An experimental study was performed for the preparation of living skin-equivalent by the using collagen gel contraction with human fibroblasts as neodermls and cultured human keratinocytes as neoderm is . The results were as follows ; 1) The rate of collagen gel contraction was dependent on the number of fibroblasts into the lattice and collagen contraction was progressed according to the increment of the number of the cells. 2) The rate of collagen gel contraction was progressed according to the decrement of the contraction of the collagen. 3) The rate of gel contraction was progressed according to the increment of serum concentration in the fixed concentration of the fibroblasts and collagen. 4) The lattice contraction was decreased according to the increment of the population doublings of the fibroblasts. 5) Macroscopically, the artificial dermis was gray white in color and tissue-like consistency and elas- ticity. 6) Microscopically, three dimensionally contracted artificial dermis showed more dense fibroblasts and its newly formed collagen fibrils in the matrix than one dimensionally contracted one. 7) Finally prepared skin-equivalent showed good attachment of living stratified keratinocytes to the dermal equivalent microscopically. It has been proposed that newly formed skin-equivalent is suitable for the graft of extensively and deeply burned patients. Shortening of the manufacturing period of skin-equivalent and development of conservation technique as a readily usable state are to be solved for our ongoing works.

In this study, we measured the anti-oxidative activity of Cirsium setidens and investigated its effect on UVA-induced MMP-1 expression in human dermal fibroblats. And then we examined possible improvement in skin elasticity by topical treatment with fomular including Cirsium setidens extract. The ethanol extract of C. setidens showed free anion radical scavenging effect(87.47 % at 1 mg/mL) and superoxide anion radical scavenging effect(61.71 % at 1 mg/mL) in the xanthine/xanthine oxidase system, respectively. At the concentration of 100 ${\mu}g/mL$, C. setidens extract showed 95.54% inhibition on lipid peroxidation of linoleic acid. UVA-induced MMP-1 expression in human dermal fibroblasts was reduced to 54.69 % by treatment with 100 ${\mu}g/mL$ of C. setidens extract. A human clinical study, in which oil-in-water emulsion with C. setidens extract was topically applied, showed significant increase in skin elasticity. These results suggest that the C. setidens extract can be effective anti-aging ingredient for cosmetics applications.

In order to investigate the effects of Saussurea involucrata on (relationship between) aging (and Saussurea involucrata), we examined the activities of antioxidation, in vitro MMP inhibition and UVA-induced MMP-1 expression in human dermal fibroblasts. S. involucrata showed scavenging activities radicals and reactive oxygen species (ROS) with the $IC_{50}$ values of $3.89{\mu}g/mL$ against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and $67.29{\mu}g/mL$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. At the concentration of $1000{\mu}g/mL$, S. involucrata showed 93.27% inhibition on lipid peroxidation of linoleic acid. S. involucrata inhibited the activities of MMP-1 in a does-dependent manner and the $IC_{50}$ value calculated from semi-log plots was $97.18{\mu}g/mL$. Also, UVA induced MMP expression in human dermal fibroblasts was reduced 42.86% by treatment with S. involucrata, and MMP-1 mRNA expression was reduced in a dose-dependent manner. Therefore S. involucrata was able to significantly inhibit MMP expression in protein and mRNA level. All these results suggested that S. involucrata might act as an anti-aging agent by antioxidation and reducing UVA-induced MMP-1 production.

Cell senescence can be identified by cellular changes that occur as a result of intrinsic aging and/or diseases. In case of skin cells, aging and cell senescence caused by external factors results in cessation of cell proliferation and cellular malfunction, which, in turn, accelerates skin aging. In this study, inhibition of cell senescence and enhancement of cell function were studied using cordycepin to evaluate the potential for skin anti-aging agent. By comparing with the number of senescence associated with ${\beta}$-galactosidase (SA-${\beta}$-gal) positive cells in young and replicative aged human fibroblasts, it was found that replicative aged cells showed higher expression of ${\beta}$-galactosidase. Treatment of cordycepin - known as an anti-oxidative and anti-inflammatory agent - reduced ${\beta}$-galactosidase expression in senescent cells and enhanced cell survival in serum-free culture condition. Cordycepin also showed superb inhibition of ROS, which is another indicator of cell senescence. The results of this study proved the anti-aging effect of cordycepin on human fibroblasts and also proposed a possibility of its use as an anti-aging cosmetic ingredient.

