동위효소 및 RAPD분석에 의한 한국재래종 누에계통의 계통학적 특성 (Phylogenetic Relationships and Characterization of Korean Native Silkworm Strains Based on RAPDs and Isozyme Analysis, Bombyx mori)

  • 집누에는 지리적인 특징에 의해 중국종, 일본종, 유럽종, 열대종 및 한국종으로 분류된다. 한국종계통은 품종수도 적을 뿐만 아니라 관련 연구도 미미한 실정이다. 본 연구는 한국재래종으로 추측되는 품종들을 국내 · 외로부터 수집하여 등위효소 및 체액단백질유전자와 RAPD다형분석을 실시하여 한국재래종 계통의 품종적 유연관계와 계통특성을 밝히고자 하였다. 1. 등위효소유전자의 분석결과, 몇 개의 유전자군에서 한국재래종과 타 지역종간에 유전자형 및 유전자 빈도의 차이가 명확히 나타났다. 2. RAPSD의 결과를 UPGMA법에 의해 분석한 결과, 짐누에군과 멧누에군으로 크게 구분되었으며 유전적 유사계수 0.6930을 기준으로 한국재래종, 일본종 및 중국종으로 그룹화되었다. 3. 이상의 결과를 종합하면, 한국재래종 누에계통은 하나의 지역종 원종계통으로 분류될 수 있는 명확한 유전적 특성을 가지는 것은 물론 한국종의 계통특성도 뚜렷한 것으로 확인되었다.

산초나무(Zanthoxylum schinifolium) 신품종의 꽃과 열매에 기반한 형태적 특성 분석 (Morphological Characteristics Based on Phenotypes of Flower and Fruit in a New Variety of Anthoxylum schinifolium)

  • 산초나무 5개 품종의 꽃과 열매에 대해 형태적 특성을 조사한 결과, 화서의 너비와 암꽃잎의 길이는 품종에 따라 유의적 차이가 있었다. 또한 각 특성에 대한 기여도 확인을 위해 주성분을 확인한 결과, 제3주성분까지의 누적 설명력은 71.0%였고, 화서의 크기, 열매 송이의 크기, 암꽃잎의 크기, 종자의 무게, 종자 수는 높은 상관관계를 보였으며, 이러한 특성들은 품종을 설명하는데 있어 중요도가 높다고 사료된다. 분산분석 결과와 주성분 분석결과에서 화서의 너비, 암꽃잎의 길이 특성이 공통적으로 유의한 결과를 보였으며, 이 특성들은 품종 특성을 설명하는 데 있어 중요 형질이라 판단된다. 유집분석 결과 거리 수준 15를 기준으로 I그룹은 한초 915호, 1020호와 1030호로 가장 근접하였으며, 나머지 930호, 10호는 각각의 그룹으로 구분되었다. 그러나 질적 특성 결과에서 개화기와 결실기 차이가 확연하게 나타남으로 품종 간 구별되는 것을 확인할 수 있었다. 본 연구를 통해 산초나무의 꽃과 열매의 형태적 특성에서 품종 간 유연관계를 확인하였으며, 산초품종 육성을 위한 기초자료로 이용될 수 있을 것으로 사료되며. 향후 다양한 품종에서 유연관계를 설명을 위해 데이터 처리에 대한 기초자료로 이용될 것을 기대한다.

Examination of Genetic Relationships of Silkworm Stocks in Korea by Additive Isozyme Analysis

  • Previously, genetic relationships among 321 silkworm races preserved in the Department of Sericulture and Entomology, NIAST, RDA were studied using six isozymes comprising 20 loci. As a part of additional studies, three additional isozymes with 7 loci [phos-phoglucomutase (PGM), glucose phosphate isomerase (GPI), malete dehydrogenase (MDH)] from hemolymphs, midgets, and eggs were employed to reevaluate the previous dendrogram (UPGMA method). Although the current study supported the recognition of the previously identified major groups, some minor changes were apparent among the selected 47 forms. The current study showed that the polymorphic loci of GPI from eggs and MDH from hemolymph appear to be responsible characters for generating a new major group in the genetic relationship. Interpreting the data with current additive isozymes may represent more robust genetic relatioships among 321 silkworm races preserved in Korea, until further evidence is available.

