  • Accumulated epidemiological and animal studies have suggested that prolonged exposure to ambient particulate matter (PM) is associated with an increased risk of cardiovascular disease and pulmonary dysfunction. While diesel exhaust particles (DEP) contain large variety of compounds, polycyclic aromatic hydrocarbons (PAHs) are a dominant component contaminated in DEP. This article reviews effects of two PAH quinones, 9,10-phenanthraquinone (9,10-PQ) and l,2-naphthoquinone (l,2-NQ), on vascular and respiratory systems.

  • In this study, we mainly focused on the PM (Particulate Matter) emission characteristics of a diesel engine. To analyze particle behavior in the tail-pipe, particle emission was measured on the engine-out (downstream of turbocharger), each upstream and downstream both of DOC (Diesel Oxidation Catalyst) and DPF (Diesel Particulate Filter). Moreover, particle emission contours on each sampling point were constructed. The reduction efficiency of particle number concentration and mass through the DOC and DPF was studied. Parameters such as EGR (Exhaust Gas Recirculation) and the main injection timing were varied in part load conditions and evaluated using the engine-out emissions. The DMS500 (Differential Mobility Spectrometer) was used as a particle measurement instrument that can measure particle concentrations from 5 nm to 1000 nm. Nano-particles of sizes less than 30 nm were reduced by oxidation or coagulated with solid particles in the tail-pipe and DOC. The DPF has a very high filtration efficiency over all operating conditions except during natural regeneration of DPF.

Chemopreventive Effect of Vegetable or Fruit Extract Against Total Diesel Exhaust Particle Extract in NIH/3T3 Cells Using Alkaline Single Cell Gel Electrophoresis (총 디젤분진의 DNA 손상작용과 야채 및 과일추출물의 보호효과)

  • In urban areas, diesel exhaust particles (DEP) are probably a major component of particulate matters, especially in Korea where drive many diesel vehicles. The aim of this study was to investigate genotoxic effects of DEP using single ceil gel electrophoresis. In order to evaluate the mechanisms of DEP genotoxicity, the rat microsome mediated and DNA repair enzyme treated comet assays together with conventional comet assay were performed. Total diesel particles (DEPT) was collected without site fractionation from diesel engine bus and dichloromethane extract was obtained. The organic extract of DEPT revealed DNA damage itself in NIH/3T3 cells. The level of DNA breaks plus oxidative DNA lesions and microsome mediated DNA damage was assessed by modified single cell gel eletrophoresis. DEPT was able to induce oxidative DNA damage as well as microsome mediated DNA damage. Vitamin C as an model antioxidant reduced DNA damage in endonuclase III treated comet assay. One of flavonoid, galangin as a CYP1A1 inhibitor. reduced DNA damage in the presence of S-9 mixture. $DEP_T$ is the sources of oxidative stress, but antioxidants can significantly reduce oxidative DNA dmage. And $DEP_T$ may contain indirect mutagens which can be inhibited by CYP1A1 inhibitors. The ethanol extracts of the mixed vegetables (BV) or the mixed fruits (BF) were evaluated for their in vitro antigenotoxic effects. BV and BF showed potent Inhibitory effects against DEPT induced DNA damage with oxidative DNA lesions and in the prescence of S-9 mixture. These results indicate that BV and BF could prevent cellular DNA damage by inhibiting oxidative stress and suppressing cytochrome P4501A1 in cell culture.

Performance and Emission Characteristics of a Diesel Engine Operated with Wood Pyrolysis Oil (목질 열분해유를 사용하는 디젤엔진의 성능 및 배기특성에 관한 연구)

  • The vast stores of biomass available in the worldwide have the potential to displace significant amounts of fuels that are currently derived from petroleum sources. Fast pyrolysis of biomass is one of possible paths by which we can convert biomass to higher value products. The wood pyrolysis oil (WPO), also known as the bio crude oil (BCO), have been regarded as an alternative fuel for petroleum fuels to be used in diesel engine. However, the use of BCO in a diesel engine requires modifications due to low energy density, high water contents, low acidity, and high viscosity of the BCO. One of the easiest way to adopt BCO to diesel engine without modifications is emulsification of BCO with diesel and bio diesel. In this study, a diesel engine operated with diesel, bio diesel (BD), BCO/diesel, BCO/bio diesel emulsions was experimentally investigated. Performance and gaseous & particle emission characteristics of a diesel engine fuelled by BCO emulsions were examined. Results showed that stable engine operation was possible with emulsions and engine output power was comparable to diesel and bio diesel operation. However, in case of BCO/diesel emulsion operation, THC & CO emissions were increased due to the increased ignition delay and poor spray atomization and NOx & Soot were decreased due to the water and oxygen in the fuel. Long term validation of adopting BCO in diesel engine is still needed because the oil is acid, with consequent problems of corrosion and clogging especially in the injection system.