We evaluated the antioxidant activity and anti-wrinkle effects of Aceriphyllum rossii leaf ethanol extract (ARLEE) in vitro using human dermal fibroblasts. The total polyphenol and flavonoid contents of ARLEE were 578.6 and 206.3 mg/g, respectively. At a concentration of $250{\mu}g/mL$, the electron-donating ability of ARLEE was 87.1%. In comparison with the vehicle, ARLEE treatment at $100{\mu}g/mL$ significantly increased type I procollagen synthesis (p < 0.01) by 50.7%. In vitro ARLEE treatment (10 mg/mL) inhibited collagenase and elastase activity by 97.1% and 99.2%, respectively. Compared with the control, ascorbic acid treatment at $100{\mu}g/mL$ significantly decreased matrix metalloproteinase (MMP)-1 protein expression (p < 0.01) by 37.0%. ARLEE treatment at $50{\mu}g/mL$ significantly decreased MMP-1 protein expression (p < 0.01) by 46.1%. Ascorbic acid and ARLEE treatments at $100{\mu}g/mL$ significantly decreased MMP-1 mRNA expression (p < 0.01) by 26.1% and 36.1%, respectively. From these results, we conclude that ARLEE has excellent antioxidant activity and even better anti-wrinkle effects than ascorbic acid in human dermal fibroblasts. These results suggest that ARLEE could be used in functional cosmetics for the prevention or alleviation of skin wrinkles induced by ultraviolet rays.

Selaginella tamariscina with the popular Korean name Keoun Back, is a traditional medicinal plant for therapy of advanced cancer patients in the Orient. In this study, we evaluated anti-aging activity of S. tamariscina using processed technology and investigated diverse biological activities of processed S. tamariscina (PST) as an anti-aging ingredient of cosmetics. PST, heated with sand, used to different purpose compared with origin in medicine. PST raises total phenol concentration and enhances the DPPH radical scavenging activity. For testing intracellular ROS scavenging activity, the cultured human dermal fibroblasts were analyzed by increase in dichlorofluorescein (DCF) fluorescence upon exposure to UVB $20 mJ/cm^2$ after treatment of PST. UVA-induced MMP-1 expression in human dermal fibroblasts was reduced in a dose-dependent manner by PST. Taken together, 4hese results suggest that PST may act as an anti-aging agent by preventing the skin cell from damage induced by UV irradiation, and imply that PST may be useful as a new ingredient for anti-aging cosmetics.
In this study, we investigated the boosting effects on collagen biosynthesis of $Anthriscus$$sylvestris$ ethanol extract (ASE) in human dermal fibroblasts. To obtain more effective fraction and subfraction for collagen biosynthesis, standard solvent partition and open column chromatography were performed. The EtOH extract, solvent fractions, and 8 EtOAc subfractions were tested for their collagen synthesis capacity by [$^3H$]Proline-incorporation and ELISA assay. ASE increased 25% of total collagen synthesis and 27% of procollagen biosynthesis. The total collagen biosynthesis was increased by EtOAc fraction and E6 subfraction to 28% and 50% respectively. Type I procollagens were also upregulated by EtOAc fraction and E6 subfraction to 30% and 47%, each. Taken together, our data suggest that potential anti-aging effect of ASE on skin is via increasing collagen biosynthesis and effective subtraction is E6 subfraction of EtOAc fraction.

