Dolichos biflorus Linn attenuate progression of renal damage in alloxan-induced diabetic rats

  • Dolichos biflorus Linn. (Fabaceae), commonly known as Horse gram is a medicinal plant, used in folk medicine for treating kidney stones and diabetes mellitus. The purpose of the present study was to investigate the effects of daily oral feeding of various doses of methanolic extract of Dolichos biflorus seeds (DB) for 42 days on blood glucose concentrations and kidney functions in Alloxan-diabetic rats. Plasma glucose levels, body weight, serum creatinine, and urinary albumin levels were monitored on $15^{th}$, $29^{th}$, $43^{rd}$ day. Renal hypertrophy was assessed as the ratio between the kidney weight and body weight of the rats. Plasma glucose concentrations in Alloxan-diabetic rats were significantly reduced by the administration of DB (350 mg/kg) and DB (700 mg/kg) on day 15 and onwards (P < 0.01). After 15 days of Alloxan administration urinary albumin levels (UAE) were over 5 fold higher in diabetic controls as compared to normal controls. Treatment with DB significantly prevented the rise in UAE levels from day 15 to 43 in comparison to diabetic controls (P < 0.01). Renal hypertrophy was significantly higher in diabetic controls as compared to non-diabetic controls. Treatments with DB (350 mg/kg) and DB (700 mg/kg) significantly prevented renal hypertrophy (P < 0.01) as compared to diabetic controls. DB (175 mg/kg) failed to modify renal hypertrophy. Thus the present study indicates that methanolic extract of Dolichos biflorus may be useful in management of hyperglycemia and kidney functions in Alloxan-diabetic rats.

방사선조사가 당뇨 백서의 발치와 치유에 미치는 영향 (Effect of irradiation on the healing of extraction sockets in diabetic rats)

  • Purpose: To observe the histologic pattern of healing in molar tooth extraction sockets of streptozotocin-induced diabetic rats following irradiation. Materials and Methods: Mature Sprague-Dawley rats were divided into three groups: control, diabetic, and diabetic-irradiated groups. Diabetes mellitus was induced by injecting streptozotocin. Control rats were injected with a citrate buffer only. After 5 days, the right maxillary first molar was extracted under general anesthesia from each of the rats. After the extraction, rats in the diabetic-irradiated group were irradiated with a single absorbed dose of 10 Gy to the head and neck region. The rats were killed at 1, 3, 7, 14, 21, and 28 days after treatment. Tissue sections were stained with hematoxylin-eosin and Masson's trichrome. Results: In the diabetic and diabetic-irradiated groups, the early healing process of the socket extraction was similar to the control group, but bone formation was delayed at 7 days after the treatment. In the diabetic-irradiated group, alveolar bone surrounding the extraction socket showed signs of necrosis at 3 days after treatment, and hemorrhage was observed in connective tissue within the extraction socket at 14 days after treatment. Conclusion: This experiment revealed that the healing process of the extraction socket was severely delayed and retarded by irradiation in the diabetic state.

선학차(仙鶴草) 추출물의 투여(投與)가 Streptozotocin으로 유발된 당뇨쥐에 미치는 영향 (Effects of Agrimoniae Herba Ledebour on steptozotocin-induced Diabetic Mellitus in Rats.)

  • Objective : The present study was carried out to investigate the preventive effect of Agrimonia pilosa Ledebour on Streptozotocin(STZ)-induced Diabetes mellitus. Method : Agrimonia pilosa Ledebour was given to rats with oral administration. The experimental animals were divided into 3groups : normal group of rats, control group of STZ-induced diabetic rats, sample group with Agrimonia pilosa Ledebour. The effect of Agrimonia pilosa Ledebour on STZ-induced diabetes was observed by measuring the survival rate of rats, weight of rats, FER, blood glucose, each organ weight of rat, total cholesterol, HDL, GOT, GPT & creatinine. Result : STZ caused hyperglycemia and hypoinsulinemia by a selectively destroying pancreatic ${\beta}-cell$. Agrimonia pilosa Ledebour treatment don't protected them from hyperglycemia and hypoinsulinemia. Organ weight only liver weight decreased but kidney, heart & spleen shows no significant changes. Agrimonia pilosa Ledebour significantly don't recoverd the increase of several biochemical parameters such as blood glucose, total cholesterol, HDL, GOT, GPT & creatinine is vice versa. Conclusion : Agrimonia pilosa Ledebour extract group did not show significant difference compared with STZ-induced Diabetes Mellitus group.

