Diallel Analysis for Cyanidin-3-glucoside Content in Pigmented Rice (이면교배에 의한 흑자색미 안토시아닌 함량의 유전분석)

  • A complete diallel cross was performed to determine the inheritance of 1,000 grain weight of brown rice and cyanidin-3-glucoside (C3G) content and combining ability of five pigmented rice that showed different level of C3G content. Parents and F1 of 20 crosses were evaluated for 1,000 grain weight of brown rice and C3G content which extracted from pigmented rice grain with 0.1% trifluoroacteic acid (TFA) in 95% EtOH. For 1,000 grain weight of brown rice and C3G content, both additive and dominant gene effects were detected and the dominance was partial and there were more dominant alleles than recessive ones. Genetic parameters pointed out that there are significant additive and dominant effects and the additive effect is bigger than the dominant one for both grain weight and C3G content. The frequency of negative alleles were higher than positive alleles. Narrow sense heritability estimates were 0.66 for grain weight of 1,000 brown rice and 0.70 for C3G contents. General combining ability (GCA) effects and specific combining ability (SCA) effects were highly significant and GCA effects were drastically higher than SCA ones. It indicates that the inheritance of the C3G contents is controlled mainly by additive genetic factor.

Genetic Analysis of Boiling-off Ratio of Amount of Sericin and Fibroin of Cocoon Layer and from Diallel Crosses of Silkworm, Bombyx mori (누에 이면교잡에 의한 견층 연감율 Sericin량 Fibroin량에 대한 유전분석)

  • The genetic analysis was caried out on the boiling-off ratio of cocoon layer, and the amounts of sericin and fibroin in the F1 generation from the diallel crosses of silkworm. The result obtained were as follows : 1. Boiling-off ratio and the amounts of sericin adn fibroin in male and female were over dominqnce and partial dominance, respectively. 2. The additive effect of gene was higher then the dominance effect in the amounts of sericin and fibroin in male and female, and in boiling off ration in female. 3. The direction of dominance in the amounts of sericin and fibroin in male and female was positive and that in boiling off ratio negative. 4. The effect of three characters on the combining ability was significant and their general combining ability was higher than the specific combining ability.

Genetic Analysis of Apoplastic Proteins in Barley Crosses

  • Antifreeze proteins (AFPs) accumulate in the leaves of barley during cold acclimation, where they may inhibit ice recrystallization and produce freezing resistance of the plant. Four parental diallel crosses of the barley varieties were used to determine the heritability of AFPs and the relationship between the accumulation level of AFPs and freezing resistance. The concentration of apoplastic proteins in the cold-acclimated leaves was increased in the mean by four-fold over as compared with that of nonacclimated. The diallel cross analyses revealed that the gene of Sacheon 6 was dominant and those of Reno and Dongbori 1 were recessive. The AFPs had high narrow-sense heritabilities. The general combining ability effects of Reno and Dongbori 1 were much higher than the other parents. The bands of 32-kD for GLP, 35-& 28-kD for CLP and 25-, 22- & 16-kD for TLP were observed in the apoplastic extracts from cold-acclimated plants, but there were no clear differences between the parents and Fl hybrids. The concentrations of AFPs were significantly correlated with the degree of freezing resistance, indicating that the concentration of AFPs in the plant is the very important factor for freezing resistance.

Relationship between Differential Gene Expression in Ovary and Heterosis of Egg Number Traits in a Chicken Diallel Cross

  • In order to understand the molecular basis of chicken heterosis in reproduction traits, mRNA differential display (DDRT-PCR) methods were used to analyze the differential gene expression of ovary tissue between hybrids and their parental lines in a 4${\times}$4 diallel cross, involving 4 chicken breeds, which were White Plymouth Rock (E), CAU Brown (D), Silkies (C) and White Leghorn (A). Total of 331 differential displayed cDNA bands from 1,161 were displayed in the 4${\times}$4 diallel cross combinations with 30 pairs of primers, which shows the differences of gene expression between hybrids and their parental lines were very obvious in quantity and quality. Seven types of differential expression patterns were found: Co-dominance expressed pattern (T1), under-expression of parental fragments in hybrids (T2), over-expression of parental fragments in hybrids (T3), hybrid-absence expressed pattern (T4), single parentspecific expressed pattern (T5), dominant expression fragments of single parent in hybrids (T6), hybrid-specific expressed pattern (T7). Correlation analysis indicated that there were significant correlations between the pattern of T3 and the heterosis percentage of egg number of 32-week and 42-week old chickens(p<0.01), while there were negative significant correlations between the pattern of T7 and the heterosis percentage of egg number of 32-week and 42 week-old birds (p<0.01).

