Resistance screening of white mustard(Sinapis alba) plant resources against clover cyst nematode, Heterodera trifolii (클로버씨스트선충에 대한 백겨자 식물자원의 저항성 스크리닝)

  • Clover cyst nematode (Heterodera trifolii, CCN) is one of the important plantparasitic nematodes in the Republic of Korea, causing serious damage to Kimchi-cabbage in Gangwon province since 2017. Soil fumigants are the preferred choice for managing CCN. However, cabbage fields in Gangwon province have a slope, making it challenging to apply soil fumigants and use plastic mulch. Consequently, alternative materials should be developed for managing CCN. Nematicidal cover crops and resistant cultivars are among the alternatives. We conducted resistance screening of 82 white mustard(Sinapis alba) resources from a genebank at RDA against CCN for use as nematicidal cover crops or breeding materials. In the first assay (1st, 2nd, and 3rd pot experiments), 15 white mustard resources were selected, while others were susceptible to CCN. To confirm the reproducibility of resistance to CCN for 13 selected resources, the second assay was performed. As a result, five white mustard resource (IT297309, IT297312, IT302951, IT302953, IT302954) demonstrated resistance to CCN. These findings indicate the potential use of these five white mustard resources as nematicidal cover crops or for breeding cultivars of Kimchi-cabbage in Republic of Korea.

A Study of New Modified Neyman-Scott Rectangular Pulse Model Development Using Direct Parameter Estimation (직접적인 매개변수 추정방법을 이용한 새로운 수정된 Neyman-Scott 구형펄스모형 개발 연구)

  • Direct parameter estimation method is verified with various models based on Neyman-Scott rectangular pulse model (NSRPM). Also, newly modified NSRPM (NMSRPM) that uses normal distribution is developed. Precipitation data observed by Korea Meteorological Administration (KMA) for 47 years is applied for parameter estimation. For model performance verification, we used statistics, wet ratio and precipitation accumulate distribution of precipitation generated. The comparison of statistics indicates that absolute relative error (ARE)s of the results from NSRPM and modified NSRPM (MNSRPM) are increasing on July, August, and September and ARE of NMNSRPM shows 10.11% that is the smallest ARE among the three models. NMNSRPM simulates the characteristics of precipitation statistics well. By comparing the wet ratio, MNSRPM shows the smallest ARE that is 16.35% and by using the graphical analysis, we found that these three models underestimate the wet ratio. The three models show about 2% of ARE of precipitation accumulate probability. Those results show that the three models simulate precipitation accumulate probability well. As the results, it is found that the parameters of NSRPM, MNSRPM and NMNSRPM are able to be estimated by the direct parameter estimation method. From the results listed above, we concluded that the direct parameter estimation is able to be applied to various models based on NSRPM. NMNSRPM shows good performance compared with developed model-NSRPM and MNSRPM and the models based on NSRPM can be developed by the direct parameter estimation method.

Development of a Pump Operation Rule in a Drainage Pump Station using a Real Time Control Model for Urban Drainage System (내배수시스템 실시간 운영 모형을 이용한 배수펌프장 운영기법 개발)

  • An urban drainage system consists of two major systems : flood drainage facilities and operating practices. The facilities are composed of sewer networks, gates, and pumping stations and the operating practice consists of pump or gate operation. Then, a real time simulation system which is able to simulate urban runoff and the pump operation and to consider the backwater effect is required to operate efficiently the pump. With this system, the efficient pump operating rule can be developed to diminish the possible flood damage on urban areas. In this study, a real time simulation system was developed using the SWMM 5.0 DLL and Visual Basic 6.0 equipped with EXCEL. Also, for developing efficient pump operating Rules, two new Rules were suggested. The first Rule is designed to operate pumps considering the condition of sewer networks such as depths of each junction. The second is to discharge all the amount of inflow at each time step. Results obtained by those Rules were compared with one by the current pump operating Rule which is able to consider only the depth of the retard basin. The developed model was applied to Joonggok retard basin and verified their applicability.

Removal of Dissolved Heavy Metals through Biosorption onto Indigenous Bacterial Biofilm Developed in Soil (토양 내 토착 미생물에 의한 바이오필름 형성과 흡착을 통한 용존 중금속 제거)

  • In situ stabilization of heavy metals through adsorption onto indigenous bacterial biofilm developed on soil particles was investigated. Biofilms were developed in soil columns by supply of various carbon sources such as acetate, lactate and glucose. During development of biofilms, acetate, lactate, and glucose solutions were flew out from the soil columns with volume ratios of 98.5%, 97.3%, and 94.7%, respectively, when compared with soil column supplied with deionized water. Decrease in effluent amounts through the soil columns amended with carbon sources over time indicated the formation of biofilms resulting in decrease of soil porosity. Solutions of Cd, Cr(VI), Cu, Pb, and Zn were injected into the biofilms supported on soil particles in the columns, and the dissolved heavy metals in effluents were determined. Concentrations of dissolved Cd, Cr(VI), Cu, and Zn in the effluents through biofilm columns were lower than those of control column supplied with deionized water. The result was likely due to enhanced adsorption of the metals onto biofilms. Efficiency of metal removal by biofilms depended on the type of carbon sources supplied. The enhanced removal of dissolved heavy metals by bacterial biofilms in this study may be effectively applied to technical development of in situ stabilization of heavy metals in natural soil formation contaminated with heavy metals.