  • Bell's palsy is an isolated facial paralysis of sudden onset caused by a neuritis of the seventh nerve within the facial canal. It occurs often in the adult man with a history of recent exposure to local cold, such as sleeping next to an open window, or in some cases it occurs after infections of the nasopharynx or masticator spaces. Especially, this neuropathy have linked with the major collagen disorders (diabetes mellitus). A segmental demyelination develops rapidly, with vascultitis in microinfarcts and ischemia to the nerve segment. The authors experienced about the bizarre neurological symptom of Bell's palsy after inferior alveolar nerve block anesthesia and TMJ dislocation in diabetic mellitus. The early and correct consultation with the multiple medical and dental departments was important to prevent the inadequate care & medicolegal problems.

제2형 당뇨모델 db/db 마우스에서 밀순 물추출물의 항당뇨 효과 (Anti-diabetic Effects of Triticum aestivum L. Water Extracts in db/db Mice as an Animal Model of Diabetes Mellitus Type II)

  • We evaluated the anti-diabetic effects of Triticum aestivum sprout water extract (TA) in diabetes mellitus type 2. For the experiments, the diabetic animal model db/db mice were divided to 3 groups: diabetic control (db/db) and two experimental groups orally treated with 25 and 100 mg/kg single dose of TA (TA-25 and TA-100, respectively). The lean mice were used as the non-diabetic normal control. All mice have free access to water and AIN-93 diet. TA was administrated to diabetic mice for 5 weeks and the diabetic clinical markers, including blood glucose level, body weight, food intake and insulin level, were measured at a time. After administration for 5 weeks, the blood glucose level was decreased 1.10 and 1.98 folds in TA-25 and TA-100 groups, respectively, compared with db/db group. The body weight and diet consumption were significantly reduced by TA treatment in dose-dependent manner. The treatments of TA-100 also significantly decreased remarkedly liver weight and slightly serum insulin levels when compared with them of the diabetic control group. However the immunohistochemical staining for insulin clearly showed high expression of insulin in the pancreatic islet cells derived from all db/db mice, even if TA was administrated. Moreover, TA-100 treatment significantly improved impaired glucose tolerance in diabetic db/db mice. The results suggest that TA has anti-hyperglycemic effect attenuating blood glucose in the animal model of type 2 diabetes and might be useful as a functional diet for human diabetic diseases.

당뇨병성 말초신경병증 환자에서 통증 정도와 당화혈색소 간의 상관성 연구 (A Study on the Correlation between Pain Intensity and HbA1c for Diabetic Peripheral Neuropathy Patients)

  • Objectives : The purpose of this study is to evaluate the correlation between HbA1c and pain intensity in diabetic peripheral neuropathy patients, and to compare the difference between two groups divided by the risk of complications. Methods : The participants were 46 men and women suffering from neuropathic pain diagnosed with diabetes mellitus who visited the Hospital of Traditional Korean Medicine, Semyung University from June, 2014 to August, 2015. Age, duration of diabetes mellitus, numeric rating scale(NRS), and Michigan neuropathy screening instrument(MNSI) were used as evaluation tools. Results : 1. In all 46 cases, there were no significant correlations between HbA1c and age, duration of diabetes mellitus, NRS, MNSIQ, or MNSIE. 2. Among 23 cases with a high risk of complications (more than 7.0 % of HbA1c, group A), there were no significant correlations between HbA1c and age, duration of diabetes mellitus, NRS, MNSIQ, or MNSIE. 3. Among 23 cases with a low risk of complications(less than 7.0 % of HbA1c, group B), there were no significant correlations between HbA1c and age, duration of diabetes mellitus, NRS, MNSIQ, or MNSIE. 4. Means of duration of diabetes mellitus, NRS, MNSIQ, and MNSIE were higher in group A than group B, but there were no significant correlations in statistics. Conclusions : This study could not find statistically significant correlations between pain intensity and HbA1c in diabetic peripheral neuropathy patients, so more studies are required in the future.

