In this research, characteristics of nitrification and denitrification using the microorganism attached on sponge and plates were examined. The denitrification and nitrification performance were investigated under the anaerobic and aerobic condition for about 2 months. Because the basins of denitrification and nitrification were connected in series, wastewater was flowed from denitrification basin to nitrification one. The 90% of influent flowrate was returned from nitrification basin to denitrification one. Most of organic material was removed in nitrification basin, wherease the only exact amount of organics required in denitrification process was removed in denitrification one. This experiment resulted in that heterotrophic bacteria existing in aerobic basin governed the removal efficiency of organic compounds. In case the influent BOD concentration into nitrification basin was 80mg/l, it did not affect to accumulation of nitrifying bacteria, the balance of heterotrophic bacteria was proved to be an important factor in nitrification/denitrification method such as anaerobic and aerobic cycling type.
Denitrification is an important biological mechanism in wastewater treatment process because this process is technically to remove nitrogen from water to air. There have been lots of study about denitrification engineering and molecular biological research about denitrifying bacteria, respectively. However, combination of these researches was unusual and rare. This study is about the correlation between quantity of denitrifying bacteria and denitrification potential, and consists of NUR batch test as analysis method of denitrification potential and quantitative molecular analysis for denitrifying bacteria. Three reactors (A/O, MLE and A/O of nitrogen deficiency) are operated to get activated sludge with various denitrification potential. All samples which were acquired from reactors were measured denitrification potential by NUR test and NUiR test. Also, Real-time PCR was conducted for quantification of denitrifying bacteria composition in activated sludge. The various denitrification potentials were measured in the reactors. The denitrifiaction potential was the highest in MLE process and the reactor of the nitrogen deficiency showed the lowest. Genomic DNA of activated sludge was obtained and consequently, real-time PCRuse the primer sets of nirK and nirS were conducted to quantify genes involving denitrification reductase production. As the result of real-time PCR, nirK gene showed more significant influence on denitrification potential comapred with nirS gene.
Nitrification and denitrification are important processes in the landfill site as they are deeply related with degradation and stabilization of refuse. Also nitrous oxide ($N_2O$) which is released from both nitrification and denitrification is known as greenhouse gas (GHG). The purpose of this study was to clarify the process by which $N_2O$ produced using $^{15}N$ isotope. Nitrate which was labeled to 10.08% with $^{15}KNO_3$ was used and $N_2O$ was analyzed with GC mass. Results was that even also when $O_2$ of bulk was 15%, $N_2O$ was released from denitrification. And as concentrations of $O_2$ increase, sum of $N_2O$ was released from denitrification. And as concentrations of $O_2$ increase, sum of $N_2O$ and $N_2$ was decreased and ratios of $N_2O$ in the reduced gases were increased. FISH technics also adaped to confirm whether which of nitrifiers existed in the substrates. When NEU was used of which the target was ammonia oxidizing bacteria, nitrifier was not detected at all. So it was confirmed that during the reaction denitrification was dominant process. Total bacteria distributions which were detected by EUB probe explained that as $O_2$ increase the number of bacteria also increase, but between the 10-15% of $O_2$ there was no any differences.

Recently, the interests on economical nitrogen removal from wastewater are growing. As a method of the novel nitrogen removal technology, nitrogen removal via nitrite pathway by selective inhibition of free ammonia and free nitrous acid on nitrite oxidizing bacteria have been intensively studied. The inhibition effects of free ammonia and free nitrous acid are low when domestic wastewater is used, however, because of its relatively lower nitrogen concentration than the wastewater from industry and landfill, etc. In this study, a sequencing batch reactor (SBR) is proposed for nitrogen removal to investigate the effect of the low nitrogen concentration on nitrite accumulation. Nitrification efficiency reached almost 100% during the aerobic cycle and the maximum specific nitrification rate ($V_{max,nit}$) reached $17.8mg\;NH_4{^+}-N/g\;MLVSS{\bullet}h$. During the anoxic cycle, average denitrification efficiency reached 87% and the maximum specific denitrification rate ($V_{max,den}$) reached $9.8mg\;NO_3{^-}-N/g\;MLVSS{\bullet}h$. From the analysis the main reason of nitrite accumulation in the SBR was free nitrous acid rather than free ammonia. Nitrite accumulation increased with the decrease of organic content in the wastewater and the mechanism is not well understood yet. From the result of fluorescent in situ hybridization, the distribution of nitrite oxidizing bacteria was in equilibrium with ammonium oxidizing bacteria when nitrite accumulation did not occur.

