A COMPARATIVE STUDY ON THE DISLODGING FORCE OF MAGNETIC ATTACHMENT TO THE DENTURE RESIN BY MAGNETIC DESIGN AND FIXING MATERIALS

  • STATEMENT OF PROBLEM: Detachment of the magnetic assembly from the denture base has been a problem in magnetic overdenture patients. PURPOSE: The objectives of this study were to compare the dislodging force by the fixing materials and the designs of the magnetic assembly, and to compare the effect between the fixing materials and the designs of the magnetic assembly. MATERIAL AND METHODS: Two fixing materials, Jet denture repair $acrylic^{(R)}$ and Super-$Bond^{(R)}$ C&$B^{(R)}$ and two types of magnetic assembly designed with or without wing were used. Each magnetic assembly was fixed in the chamber of the denture base resin block ($Lucitone^{(R)}$199) with each fixing material respectively. These specimens were thermocycled 2,000 cycles in the water held at $4^{\circ}C$ and $60^{\circ}C$ with a dwell time of 1 min each time. Each specimen was seated in a testing jig and then a push-out test was performed with a universal testing machine at a cross head speed of 0.5 mm/min to measure the maximum dislodging forces. RESULTS: Comparing the fixing materials, Super-Bond C&$B^{(R)}$ showed superior dislodging force than Jet denture repair $acrylic^{(R)}$. Comparing the design of the magnetic assemblies, the wing design magnetic assembly showed better dislodging force. Combination of the Super-Bond C&$B^{(R)}$ as a fixing material and wing design magnetic assembly revealed a greatest dislodging force. The kind of fixing material was more influential than the type of magnetic assembly. CONCLUSION: The dislodging force of Super-Bond C&$B^{(R)}$ was significantly higher than Jet denture repair $acrylic^{(R)}$. And the dislodging force of magnetic assembly which have wing design was significantly higher than magnetic assembly which have no wing design.

Bead-to-Bead Cell Transfer by Induction of Detachment of Anchorage Dependent HeLa Cells Grown on Macroporous Microcarriers (부착성 HeLa 세포의 탈리 유도에 의한 다공성 미립담체의 담체간 전이 배양)

  • Using a cellulose macroporous microcarrier, HeLa cells were cultivated in 100mL spinner flask(Bellco Co., USA) and confluent cell laden microcarriers were subcultured by bead-to-bead cell transfer method. In macroporous mcirocarrier-HeLa system viable suspended cells played an important role in bead-to-bead cell transfer and that could be increased by use of RPMI-1640, a calcium-ion-reduced-media and high speed agitation. Successful bead-to-bead cell transfers were performed continuously three time in spinner flask. We applied this technique to produce recombinant Vaccinia virus which express $\beta$-galactosidase. Recombinant protein yield of bead-to-bead transferred culture was comparable to conventional microcarrier cultures that were inoculated by cells detached from T-flask. Although trypsinization is a useful method for subculturing microcarriers in some cases, that process adds quality control problem and handling steps to large scale cell production. There fore, bead-to-bead cell transfer technique offers another convenient and efficient scale-up method for continuous microcarrier cultures.

  • This paper considers the issue of LSS(Liquid Spill Stopper)'s performance verification testing against external resisting force. The importance of the test is to ascertain the possibility of liquid spill stop quickly and efficiently by LSS. The method adopted in this research is the experimental device for puncture's inflow pressure and test the adhesive (magnetic) force on shell plates during the sailing. The major results of the paper are the followings : (a) The performance of LSS against 40 and 100mm diameter puncture was confirmed under 1.0bar. (b) LSS near bow detached first at 20.2 knots. This indicates that LSS can recover the damaged movability needed to naval warships' operation as well as merchant ships.

