Purification and Characterization of Cold Active Lipase from Psychrotrophic Aeromonas sp. LPB 4

  • A lipase from Aeromonas sp. LPB 4, a psychrotophile isolated from a sea sediment was purified and characterized. The lipase was purified 53.5 fold to a homogeneous state by acetone precipitation and QAE sephadex column chromatography and its molecular weight was determined to be 50 kDa by SDS-PAGE. The enzyme exhibited maximum activity at 10$^{\circ}C$ and was stable at temperatures lower than 50$^{\circ}C$. This lipase favored substrates containing medium carbon chain of acyl group, while too low and high carbon chain decreased its activity. The lipolytic activity of purified lipase was slightly increased by the addition of 0.1% detergent, but decreased by 1% of detergent. Butanol severely decreased the lipase activity while methanol increased the activity about 15%.

Biodegradation of and comparison of adaptability to dectergents (미생물에 의한 계면활성제의 분해능과 적응력의 비교)

  • Microorgansims utilizing anionic detergent as their carbon and sulfur sources were isolated from soils and sewages. Alkyl benzene sulfonate (Hiti) and sodium dodecyl sulfonate (SDS) were the detergent compound tested. Three of these isolated microorganisms were identified as Pseudomonas spp. and the others asKlbsiella, Enterobacter and Acinetobacter. Biodegradation rate of the detergents and growth rate of Acinetobacter Strain II-8, Pseudomonas strain H-3-1 and 554 among six isolated microorganisms were investigated with colorimetric, warburg manometric, and ultraviolet absorption analyses. By performance of 4 serial successive tranfer to new culture broth for the purpose of adaptation method, ABS and SDS could be degraded to far more than 40%-60% and 70%-75%, respectively. However the employment of nonadaptation method, ABS and SDS were degraded to 30%-45% and 45%-65%, respectively. In another words, detergents degradation ability was increased to a certain extent by successive transfer to the new minimal media. We would conclude that the development of adaptation was effective in the removal of recalcitrant compounds.

Quantitative Analysiss and Physicl Properties of Dietary Fibers in Vegetables (채소류의 식이섬유 함량 및 물리적특성)

  • The contents of dietary fiber and its physical properties of vegetables were determined. The samples selected in this study were soybean sprouts (Glycine max MERILL sprouts) butterber (Petasites japonicus MIQ), taro stem(Colcasia antiquorum SCHOTT stem), bracken (Pteridium aquilinum KUHN)and dried Korean cabbage leaves(Brassica compestris L.leaves). The contents of dietary fiber were 20.96~50.62% of neutral detergent figer, 10.60-38.19% of acid detergent fiber, 1.74-10.88% of lignin, 5.79~33.29% of hemicellulose and 5.79-33.29% of cellulose. Butterber had the highest dietary fiber content. Mean waterholding capacity of samples was 5.65-12.26g.water/g.acetone dried powder and butterber and dried Korean cabbage leaves had higher water holding capacity value than the others. Densities were 140-250mg/ml of direct density and 385-417 mg/ml of bulk density.
