cholate. Total serum cholesterol contents of the rats fed Houttuynia cordata extracts were decreased compared to the control. On the other hand, HDL cholesterol contents were increased along with the decrease of athrogenic index. When high fat diet was fed, total serum cholesterol contents were significantly increased(p<0.01) with the athrogenic index increase of four times of the control. With the administration of Houttuynia cordata extract HDL cholesterol was increased by 53% in the high fat diet group. Antioxidant enzymes including GST and catalase activities were increased comparing the control. On the otherhand, the extracts lowered phospholipid(p<0.01), GOT, GPT, Cu,Zn SOD and alkaline phosphatase enzyme activities in the serum which are related to the liver functions. Therefore, the above results suggest that Houttuynia cordata ethanol extracts can help to maintain normal liver functions and help to protect from peroxidative damages caused by excess dietary fat intake.

Effect of Cnidi rhizoma Water-Extracted Solution on Fat Contents in Plasma, Liver and Adipose, and Fecal Steroids of Fatted Rats (천궁의 열수 추출액이 고지방식이에 의한 흰쥐의 혈장, 간 및 지방조직의 지질함량과 분변 Steroids에 미치는 영향)

  • This study was carried out to evaluate the effect of Cnidi rhizoma (CR) water extract on fat accumulation in fatted rats induced by the oral high fat administration for six weeks. Total cholesterol, neutral lipid and epididymal fat pad of CR groups were lower than the control group. On the other hand, high density lipoprotein(HDL) -cholesterol was somewhat higher In CR than In the control group. In addition, the same result as the above case was obtained when examining adipose for liver and epididymal fat components. During the feeding experiment, level of low density Lipoprotein(LDL) and very low density Lipoprotein (VLDL) increased while HDL was decreased. The degree of LDL and VLDL was decreased however, and HDL was, increased In CH groups as compared with the control group. Insulin and cortisol were higher than the control group, due to the fat accumulation. Experiment group showed the increased excretion as compared with the control group, in the excretion of bile acid and neutral sterol. The liver of the control group observed by the light microscope appeared to the fatty liver, but CR groups showed some improvement of the fatty liver. Based on the above results, it was shown that It Is possible to improve fat accumulation induced, by high fat dietary through using the oral administration of Cnidi rhizoma boiling extract solutions.

Effects of Methanol Extract of Ma-huang on Adipocyte of Epidiymal Fat in Rat Fed High Fat Diet (고지방 식이를 섭취한 흰쥐에서 마황의 메탄올 추출물이 체지방조직 세포에 미치는 영향)

  • Objectives : Ma-huang is a traditional Chinese medicinal herb, derived from Ephedra sinica Stapf and other Ephedra species, used to treat asthma, nose and lung congestion, and fever with anhidrosis. It contains 0.5-2.5% by weight of total alkaloids, of which ephedrine accounts for 30 to 90%. Recently, Ma-huang has been used as a source of ephedrine in many dietary supplements formulated for the treatment of obesity, since ephedrine has been found to be effective in inducing weight loss in the obese. In this study the effects of the methanol extract of Ma-huang on the adipocyte of epididymal and brown fat pads in rats fed a high fat diet for six weeks were studied. Methods : Male Sprague Dawley rats weighing an average 94g (4 weeks old) were fed either a regular diet (RE) or a high fat diet (HF), and the HF group was subdivided into a Ma-huang methanol extract (30mg/100g body weight) group (HF+MH). The weight of epididymal fat pad and brown adipose tissue were measured. The cell size and cell number per unit area of epididymal fat pad were investigated. Results : The yield weight of methanol extract of Ma-huang was 3.63mg per l00g of Ma-huang. The body weight gain of the HF group was similar with that of the HF+ MH but higher than that of the RE. The weights of the epididymal fat pads and brown adipose tissue of the RE group were lower than those of HF and HF+MH groups. The cell sizes and numbers per unit area of epididymal fat pads of the RE and HF+MH groups were larger than those of HF group. The cell numbers per unit area size of epididymal fat pads were the smallest in the RE group. Conclusions : It could be concluded that the Ma-huang extract has no effect on the epididymal fat pads in rats fed a high fat diet and the clinical application of Ma-huang for the treatment of obesity should be re-considered.

  • In this study the sensitivity subjected to housing environmental as well as actual life and subjected to dietary living using experimental mice were investigated. In order to invest sensitivity for dietary life of the mice, using the dropwort(MINAISU) juice and the fermented with the extracted dropwort for change of weight on the mice, this was examined. The result of thus, group of infection through the mouth on dietary treatment for the high-fat foods and the dropwort more than compared with group of dietary treatment for the high-fat foods tended to be decreased. On fermented secretion group of the dietary treatment total cholesterol number also was decreased, the concentration of HDL cholesterol was shown a slight increased. The result of studying for the sensitivity controlled architectural structural material for the mice, coming under wooden living environment, making appearance with increasing weight as well as taking food and drink for psychological rest was considered. To examine changing weight through the mice in regard to the sensitivity for dietary life and living environment, in connection with promotion of health as well as diseases control the caused by those, would be achieve new balanced growth on basic study. Also that would be anticipated to making good use of thus results.