Improvement of Adhesion Strength of High Temperature Plasma Coated Aluminum Substrate with Aluminum-Alumina Powder Mixture (알루미늄 기지에 알루미늄-알루미나 혼합분말을 이용한 고온플라즈마 열분사 코팅층의 밀착강도 향상기구)

  • High temperature plasma coating technology has been applied to recover damaged aluminum dies from wear by spraying pure aluminum and alumina powder. However, the coated mixed powder layer composed of aluminum and alumina often undergoes a detachment from the substrate, making the coated substrate die unable to maintain its expected life span. In this study, in order to increase the bonding strength between the substrate and the coating layer, a pure aluminum layer was applied as an intermediate bond layer. In order to prepare the specimen with variable bond coating conditions, the bond coat layers with a various gun speed from 10 cm/sec to 30 cm/sec were prepared with coating cycle variations ranging from three to nine cycles. The specimen with a bond coat layer coated with a gun speed of 20 cm/sec and three coating cycles exhibited ~13MPa of adhesion strength, while the specimen without a bond coat layer showed ~6 MPa of adhesion strength. The adhesion strength with a variation of bond coat layer thickness is discussed in terms of coating parameters.

Evaluation of Macroporous and Microporous Carriers for CHO-K1 Cell Growth and Monoclonal Antibody Production

  • The emergence of microcarrier technology has brought a renewed interest in anchorage-dependent cell culture for high-yield processes. Well-known in vaccine production, microcarrier culture also has potential for application in other fields. In this work, two types of microcarriers were evaluated for small-scale monoclonal antibody (mAb) production by CHO-K1 cells. Cultures (5 ml) of microporous Cytodex 3 and macroporous CultiSpher-S carriers were performed in vented conical tubes and subsequently scaled-up (20 ml) to shake-flasks, testing combinations of different culture conditions (cell concentration, microcarrier concentration, rocking methodology, rocking speed, and initial culture volume). Culture performance was evaluated by considering the mAb production and cell growth at the phases of initial adhesion and proliferation. The best culture performances were obtained with Cytodex 3, regarding cell proliferation (average $1.85{\pm}0.11{\times}10^6$ cells/ml against $0.60{\pm}0.08{\times}10^6$ cells/ml for CultiSpher-S), mAb production ($2.04{\pm}0.41{\mu}g/ml$ against $0.99{\pm}0.35{\mu}g/ml$ for CultiSpher-S), and culture longevity (30 days against 10-15 days for CultiSpher-S), probably due to the collagen-coated dextran matrix that potentiates adhesion and prevents detachment. The culture conditions of greater influence were rocking mechanism (Cytodex 3, pulse followed by continuous) and initial cell concentration (CultiSpher-S, $4{\times}10^5$ cells/ml). Microcarriers proved to be a viable and favorable alternative to standard adherent and suspended cultures for mAb production by CHO-K1 cells, with simple operation, easy scale-up, and significantly higher levels of mAb production. However, variations of microcarrier culture performance in different vessels reiterate the need for optimization at each step of the scale-up process.

Evaluation of static fracture resistances and patterns of pulpless tooth restored with poly-ether-ketone-ketone (PEKK) post (Poly-ether-ketone-ketone (PEKK) 포스트로 수복한 근관 치료 치아의 정적 파절 저항성 및 파절 형태에 관한 평가)

  • Purpose: The purpose of present study was to investigate fracture strength and mode of failure of endodontically treated teeth restored with metal cast post-core system, prefabricated fiber post system, and newly introduced polyetherketoneketone (PEKK) post-core system. Materials and methods: A total of 21 mandibular premolar were randomly grouped into 3 groups of 7 each according to the post material. Group A was for metal cast post core; Group B for prefabricated glass fiber post and resin core; and Group C for milled PEKK post cores. All specimens were restored with metal crown. The fracture strength of each specimen was measured by applying a static load of 135-degree to the tooth at 2 mm/min crosshead speed using a universal testing machine. After the fracture strength measurement, the mode of failure was observed. The results were analyzed using Kruscal-Wallis test and post hoc Mann-Whitney U test at confidence interval ${\alpha}=.05$. Results: Fracture resistance of PEKK post core was lower than those of cast metal post and fiber reinforced post with composite resin core. In the aspect of fracture mode most of the root fracture occurred in the metal post core, whereas the post detachment occurred mainly in the fiber reinforced post. In the case of PEKK post core, teeth and post were fractured together. Conclusion: It is necessary to select appropriate materials of post for extensively damaged teeth restoration and clinical application of the PEKK post seems to require more research on improvement of strength.
