한국인 상용 식품 중 수은 함량 데이터베이스 구축 (Development of a mercury database for food commonly consumed by Koreans)

  • 본 연구는 문헌검색을 통해 우리나라 상용 식품의 수은 함량 데이터베이스를 구축하기 위해 수행하였고, 식품 섭취량의 95%를 포함하는 완성도가 높은 데이터베이스를 구축하였다. 2007~2009년 국민건강영양조사 식품 섭취량 조사 결과를 기반으로 대상 식품을 선정하였고, 국내에서 식품 중 수은 함량을 분석한 문헌을 고찰하여 수은 함량값을 수집하였다. 미국 농무성의 데이터 질적 평가시스템을 적용하여 수집된 문헌의 함량자료를 선택했고, 함량값이 없는 식품은 식재료의 수분함량 차이를 고려하여 대체값을 적용하였다. 구축된 데이터베이스는 총 702개의 식품 중 356개의 식품에 대한 수은 함량값을 포함하였고 (50.7%), 대상자들이 섭취한 식품 섭취량으로는 95.1%에 달하는 식품을 포함하는 것으로, 질적으로 양적으로 완성도가 높은 것으로 평가되었다. 본 연구의 결과는 향후 식품을 통한 수은 노출량 추정 및 위해성 평가에 활용될 수 있으며, 본 연구의 데이터베이스 구축 방법론은 식품을 통해 노출되는 다른 유해물질의 함량데이터베이스를구축하는 데에 적용할 수 있을 것이다.

Urinary Arsenic Concentrations and their Associated Factors in Korean Adults

  • Arsenic (As) is a well-known human carcinogen and its dietary exposure has been found to be the major route of entry into general population. This study was performed to assess the body levels of As and their associated factors in Korean adults by analyzing total As in urine. Urine and blood samples were collected from 580 adults aged 20 years and older, who had not been exposed to As occupationally. Demographic information was collected with the help of a standard questionnaire, including age, smoking, alcohol intake, job profiles, and diet consumed in the last 24 hrs of the study. Total As, sum of As(III), As(V), monomethylarsonic acid (MMA), dimethylarsinic acid (DMA), in urine was determined using atomic absorption spectrometer involving hydride generation method. The geometric mean concentration of total As in urine was $7.10{\mu}g/L$. Urine As was significantly higher in men ($7.63{\mu}g/L$) than in women ($6.75{\mu}g/L$). Age, smoking, alcohol consumption, and job profiles of study subjects did not significantly affect the concentration of As in urine. No significant relationship was observed between body mass index (BMI), Fe, and total cholesterol in serum and urinary As. Urine As level was positively correlated with seaweeds, fishes & shellfishes, and grain intake. A negative correlation between urinary As level and HDL-cholesterol in serum and meat intake was observed. Overall, these results suggest that urinary As concentration could be affected by seafood consumption. Therefore, people who frequently consume seafood and grain need to be monitored for chronic dietary As exposure.

  • Recent years have seen steadily increasing concerns about human exposure to heavy metals through seafood consumption. In Korea, the diverse dietary patterns of different bivalve shellfish necessitate tissue-specific evaluation of heavy metal contents for risk assessment. We analyzed the heavy metal contents of various tissues in scallops (Patinopecten yessoensis) and pen shells (Atrina pectinate). In both species, we found higher levels of cadmium (Cd) in the mantle than in the adductive muscle, while there was no significant difference between the two tissue types in total mercury (THg) and lead (Pb) (P<0.05). Levels of THg and Pb were acceptable by Korean standards. However, the level of Cd in pen shell mantle tissue exceeded the standard by 40%. The rate of heavy metal exposure from consumption of these species was shown to be 0.003-0.010% of the provisional tolerable weekly intake of THg and 0.312-0.600% of the provisional tolerable monthly intake (PTWI) of Cd as recommended by the Joint FAO/WHO Expert Committee on Food Additives. These findings on the tissue-specific heavy metal contents of these species will be useful in assessing the health risks of Korean dietary habits.

