Screening of White Rot Fungi with Selective Delignification Capacity for Biopulping (백색목재부후균중 Biopulping에 이용가능한 선택적 리그닌분해균의 스크리닝)

  • To obtain white rot fungi which have selective delignification capacity and can be used in biopulping processes, 94 different wood rotting fungi were screened and the capabilities of selected species were evaluated on deciduous and coniferous wood blocks. White rot fungi, first of all, were selected by simple enzyme tests, i.e., cellulase activity test; phenol oxidase activity test; laccase and peroxidase activity test. Most organisms that gave a positive Bavendamm gave a strongly positive laccase test with syringaldazine whereas most of those that gave a negative Bavendamm test also negative test for laccase and peroxidase, even if some exceptions were noted. Wood decay experiement were carried out to select fungal species with selective lignin-degrading ability by inoculating selected fungi to both wood blocks of Populus tomentiglandulosa and Larix leptolepis. After 12 weeks of incubation, weight losses, lignin losses, and morphological characteristics of the decayed wood were investigated. Almost all fungi tested caused 2 or more times of weight losses in P. tomentiglandulosa than in L. leptolepis, while no weight losses were detected from the un-inoculated wood blocks. Ceriporiopsis subvermispora and Phanerochaete chrysosporium were the best delignifiers for both hardwood and softwood. P. chrysosporium, however, was less effective than C. subvermispora. Bjerkandera adusta and two unidentified spp. caused delignification for only P. tomentiglandulosa. B. adusta caused simultaneous rot of all cell wall components, resulted in thinning of the secondary cell wall layers. Other fungi caused selective delignification resulting in the removal of lignin from middle lamella and separation of cells from each other.

The Effects of the Biodiversity Increase after Creation of the Artificial Wetland -The Case of Ecological Pond at Seoul Technical High School- (인공습지 조성후 생물다양성 증진 효과에 관한 연구 -서울공고 생태연못을 중심으로-)

  • The purpose of this study is to evaluate the creation techniques of artificial wetland, one of biotopes developed to promote biodiversity in urban areas, and to look for improvement steps. Specifically, artificial wetland creation techniques were categorized into living environment and living creature classification. Being living conditions for creations, habitat environment was reviewed with a focus on water and soil environments. Living creatures were classified into plants, insects, fish, and birds. The evaluation of creation techniques was done in post-construction evaluation while considering the creation of habitats for living creatures. Intervention by users, changes in living environment and living species, and relevance of creation techniques were reviewed. Key results of this study are as follows. (1) Water environment for the living environment of creatures provides a suitable environment conditions for the living of creatures through a process easing the use of piped water. Various water depths and embankment appear to have a positive impact on the living of aquatic life. In particular, embankment covered in soil naturally played an important role as a place for the activities of aquatic insects and young fish as well as the growth of aquatic plants. (2) Various aquatic and ground plants to promote insect-diversity, shallow water, and old-tree logs had contributed greatly in increasing the types and number of insects. Aquatic insects. Aquatic insects were seen much particularly in areas where aquatic plants are rich but water is shallow than any other areas. (3) A space piled with stone to provide habitats for fish was not much used. However, it was observed that fish used embankment built with natural stones and embankment using logs in areas where water is deep. In addition, it was confirmed that 1,500 fish that had been released propagated using various depths and places for birth. (4) It was analyzed that techniques (creation of island, log setting, and creation of man-made bird nests) to provide habitats and to attract birds are not serving their roles. In such a case, it is believed that species had not increased due to the smallness as well as isolated features of the area. Based on theoretical review, they are judged to be areas that are likely to be used when a greater variety of birds is introduced. It is judged that attracting and keeping more birds at the site, such spaces need to be linked systematically in the future in terms of building eco-network while ensuring an adequate living areas. (5) In the study areas, users intervened greatly. As a result, a blockage was created preventing the normal growth of plants and non-indigenous plants were introduced. Physiological Responses of the Three Deciduous Hardwood Seedlings Growing Under Different Shade Treatment Regimes (상이한 피음조건에서 자라는 3개 활엽수의 생리적 반응)