This study focused on the anti-oxidative and collagenase- and elastase inhibition effects of low molecular weight peptides (LMP) from commercial Jeju horse leg bone hydrolysates (JHLB) on pancreatin, via enzymatic hydrolysis. Cell viability of dermal fibroblasts exposed to UVB radiation upon treatment with LMP from JHLB was evaluated. Determination of the antioxidant activity of various concentrations of LMP from JHLB were carried out by assessing 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azino-bis-3-ethybenzothiazoline-6-sulphonic acid (ABTS) radical scavenging activity, ferric reducing antioxidant power (FRAP), and oxygen radical absorbance capacity (ORAC). The DPPH radical scavenging activity of LMP from JHLB (20 mg/mL) was 92.21% and ABTS radical scavenging activity (15 mg/mL) was 99.50%. FRAP activity (30 mg/mL) was $364.72{\mu}M/TE$ and ORAC activity (1 mg/mL) was $101.85{\mu}M/TE$. The anti-wrinkle potential was assessed by evaluating the elastase- and collagenase inhibition potential of these LMP. We found that 200 mg/mL of LMP from JHLB inhibited elastase activity by 41.32%, and 100 mg/mL of LMP from JHLB inhibited collagenase activity by 91.32%. The cell viability of untreated HS68 human dermal fibroblasts was 45% when exposed to a UVB radiation dose of $100mJ/cm^2$. After 24 h of incubation with $500{\mu}g/mL$ LMP from JHLB, the cell viability increased to 60%. These results indicate that LMP from JHLB has potential utility as an anti-oxidant and anti-wrinkle agent in the food and cosmetic industry. Additional in vivo tests should be carried out to further characterize these potential benefits.
Background: Human skin undergoes distinct changes throughout the aging process, based on both intrinsic and extrinsic factors. In a process called photoaging, UVB irradiation leads to upregulation of matrix metalloproteinase-1, which then causes collagen degradation and premature aging. Mixtures of medicinal plants have traditionally been used as drugs in oriental medicine. Based on the previously reported antioxidant properties of Panax ginseng Meyer and Crataegus pinnatifida, we hypothesized that the mixture of P. ginseng Meyer and C. pinnatifida (GC) would have protective effects against skin aging. Methods: Anti-aging activity was examined both in human dermal fibroblasts under UVB irradiation by using Western blot analysis and in healthy human skin by examining noninvasive measurements. Results: In vitro studies showed that GC improved procollagen type I expression and diminished matrix metalloproteinase-1 secretion. Based on noninvasive measurements, skin roughness values, including total roughness (R1), maximum roughness (R2), smoothness depth and average roughness (R3), and global photodamage scores were improved by GC application. Moreover, GC ameliorated the high values of smoothness depth (R4), which means that GC reduced loss of skin moisture. Conclusion: These results suggest that GC can prevent aging by inhibiting wrinkle formation and increasing moisture in the human skin.
To evaluate the protective effect of collagen peptide-coated coffee extract on skin aging, cell viability was measured with a MTT assay using cultured CCD-986sk fibroblasts, and its effect on wrinkles in the skin of hairless mice induced by UVB-irradiation was examined. In addition, its effect on procollagen synthesis and anti-oxidative, and its inhibitory activity against collagenase, elastase, tyrosinase and MMP-1 were analysed. After the 30-minute topical treatment, the animals were exposed to UVB irradiation (60-100 mJ/cm2) for 4 weeks and its intensity increased during the period. Under the experimental conditions set in this study, the skin thickness of hairless mice significantly decreased (11.8-21.3%) compared to the control group. Based on these results, the prolonged oral intake of a collagen peptide mixture with coffee is expected to significantly increase the synthesis of procollagen in dermal fibroblasts, thereby contributing to the alleviation of wrinkling and lowered elasticity due to structural damage to the dermal layer caused by UV. The oral intake of collagen-coated coffee contributes to increasing collagen biosynthesis in a dose-dependent manner and alleviates the symptoms of thickened keratin caused by UV irradiation. However, it did not inhibit the enzymes involved in skin aging, whitening, wrinkle improvement, and antioxidation. Based on the these results, it can be concluded that the intake of collagen peptide-coated coffee extract can be utilized as an alternative material for the prevention or treatment of diseases associated with photoaging.
