Trichoderma viride 변이균주 분리를 위한 Cellulase Semiquantitative Plate Assay (Plating Technique for Cellulase Mutants of Trichoderma viride)

  • Trichoderma viride QM 9414의 생육을 저해하여 colony를 형성 시키고 배지에 첨가된 cellulose가 균이 분비한 cellulase에 의해서 분해되어 생긴 clear zone으로서 육안으로 cellulase 역가를 쉽게 측정할 수 있는 방법과 repressor를 첨가하여 constitutive mutant를 선별할 수 있는 방법에 관한 실험을 하였든 바 그 결과를 요약하면 다음과 같다. 1) Colony를 형성시키고 그 size를 제한 하는데는 deoxycholate를 배지에 0.15% 첨가 하였을 때 가 가장 좋았다. 2) Saponin을 배지에 0.l% 첨가하면 뚜렷한 clear zone을 형성하였다. 3) Constitutive mutant를 선별하기 위해서는 기질로 pulp를 사용하였을 경우 glucsoe를 1% 첨가하거나 glycerol이나 cellobiose를 0.5% 첨가하면 가능하다고 생각되며 기질로 pure cellulose를 사용하였을 경우는 그대로 배지에 spore를 plating하면 가능하다고 생각된다. 4) Cellulase 역가가 높은 균주는 clear zone의 크기로서 쉽게 분리할 수 있다.

Studies on the Formation and Stability of Colloids (I): Perturbation of Micelle Formation of Sodium Deoxycholate by Amides

  • The critical micelle concentration (CMC) of sodium deoxycholate (NaDC) and the effects of amides on the micellization processes have been studied by fluorometric technique using pyrene as a probe. The addition of amides as cosolvent destabilized the NaDC micelle and increased the CMC. The order of effectiveness for the perturbation of NaDC micelle was N-methylacetamide ${\ge}$ DMF > acetamide > formamide, which is the order of hydrophobicity of the amines. This indicated that the effect of amides on the micellization processes of NaDC arises from diminution of the hydrophobic effect. The electrostatic repulsion between ionic head groups in the NaDC micelle appeared to be much less than that in aliphatic ionic micelle. This was also revealed in the weaker dependence of the CMC on ionic strength. The premicellar association of NaDC was not significantly involved in the micellization processes of the bile salt.

  • Aqualyx (Marllor International Ltd, Rimini, Italy) was originally developed in Italy by Professor Pasquale Motolese and has been commercially available since 2009. It is a deoxycholate, aqueous gelatinous solution mixed with saline and buffering compounds. It is the only drug approved by the European Union for the reduction in localized fat. Aqualyx is sold exclusively to doctors and nurses trained in intralipotherapy. In the case of our patient, the product administered was advertised as Aqualyx, but was not administered by a trained health professional and was administered too superficially. The patient developed severe pain following the injection and was unable to sit for several weeks. There was localized skin necrosis, and palpable collections where the injection was administered. Our initial suspicion was development of an abscess or hematoma. To characterize further, we arranged an ultrasound scan that showed a "superficial hypoechoic lesion" but no deeper infection or spread. The numerous painful nodules ruptured onto the skin surface, resulting in purulent and bleeding lesions. This case demonstrates the importance of appropriate training and competence in performing cosmetic procedures including injections and fat dissolving treatments.

인삼(人蔘) 사포닌을 비롯한 계면활성제(界面活性劑)가 위장관내(胃腸管內)의 단백질(蛋白質) 가수분해효소(加水分解酵素) 반응(反應)에 미치는 영향(影響) (The Effects of Surfactants Including Ginseng Saponins on the Gastric Enzyme-Catalyzed Hydrolysis)

  • Objectives : This study was conducted to investigate the effects of ginseng saponins and commercial surfactants such as Triton X-100, sodium deoxycholate, and sodium dodecyl sulfate on the gastric enzyme-catalyzed hydrolysis. Methods : Saponins (a surface-active plant component) from fresh ginseng root were extracted to examine its effect on the gastric enzyme-catalyzed hydrolysis. Commercial surfactants such as Triton X-100, sodium deoxycholate, and sodium dodecyl sulfate were also employed in the hydrolysis system to compare their effects with that of the ginseng saponins. The effects of surfactants on the gastric enzyme-catalyzed hydrolysis were measured by using a spectrophotometer. A spectropolarimeter was used to examine the conformational change of enzymes and substrates by the addition of ginseng saponins into the system. Results : Both the tryptic and the peptic digestion of milk casein or eggalbumin were slightly improved with an increase in the amount of ginseng saponins in the system. Triton X-100 showed an effect similar to that of ginseng saponins, while sodium dodecyl sulfate and sodium deoxycholate diminished the hydrolysis. Circular dichroism spectra of enzymes and substrates was significantly changed by the addition of ginseng saponins into the system. Conclusions : These results show that ginseng saponins affect positively the gastric enzyme-catalyzed hydrolysis, and suggest that the digestion of substrates by gastric enzymes is affected by the change of enzyme conformation by ginseng saponins.