  • This study was performed to investigate the effects of peking-duck extracts added with medicinal herbs(DJ) on the intoxication of heavy metals (Hg, Pb, Cd, As) in rats. Sprague-Dawley rats weighing 150$\pm$15g, were randomly assigned to groups: basal diet only in normal control group(NCG): basal diet and heavy metals without DJ injection in heavy metal control group(HMC); basal diet, heavy metals and DJ(3mg/ml) injection in heavy metal low duck-juice group(HMLD), basal diet, heavy metals and DJ(30mg/ml) injection in heavy metal middle duck-juice group(HMMD): basal diet, heavy metal and DJ(300mg/ml) injection in heavy metal high duck-juice group(HMHD). Hg and As was injected by 50ppm and Cd and Pb by 25ppm for 17days. Also DJ oral feeding was conducted for 28days. The result of this study were as follows: Food intake and body weight gain in heavy metal administered groups were lower than those of NCG. Liver, kidney and testis weights were not significantly different among 5 groups. GOT, GPT and BUN activities were significantly reduced in DJ treated groups as compared to HMC. DJ showed the suppressing effect on the accumulation of Hg, Pb and Cd in serum, liver and kidney. Fecal Hg and Cd excretions increased with DJ feeding. The results suggested that DJ may have some protective effects on Hg, Cd and Pb intoxication by reducing the accumulation in tissues and increasing excretion. This study also showed the effective way of using duck-extract and its application to the oriental medicine.

Protective Effect of Dietary Buchu (Chinese chives) Against Oxidative Damage from Aging and Ultraviolet Irradiation in ICR Mice Skin

  • Protective effect of skin by antioxidative dietary buchu (Chinese chives, Allium tuberosum Router), was evaluated in ICR mice fed diets containing 2% or 5% buchu for 12 months. Lipid peroxidation and protein oxidation in skin, with or without ultraviolet B (UVB) irradiation, activities of antioxidative enzymes, total glutathione concentrations, and non-soluble collagen contents were measured. Dietary buchu decreased significantly in TBARS and protein carbonyl levels in skin compared to the control group, and were lower in those fed 5% than 2% buchu diet group. ICR mice exhibited an age-dependent decrease in antioxidative enzyme activities and total glutathione concentrations on the control diet, but in the groups fed buchu diet the enzyme activities and glu-tathione concentrations remained at youthful levels for most of the study. SOD, glutathione peroxidase, and catalase activities as well as total glutathione concentrations increased with time in the skins of the mice fed buchu diets. Lipid peroxidation and protein oxidation provoked by UVB irradiation on ICR mice skin homogenates were also significantly inhibited by dietary buchu. The buchu diets also decreased the formation of non-soluble collagen in mice skin, compared to the control group. These results suggest that antioxidative components and sulfur-compounds in buchu may confer protective effect against oxidative stress resulting from aging and exposure to ultraviolet irradiation.

Effect of Dietary Folate on Hyperhomocysteinemia and Cellular Toxicity Induced Alcohol Administration in Rat Liver

  • The critical role of folate in the remethylation pathway for methionine synthesis from homocysteine has been well documented. Hyperhomocysteinemia resulting from inadequate folate nutrition has been implicated in increased incidence of macrovascular diseases, colorectal cancer, neural tube defects, etc. Chronic exposure to ethanol impairs folate nutrition and one-carbon metabolism in the liver, which often results in fatty liver due to a defective remetylation process. This study was carried out to investigate the chronic effects of moderate levels of alcohol and dietary folate on plasma homocysteine levels, and on histopathology and biochemical functions of the liver. Rats were raised on experimental diets with three levels of folate (0, 2, 8 mg/kg diet), and 50% ethanol (1.8 ml/kg body weight) was administered intragastically by intubation tubes three times a week for 10 weeks. Plasma homocysteine concentrations were found to be significantly influenced by dietary folate intake and alcohol administration. Among all treatment groups, plasma homocysteine levels were the highest in the animals receiving a combined treatment of folate deficient diet and alcohol administration. Plasma homocysteine concentrations were negatively correlated with folate concentration in the plasma (p<0.01) and liver (p<0.05). Among alcohol treated rats, increase in plasma homocysteine values due to macrovascular and microvascular fatty changes and spotted necrosis were observed more frequently in folate-deficient animals diet than those on folate-adequate and folate supplemented diets in alcohol-treated rats. These results indicate that folate supplementation above the recommended level might be beneficial in the prevention of alcohol-related hyperhomocysteinemia and abnormal histologic changes in the liver.

