An about 5-month-old, female rabbit was presented with pruritus, alopecia and mucopurulent ocular discharge. On physical examination mild to moderate scales on whole body were observed. Dermatological lesions such as alopecia, erythema, papules, pustules and crusts were observed in the eyelids, nose, inner pinna, medial sites of four feet, metacarpal and metatarsal areas, and cranial site of left stifle joint. Also, mild conjunctivitis, blepharitis and keratitis were found. For extensive dermatologic diagnostic evaluation skin scraping, tape stripping, impression smear, combing, wood' light, bacterial culture and fungal culture were performed. Finally, Cheyletiella spp. was found by combing. Many heterophils and eosinophills were appeared in impression smear. The result of fungal culture was negative. Pasteurella spp was cultured. Definitive diagnosis of Cheyletiellosis and secondary Pasteurella spp infection were established. The rabbit was treated with 6 mg/kg of selamectin topically every two weeks and restricted in cage for one month. During one month of initial treatment, clinical signs such as pruritus, alopecia, scales, papules, pustules and crusts were remarkably improved. However, in spite of good therapeutic response of selamectin the rabbit was dead suddenly at 22 days after second administration of selamectin. This case showed that selamectin was possibly effective for cheyletiellosis in rabbit; safety of selamectin for rabbit was not identified.
Codonopsis lanceolata ("Deoduck" in Korea) is a perennial herb, and belongs to family, Campanulaceae. Its taproot is used a good source of a wild vegetable as well as an herbaceous medicine. In this study, to develop a bialaphos-resistant transgenic Codonopsis, seed germination mechanism and somatic embryogenesis of the plant were investigated, and Agrobacterium-mediated transformation with bar gene encoding phosphinothricin acetyltransferase (PAT) was performed. Attempt were made to regenerate plant from cells via somatic embryogenesis. When the cotyledons, nodes and leaf disks were cultured on MS medium containing 2,4-D and zeatin, embryogenic calli were induced. Upon transferring the somatic embryos to N6 solid medium without plant growth regulators, they developed into plantlets under continuous illumination. All plants were dead on MS basal medium containing 10 mg/L phosphinothricin (PPT) and Basta, respectively. The explants did not produce calli in the medium containing 200 mg/L kanamycin. The explants were cocultured with Agrobacterium tumefaciens for 2 days, and transformants were selected in MS basal medium containing 1.0 mg/L 2,4-D, 100 mg/L kanamycin and 500 mg/L carbenicillin. After the selection, embryogenic calli were induced and then somatic embryos were produced by subsequent subculturing. The somatic embryos were germiated on N6 basal medium containing 200 mg/L kanamycin and 500 mg/L carbenicillin. PCR analysis showed that nptII and bar genes were introduced in the Deoduck transformants. After the confirmation of bar gene expression in RNA and protein level, the transgenic Deoduck will be used to study the genetics of filial generation with the herbicide control gene, bar.gene, bar.
In this study we investigated the effect of gametocides on the number of larger-sized pollen in anther, and also induced callus from the anther culture of Zoysia japonica Steud. Before culturing, we have observed pollens in anther through fluorescence and electron microscopes to know pollen dimorphism. There were two types of pollens observed. One type (30-36 ${\mu}{\textrm}{m}$ in diameter) consisted of vacuolated, larger-sized pollens and the other (15-20 ${\mu}{\textrm}{m}$ in diameter) smaller-sized ones with dense cytoplasm and plenty of amyloplasts. Within few hours, all the smaller-sized pollens were dead, while larger-sized ones were viable for one or two days. To induct larger-sized pollens, various gametocides were leaf-sprayed on three booting stages cultured under 4$0^{\circ}C$ /15$^{\circ}C$ (day/night) before anther culturing. Number of these larger pollens were few (less than 1%) in anther without spraying gametocides. GA$_3$increased the number of larger-sized pollens when applied at mid-booting stage. GA$_3$ with 50 mg/L treatment caused the highest percentage (25.4%) of the larger-sized pollen. Anthers with GA$_3$ treatment were only produced calli on AA medium (modified B$_{5}$+8.0 mg/L 2,4-D +0.2 mg/L kinetin), but callus formation was quite low (less than 1%).).