Streptozotocin유발 당뇨쥐에서 세사민과 세사몰린의 항산화효과 (Antioxidant Effects of Sesamin and Sesamolin in Streptozotocin-induced Diabetes Mellitus Rat)

  • 산화적 스트레스는 당뇨병과 관련된 맥관계병변의 전개에 아주 중요한 역할을 한다고 알려져 있다. 따라서 본 실험에서는 streptozotocin(STZ)투여에 의한 당뇨유발쥐를 사용하여 참께함유 항산화성분인 sesamin과 sesamolin의 혈당과 생체내 항산화 방어효소에 미치는 효과에 대하여 알아보고자, 당뇨유발쥐에 0.2% sesamin또는 0.2% sesamolin 함유사료를 3주간 급여하여, 혈당농도, 뇨당농도, 혈청 GOT와 GPT활성, 혈청 및 간조직의 지질과산화물,그리고 GSH함량과 GST활성에 미치는 영향을 조사하였다. STZ투여군의 혈당량은 319.4 $\pm$105.74mg/dL였는데, sesamin급여군은 146.56$\pm$23.99mg/dL, sesamolin급여군은170.74$\pm$105.94mg/dL로 유의하게 감소하였다. 당뇨병쥐의 뇨당농도가 21.16$\pm$13.72 mg/dL였는데, 세사민과 세사몰린 급여시에는 19.52 $\pm$ 8.99 mg/day와 10.93 $\pm$ 7.79 mg/day로 유의하게 감소하였다. STZ투여에 의하여 증가된 GOT와 GPT활성도는 세사민과 세사몰린의 급여에 의하여 GPT활성도만이 유의하게 감소되었다. STZ투여에 의해 증가한 간조직 중의 지질과산화물 생성은 세사민과 세사몰린 급여에 의해 유의하게 감소하였다. STZ투여에 의해 감소된 신장의 GSH함량과 GST활성은 세사민과 세사몰린에 의해 회복되었다. 이상의 결과 세사민과 세사몰린이 신장조직에서 일차적으로 산화된 산화물을 포집하고 GSH를 이용하여 체내의 독성 물질을 전이 분해시키는 작용을 하여 STZ에 의해 유도된 각종 독성을 무독화시켜 신장의 손상을 보호하여 당뇨억제 작용과 함께 항산화 효과를 나타낸 것으로 생각된다.

Overexpression of KAI1 Protein in Diabetic Skin Tissues

  • Background Patients with diabetes mellitus often have a difficult life, suffering from foot ulceration or amputation. Diabetes is characterized by chronic inflammation, and one of the features of inflammation is hypoxia. Recently, it has been reported that KAI1 is a hypoxia target gene. There is no published research on hypoxia-related KAI1 protein levels in human diabetic skin. Therefore, we have investigated the expression of KAI1 protein in diabetic skin tissue in vivo. Methods The expression of KAI1 protein was evaluated by western blotting in 6 diabetic skin tissue samples and 6 normal skin samples. Immunohistochemical staining was carried out to identify KAI1 expression. Results The western blotting revealed significantly increased expression of the KAI1 protein in diabetic skin tissues as compared to normal skin tissues. Immunohistochemical examination demonstrated that KAI1 was expressed in all diabetic skin tissues with moderate-to-strong positivity and weakly expressed in normal skin tissues. Conclusions Our data suggest that a high expression of the KAI1 protein can be observed in diabetic skin tissue. To the best of our knowledge, this is the first report suggesting that KAI1 protein expression in diabetic skin tissues may be associated with chronic inflammatory states and hypoxia.

Diabetes is an Important Risk Factor for Metastasis in Non-Muscle-Invasive Bladder Cancer

  • Background: Epidemiological evidence indicates that individuals with diabetes mellitus (DM) may have a modestly increased risk of bladder cancer. In the present study, we aimed to show any association between DM and risk of metastasis in patients with non-muscle-invasive bladder cancer (NMIBC). Materials and Methods: We retrospectively analyzed 698 patients between January 2007 and December 2014 who were diagnosed with and underwent transurethral resection of bladder tumors (TUR-BT). Comparisons of means was conducted by independent samples t test, and relations between categorical variables were investigated by non-parametric chi-square test. A p value of 0.05 was accepted as statistically significant in comparisons. Results: We analyzed 418 patients with non muscle invasive bladder cancer. 123 of whom were diabetic and 295 non-diabetic. In diabetic patients, 13 were N1 stage and 11 M1 stage. When compared with non diabetic patients that was statistically significant (p<0.001). TNM stages were more advanced in diabetic patients (p<0.001), but concurrent CIS (p=0.1) and squamous metaplasia did not significantly differ between diabetic and non-diabetic cases (p=1). Conclusions: Diabetic patients with non-muscle-invasive bladder cancer may suffer metastases earlier than expected although they are non invasive. Therefore such patients must be followed-up carefully and early cystectomy decision may be necessary. Further prospective studies with more patients are needed to confirm these findings.