  • This present study was conducted to investigate photosynthetic characteristics, chlorophyll fluorescence, chlorophyll contents of Betula platyphylla var. japonica, Prunus leveilleana, Magnolia sieboldii, growing under four different light intensity regimes (full sun, and 64~73%, 35~42%, 9~16% of full sun). As result, Betula platyphylla var. japonica showed outstanding photosynthetic capacity and apparent quantum yield in full sun and showed low shade tolerance. Prunus leveilleana showed good photosynthetic capacity and apparent quantum yield in 64~73% or 35~42% of full sun and showed common shade tolerance. However, Magnolia sieboldii showed good photosynthetic capacity and apparent quantum yield in 35~42% of full sun, while the lowest in full sun. Magnolia sieboldii showed the highest shade tolerance compared to the other species. As the shading level increased, the total chlorophyll contents of all species increased with significant difference.

Nutrient Dynamics in Litterfall and Decomposing Leaf Litter at the Kwangneung Deciduous Broad-Leaved Natural Forest (광릉 천연활엽수림의 낙엽낙지와 낙엽분해에 따른 양분동태)

  • Litterfall and litter decomposition represent a major contribution to the carbon and nutrient inputs in a forest ecosystem. We measured litterfall quantity and nutrient dynamics in decomposing litter for two years at the Kwangneung broadleaf natural forest (DK site) in Korea. Litterfall was collected in circular littertraps (collecting area : 0.25 $m^2$) and mass loss rates and nutrient release patterns in decomposing litter were estimated using the litterbag technique employing 30 cm ${\times}$ 30 cm nylon bags with 1.5 mm mesh size. Total annual litterfall was 5,627 kg/ha/yr and leaf litter accounted for 61 % of the litterfall. The leaf litter quantity was highest in Quercus serrata, fallowed by Carpinus laxiflora and C. cordata, etc., which are dominant tree species in the site. Mass loss from decomposing leaf litter was more rapid in C. laxiflora and C. cordata than in Q. serrata litter. About 77% of C. laxiflora and 84% of C. cordata litter disappeared, while about 48% in Q. serrata litter lost over two years. Lower mass loss rates of Q. serrata litter may be attributed to the difference of substrate quality such as lower nutrient concentrations compared with the other litter types. Nutrient concentrations (N, P, Mg) of three litter types except for potassium (K) increased compared with initial nutrient concentrations of litter over the study period. Compared with Q. serrata litter, nutrients (N, P, K, Ca, Mg) in C. laxiflora and C. cordata litter were released rapidly. The results suggest that litter mass loss and nutrient dynamic processes among tree species vary considerably in the same site conditions.

Antioxidative Activities and Whitening Effects of Ethyl Acetate Fractions from The Immature Seeds of Abeliophyllum distichum (미선나무 미성숙 종자의 항산화 및 미백 활성)

  • Abeliophyllum distichum Nakai is deciduous shrubs of flowering plant in Oleaceae. It is important plant resources and consists of one species in the world. Also the endemic plant of A. distichum has been protected and designed endangered plant in Korea. For this reason, study on the immature seeds of A. distichum (ADS) hasn't progressed. In the present study, we evaluated the antioxidant activity and inhibitory effects on proteins and mRNA levels were related in the whitening effect in B16F10 cells. ADS was effective for reaction oxygen species (ROS). ROS causes various diseases such as aging, inflammation, cancer, and etc. Antioxidant properties were evaluated DPPH, ABTS radical scavenging activity and Reducing power. Plants were known that contained phenolic compounds were related in antioxidant activity. Phenolic compounds were phytochemicals commonly named natural polyphenols. These are secondary metabolites of plants involved in the defense against different types of stresses. In results, ADS suppressed the expression and transcription of Tyrosinase, TRP-1, TRP-2, and Microphthalmia-associated transcription factor (MITF). Tyrosinase, tyrosinase-related protein 1 (TRP-1), tyrosinase-related protein 1 (TRP-2) are known to play an important role in melanin biosynthesis. MITF regulated the expression and transcription of Tyrosinase, TRP-1, and TRP-2. In conclusion, ADS was effective in both antioxidant activities and whitening effects. Also, they were associated with the content of phenolic compounds. We suggested that ADS can be use antioxidants and skin-whitening functional cosmetics material derived from natural plant resources.