Epidemiological and statistical observations were made of fire hazards that occurred during the past 18 years, 1948 to 1965. Injury and mortality rates for all ages were computed chronologically. For the years of 1955, 1961 and 1965, all fire accidents were epidemiologically analysed to draw characteristic patterns in relation to the seasonal and 24 hour distribution, causes and sites of accidents etc.. Fire hazards observed herein are the categorys E 916 of the International Classification of Causes of Death, 1955, and includes all accidents caused by fire and explosion of combustible materials. The following conclusion was made: 1. The average number of annual deaths due to fire was 183 and the number of the in jured due to the same cause was 335. The mortality rate per 100,000 population was 0.8 and the ratio of injuries per death was 1.83. 2. The casually rate including both the dead and injured was 5.0 per 100,000 in Seoul, the highest among the provinces and followed by 3.4 in Cheju -Do, 2.1 in Kangwon-Do, 1.7 in Kyunggi-Do accordingly. The other provinces had a range of 0.6 to 1.2. 3. The monthly distribution of fro accidents showed that the winter months, December through February, had more frequent accidents, while the summer season, June through August had less. The 24 hour distribution of accidents showed more cases from 12:00 to 18:00 and less from 4:00 to 10:00 hours. 4. The per cent distribution of causes of accidents showed; 90.0% for careless, 10.0% for arson. The cause of carelessness was further breakdown into; 15.0% for kitchen fire places, 13.8% for fire playing, 9,4% for electrical heating and wires, 8.3% for fuels, 6.3% for matches, 5.2% for ash dumps and the remaining for others. 5. The accidents as classified by place revealed that 56.8% of the total occurred at the common dwelling houses, 11.3 at various industrial workshops, 9.3% at the street shops and the remaining at the miscellaneous places.

The objective of this study is to characterize a toxicity of Polygalae Radix (PR) in F344 rats and to find a dose levels for the 13 weeks toxicity study. PR is well known as medicinal herb in many Asian countries for treatment of expectorant, tonic, tranquillizer, antipsychotic agent and functional diet for improving memory. However, there is insufficient background information on toxicological evaluation of PR extract to support its safe use. Therefore, we conducted toxicological evaluation of this drug in compliance with OECD and KFDA guideline in this study. The extract of PR was administered orally to F344 rats at dose levels of 0, 500, 1000, 2000, 3500 and 5000 mg/kg/day for 2 weeks. Each group was composed to five male and five female rats. In the result, there were no treatment PR-related adverse changes in food consumption, hematology, clinical chemistry, urinalysis, gross finding at necropsy, organ weight examination. Four males at 5000 mg/kg/day were found dead during the treatment period. These animals showed salivation. The cause of death is still under investigation. The animals treated at 500, 1000, 2000, 3500 and 5000 mg/kg/day showed salivation and all animals at 5000 mg/kg/day exhibited lower body weight and cumulative weight gain in compared to those of control animals. Therefore, we recommend that a dose group of 3500 mg/kg/day is a highest treatment group in 13-week exposure study.
'Yumyeong' peach has desirable characteristics of long shelf-life and specific non-melting nature with long harvesting period. However, some fruits harvested too late tend to show fruit pithiness symptom or internal breakdown. The study was conducted to investigate the differences between fruit flesh pithiness and internal breakdown symptoms, and to find out inducing factors of flesh pithiness in 'Yumyeong' peach. The incidence of flesh pithiness was higher in the fruits harvested late. Sugar and malic acid contents were not different between normal fruits and flesh pithiness fruits, but the acidity was significantly lower affected by low citric acid content in flesh pithiness fruit. In flesh pithiness fruits, calcium contents were low in both skin and flesh. Occurrence of flesh pithiness fruits was high in the years with low precipitation and high temperature for 2 months before harvest. In observations on morphological characteristics, the parts showing flesh pithiness consisted of smaller cells than the normal parts. Tonoplasts were disintegrated and the number of dead cells was high in internal breakdown fruits, while the tonoplasts were intact with contracted vacuoles in flesh pithiness fruits. Tylosises were observed in vascular tissues around the flesh pithiness, therefore, it was assumed that those tylosises restricts flesh tissue development resulting in flesh pithiness. Other varieties such as 'Fantasia' and 'Wolmi' also showed tylosis and smaller cells were observed in the flesh tissue of these cultivars, indicating abnormal growth of the flesh part. These results suggested the possibility of the occurrence of pithiness like symptoms in other peach varieties.
Pathological symptoms and hematological parameters of red sea bream, Pagrus major and water temperature in the culture ground was investigated to clarlify the cause of winter moratility. Dead fish showed green liver and accumulation of ascites in the cavity. A few Bivagina tai were also found on the gill but either bacteria or virus were not. When hematological parameters from fish taken before/after the winter mortality were compared, blood glucose, serum AST and ALT, and total cholesterol, trigyceride and total protein were significantly decreased in the fish after the winter mortality. These results may explain that the nutritional level of fish was decreased because fish could not fed during winter season. According to the CORI monitoring system operating by KODC, NFRDI a long term water temperature from Dec. 25, 2005 to Feb. 24, 2006 (60 days) were exposed the low water temperature environments to the red sea bream. First of all, mass mortality began at Sarayngdo where low temperature below 8℃ continued for 42 days. The winter mortality did not occured in the depths of 9 m to 19.2 m where difference of water temperature in the surface and bottom was only within 0.1℃. But in the depth of 7.5 m to 11 m winter mortality occured where water temperature in the surface and bottom showed much variation ranged from 0.2℃ to 1.4℃. From these results, great difference of water temperature in the surface and bottom of the culturing area might results in winter mortality of red sea bream.