The Effects of n-6/n-3 and P/S Ratio of Dietary Lipid on Lipid Metabolism of Rats at Different Age (n-6/n-3 비율과 P/S 비율을 변화시킨 식이지방이 나이가 다른 흰쥐의 체내 지방대사에 미치는 영향)

  • The effects of age and dietary fatty acid composition on lipid metabolism were investigated in Sprague-Dawley strain male rats. These animals weighing 88.6$\pm$2.2g were fed 10% dietary fat(W/W, 20% of total energy) with 0.5, 1, 2 P/S ratio and in each P/S ratio there were three different levels of n-6/n-3 fatty acid ratio ; 2, 4, 8. The experimental period was 1 month, 6 months and 12 months. The results of this study were as follows. The body weight of rats increased rapidly for the first two months, then increased slowly until 7 to 8 months. After 10 months of dietary regimen their weight decreased. The weight of liver, kidney and epidydimal fat pad increased along with the body weight and then decreased in the 12 months. Plasma total lipid increased with age and it decreased significantly when P/S ratio of dietary fatty acid was high. In creased with age and it decreased significantly when P/S ratio of dietary fatty acid was high. In creasing n-3 fatty acid intake in each P/S ratio resulted in lower plasma total lipid although was not statistically significant. The amount of plasma total cholesterol increased at 6 months, but decreased at 12 months. In case of 1, 12 months, increasing P/S ratio significantly plasma total cholesterol and LDL-cholesterol were decreased and hepatic cholesterol was increased, VLDL-HDL-cholesterol did not changed. The n-6/n-3 ratio did not affect any of theses. The amount of plasma triglyceride and VLDL-triglyceride increased at 6 month then decreased. When the rats consumed higher amount of n-3 fatty acid in each P/S ratio, their plasma triglyceride and hepatic triglyceride increased at 1, 12months.

  • The effects of dietary cellulose to utilize the intake nutrients in the albino rats were investigated by the two consecutive experiments in this study. The author wanted to observe the effects of the age of animals and different level of dietary cellulose, high and low in the first experiment and the effects of dietary cellulose from three different sources, rice bran, vegetable and pure-cellulose at high level in the second experiment. The results were summarized as follows. (1) The obvious effect of dietary cellulose caused to increase feces weight. Consequently apparent digestibilities of intake nutrients in the diet decreased. It was noteworthy that fecal glucose excretion was significantly affected by cellulose intake. Since three nutrients, protein, fat and carbohydrate serve as energy sources in the body, this result can be interpreted to be lowered in the efficiency of energy utilization out of intake from the diet. (2) The nutrients loss due to dietary cellulose resulted in reduction of skeletal muscle and bone weights which lead eventually to affect in reduction of body weight. (3) The effects of dietary cellulose showed differently according to the amount of cellulose ingested, dietary nutrients composition, the kinds of dietary cellulose supplemented and the age of experimental animals. When the intake diet was proportionately imbalanced in three different energy sources, carbohydrate, fat and protein, the cellulose intake was adversely affected in the utilization of the nutrients in the body. Cellulose from rice bran decreased body weight gain, cellulose from vegetable maintained medium level in body weight and pure-cellulose increased body weight. Growing rats showed more sensitive effects of cellulose tolerance in the body than grown rats in every concerns of this experiment.

Trends in Food and Nutrient Intake of High School Students based on the Korea National Health and Nutrition Examination Survey 2007~2015 (우리나라 고등학생의 식품 및 영양소 섭취 변화 추이 - 2007~2015 국민건강영양조사를 이용하여 -)

  • This study assessed yearly trends of food and nutrient intake among high school students aged 16 to 18 years (n=2,377) using the 2007~2015 Korea National Health and Nutrition Examination Survey (KNHANES). Yearly trends of food or nutrient intake were analyzed via logistic regression analysis. The results showed that consumption of sugars & sweets, and beverages & alcohols was increased rapidly during this period (p<0.0001). Intake of meat & meat products, and fish & shellfish also was increased (p=0.0008). Intake of grains and grain products was increased until year 2011 but declined after 2012 (p=0.0025). Consumption of vegetables, and milk & milk products was decreased (p=0.0395). Intake of protein, fat, thiamin, riboflavin, niacin, and iron was increased (p=0.0445). Carbohydrate energy ratio was decreased, whereas fat energy ratio was increased (p=0.0235). Most nutrient intakes satisfied the dietary reference intakes for Koreans except dietary fiber (19.6~26.2%), calcium (46.9~55.2%) and sodium (more than 221.4%) during this period. There was a significant positive correlation between most food group intakes and most nutrient intakes (p=0.0468). Therefore, it is crucial to increase dietary fiber and calcium intake and decrease consumption of sugars, fats and sodium through diverse eating of food groups to ensure balanced nutrition of subjects.

Effects of Genistein Supplementation on Fatty Liver and Lipid Metabolism in Rats Fed High Fat Diet (고지방식이를 섭취하는 흰 쥐에서 제니스테인 보충이 지방간 및 지질대사에 미치는 영향)

  • This study was performed to investigate the effects of genistein, a kind of soy isoflavones, on fatty liver and lipid metabolism in rats fed high fat diet. Twenty four male Sprague-Dawley rats were divided into four groups by dietary fat and genistein contents then raised for six weeks. The rats(n=6/group) were fed normal fat diet(NOR), high fat diet (HF), high fat with 0.1% genistein(HF+0.1%G) or high fat with 0.2% genistein(HF+0.2%G). Hepatic total lipid, triglyceride, total cholesterol and Serum GPT, as a marker for fatty liver, were significantly increased by high fat diet. Also, serum total lipid, triglyceride, total cholesterol, glucose and insulin concentration, hepatic lipogenic enzyme (fatty acid synthase and malic enzyme) activities were significantly increased by high fat diet. However, hepatic total lipid, triglyceride, total cholesterol and Serum GPT were significantly decreased by genistein intake. Also, genistein supplementation decreased serum total lipid, triglyceride, glucose and insulin concentration, hepatic lipogenic enzyme (fatty acid synthase and malic enzyme) activities. There were no differences by genistein level except for serum insulin. These results suggest that fatty liver induced by high fat diet was caused by increased serum lipid profiles and hepatic lipogenesis, whereas, genistein may be useful in inhibiting of fatty liver by reducing serum lipid profiles and hepatic lipogenesis.
