In the present work, current knowledge on the potential fate, microbial degradation, and toxicity of hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) was thoroughly reviewed, focusing on the toxicological assessment of a variety of potential RDX degradation pathways in bacteria and fungi. The present review on microbial degradation pathways and toxicities of degradation intermediates suggests that, among aerobic RDX degradation pathways, the one via denitration may be preferred in a toxicological perspective, and that among anaerobic pathways, those forming 4-nitro-2,4-diazabutanal (NDAB) via ring cleavage of 1-nitroso-3,5-dinitro-1,3,5-triazinane (MNX) may be toxicologically advantageous owing to its potential mineralization under partial or complete anoxic conditions. These findings provide important information on RDX-degrading microbial pathways, toxicologically most suitable to be stimulated in contaminated fields.
Tetracycline is one of the most commonly used antibiotics in domestic and foreign livestock industries to suppress the growth of pathogens. Tetracycline has been reported as a non-biodegradable compound. Therefore it has been not completely removed in the sewage treatment process. In this study, tetracycline was degraded using liquid ferrate (VI). Based on these experiments, the optimal water condition (pH and water temperature) were selected, appropriate liquid ferrate (VI) dosage was calculated, and finally the degradation pathway was estimated with the intermediate products detected by LC/MS/MS. All degradation experiments were completed within 30 seconds and the optimal condition was obtained in basic condition (pH 10) at room temperature (20℃). And the appropriate molar ratio between tetracycline and liquid ferrate (VI) was 12.5:1. Finally, 12 intermediate products were detected with LC/MS/MS and the degradation pathways and the degradation pathways and proposed the degradation pathways.

Cellular protein homeostasis is maintained by two major degradation pathways, namely the ubiquitin-proteasome system (UPS) and autophagy. Until recently, the UPS and autophagy were considered to be largely independent systems targeting proteins for degradation in the proteasome and lysosome, respectively. However, the identification of crucial roles of molecular players such as ubiquitin and p62 in both of these pathways as well as the observation that blocking the UPS affects autophagy flux and vice versa has generated interest in studying crosstalk between these pathways. Here, we critically review the current understanding of how the UPS and autophagy execute coordinated protein degradation at the molecular level, and shed light on our recent findings indicating an important role of an autophagy-associated transmembrane protein EI24 as a bridging molecule between the UPS and autophagy that functions by regulating the degradation of several E3 ligases with Really Interesting New Gene (RING)-domains.
Tetracycline is one of the most commonly used as antibiotics for the livestock industry and it is still widely used nowadays. Tetracycline and its metabolites are excreted with excrement, which is difficult to completely removed with conventional sewage treatment, therefore it is apprehended that the tetracycline-resistant bacteria occurs. In this study, the oxidant named ferrate(VI) was used to degrade the tetracycline and investigate the reaction between ferrate(VI) and tetracycline under various aqueous conditions. The highest degradation efficiency of tetracycline occurred in basic condition (pH 10.1 ± 0.1) because of the pKa values of tetracycline and ferrate(VI). The results also showed the effect of water temperature on the degradation of tetracycline was not significant. In addition, the dosage of ferrate(VI) was higher, the degradation of tetracycline and the self-degradation of ferrate(VI) also higher, finally the efficiency of ferrate(VI) was lower. The results said that the various mechanisms effects the reaction of ferrate(VI) oxidation, it required the consideration of the characteristics of the target compound for optimal degradation efficiency. Additionally, intermediate products were detected with LC/MS/MS and three degradation pathways were proposed.
The degradation of 3-chlorophenol(3-CP) by various AOPs(Advanced Oxidation Processes) including the ultraviolet / hydrogen peroxide, the Fenton and the ultraviolet(UV)-Fenton process has been conducted. The highest removal efficiency for 3-CP in the aqueous phase was obtained by the UV-Fenton process among the AOPs. In the UV-Fenton process, The removal efficiency of 3-CP decreased with increasing pH in the range of 3 to 6, and it decreased with increasing initial concentration. As the intermediates of 3-CP by UV-Fenton reaction, 3-chlorocatechol, 4-chlorocatechol, and chlorohydroquinone were detected thus the degradation pathways were proposed.
The degradation of 2,3-dichlorophenol(2,3-diCP) by various advanced oxidation systems with continuous feeding of hydrogen peroxide including the ultraviolet/hydrogen peroxide, the Fenton and the photo-Fenton process has been conducted. The highest removal efficiency for 2,3-diCP in the aqueous phase was obtained by the photo-Fenton process among the advanced oxidation systems. In the photo-Fenton process, The removal efficiency of 2,3-diCP decreased with increasing pH in the range of 3 to 6, and it decreased with increasing initial concentration. As the intermediates of 2,3-diCP by photo-fenton reaction, 3,4-chlorocatechol and 2,3-dichlorohydroquinone were detected, thus the degradation pathways were proposed.
The degradation efficiency and catabolism pathways of the different methylxanthines (MXs) in isolated caffeine-tolerant strain Pseudomonas putida CT25 were comprehensively studied. The results showed that the degradation efficiency of various MXs varied with the number and position of the methyl groups on the molecule (i.e., xanthine > 7-methylxanthine ${\approx}$ theobromine > caffeine > theophylline > 1-methylxanthine). Multiple MX catabolism pathways coexisted in strain CT25, and a different pathway would be triggered by various MXs. Demethylation dominated in the degradation of N-7-methylated MXs (such as 7-methylxanthine, theobromine, and caffeine), where C-8 oxidation was the major pathway in the catabolism of 1-methylxanthine, whereas demethylation and C-8 oxidation are likely both involved in the degradation of theophylline. Enzymes responsible for MX degradation were located inside the cell. Both cell culture and cell-free enzyme assays revealed that N-1 demethylation might be a rate-limiting step for the catabolism of the MXs. Surprisingly, accumulation of uric acid was observed in a cell-free reaction system, which might be attributed to the lack of activity of uricase, a cytochrome c-coupled membrane integral enzyme.