This study was conducted to evaluate whether \"Royal jelly\" (RJ) added to Tris-buffer dilute contributed to supporting post-thaw viability and longevity of frozen canine spermatozoa. Two Japanese spitzs (2 to 4 years of age) were used as a semen donor. Semen was collected by manual masturbation and separated into 3 fractions. Only the sperm-rich fraction having sperm motility of more than 70%, containing sperm concentration of 2~4$\times$10$^{8}$ cells/ml and having dead or abnormal spermatozoa of less than 15% was used for the experiment. Each ejaculated semen was centrifuged at 400 $\times$ g for 5 min and then diluted in a Tris-buffer supplemented with 20 ml egg yolk (Ext I), 4% glycero1 and 1% Equex STM Paste (Ext II) or g1ycero1, Equex STM paste and RJ of various concentrations (Ext II-RJ). After freezing and thawing, viability of spermatozoa in Ext II -RJ containing 1% RJ immediately after thawing (67.5$\pm$9.6) was significantly lower than that of Ext II , Ext II -RJ containing 0.01 or 0.1% RJ (77.5$\pm$12.5, 78.7$\pm$8.2 and 80.0$\pm$6.3). However, Ext II-RJ containing 0.1% RJ yielded higher viability than Ext II, Ext II-RJ containing 0.01% at or 1% 1 h after thawing (69.5$\pm$8.1 vs. 55.0$\pm$12.9, 57.5$\pm$9.6 and 41.5$\pm$12.6; P<0.05). At 1 h after thawing, the viability of spermatozoa thawed in 7$0^{\circ}C$ (68.8$\pm$12.5) was significantly higher than that of spermatozoa thawed in 38$^{\circ}C$ (48.8$\pm$16.3), although there was no difference in the viability between both groups immediately after thawing (77.5$\pm$9.6 and 81.3$\pm$8.1). Post-thaw viability and longevity of post-thaw spermatozoa in Ext II-RJ containing 0.1% RJ was higher in those in Ext II at 1 h (65.0$\pm$12.9 vs. 42.5$\pm$12.6), 2 h (52.5$\pm$12.6 vs. 27.5$\pm$17.1) and 3 h (40.0$\pm$14.1 vs. 20.0$\pm$12.1) after thawing. These results indicated that addition of 0.1% af to Tris-buffer enhanced post-thaw viability and longevity of canine spermatozoa and this additive can be used for increasing the possibility of collision between spermatozoa and ova during insemination.emination.
A new nutritional disease has occurred among the hatchery-reared Korean bullhead fingerlings (Pseudobagrus fulvidraco) in the Chonbuk Province in September 1997. Diseased fish were all dead within 3-7 days, showing sluggish behavior, head up and tail down swimming. Most characteristic clinical signs were anaemia, clubbed and fused gill, skin desquamation. haemorrhage around the mouth and at the base of pectoral fins. Any causative bacteria and parasites were not isolated from the lesions and internal organs of the diseased fish. The hepatosomatic index, red blood cell count, hematocrit, hemoglobin and erythrocytes size of peripheral blood in the diseased fish were remarkably decreased compared with those of normal fish. In the histopathological observations, epithelial hyperplasia of the gill filaments initiated at the base of the gill was pronounced. This symptom was the characteristic appearence of all the diseased fish. A 0.6% saline bath and feeding a pantothenic acid-supplemented diet were conducted to decrease the mortality. Ten days after 0.6% saline bath or 25 days after feeding a pantothenic acid supplemented diet resulted in decreasing in the mortality. Microscopic appereance of the gill from the recovered fish was similar to that of the gill from healthy fish. These results indicate that the disease was caused by deficency of pantothenic acid in their diet and that 0.6% saline bath or supplementation of pantothenic acid in the diet was an effective way to decrease the mortality.
CT(Sodium N-chloro-para toluenesulfonamide, Chloramine-T) known to be a strong oxidative agent was investigated to use as a candidate of disinfectant in the marine aquaculture industry by the analysis of the bactericidal activity against different aquatic microorganisms. One hundred percent mortality appeared at and above 16 ppm CT in sea water in flounder, the predominant species in the marine aquaculture of Korea, when exposed for 48hr. However, bactericidal activity was appeared to be very effective, and all different species of the fish pathogenic bacteria exposed to CT of less than 2 ppm in sea water were dead within 15 min. It allowed us to confirm that CT could be a very effective disinfectant to protect the spread of fish pathogenic bacteria derived from diseased fish or sea water in marine aquaculture. High concentration(10 ppm) with longer exposing time(24 hrs) was required for the algicidal activity of CT, at which concentration might induce the acute toxicity against fish, however, restricted the expanded use of CT for the elimination of zooplanktons at phytoplanktons in marine farms. Moreover the bactericidal activity of CT inhibited almost completely in the present of more than 10 ppm of organic materials pointed out that quality of the culturing sea water should be considered carefully for the application CT to the aquatic farms as a therapeutic agent.