The Effect of Phosphatidylcholine and Deoxycholate Compound Injections to the Localized Adipose Tissue: An Experimental Study with a Murine Model

  • Background Phosphatidylcholine (PPC) and deoxycholate (DCA) compound has been recently used for the purpose of partial lipolysis and is valued for its efficacy and lower invasiveness compared to liposuction and dermolipectomy used previously. In this article, the authors discuss the efficacy of the PPC dissolved in DCA via an experimental rat study model, along with suggesting a useful animal experimental model for the study of adipose tissue and lipolysis. Methods Bilateral inguinal fat pads of an experimental rat were elevated with the deep inferior epigastric vessel as the sole vascular pedicle. Normal saline was injected on one side as a control group and a PPC and DCA compound was injected on the other side. After 4 days, the rats were euthanized for microscopic tissue examination. The pathology was scored by a sem-iquantitative system in 4 categories: normal fat amount, fat necrosis, inflammatory activity, and stage of fibrosis. A Wilcoxon signed-rank test powered by SPSS packet program was used for statistical analysis and to determine significance. Results Microscopic examination was performed on the obtained samples, and the experimental data of all four categories showed significant histologic differences compared to the control group. All of the data also showed statistical significance by the Wilcoxon signed-rank test (P<0.01). Conclusions In the inguinal fat pad rat model, the control group and the experimental group had a differed significantly in the amount of normal fat tissue, inflammation, necrosis, and fibrosis. We recommend the rat inguinal fat pad model used in this study, as it is likely to be useful in related research.

계면활성제 첨가배양에 따른 Rhizopus oryzae의 $\alpha$-Amylase와 Phosphatase분비촉진 (Enhancement by Surfactant on Release of $\alpha$-Amylase and Phosphatase in Submerged Culture of Rhizopus oryzae)

  • Rhizopus oryzae를 액체배양하는 동안, $\alpha$-amylase, acid phosphatase 그리고 alkaline phosphatase와 같은 분비효소의 분비능에 대한 계면활성 제의 촉진효과를 조사하였다. 그 결과 집락의 형태적인 변화를 볼 수 있었는바, 0.18mM SDS 처리구에서는 작은 pelletal형, 0. 48mM DOC 처리구에서는 pulpy형, 계면활성제를 처리하지 않은 시험구에서는 사상형의 집 락을 나타내었다. SDS를 첨가함으로써 $\alpha$-amylase는 9배, acid phosphatase는 25배의 활성도 증가가 유도되었다. DOC첨가 시험구에서 alkaline phosphatase 활성도는 배양액에서 11배 증가가 나타났을 뿐 아니라 세포질에서도 역시 상승하였다.

  • The effect of substrate micellization on the hydrolysis rate in the production of lysopho-sphatidylcholine (LPC) from phosphatidylcholine (PC) using hog pancreas phospholipase A2(PLA2) was studied. The optimal temperature and pH for the reactions in aqueous phase was found 42C and 7.2, respectively. For a given PC concentration, initial reaction rate was progressively increased with the addition of sodium deoxycholate (DOC), which could transform the bilayer of phospholipids into micellar structure.

  • To improve the solubility properties of indomethacin, a poorly water-soluble drug, some attempts were made with surfactants, such as sodium deoxycholate, sodium laurylsulfate and polysorbate-80. The enhancement effect in solubility was found to be due to the micellar solubilization and dependent on the concentration of surfactants, pH and temperature. The mechanism of interaction was suggested to be attributed to the formation of mixed micelles between the drug and solubilizers.

  • The micelle formation of NaDC was studied by fluorometric and viscometric measurements. The thermodynamic parameters of the primary and secondary micellization of the bile salt were evaluated. The primary micelle formation was appeared to be an entropy driven process due to hydrophobic effect, while the major driving force for secondary micelle formation of the bile salt is the large negative enthalpy. The secondary micelle provides less hydrophobic environment to pyrene than the primary micelle does. The cooperative aggregation of primary micelles via hvdrogen bond formation was proposed for the secondary micelle formation.

  • The bile secretion was accelerated generally by the administration of all derivatives tested: chemodeoxycholate, deoxycholate, cholate, dehydrocholate, 7-ketochenodeoxycholate, and 3, 7-diketochenodeoxycholate in decreasing order. Bile acids content in bile from animals administered with cholate was increased, however, other derivatives did not alter the contents of bile acids and bilirubin. In view of pharmacological point, all derivatives have hydrocholeretic action, however, only cholate exhibits typical choleretic action.