The hypoxia-inducible factor-1α (HIF-1α) is a key regulator of hypoxic stress under physiological and pathological conditions. HIF-1α protein stability is tightly regulated by the ubiquitin-proteasome system (UPS) and autophagy in normoxia, hypoxia, and the tumor environment to mediate the hypoxic response. However, the mechanisms of how the UPS and autophagy interplay for HIF-1α proteostasis remain unclear. Here, we found a HIF-1α species propionylated at lysine (K) 709 by p300/CREB binding protein (CBP). HIF-1α stability and the choice of degradation pathway were affected by HIF-1α propionylation. K709-propionylation prevented HIF-1α from degradation through the UPS, while activated chaperon-mediated autophagy (CMA) induced the degradation of propionylated and nonpropionylated HIF-1α. CMA contributed to HIF-1α degradation in both normoxia and hypoxia. Furthermore, the pan-cancer analysis showed that CMA had a significant positive correlation with the hypoxic signatures, whereas SIRT1, responsible for K709-depropionylation correlated negatively with them. Altogether, our results revealed a novel mechanism of HIF-1α distribution into two different degradation pathways.

Rhodococcus sp. strain T104, which is able to grow on either biphenyl or limonene, was found to utilize phenol as sole carbon and energy sources. Furthermore, T104 was positively identified to possess three separate pathways for the degradation of limonene, phenol, and biphenyl. The fact that biphenyl and limonene induced almost the same amount of catechol 1,2-dioxygenase activity indicates that limonene can induce both upper and lower pathways for biphenyl degradation by T104.
To study the feasibility of transmucosal delivery of leucine enkephalin (Leu-Enk) and $[D-ala^2]$-leucine enkephalinamide (YAGFL), their degradation extents and pathways in various rabbit mucosa extracts were investigated by high performance liquid chromatography. The degradation of Leu-Enk and YAGFL was observed to follow the first-order kinetics. The degradation half-lives of Leu-Enk in the nasal, rectal and vaginal mucosal extracts were 1.62, 0.37 and 1.12 hrs and those of YAGFL were 30.55, 9.70 and 6.82 hrs, respectively, indicating Leu-Enk was degraded in a more extensive and rapid manner than YAGFL. But the mucosal and serosal extracts of the same mucosa showed the similar degradation rates for both pentapeptides. The degradation was most rapid in the neutral pH and increasing concentrations of substrates retarded the degradation rates. The maior hydrolytic fragments of Leu-Enk were Des-Tyr-Leu-Enk and tyrosine, indicating the enzymatic hydrolysis by aminopeptidases. However, the data also suggested endopeptidases such as dipeptidyl carboxypeptidase and dipeptidyl aminopeptidase could play some role in the degradation of Leu-Enk. On the other hand, the hydrolytic fragments of YAGFL in all the mucosa extracts were mainly Tyr-D-Ala-Gly and Phe-Leu-Amide, demonstrating the hydrolytic breakdown by endopeptidases. The degradation pathways were further explored by concomitantly determining the formation of smaller metabolites of primary hydrolytic fragments of Leu-Enk and YAGFL in the mucosa extracts.