한국산 또아리물달팽이과 ( family Planorbidae ) 3종에서의 동위 효소 변이 (Isozyme Variability in Three Species of Freshwater Planorbid Snails in Korea : Gyraulus convexiusculus , Hippeutis cantori and Segmentina hemisphaerula)

  • A horizontal starch gal electrophoresis for enzyme proteins extracted from 3 species of Korean planorbid snails; Gyraulus convexiusculus, Hippeutis cantori and Segmentina hemisphaerula was carried out in order to elucidate their genetic relationships.The results from 12 enzymes employed in three different kinds of buffer systems are summarized as follows:1) Two loci from each enzyme of aldehyde oxidase, esterase, glucose phosphate isomerase. isocitrate dehydrogenase, leucine aminopeptidase, malate dehyogenase, peptidase and xanthine oxidase were detected, and only one locus was observed from each of the following four enzymes: 6-phosphogluconate dehydrogenase, glyceraldehyde-phosphate dehydrogenase, glutamate oxaloacetate transaminase and glycerol-3-phosphate dehydrogenase.2) Most of loci in 3 species of planorbid snails employed showed homozygous and monomorphic banding patterns and some of them were specifis as genetic markers among different species. However, a few of loci (EST-1. EST-2 and GPI-2)showed polymorphic banding patterns. 3)Hippeutis cantori and Segmentina hemisphaerula were more closely clustered in a dendrogram with the genetic iddentity value of 0.431, and these two species were lineated with Gyraulus convexiusculus as another cluster at the value of 0.294.In summarizing the above results, three species of Korean planorbid snails employed in this study mostly showed monomorphic enzyme protein banding patterns and genetic differences specific among 3 species.

마산만 준설사업 및 하수처리장 가동에 따른 진해만의 수질변동 (Water Quality Variations in Jinhae Bay by Dredging & Operating the Sewage Disposal Plant)

  • This study investigates the spatial and temporal distribution characteristics and relationships among water quality parameters, which based on 6 years' data(from 1989 to 1994) measured at 16 stations on Jinhae Bay. The results of these analysis, monthly variations range between surface and bottom layer of water quality had a tendency to increase and decrease, and appeared to be at the maximum value in August. The relationships between concentration of COD ana nutrients(DIN and DIP), which obtained by correlation analysis of water quality, were shown $85\%$ and $74\%$, respectively. Using the cluster analysis to develop the division of the sea basin by the dendrogram, before and after dredging of Masan bay and operating a sewage disposal plant, the variation characteristics of water quality of Jinhae Bay were discussed. Through it, we can see the serious pollution of northen sea basin of Jinhae Bay(B2) although dredging Masan bay and operating the sewage disposal plant. As the results, it doesn't appear the improvement effect of water quality in spite of carrying out the effort of water quality improvement.

The Hierarchy of Images according to Construction Factors of the Flared Skirts

  • This study analyzed hierarchy of image for visual evaluation of flare skirt. This study analyzed expression words about flare skirt with frequency data of image expression words with different length and volume of flare. Stimuli for the study were set to be 4 different volume of flare ($90^{\circ}$, $180^{\circ}$, $270^{\circ}$, $360^{\circ}$) and 3 different length of skirt(48cm, 58cm, 68cm). Stimuli were made by using I-Designer which is Virtual Sewing System. From simulation of flare skirt, the subjects were asked to write down suggested adjective freely and selected 210 adjectives. With this, we chose total 38 adjectives considering frequencies in the pre-study. And we analyzed the combination process of expression words according to construction factor of flare skirt and hierarchy of image from dendrogram which was resulted by hierarchical cluster analysis. 'Feminine' got high score in all 12 flare skirt. When the skirt was short, it was vivid, and as the skirt got longer, ordinary and pure image showed. Also, as the volume of flare got larger, the average of visual effect was higher than visual image. Visual hierarchy construction according to construction factors of flare skirt could be divided into visual image and visual effect, and visual image was shown to be form 'A type - large volume of flare and short skirt length', 'H type-small volume of flare and short skirt length' and 'X type - large volume of flare and long skirt length'.