The Genetic Analysis of Quantitative Characters in the Silkworm by Diallel Cross of Four Inbred Lines Differing in Silk Yield (견생산력이 다른 계통간의 Diallel Cross에 의한 누에 양적 형질의 유전분석)

  • Extensive studies were undertaken to analyze the genetic basis of economically important quantitative characters in the silkworm by diallel crosses of four inbred lines differing widely in silk yielding ability. Some differences between the reciprocal corsses were detected in cocoon weight, cocoon shell weight, cocoon filament length and cocoon filament weight in case the parental lines were greatly differenent each other in silk yielding ability. The general combining ability (GCA) varied with the inbred lines and M242, a Chieese sexlimited larval marking variety showed high GCA value in the economic characters, such as cocoon yield, cocoon weight, cocoon filament length and weight, and raw silk percentage. The highest heterosis effect, about 13% to 14% was seen in cocoon and raw silk yield and it was low in cocoon reelability and raw silk percentage with less than 1%. It is advisable to improve highly heritable quantitative characters such as larval duration, cocoon shell weight, cocoon filament length and raw silk percentage by means of selection, and to select single crosses with high heterosis effect for cocoon weight and cocoon yield which show overdominance. Genetic correlation should be considered when more than two characters are targets for improvement and selecting high cocoon shell weight is effective to breed high silk yielding varieties. It is difficult to improve cocoon reelability because of low heritability (0.11) and its negative correlation with cocoon-silk quality.

8-Parents Diallel Cross Analysis on Heading Date of Barley (이면교잡에 의한 대맥품종의 출수기 유전에 관한 연구)

  • In order to obtain the basic information on the response of different day-length to heading date and on the inheritance of heading date of barley varieties. the 8 parents F$_1$ diallel crosses were tested with 4 different day-length. Varietal differences in photo-sensitivity were observed and Haganemugi was the most photo-insensitive variety. The inheritance of earliness were appeared to be partial dominance or partial ressesive and the inheritance of photo-insensitivity over photo-sensitivity were controlled by the single ressesive factor.

Genetic Analysis for Weight of Matured Silkworm and Number of Eggs Laid in Hybrid Population of the Silkwom, Bombyx mori (누에의 숙잠체중과 산란성에 대한 유전분석)

  • The genetics analysis for weight of matured silkworm(WMS) and number of eggs produced per moth(NEM) was studied by the seven parents diallel. Mean squares of additive effect, dominant effect, maternal effect and reciprocal effect were significant for two characters observed. The component of genetic variance analysis for WMS and NEM showed that dominant effect was higher than additive effect. Narrow sense heritability(h2ns) estimates were 0.773 and 0.228, in the WMS and NEM. The estimate of broad sense heritability(h2bs) value was higher than that of h2ns because of the low importance of dominance effect. Incomplete dominance was shown by Vr-Wr graphic analysis in the weight of matured silkworm and overdominance in the number of eggs produced per moth. In general combining ability effect, Jam 107 and Jam 124 was showed positively high for WMS and Jam 107 and S1 was expressed positively high for NEM. In specific combining ability effect, hybrids in S1XC51, S1XJam124 and Jam 107XJam 108 were exhibited positively high for WMS and Jam 107XN63, S1XC51, N74XJam 108 and Jam 107XJam 108 were found positively high for NEM.

Inheritance of Grain Filling Duration in Corn (옥수수 이면교잡에 의한 등숙기간의 유전 분석)

  • This study was conducted to clarify the genetic effect on the duration of grain filling with using the eight corn inbreds. In diallel cross analysis, the grain filling during the lag period showed partial dominance with great additive effects. Inbreds FR14A and A508 showed greater recessive gene effects for lag period, while FR25 showed greater effects of dominant genes. The genetic analysis for the effective filling period(EFP) showed over dominance without additive gene effects. FR25 of 8 inbreds showed greatest effects of dominant genes for EFP, while YUBC208 showed greater recessive gene effects for EFP than other inbreds. The genetic analysis for total grain filling period(TGFP) seemed to be due to partial dominance with greater additive effects. Early inbred line, YUBC208 especially showed greater recessive gene effects for TGFP than others. No. of effective genes related to EFP and TGFP were estimated by at least 5.