Genetic Diversity and Spatial Genetic Structure of Populus koreana Population in Mt. Odae, Korea (오대산 물황철나무(Populus koreana) 집단의 유전다양성 및 공간적 유전구조 분석)

  • This study describes analysis of genetic diversity and spatial genetic structure of Korean poplar (Populus koreana Rehder) in Mt. Odae using I-SSR markers. P. koreana is a deciduous broad-leaved tree species that primarily grows in the alpine valleys of China, Russia and North Korea. In South Korea, P. koreana is found limitedly in Gangwon province. Especially, the population in Mt. Odae is located on the southern limit line, its importance is emphasized from the genetic resource conservation perspective. The Shannon's diversity (I=0.230) and the expected heterozygosity (He=0.151) were relatively low as compared with those of other plant species. Spatial autocorrelation analysis using Tanimoto's distance showed that the genetic patch was founded within 400 m. It is suggested that individual trees for ex situ conservation should be sampled with a minimum distance of 400 m between trees.

Taxonomic identity of Crepidiastrum ×nakaii recorded on Hongdo Island (홍도고들빼기의 분류학적 실체)

  • The plant known as "Hong-do-go-deul-ppae-gi" on Hongdo Island, belonging to the genus Crepidiastrum, has been recorded as C. ×nakaii, a hybrid between C. denticulatum and C. platyphyllum. During a survey of the flora of Hongdo Island, we reexamined the taxonomic identity of "Hong-do-go-deul-ppae-gi". The morphological traits, geographical distribution, and internal transcribed spacer (ITS) sequences of "Hong-do-go-deul-ppae-gi" were compared to related species within the genus. A morphological examination revealed that the plant was similar to C. lanceolatum in that it has subshrub stems with caudex stout, 8 inner involucrals bracts, and 10 to 12 florets. In contrast, similarities to C. denticulatum were observed in terms of radical leaves deciduous in the flowering period, cauline leaves membranous with acute serrations, and beaked achenes. Furthermore, C. denticulatum and C. lanceolatum grew on Hongdo Island and nearby areas, but C. platyphyllum was not distributed in these areas. The results of morphological and distributional investigations revealed that the Hongdo Island plant should be regarded as C. ×muratagenii, a hybrid between C. denticulatum and C. lanceolatum, not C. ×nakaii, which is a hybrid between C. denticulatum and C. platyphyllum. However, the resolution of the ITS sequences was insufficient such that we could not separate the examined species.

Ecological Characteristics of Buxus sinica Habitat Distributed on the Ridge of Gyeokjabong(Mt.), Bogildo (보길도 격자봉 일대 능선부에 분포하는 회양목 생육지의 생태적 특성)