  • In this study, non-flammable mineral foam board using waste glass that can be produced to standardized specifications were developed and evaluated for the performance. In addition to the physical and mechanical performance, the environmental properties such as insulation, non-combustibility, gas hazard, sound absorption, etc. were tested to verify the use as interior and exterior building materials. Through the structural review, the validity was verified for the application of the office and restaurant building.

  • Recently, significant attention is given to the wastewater treatment using Constructed wetland. This is because the wetland system is a kind of natural taeatment system, simple to maintain and it has relatively fewer technical difficulty. Thus, it would be a practical method to employ especially in rural area in Korea. In this paper, the authors dscuss the design criteria of construted wetland developed in USA as an initial feasibility study to adopt it in Korea. We discuss about especially types of vegetation, natural succession and management, planting techniques, seeding teckniques and management after censtruction.

The Role of the Immune System in the use of Probiotic Lactic Acid Bacteria in Preventing and Treating Allergic Diseases

  • The immune system is generally divided into the innate and the adopted immune systems, both protecting the body from pathogens. Recently, allergies, a disease associated with an imbalanced immune system, have increased rapidly in developed countries. Prevailing symptoms of allergic diseases are eczema, allergic rhinitis, asthma, inflammatory bowel disease, and food allergy. Probiotic bacteria, mainly consisting of lactic acid bacteria, are used in the prevention and treatment of allergic diseases. The function of them is to stimulate the intestinal immune cells and form a complex signal network to activate other immune cells. Beneficial health effects of probiotics are based on the hygiene hypothesis, which suggests that sanitary environment is important for health, but limited exposure to environmental factors increases allergic diseases. An immunoregulatory effect of probiotic bacteria is demonstrated by controlled trial, animal model, in vitro, in vivo and ex vivo designs. However, the immunoregulatory effect of probiotic bacteria is controversial because it depends on probiotic strains, a dose and a type of diseases. In this review, we discussed clinical evidences on immunoregulatory effects of probiotic bacteria.

The Development of the Automatic Discharge Acquisition & Management System (ADAMS) using Ubiquitous Technique

  • Accurate river discharge is the most important factor in managing river basins and for successfully maintaining total maximum daily loads in Korea. It is not easy to measure the discharge directly in large rivers owing to physical and environmental constraints, even after investing much time and money. Recently, to overcome these historical drawbacks in river discharge measurement, we have developed the Automatic Discharge Acquisition & Management System (ADAMS) that scans the river cross-section and measures each cell $(1m{\times}1m)$ velocity using HADCP. The hardware system is composed of an HADCP sensor and winch, as well as a PC and software system for the discharge calculation module and hardware control module. It is controlled remotely via the internet and uses the velocity-depth integration method and the velocity-contour method for calculating river discharges. The characteristics of ADAMS are a ubiquitously accessible system, featuring real time automatic discharge measurement, remote control via the internet. The results using ADAMS at the Jindong stage site show less than 5% uncertainty and are 4 times more efficient than the ADCP & Q-boat system. This system can be used to measure any large river, river mouth or tributary river affected by backwater, all of which have a very difficult measuring real time discharge. The next generation of ADAMS will feature an upgrade to increase portability and GPS integration.

Rainwater Harvesting System as an Alternative Water Source

  • The objectives of this study were to install RCR systems at a typical single family house and a school in Alabama, and evaluate the feasibility and efficiency of using the RCR systems for water harvesting in Alabama. The RCR systemswere equipped with a control system and a CR10X data logger to monitor the system operation and to collect data on precipitation, temperature, overflow, water depth in the storage tank and daily uses of toilet flushing. Daily average water use of the home for toilet flushing was 95 liter and 2100 liter was used at the school during the school days. Rainwater harvesting efficiency was 83.3 and 89 percent and RCR use efficiency was 18 and 98 percent from the home and the school, respectively. A computer program was developed to estimate potential effectiveness of RCR systems. From the analysis result with 10 years rainfall data, a total of 67,000 liters of rainwater could be harvested for domestic uses from a typical single family house which supplies 190 liters per day.

  • Destruction of HFC-134a from ELV (End of Life Vehicle) were determined in a gasification-melting demonstration system of municipal solid waste (100ton/day). The injection system has been developed for the uniform injection of HFC-134a to the gasification-melting system. The destruction characteristics of HFC-134a and analysis of exhaust gases have been performed. The destruction efficiency was 99.995% for HFC-134a feeding of 3 kg/hr and the exhaust gases such as CO, SOx, NOx, HCl and HF satisfied the environmental standards.