  • Diesel exhaust particle (<2.5 ${\mu}{\textrm}{m}$, DEP$_{2.5}$) is known to be probarbly carcinogenic (IARC group 2A). DEP$_{2.5}$ contains organic compounds such as polycyclicaromatic hydrocarbon (PAH), heterocyclic compounds, phenols, and nitroarenes. Reactive oxygen species (ROS) are generated by DEP$_{2.5}$ without any biological activation system. Therefore, an alternative mechanism by which DEP$_{2.5}$ could be carcinogenic is known by the generation of oxidative DNA damage. The aim of this study was to investigate genotoxic effects of DEP$_{2.5}$ using single cell gel electrophoresis. Saponin attenuates diesel exhaust particle (DEP)-induced MUC5AC expression and pro-inflammatory cytokine upregulation via TLR4/TRIF/NF-𝛋B signaling pathway in airway epithelium and ovalbumin (OVA)-sensitized mice

  • Background: Diesel exhaust particle (DEP) is a harmful kind of particulate matter known to exacerbate pre-existing respiratory diseases. Although their adverse effects on airway pathologies have been widely studied, the mechanistic analysis of signaling pathways and potential targets in reducing DEP-induced mucin secretion and pro-inflammatory cytokine production remain elusive. We, for the first time, investigated the effects of Korean Red Ginseng (KRG) extracts on mucin overproduction and airway inflammation induced by DEP. Methods: The effects of KRG and saponin on DEP-induced expression of MUC5AC and interleukin (IL)-6/8 were examined by real-time polymerase chain reaction (PCR) and enzyme-linked immunosorbent assay (ELISA) in human airway epithelial NCI-H292 cells. We conducted Western blotting analysis to analyze the associated signaling pathways. To evaluate the effects of saponin treatment on DEP-induced MUC5AC expression and inflammatory cell infiltrations in ovalbumin (OVA)-sensitized mice, immunohistochemical (IHC) staining and real-time PCR were implemented. Results: The KRG extracts markedly attenuated DEP-induced MUC5AC expression in vitro by inhibiting the TLR4/TRIF/NF-𝛋B pathway. Furthermore, KRG and saponin inhibited DEP-induced pro-inflammatory cytokine IL-6/8 production. The in vivo study revealed that saponin blocked DEP-induced inflammation, mucin production and MUC5AC expression. Conclusion: Our study revealed that KRG extracts have inhibitory effects on DEP-induced expression of MUC5AC and the production of pro-inflammatory cytokines. This finding provides novel insights into the mechanism by which saponin alleviates diesel-susceptible airway inflammation, elucidating its potential as a phytotherapeutic agent for inflammatory pathologies of airway.

  • Diesel particulate trap is a core technology for the reduction of PM from diesel vehicles This study presents the features and the characteristics of DPF system when using pellet type filters. In comparison with wall-flow filter, the pellet filter has the advantages of cracking free during regeneration and shape flexibility. Experiments are conducted in a test bench simulated as diesel engine exhaust condition. Pressure drop and particle loading rate was compared by using two pellet filters having the porosity of 70% and 0%. Also its regeneration was tested.

  • This paper presents the atomization characteristics of single hole injector in the direct injection type diesel engine. The spray characteristics of fuel injector such as the droplet size and velocity were measured by phase Doppler particle analyzer. In this paper, the atomization characteristics of fuel spray are investigated for the experimental analysis of the measuring data by the results of mean diameter and mean velocity of droplet. The effect of fuel injection pressure on the droplet size shows that the higher injection pressure results in the decrease of mean droplet diameter in the fuel spray. The minimum size of fuel spray droplet appears on the location of 40mm axial distance from nozzle exit of diesel injector. Based on the experimental results, the correlation between the droplet diameter and mean velocity of the diesel spray due to the change of axial and radial distance from the nozzle tip were investigated.