  • This study was intended to investigate and analyze the current status of the plant community structure in order to provide basic data for the systematic conservation and management of the Buxus sinica area appearing on the ridgeline connecting the Gyeokjabong(Mt.) and Keungiljae, on Bogildo Island. In consideration of location characteristics such as density, topography, and direction of B. sinica appearing in the shrub layers, a total of 26 sites were set as the survey area for representative vegetation communities or areas with changes in topography. According to DCA and TWINSPAN, it was classified into 6 communities, namely Carpinus turczaninovii-Deciduous broad-leaved, C. turczaninovii, C. turczaninovii-B. sinica, C. turczaninovii-Evergreen broad-leaved, Q. acuta, and D. trifidus-Q. acuta. The community dominated by C. turczaninovii maintains the status quo for the present moment, however, Q. acuta will dominate the surrounding area in the long term, so Q. acuta has high succesion potential. In the case of the Q. acuta and D. trifidus-Q. acuta communities, Q. acuta, known as the climax species of warm-temperate forests, will maintain dominant status. In a case of B. sinica, the community dominated by C. turczaninovii will remain in good status due to the topographical conditions, but the community dominated by Q. acuta growth difficulties are expected due to the high coverage. In the relationship between environmental factors and vegetation distribution, analysis showed that only soil pH affect vegetation distribution. Furthermore, the soil acidity (pH) was 3.78-5.30, the electrical conductivity was 0.186-0.543 dS/m, and the organic matter content was 2.25-2.89%.

Floristic study of Mt. Seongdeok (Ganghwa-gun) in Korea (성덕산(강화군)의 관속식물상)

  • This study was conducted to investigate the flora of Mt. Seongdeok (Ganghwa-gun) in Korea. From the results of nine field surveys from March 2020 to July 2022, a total of 445 total taxa, representing 397 species, nine subspecies, 35 varieties, two forms and two hybrids were identified, which were placed in 273 genera and 93 families. The plant formation of Mt. Seongdeok is a deciduous broad-leaved and conifer mixed forest, which is common in the middle part of the Korean Peninsula. Most of the mountain is covered by a young secondary forest, which is mainly composed of Quercus ssp. and Pinus ssp. Among them, five taxa were endemic to Korea, one taxon were endangered plant and two taxa were red list plants. The floristic target plants amounted to 27 taxa, specifically one taxon of grade V, four taxa of grade III, seven taxa of grade II, and 15 taxa of grade I. And 43 taxa were northern lineage plants. A total of 46 species of alien plants were identified, with a Naturalized Index of 10.3%, an Urbanization Index of 11.7%, and six plants that disturbed the ecosystem. Our results provide basic data on vascular plants flora, and plant diversity and distributional changes.

Analysis of Water Relations of Economic Oak Species by Hydraulic Architecture Method (Hydraulic architecture를 이용한 참나무속 주요 수종의 수분 특성 분석)

  • Several parameters of hydraulic architecture relating to hydraulic conductance in xylem vessels were investigated in the current-year shoots of six species of deciduous oak trees. The above parameters were also investigated in the sprouts of Quercus mongolica and Q. variabilis, as well as in the seedlings of Q. mongolica and Q. acutissima. The values of specific conductivity, leaf specific conductivity and Hagen-Poiseuille's relative hydraulic conductivity relating to vessel diameter of Q. dentata were the highest in all of the species studied. The above values of most of the species studied were higher in May-June than in September-October because of increasing the vessel embolism by cavitation and so on through the growing season. The estimated values of relative hydraulic conductivity of vessel by Hagen-Poiseuille's empirical equation and the real values of hydraulic conductivity presented positive relationships in most of the species studied. Huber value and leaf specific conductity using leaf area or leaf weight generally exhibited similar patterns each other even if having some exceptions. The hydraulic conductances of sprouting shoots were much better than those of normal growing shoots in Q. rnongolica and Q. variabilis. The specific conductivity and leaf specific conductivity were rapidly decreased by the vessel embolism through cavitating just after cutting the shoots in Q. mongolica and Q. acutissima seedlings. Diurnal changes of the conductivities in the seedlings of Q. mongolica and Q. acutissima presented the possibility of their self-controlling of conductance by active moisture absorption under mild water stress. Specific conductivity and leaf specific conductivity, and so on of Q. acutissima seedlings subjected to periodical moisture stress or not have decreased through the growing season, but the influences of moisture stress to the conductance were not proved definitely because of influencing similarly and simultaneously to the development of xylem and leaf having inverse relation in the influences. The values of conductivities were higher generally in middle or upper parts of stems than root collar in the seedlings.