Excessive dietary lead reduces growth performance and increases lead accumulation in pigs

  • Objective: The objective of this study was to investigate the influence of dietary lead (Pb) supplementation and feeding period on growth performance, organ weight, and Pb accumulation in pigs. Methods: In a 56-day feeding experiment, a total of 48 barrows with initial body weight 10.4±0.6 kg were allotted to 2 dietary treatments (0 and 200 mg/kg of supplemental Pb) in a completely randomized design with 6 replicates. Body weight and feed intake were recorded to calculate growth performance. At the end of each 14 day-period (on days 14, 28, 42, and 56), an animal was randomly selected from each pen and slaughtered to collect blood samples, hair samples, left 5th rib, heart, liver, kidneys, lungs, and longissimus dorsi muscle samples. Results: Average daily gain and average daily feed intake were reduced (p<0.05) by supplemental Pb during the day 42 to 56. Relative kidney weight to body weight was linearly increased with increasing feeding period in pigs fed the Pb-supplemented diet, but not in pigs fed the control diet (p<0.05). The Pb concentrations in hair, left 5th rib, kidneys, and lungs were linearly increased with longer feeding period in pigs fed the Pb-supplemented diet, but not in pigs fed the control diet (p<0.01). Conclusion: Dietary Pb supplementation caused growth retardation and Pb accumulation in most organs, particularly in hair, bone, and kidneys in a time-dependent manner.

  • Objectives: The aim of this study is to compare exposure levels of PAHs and benzene metabolites in the urine of nationally representative adults in Korea and identify exposure factors in relation to questionnaire results. Methods: The study analyzed PAHs and benzene metabolites in the urine of adults aged 19 and older recruited in Cycles 1-3 of the Korean National Environmental Health Survey (KoNEHS). Participants were administered questionnaires on demographic characteristics, lifestyle, and dietary habits to identify the association with exposure levels to environmental chemicals. Results: 1-hydroxypyrene and t,t-muconic acid levels in this study were higher than those in large-scale biomonitoring conducted in other countries (the US and Canada), while 2-hydroxynaphthalene levels were lower. Most environmental chemicals exhibited significant differences depending on gender and age, and exposure levels tended to increase with grilled food intake. In addition, variables such as smoking, distance between home and a road, travelling on public transportation, and ventilation time had a significant effect on exposure to environmental chemicals. Conclusions: The importance of this study lies in utilizing the representative results of KoNEHS, large scale biomonitoring conducted in Korea over roughly ten years. Variables including smoking and roasted food intake were found to affect exposure to PAHs and benzene, which can be used to establish baseline exposure levels of environmental chemicals in Korea.

Effects of 4-tert-octylphenol on glutathione-related antioxidant status in olive flounder Paralichthys olivaceus

  • Effects on glutathione-related antioxidant parameters were examined after a chronic exposure of olive flounder, Paralichthys olivaceus to dietary 4-tert-octylphenol (4-tert-OP). Fish were fed diets containing 4-tert-OP at 0, 1, 5 and 10 mg/kg diet for 6 weeks. Antioxidant parameters examined were reduced glutathione (GSH) contents and enzyme activities of glutathione reductase (GR), glutathione S-transferase (GST) and glutathione peroxidase (GPx) in tissue homogenates of the liver, kidney and gill. It was observed that all parameters examined increased although there were some differences in dose responses and temporal patterns in the increase. GSH contents increased after exposure to 4-tert-OP in the three organs examined. However, the GSH increase was evident only after 4 weeks in the liver whereas it was elevated after 2 weeks in the kidney and gill. GR activity exhibited a significant increase in response to 4-tert-OP at 1 mg/kg in all three organs, however, its activity returned to control levels when exposed to 5 and 10 mg/kg. Hepatic GST activity showed an earlier increase at week 2 in contrast to the kidney and gill where they increased after 4 weeks of 4-tert-OP exposure. Temporal patterns in GPx activity changes to 4-tert-OP exposure were dissimilar among the organs: hepatic activity increased from week 2 through week 6; renal activity increased transiently at week 2; gill levels were higher through weeks 4 - 6. The results suggest that elevation of several GSH-related antioxidant parameters can be considered as evaluation criteria for 4-tert-OP-induced oxidative stress in a fish.