Potential Biotypes in Korean Isolates of Bipolaris cactivora Associated with Stem Rot of Cactus

  • A total of 62 isolates of Bipolaris cactivora causing cactus stem rots were isolated from major cactus-growing areas in Korea. Colony morphology of the isolates on potato-dextrose agar was differentiated into aerial (CA) and non-aerial mycelial types (CB). CA had profound aerial mycelium with grayish brown (CA-l), light brownish (CA-2), and brownish (CA-3) pigmentations; respectively, while CB had dark brownish pigmentations. CA had conidia of less dark pigmentation and acute terminal end. CB had darker and more round-end conidia. Twenty-eight amplified fragments were produced by polymerase chain reaction (PCR) with a set of 2 random primers. The sizes of amplified DNA fragments ranged approximately from 0.1 to 2.3 kb. The isolates were classified into 2 major genomic DNA random amplified polymorphic DNA (RAPD) groups at the genomic similarity of 97.7% and 95.1%, respectively. Cluster analysis of genetic similarity among the isolates generated a dendrogram that clearly separated all isolates into SA or SB. This result suggests that there may be two morphotypes of B. cactivora in Korea that may differ in their genetic constitutes.

Detection of tube defect using the autoregressive algorithm

  • Easy detection and evaluation of defect in the tube structure is a continuous problem and remains a significant demand in tube inspection technologies. This study is aimed to automate defect detection using the pattern recognition approach based on the classification of high frequency stress wave signals. The stress wave signals from vibrational impact excitation on several tube conditions were captured to identify the defect in ASTM A179 seamless steel tubes. The variation in stress wave propagation was captured by a high frequency sensor. Stress wave signals from four tubes with artificial defects of different depths and one reference tube were classified using the autoregressive (AR) algorithm. The results were demonstrated using a dendrogram. The preliminary research revealed the natural arrangement of stress wave signals were grouped into two clusters. The stress wave signals from the healthy tube were grouped together in one cluster and the signals from the defective tubes were classified in another cluster. This approach was effective in separating different stress wave signals and allowed quicker and easier defect identification and interpretation in steel tubes.

바다빙어과 2종 (Hypomesus nipponensis와 Mallotus villosus)의 변이 (Variability in Two Species of Osmeridae (Hypomesus nipponensis and Mallotus villosus))

  • The variability within and between Korean pond-smelt (Hypomesus nipponensis; KPS) and Canadian capelin (Mallotus villosus; CCP) were studied in order to clarify the genetic distances and differences. The dendrogram obtained by the seven primers indicates cluster 1 (KOREAN 01$\sim$KOREAN 11) and cluster 2 (CANADIAN 12$\sim$CANADIAN 22). The longest genetic distance displaying significant molecular differences was found to exist between individuals in the two geographic species of Osmeridae, between individuals' no. 10 of Korean and no. 18 of Canadian (0.686). 121 unique shared loci to each species, with an average of 17.3 per primer, were observed in the KPS species, and 264 loci, with an average of 37.7 per primer, were observed in the CCP species. 77 shared loci by the two species, with an average of 11.0 per primer, were observed in the two fish species. RAPD analysis showed that the KPS species was more genetically diverse than the CCP species. KPS species may have high levels of genomic DNA variability owing to the introduction of the wild individuals from the other sites to sampling sites although it may be the geographically diverse distribution of this species. As stated above, the existence of species discrimination and genetic variability between the KPS and the CCP species was identified by RAPD analysis.

Evaluation of genetic affinities among Fusarium oxysporum f. sp. fragariae by RAPD and rDNA RFLP

  • Fusarium oxysporum f. sp. fragariae is a fungal pathogen causing wilt disease on strawberry. The RAPD and RFLP of IGS region of rDNA were used to identify genetic affinity among 22 isolates of F. oxysporum f. sp. fagariae obtained from various location of major strawberry cultivating areas in Korea. Approximately 2.6kb DNA fragment was amplified with primer CNS1 and CNL12, and polymorphisms were observed with Avail and HinfI. A dendrogram was constructed using the UPGMA for cluster analysis. Eight distinct groups were clustered based on the banding pattern obtained from RAPD and rDNA RFLP. There was high level genetic variation among Korean isolates of Fusarium of sporum f. sp. fragariae.