Water pollution by ammonium ion and nitrate is a common and growing problem in the ecosystem. Process of biological removal consists of nitrification and denitrification by bacteria. Ammonium is oxidized generally to nitrate by nitrification and nitrate is reduced to dinitrogen gas in the subsequent denitrification process. Although natural zeolite is well known for its ability to preferentially remove ammonium, it is not sufficiently removing ammonium ion and nitrate by adsorption. In order to overcome this problem, a method of biological removal with zeolite is used for simultaneous removal of ammonium and nitrate. As a result, in case of shaking culture with 1% seed and passing through zeolite column, the process revealed that ammonium ion could be removed completely after 14 hours. The removal of nitrate using columns with naturally adsorbed bacteria onto zeolite reached approximately 100% after 4 hours.
Groundwater samples were collected at 6 sites in Seoul area. DNA extraction from the sample was performed by the boiling method. Samples were boiled with guanidinium thyocyanate and phenol-chloroform. One set of primer was designed for amplification of 16S rDNA. For detection of denitrifying bacteria in groundwater sample, the author used primer sets consensus regions in gene sequences encoding the two forms of nitrite reductase (NIR), a key enzyme in the denitrification pathway. Two sets of PCR primer were designed to amplify $cd_1$-and Cu-nir. We confirmed the existence of denitrifying bacteria in 3 sites using $cd_1$-nir primer and in 4 sites using Cu-nir primer.
To remove nitrogen from wastewater, ammonia nitrogen has to be oxidized to nitrate nitrogen before denitrification reaction which converts nitrate nitrogen to nitrogen gas. In order to understand nitrification, several mathematical models had been proposed and Monod type model has been accepted internationally. Since Monod type model consists of maximum substrate utilization rate, substrate concentration and half-saturation coefficient, these values have to be addressed before using Monod type model. Several experimental procedures to determine half-saturation coefficient have been developed, however, Infinite dilution method was known to be time saving procedure. In this study, the mathematical equations and experimantal procedures for Infinite dilution method are presented and this method is used to determine half-Saturation coefficient for nitrifying bacteria. As results, Infinite dilution method is proved that this coefficient can be determined within 8 hours and the values of half-saturation coefficient has a range of 0.728 and $0.455gNH_4{^+}-N/m^3$ and the average has $0.580gNH_4{^+}-N/m^3$ through 5 sets of experiments.

A sulfur utilizing nitrite denitrification process could be placed after the shortcut biological nitrogen removal (SBNR) process. In this study, removal of nitrite using sulfur oxidizing denitrifier was characterized in batch tests with granular elemental sulfur as an electron donor and nitrite as an electro acceptor. At sufficient alkalinity, initial nitrite nitrogen concentration of 100 mg/L was almost completely reduced in the batch reactor within a incubation time of 22 h. Sulfate production with nitrite was 4.8 g ${SO_4}^{2-}/g$${NO_2}^-$-N, while with nitrate 13.5 g ${SO_4}^{2-}/g$${NO_3}^-$-N. Under the conditions of low alkalinity, nitrite removal was over 95% but 15 h of a lag phase was shown. For nitrate with low alkalinity, no denitrification occurred. Sulfate production was 2.6 g ${SO_4}^{2-}/g$${NO_2}^-$-N and alkalinity consumption was 1.2 g $CaCO_3/g$${NO_2}^-$. The concentration range of organics used in this experiment did not inhibit autotrophic denitrification at both low and high alkalinity. This kind of method may solve the problems of autotrophic nitrate denitrification, i.e. high sulfate production and alkalinity deficiency, to some extent.
Nitrogen (N) loading from domestic, agricultural and industrial sources can lead to excessive growth of macrophytes or phytoplankton in aquatic environment. Many studies have used stable isotope ratios to identify anthropogenic nitrogen in aquatic systems as a useful method for studying nitrogen cycle. In this study to evaluate the precision and accuracy of denitrification bacteria method (Pseudomonas chlororaphis ssp. Aureofaciens ($ATCC^{(R)}$ 13985)), three reference (IAEA-NO-3 (Potassium nitrate $KNO_3$), USGS34 (Potassium nitrate $KNO_3$), USGS35 (Sodium nitrate $KNO_3$)) were analyzed 5 times repeatedly. Measured the ${\delta}^{15}N-NO_3$ and ${\delta}^{18}O-NO_3$ values of IAEA-NO-3, USGS 34 and USGS35 were ${\delta}^{15}N:4.7{\pm}0.1$‰ ${\delta}^{18}O:25.6{\pm}0.5$‰, ${\delta}^{15}N:-1.8{\pm}0.1$‰ ${\delta}^{18}O:-27.8{\pm}0.4$‰, and ${\delta}^{15}N:2.7{\pm}0.2$‰ ${\delta}^{18}O:57.5{\pm}0.7$‰, respectively, which are within recommended values of analytical uncertainties. Also, we investigated isotope values of potential nitrogen source (soil, synthetic fertilizer and organic-animal manures) and temporal patterns of ${\delta}^{15}N-NO_3$ and ${\delta}^{18}O-NO_3$ values in river samples during from May to December. ${\delta}^{15}N-NO_3$ and ${\delta}^{18}O-NO_3$ values are enriched in December suggesting that organic-animal manures should be one of the main N sources in those areas. The current study clarifies the reliability of denitrification bacteria method and the usefulness of stable isotopic techniques to trace the anthropogenic nitrogen source in freshwater ecosystem.

A combined process consisted of a biofilm reactor and a continuously stirred-tank reactor (CSTR) was investigated for highly loaded ammonium wastewater treatment via nitrite accumulation. To enhance ammonium oxidizing bacteria over nitrite oxidizing bacteria on the surface of carriers, the biofilm reactor was operated at temperature of $35^{\circ}C$ for more than three months but the influent ammonium (500 mg-N/L) was partially oxidized to nitrite (240 mg-N/L). As pH was increased from 7.5 to 8.0, nitrite accumulation was fully achieved due to the inhibition of nitrite oxidizing bacteria under high free ammonia concentration. The biofilm reactor performance was severely deteriorated at the hydraulic retention time of 12 hr, at which incomplete nitrification of ammonia was observed. Various solubilization methods were applied to sewage sludge for enhancing its biodegradability and the combined method, alkaline followed by ultrasonic, gave the highest solubilization efficiency (58%); the solubilized solution was used as the external carbon source for denitrification reaction in CSTR. FISH analysis showed that the dominant microorganisms on the carriers were ammonium oxidizing bacteria such as Nitrosomonas spp. and Nitrospirar spp. but low amounts of nitrite oxidizing bacteria as Nitrobacter spp. was also detected.
